  • choChorion(egg-shell) morphology of the F1 hybrid between Bombyx mori and Bombyx mandarina has been observed by scanning electrom microscope and chorion protein was analyzed by electrophoresis. The chorion surface structure of F1 hybrids in the lateral(flat) region was similar to that of maternal line. The F1 hybrids chorion was found to have basically a three layer structure. The middle and inner layer were very much like those of the Bombyx mandarina and Bombyx mori. There were many conic pillar structures in the outer layer of the F1 hybrid, which was similar to Bombyx mandarina. This conic pillar structure had a thin cover layer was more clear in the dorsal and ventral side of the F1 hybrid chorion. The conic pillar structure of Bombyx mandarina was found to be dominant in F1 hybrid chorion irrespective of their maternal line. Major components of chorion protein were analyzed by two-dimensional electrophoresis and found to have isoelectric points in the range of pH 4.0-6.5 and molecular weight 10 to 50 kd. F1 hybrid chorion protein components related directly to those of the maternal line. The conic pillar structure was dominat characteristic and it was present in all F1 hybrid.

  • The surface patterns and the structures of transverse section of the egg-shell of the sikmoth, Bombyx mandarina, have been described by scanning electron microscope. Three spatially differentiated cross section, called lamellar, conic pillar and cover layers, are found on the mature eg-shell. Silkmoth chorion proteins were detected more than 80 components from a single chorion by two-dimensional electrophoresis. Major protein components of the egg-shell have bee identified on the basis of their isoelectric points and molecular weights, pH 4-6 and 6-30 kd. Several protein components are found entirely or predominantly in th cover layers.

  • Bovine yolk sac at day 24 of pregnancy, and placental membranes (chorion and allantois) from days 70 and 100 of pregnancy were isolated and cultured in a modified minimum essential medium in the presence of $[^{35}S]$methionine. Proteins synthesized and secreted by isolated bovine yolk sac, chorion and allantois were analyzed by fluorography of two-dimensional polyacrylamide gel electrophoresis. Serum-like proteins, transferrin, ${\alpha}$-fetoprotein, ${\alpha}$1-antitrypsin and ${\alpha}$1-acid glycoprotein, were the major protein products of yolk sac. A 21 kDa protein produced by yolk sac was identified immunochemically as retinol-binding protein (RBP). Chorion and allantios from days 70 and 100 of pregnancy were active in protein synthesis and secretion. Both chorion and allantois did not secret serum-like proteins but secreted a number of neutral-to-acidic proteins including RBP. Secretory proteins produced by the yolk sac, chorion and allantois may play important roles in the embryonic development and the successful outcome of pregnancy. Antiserum against bovine placental RBP was employed to the immunocytochemistry by immunoperoxidase method. Immunoreactive RBP was localized in epithelial cells and island-like cell clones of yolk sac. Immunostaining for RBP was detected in simple columnar epithelium of chorion and in simple squamous epithelium of allantois. In the present study, proteins synthesized and secreted by yolk sac at day 24 of pregnancy, chorion and allantois from days 70 and 100 of pregnancy were characterized In addition, RBP was localized in yolk sac, chorion and allantois by immunoperoxidase method. The immunoperoxidase method has been proven to be a very effective technique to identify the cellular source of protein synthesis in extraembryonic membranes.

On the basis of protem synthetic profiles, 8 stages of chorwgenesis are defined. Detail a analysis of the proteins of chorion of Bombyx mandarina, reveals more than 17 components by eletrophoretic mobility and synthetic kinetics. We have defined three abbreviated synthetlc s stages, based on the pattern of protein synthesized at early, middle, and late synthetic stages

  • The eggs of most animal species are surrounded by an extracellular matrix known as chorion, egg envelope, egg coat, or zona pellucida. Development of fish embryo usually takes several days in an aquatic environment. During embryonic development, the chorion must protect embryo from physical damage and microbial infection in the exposed aquatic environment. (omitted)

  • The developing ovarian oocyte of Dendrolimus spectabilis has been studied by using electron microscopical techniques. After yolk formation the vitelline membrane was laid down in the intercellular space between the follicle cell and the oocyte. But before the vitelline membrane formation the granules with high electron density that the vitelline membrane precusor are observed in the follicle cell. At the late vitellogenesis stage these granules were transported into the intercellular space between the follicle cells and the oocyte. These granules fuse to each other and larger bodies which eventually produce the vitelline membrane. The vitelline membrane was distinguished into the light inner and dark outer membrane. Next the chorion was laid down. It was apparent that the chorion was laid down in the intercellular space immediately adjacent to the vitelline membrane, and that it was formed by the follicle cells only.

  • Oocyte maturation of the swordtail (Kiphophorus hellerii) was investigated by light and electron microscopy. In the ovary of the swordtail, various staged oocytes were observed, Mature oocytes were located in ovarian cortex, meanwhile immature ones were positioned in ovarian medulla. The oocyte was surrounded by several structures or cells such as chorion, follicle cells, follicular theaca and ovarian epithelium, respectively, from the inside toward outside. Growing and maturing oocytes healed numerous microvilli which interconnected the oocyte and the follicle cells to communicate each other. The mature oocyte had the electron dense chorion which appeared to be ultrastructure of two layers and contained pore canals. Oocyte maturation was characterized by not only the enlarged cell size and well differentiated cell organelles, brit also the increases of fat droplets, pinocytotic vesicles and yolk granules.
