Effects of Didecyldimethylammonium Chloride (DDAC) on Sprague-Dawley Rats after 13 Weeks of Inhalation Exposure

  • Didecyldimethylammonium chloride (DDAC) is used in many types of biocidal products including tableware, carpets, humidifiers, and swimming pools, etc. In spite of increased chances of DDAC exposure through inhalation, studies on the inhalation toxicity of DDAC are not common even though the toxicity of DDAC might be significantly higher if it were to be administered through routes other than the respiratory system. DDAC aerosols were exposed to Sprague-Dawley rats in whole body exposure chambers for a duration of 13 weeks. The Mass Median Aerodynamic Diameters of the DDAC aerosol were $0.63{\mu}m$, $0.81{\mu}m$, and $1.65{\mu}m$, and the geometric standard deviations were 1.62, 1.65, and 1.65 in the low ($0.11{\pm}0.06mg/m^3$), the middle ($0.36{\pm}0.20mg/m^3$) and the high ($1.41{\pm}0.71mg/m^3$) exposure groups, respectively. Body weight was confirmed to be clearly influenced by exposure to DDAC and mean body weight was approximately 35% lower in the high ($1.41{\pm}0.71mg/m^3$) male group and 15% lower in the high ($1.41{\pm}0.71mg/m^3$) female group compared to that of the control group. In the bronchoalveolar lavage fluid assay, the levels of albumin and lactate dehydrogenase had no effect on DDAC exposure. The lung weight increased for the middle ($0.36{\pm}0.20mg/m^3$) and the high ($1.41{\pm}0.71mg/m^3$) concentrations of the DDAC exposure group, and inflammatory cell infiltration and interstitial pneumonia were partially observed in the lungs of the middle ($0.36{\pm}0.20mg/m^3$) and the high ($1.41{\pm}0.71mg/m^3$) exposure groups. However, severe histopathological symptoms, including proteinosis and/or fibrosis, were not found. Based on the results of the changes in the body weight and lung weight, it is considered that the NOAEL (no-observed adverse effect) level for the 13-week exposure duration is $0.11mg/m^3$.

Toxicity of agricultural chemicals on Lymnaea viridis the intermediate host of Fasciola hepatica (간질(肝蛭)의 중간숙주(中間宿主)인 애기물달팽이에 대한 몇가지 농약(農藥)의 독성시험(毒性試驗))

  • In the present study, the effects of 4 agricultural chemicals commonly used in this conuntry were experimentally assessed on Lymnaea viridis the intermediate host of Fasciola hepatica, which in non-target organism of these chemicals. The major habitat of the snail is rice paddies in Korea and many agricultural chemicals are used for weed, fungi or insect control in rice paddies and there is a general concern that certain levels of these chemicals could reach the aquatic ecosystem and possible alter the snail life. Agricultural chemicals used in this study included two herbicides, an insecticide and a fungicide. The tenth generation of laboratory reared snails were selected and exposed to the varying concentrations(0-100 ppm) of these chemicals. As concentrations and time of exposure increase, the per cent mortality increases(p<0.01). $LC_{50}$(lethal concentration for 50% mortality) values of these chemicals on snail after 96-hour exposure were variable; iprobenfos showed the highest acute toxicity(12.6 ppm), while carbofuran showed the lowest acute toxicity(74.5ppm). Sublethal concentrations of chemicals after 96-hour exposure were also variable ; bentazone showed the highest chronic toxicity(0.81ppm), while carbofuran showed the lowest chronic toxicity(5.04 ppm).

Toxicity characteristics of sewage treatment effluents and potential contribution of micropollutant residuals

  • Background: A typical sewage treatment plant is designed for organic and nutrient removal from municipal sewage water and not targeted to eliminate micropollutants such as pesticides, pharmaceuticals, and nano-sized metals which become a big concern for sustainable human and ecological system and are mainly discharged from sewage treatment plant. Therefore, despite contaminant removal by wastewater treatment processes, there are still remaining environmental risks by untreated pollutants in STP (sewage treatment plant) effluents. This study performed aquatic toxicity tests of raw wastewater and treated effluents in two sewage treatment plants to evaluate toxicity reduction by wastewater treatment process and analyze concentration of contaminants to reveal potential toxic factors in STP effluents. Methods: Water samples were collected from each treatment steps of two STPs, and acute and chronic toxicity tests were conducted following USEPA (United States Environmental Protection Agency) and OECD (Organization for Economic Cooperation and Development) guidelines. Endpoints were immobility for mortality and reproduction effect for estrogenicity. Results: Acute $EC_{50}s$ (median effective concentration) of influents for Seungki (SK) and Jungnang (JN) STPs are $54.13{\pm}32.64%$ and $30.38{\pm}24.96%$, respectively, and reduced to $96.49{\pm}7.84%$ and 100%. Acute toxicity reduction was clearly correlated with SS (suspended solids) concentration because of filter feeding characteristics of test organisms. Chronic toxicity tests revealed that lethal effect was reduced and low concentration of influents showed higher number of neonates. However, toxicity reduction was not related to nutrient removal. Fecundity effect positively increased in treated wastewater compared to that in raw wastewater, and no significant differences were observed compared to the control group in JN final effluent implying potential effects of estrogenic compounds in the STP effluents. Conclusions: Conventional wastewater treatment process reduced some organics and nutritional compounds from wastewater, and it results in toxicity reduction in lethal effect and positive reproductive effect but not showing correlation. Unknown estrogenic compounds could be a reason causing the increase of brood size. This study suggests that pharmaceutical residues and nanoparticles in STP effluents are one of the major micropollutants and underline as one of estrogenic effect factors.

Derivation of Ecological Protective Concentration using the Probabilistic Ecological Risk Assessment applicable for Korean Water Environment: (I) Cadmium

  • Probabilistic ecological risk assessment (PERA) for deriving ecological protective concentration (EPC) was previously suggested in USA, Australia, New Zealand, Canada, and Netherland. This study suggested the EPC of cadmium (Cd) based on the PERA to be suitable to Korean aquatic ecosystem. First, we collected reliable ecotoxicity data from reliable data without restriction and reliable data with restrictions. Next, we sorted the ecotoxicity data based on the site-specific locations, exposure duration, and water hardness. To correct toxicity by the water hardness, EU's hardness corrected algorithm was used with slope factor 0.89 and a benchmark of water hardness 100. EPC was calculated according to statistical extrapolation method (SEM), statistical extrapolation $method_{Acute\;to\;chronic\;ratio}$ ($SEM_{ACR}$), and assessment factor method (AFM). As a result, aquatic toxicity data of Cd were collected from 43 acute toxicity data (4 Actinopterygill, 29 Branchiopoda, 1 Polychaeta, 2 Bryozoa, 6 Chlorophyceae, 1 Chanophyceae) and 40 chronic toxicity data (2 Actinopterygill, 23 Branchiopoda, 9 Chlorophyceae, 6 Macrophytes). Because toxicity data of Cd belongs to 4 classes in taxonomical classification, acute and chronic EPC (11.07 ${\mu}g/l$ and 0.034 ${\mu}g/l$, respectively) was calculated according to SEM technique. These values were included in the range of international EPCs. This study would be useful to establish the ecological standard for the protection of aquatic ecosystem in Korea.

Folic acid supplementation reduces oxidative stress and hepatic toxicity in rats treated chronically with ethanol

  • Folate deficiency and hyperhomocysteinemia are found in most patients with alcoholic liver disease. Oxidative stress is one of the most important mechanisms contributing to homocysteine (Hcy)-induced tissue injury. However it has not been examined whether exogenous administration of folic acid attenuates oxidative stress and hepatic toxicity. The aim of this study was to investigate the in vivo effect of folic acid supplementation on oxidative stress and hepatic toxicity induced by chronic ethanol consumption. Wistar rats (n = 32) were divided into four groups and fed 0%, 12%, 36% ethanol, or 36% ethanol plus folic acid (10 mg folic acid/L) diets. After 5 weeks, chronic consumption of the 36% ethanol diet significantly increased plasma alanine transaminase (ALT) (P < 0.05) and aspartate transaminase (AST) (P < 0.05), triglycerides (TG) (P < 0.05), Hcy (P < 0.001), and low density lipoprotein conjugated dienes (CD) (P < 0.05) but decreased total radical-trapping antioxidant potential (TRAP) (P < 0.001). These changes were prevented partially by folic acid supplementation. The 12% ethanol diet had no apparent effect on most parameters. Plasma Hcy concentration was well correlated with plasma ALT (r = $0.612^{**}$), AST (r = $0.652^*$), CD (r = $0.495^*$), and TRAP (r = $-0.486^*$). The results indicate that moderately elevated Hcy is associated with increased oxidative stress and liver injury in alcohol-fed rats, and suggests that folic acid supplementation appears to attenuate hepatic toxicity induced by chronic ethanol consumption possibly by decreasing oxidative stress.

Chronic Toxicity(Mortality) of Freshwater Amphipod Diporeia spp. for Zn in Sediment Microcosm

  • Sediment microcosm experiments were conducted for 14 and 28 days using Zn spiked sediment to examine chronic toxicity (mortality) of Diporeia spp. as a function of density and time. Mean cumulative Diporeia mortality in 28 day sediment microcosms was 25% at $1,800\;{\mu}g\;g^{-1}$ total Zn in sediment. Although a certain fraction $(20{\sim}40%)$ of Diporeia was dead, its mortality was attributed by handling stress within 4 days and was not significantly encreased with increasing within the range of Zn concentrations examined in this study. These results suggest that Diporeia can tolerate Zn contaminated sediment and may be useful as a biomonitor for Zn contamination in freshwater environments.

A Study on the Effects of Radix Menispermi Extracts Against Cadmium Chloride Sub-chronic Toxicity in Rats (카드뮴 중독에 대한 산두근 추출액의 해독효과에 관한 연구)

  • This study was conducted to investigate the antitoxic effects of Radix Menispermi extracts against cadmium toxicity. The experimental rats were divided into 5 groups such as control group, cadmium alone treatment group and simultaneous treatment groups of cadmium and three doses of Radix Menispermi extracts. Each group was administered with different dose of Radix Menispermi extracts such as 0.55 mg, 1.10 mg, 1.65 mg/kg wet weight in pallets for 12 weeks. Cadmium Chloride $(CdCl_2)$ was administered by 4 mg/kg body weight. The results were summarized as follows: 1. The simultaneous administration of cadmium and Radix Menispermi significantly more decreased cadmium concentration in liver tissues compared to the administration of cadmium only (P < 0.05). 2. When blood were measured, no significantly difference in haemoglobin, haematocrit, erythrocyte values compared to the administration of cadmium only. 3. The simultaneous administration of cadmium and Radix Menispermi more increased metallothionein concentration in liver than the administration of cadmium only (P < 0.05). 4. There were the histopathological slight changes in the liver and kidney tissues of rats.

The Long-Term Safety and Efficacy of Intrathecal Therapy Using Sufentanil in Chronic Intractable Non-Malignant Pain

  • This report describes the long term safety and efficacy of intrathecal therapy using Sufentanil for the management of chronic intractable neuropathic pain in 12 chronic pain patients. Standardized psychological screening was used to determine treatment suitability. Evaluation data included the Visual Analog Scale (VAS), Wong-Baker Faces Scale, Brief Pain Inventory (BPI), Disability of Arm, Shoulder, and Hand (DASH), McGill Quality of Life Questionnaire, and complications (granulomas, toxicity, withdrawal, or deaths). SPSS version 18 was used for data analysis. Pre- and post- treatment BPI measures and pain scale scores showed a statistically significant difference. There were no complications directly related to drug toxicity, nor drug withdrawals, granulomas, or deaths. Intrathecal therapy with Sufentanil therapy offers a good treatment alternative for those cases that have failed both surgery and standard pain treatment. Strict patient selection based on psychological screening, control of co-morbidities, a proper pain management may contribute to successful outcome.

Subchronic Oral Toxicity Study of Acanthopanax divaricatus var. albeofructus in Rats

  • Acanthopanax divaricatus (Siebold & Zucc.) Seem. var. albeofructus (ADA), a traditional medical herb, has been used to treat arthritis and muscular injury, to strengthen muscle and bone, and to get vital energy. However, information regarding its toxicity is limited. ADA was administered by oral gavage to groups of rats at doses of 0 (control), 1,000, 1,500, 2,000, 2,500, and 3,000 mg/kg five times per week for 13 weeks. Mortality, clinical signs, body weights, food consumption, hematology, serum chemistry, urinalysis, organ weights, necropsy, histopathological finding, vaginal cytology, and sperm morphology were compared between control and ADA-treated groups. Salivation was intermittently observed in both sexes receiving 2,500 and 3,000 mg/kg directly after dosing. Absolute liver weights increased in females receiving 2,000, 2,500, and 3,000 mg/kg ADA (P < 0.05, P < 0.01, and P < 0.01, respectively) and so did the relative liver weights (P < 0.001). Salivation and increased liver weight were ADA-related changes but not considered to be adverse effects. Salivation was intermittent and transient, and the liver weight increase was minor and not accompanied by other changes such as hepatic morphological or functional alterations. The no-observed-adverse-effect-level was determined to be at least 3,000 mg/kg in both sexes of rats.

The Acute and Chronic Toxicity Studies of Herbicide, Molinate to Waterfleas (Molinate의 물벼룩에 대한 급성 및 만성독성 연구)

  • To assess the impact of molinate on freshwater aquatic organisms, acute and chronic toxicity studies for waterfleas were conducted. In acute toxicity studies for Daphnia magna, and Moina macrocopa, the 48-h $EC_{50}$ values were 11.4 and 8.3 mg/L respectively. And in reproduction toxicity studies for the same species, the NOEC's were 2.5 and 2.0 mg/L respectively. These results suggest that waterfleas have simillar sensitivity to molinate. On the other hand, the NOEC for 3-generation toxicity of moina macrocopa 0.16 mg/L, was much lower than those of acute values. This studies concludes that molinate has minimal risk to waterfleas in river.
