Effect of Nutrient Solution Composition Modification on the Internal Quality of Some Leaf Vegetables in Hydroponics (수경재배시 양액 조성 처리가 몇가지 엽채류의 내적 품질에 미치는 영향)

  • This study was conducted to find out the change of infernal quality, such as vitamin C and nitrate contents in some leaf vegetables grown hydropoincally in different nutrient conditions. Pak-choi (Brassica camperistis L. spp. chinesis Jusl.), chungchima (Lactuca sativa L. var. crispa cv. Chungchima) and romaine (Lactuca sativa L. var. longifolia Lam.) lettuces were cultivated for 2 weeks in 4 different nutrient solutions, such as tap water; no-nutrient, added $NH_4$, discarded $NO_3$, and supplied Yamazaki' solution for lettuce as a control. The growth of leaf vegetables was not different among nutrient solution treatments except tap water. The nitrate content showed the highest in control, and followed by $+NH_4$ treatment, $-NO_3$ and tap water treatment, regardless of kind of vegetables. The vitamin C content in 3 different vegetables showed the opposite result against nitrate content so that the treatment that showing the highest vitamin C content was tap water in romaine and chungchima lettuces, and $-NO_3$ treatment in pak-choi. The vitamin C and the nitrate content showed high correlations; $r=-0.614^*$ in pak-choi, $-0.651^*$ in romaine lettuce, and $-0.804^{**}$ in chungchima lettuce.

Transformation of Lettuce (Lactuce sativa L.) Using Iron Storage Protein Ferritin Gene (철 저장단백질 관련 Ferritin 유전자를 이용한 상추의 형질전환)

  • Explants of Lactuce sativa cultivar, chungchima, were co-cultivated with Agrobacterium tumefaciences LBA4404, EHA101 strains containing nptll gene and ferritin gene encoding iron storage protein from soybean for transformation. Through initial selection of regenerated explants by culturing on a kanamycin and carbenicillin containing MS medium, multiple shoots were obtained after 2 months of culture. For a complementary step of selection, putative transgenic shoots were transferred to 1/2 MS basal medium supplemented with 100 mg/L kanamycin and 500 mg/L carbenicillin. The selected shoots were tested with PCR analysis using nptll, ferritin specific primers whether ferritin gene was introduced to genome of the plants. These results confirmed that produced the specific PCR bands in the putative transgenic lines. Additionally the Northern blot showed that transcripts of ferritin gene were detected in mature leaf of the transgenic lines. These results suggest that ferritin gene be successfully integrated and transcribed in the putative transgenic lettuce plants.

Efficient plant regeneration from cotyledon and primary leaf explants of lettuce (Lactuca sativa L.) (상추의 자엽 및 제 1엽 절편체들로부터 효율적인 식물체 재분화)

  • The efficient system for plant regeneration from cotyledon and primary feat explants of lettuce was established. Plant regeneration efficiency was shown 91.3% from cotyledon and 85.9% from primary leaf explants of variety 'Jungtongpogi' in KN medium. Plant regeneration efficiency was also estimated with various plant regeneration media in variety' Chungchima', which was lowest plant regeneration efficient showing 35.4% from cotyledon and 30.3% from prima leaf explants in KN medium. Kl medium increased 77.9% and 80.7% of plant regeneration efficiencies from cotyledon and primary leaf explants of variety 'Jungtongpogi' were cultured on KN medium. In case of varie쇼 ‘Chungchima', efficient plant regeneration was shown when primary leaf explants were cultured on SH and KI media.

Effect of Remained Leaf Number on the Growth and Yield of Hydroponically Grown Leaf Lettuce (잔존엽수가 수경재배 잎 상추의 생육 및 수량에 미치는 영향)

  • This study was conducted to investigate the effect of remained leaf number on leaf, stem and root growth of leaf lettuce (Lactuca sativa L. var. Chungchima). Plants were grown hydroponically in $0.45\pm0.15\pm3.0m$ growth channels using a recirculating DFT (Deep Flow Technique) system. The growth response to remained leaf number was evident in leaf and stem than root of leaf lettuce. Leaf fresh weight increased and leaf quality was lowered with increasing of remained leaf number. Results indicate that 4 to 6 leaves could be optimum remained leaf number of hydroponically grown leaf lettuce.

Suppression of Meloidogyne incognita in Lettuce and Oriental Melon by Pasteuria penetrans KW1

  • Pasteuria penetrans KW1 (PP), parasitic bacterium of nematode, was isolated from oriental melon greenhouse soil in Korea and evaluated for the suppression effect on the reproduction of southern root-knot nematode, Meloidogyne incognita (MI), in lettuce (Lactuca sativa L. var. Chungchima) and oriental melon (Cucumis melo L. var. Eunchun). Pot experiments were conducted by planting the lettuce seedlings in medium inoculated with 5,000 MI juveniles/pot (J), J +100,000 PP endospores/l g medium, and J +200,000 PP endospores/1 g medium. After 11 weeks of plantation, number of root galls in J +200,000 PP endospores/1 g medium was decreased to 92/root (38.9%, control effect), compared to the J of 150/root. In the second plantation of lettuce in the same pots, the numbers of root gall were significantly decreased in PP treated pots with 75 (77.2%, control effect) and 150/root (54.4%, control effect) in J +200,000 and J +100,000 PP endospores/1 g medium, respectively, compared to the J of 330/root when harvested at 10 weeks after planting. In oriental melon, root gall percentages were 32.1 (60.2%, control effect) and 52.9% (34.5%, control effect) in J +200,000 and J + 1(10,000 endospores/l g medium which were significantly lower than that of 80.7% in J.

Effect of Covering Method of Ventilating Non-Woven Fabric and $\textrm{GA}_3$ Treatment on the Growth and Yield of Leaf Lettuce during Low Temperature Season (통기성 부직포의 피복방법 및 $\textrm{GA}_3$ 처리가 저온기에 잎상추의 생육 및 수량에 미치는 영향)

  • This study was conducted to estimate stimulation of plant growth of green and red leafy lettuces (Lactuca sutiva L. cv. Chungchima and Jugchima) at affected by covering method and time with protected film materials containing concentrated PO film and GA$_3$ foliar spray. Plants were covered with ‘Pasraido’ material from October 26 to November 31 (direct 1), from November 5 to November 31 (direct 2), or with a tunnel, or direct 2+ tunnel. Mean air temperature and relative humidity under the covering were higher in direct and tunnel, direct and non-covering, in that order. The fresh weight and dry weight of red and green lettuces increased in direct and tunnel followed by direct 1, direct 2, tunnel and the control. GA$_3$ treatment promoted the growth in all covering methods, but decreased the chlorophyll contents. Fresh weight and dry weight were higher in red lettuce than in green lettuce in the control.
