$\beta$-cyclodextrin adsorption and saponification methods were applied to isolate and purify cholesterol from the by-product of the low-cholesterol egg yolk product. They by-product was prepared from processing low-cholesterol egg yolk followed by extracting with chloroform to remove $\beta$-cyclodextrin and concentrated to 3,069 mg% cholesterol. When $\beta$-cyclodextrin method between two purification methods was applied, 50% ethanol as a solvent showed higher cholesterol concentration of 5.82% rather than the other solvents. Repeated purification of 3 times could not improve the cholesterol concentration significantly(p<0.05). In case of purification using saponification method, hexane as a solvent for extraction of unsaponificated materials was more efficient to increase cholesterol concentration than chloroform and ether. 60 times(v/w) saponification solution (95% ethanol:33% KOH = 94:6) of sample weight was most effective to increase the cholesterol concentration of 35.7%. Repeated purification process by saponification method could increase cholesterol concentration to 95.7% by 4 times repetition.
The effects of diet induced hyperlipidemia on kidney function were studied in Sprague-Dawley rats fed high fat diet containing 20% beef tallow and high cholesterol diet containing 5% cholesterol for 8, 12, 16 weeks, respectively. The concentrations of serum total lipid, total cholesterol and LDL-cholesterol were significantly high cholesterol diet groups during all experimental periods (p<0.05). HDL-cholesterol concentration was the lowest value in high cholesterol diet group of 16 weeks(p<0.05). Triglyceride concentration was not affected by experimental diets. Serum total protein, albumin and creatinine concentration tended to higher in high cholesterol diet groups than those in high fat diet groups. And serum urea-N concentration was higher in high fat diet group of 16 weeks than that in other diet groups. Urinary total protein and urea-N were higher in high cholesterol diet groups than those in other diet groups regardless of experimental period period. There was no significant difference in urinary creatinine concentratin among diet groups(p<0.05). GFR was lower in high cholesterol diet groups than that in high fat diet groups at 8, 16 weeks, respectively. Wet weight per body weight, total lipid, triglyceride, total cholesterol concentations of liver tissue were apparently high in high cholesterol diet groups(p<0.05). Kidney wet weight per body weight wer not affected by experimently diets, total lipid concentration of kidney tissue was significantly high in high fat diet groups of 12 weeks(p<0.05), kidney tissue triglyceride concentrations of high cholesterol diet groups of 12, 16 weeks apparently low, and total cholesterol concentration of kidney tissue was higher in experimental diet groups than that of control groups at 12, 16 weeks(p<0.05). Fecal excretion, total lipid, triglyceride and cholesterol concentrations of feces were markedly high in all high cholesterol diet groups except high fat diet group of 16 weeks. The results of light microscopic examination indicated that glomerulosclerosis was not obsrved in rats fed experimental diets.
In this study, a new system to determine the concentration of cholesterol using a color change was developed. The system comprised diacetylene vesicles and cholesterol oxidase (ChOx). 10,12-Pentacosadiynoic acid (PCDA) was used as the diacetylene compound, and PCDA vesicles were formed using sonication. The H2O2 produced during the reaction between cholesterol and ChOx was used to initiate the polymerization of the PCDA in the vesicles. During polymerization, the vesicles changed from colorless to blue. Therefore, the cholesterol concentration was proportional to the intensity of the blue color. The absorption at 665 nm indicated that the blue color was directly proportional to the cholesterol concentration. This indicates that the system can be used for cholesterol detection. The minimum cholesterol concentration detected using this system was 1.0 mM.
This study was designed to observe the effects of the feeding of Hijikia fusiforme(Harvey) Okamura juice extract on the improvement of lipids in the serum of cholesterol-supplemented diet induced dietary hyperlipidemic rats fed experimental diets for 4 weeks. Concentration of total cholesterol and LDL-choloesterol in serum was significantly lower in the Hijikia fusiforme juice extract group than in the cholestero diet group. Concentration of HDL-cholesterol in serum was higher in the Hijikia fusiforme juice extract group than the cholesterol supplemented diet group. Those in the Hijikia fusiforme juice extract group were higher than those in the cholesterol diet group. In the ration of HDL-cholesterol concentration to total cholesterol concentration, Hijikia fusiforme juice extract administration group was higher percentage than the other groups. Atherosclerotic index was lower in the Hijikia fusiforme juice extract group than in the cholesterol diet group. Concentration of free cholesterol and cholesteryl ester in serum was rather lower in the Hijikia fusiforme juice extract group than the other groups. Triglyceride concentration in serum was significantly lower in the Hijikia fusiforme juice extract group than in the cholesterol diet group. Concentration of phospholipid in serum was more decreased in the Hijikia fusiforme juice extract group than in the cholesterol diet group. The activities of aspartate aminotransferase and alanine aminotransferase in serum were rather lower in the Hijikia fusiforme juice extract group than the cholesterol supplemented diet group. From the avove research, the Hijikia fusiforme juice extracts were effective on the improvement of the lipid compositions in serum of high fat diet induced dietary hyperlipidemic rats.
In order to investigate the hypocholesteremic effect of soybean perptides, soybean protein(ISP), casein(CNP), their peptic hydrolyzates fractionated by acid precipitation at different pH's(SHT, SH8, SH6, SH4, CHT, CH5, CH4) and amino acid mixtures of the same composition as the proteins(SAA, CAA) were fed to rats and the concentration of serum cholesterol was measured. Then in vitro digestibility and molecular weight distribution of the peptides by pepticpancreatic hydrolysis was measured by FPLC. The lower the in vitro digestibility of peptides is, the lower the concentration of serum cholesterol becomes(r=0.986) and the higher the ratio of macropeptides is, the lower the concentration of serum cholesterol becomes(r=-0.932) in rats. These results suggest that the in vitro digestibility of peptides has close relationship to the concentration of serum cholesterol in rats and non-digestible meacropeptides or polypeptides especially more than 1 kDa, formed through digestion in gut, may lower the serum cholesterol in rats.

Influence of dietary sesame meal on plasma glucose, non-esterified fatty acid (NEFA), triglyceride, total cholesterol, high-density lipoprotein (HDL)-cholesterol and urea concentrations in goats was examined. Goats were fed a control diet (50% timothy hay and 50% concentrates) (CD) or a sesame meal diet (50% timothy hay, 25% concentrates and 25% sesame meal) (SMD) during 12 days. Blood samples were taken after overnight fasting and afternoon every day. Body weight was not changed by feeding either CD or SMD. The concentrations of plasma triglyceride and urea were higher (p<0.05) in goats fed SMD than those fed CD. Plasma NEFA concentration was higher in plasma samples after overnight fasting. Plasma glucose concentration in plasma samples collected afternoon was higher than those after overnight fasting. Plasma total cholesterol concentration was significantly increased by feeding SMD but not by feeding CD, which was due to the remarkable increase of plasma HDL-cholesterol concentration. In conclusion, dietary sesame meal brought about an increase in plasma total cholesterol concentration accompanied with an increment in plasma HDL-cholesterol consentration in goats.
The present study examined the effect of soy isoflavones on lipid metabolism in growing female rats. Rats were randomly assigned to three different groups and provided experimental diets for 9 weeks. The experimental groups were classified into 1) a control group, 2) a soy protein isolate group: soy (+)) group and 3) a soy protein concentration group: soy (-)) group. Diets contained either casein or one of two soy proteins with (soy (+)) or without isoflavones (soy (-)). Serum triglyceride concentration showed no significant differences among the experimental groups. Serum total cholesterol concentration was significantly lower in both the soy (+) and soy (-) groups than in the control group and LDL-cholesterol concentration was significantly lower in the soy (+). Serum HDL-cholesterol concentration was significantly higher in the control group than in the soy protein groups but the HDL-cholesterol share rate in total cholesterol tended to be lower in the control group than in the soy protein groups, insignificant as it was. Hepatic IDL receptor mRNA level was significantly increased in the soy (+) group when compared to the other two groups to be 20% higher than the control group. In conclusion, soy protein isolate, soy protein rich with isoflavones reduced serum total cholesterol and LDL-cholesterol concentration and increased hepatic IDL receptor mRNA expression in growing female rats. Therefore, it is considered that the intake of soy isoflvones during puberty can be advantageous in terms of the long-tenn control of serum lipid.
This study was conducted to investigate the effect of fructooligosaccharide on intestinal flora, lipid metabolism and immune response. Thirty two male rats of Sprague-Dawley strain were divided into two groups according to body weight. Each group was fed the diet containing 2% cholesterol or the normal diet, respectively for 4 weeks. Each group was again divided into two sub-groups and they were fed with the diet containing 5% of sucrose and fructooligosaccharide, respectively for 8 weeks. The number of bifidobacteria slight increased, but not significantly, in oligosaccharide groups. Plasma total lipid concentration in cholesterol group was signficantly increased compared to the value in normal group. Dietary oligosaccharide decreased plasma total lipid concentration and triglyceride concentrations in normal group, but not in cholesterol group. Fecal lipid excretion was higher in cholesterol group than in normal group. Fecal cholesterol concentration in cholesterol-oligosaccharide group was significantly increased compared to other groups. Cholesterol had slight effects on TBARS and the immune status, but dietary oligosaccharide had no effects on these variables. The results in this study suggest that dietary oligosaccharide had no effect on serum and liver profiles of rats fed high cholesterol diet but increases the fecal cholesterol excretion.
The chemical composition and hypocholoesterolaemic effect of Nostoc commune were studied. Proximate analysis data, composition of dietary fiber and oxalic acid-ammonium oxalate soluble substance of Nostoc commune were presented. Thirty rats were fed with the one of the following diet for three weeks: The control diet contained 0.75% cholesterol, 0.25% sodium cholate and 5% cellulose powder, CF was cholesterol free, PNC, ONC and RNC diet contained 7.2% powder, 2.2% extracts and 5% residue of Nostoc commune respectively. The lipid components of serum were assayed. The concentration of the total cholesterol was significantly lower in PNC, ONC than the control. The concentration of the total cholesterol was significantly lower in PNC, ONC than the control. The concentration of HDL-choloesterol was significantly higher in PNC than the control. The concentration of LDL, VLDL and chylomicron were comparatively lower in CF and PNC. The concentration of seum triglyceride and phospholipid was lower in PNC group than in the other group.

We analyzed and compared the concentration of total cholesterol (CHOL), high density lipoprotein (HDL) cholesterol, low density lipoprotein (LDL) cholesterol and triglyceride (TG) in serum and the serum protein electrophoresis fractions of thyroid disease patients. In comparison with the average of reference, our data showed that the average concentration of CHOL, LDL cholesterol and TG in hyperthyroidism patients were decreased significantly, but HDL cholesterol was increased significantly. In hypothyroidism patients, CHOL, HDL cholesterol, LDL cholesterol and TG were all increased significantly. In comparison of the concentration of lipids in each patient to reference range, 28.3% of hyperthyroidism patients showed abnormally low level of total cholesterol. In the patients with hypothyroidism, the percentage of patients showed abnormally high level of CHOL, HDL cholesterol, LDL cholesterol and TG were 37.7%, 10%, 68.8% and 49.1%, respectively. In our studies of serum protein electrophoresis, the average of ${\alpha}_2$-globulin and $\gamma$-globulin in hyperthyroidism patients were increased and $\beta$-globulin was decreased significantly. In hypothyroidism patients, the average of $\gamma$-globulin was increased and $\beta$-globulin was decreased significantly. In comparison of protein fractions of each patient to reference range, 38.3% and 50.0% of hyperthyroidism patients showed abnormally high levels of ${\alpha}_2$-globulin and $\gamma$-globulin, but 73.3% of patients showed abnormally low level of $\beta$-globulin. In hypothyroidism patients, 70.4% of patients were abnormally decreased in $\beta$-globulin and 63.9% of patients were abnormally increased in $\gamma$-globulin. These data suggest that the concentrations of CHOL, HDL cholesterol, LDL cholesterol and TG are not critical data for clinical interpretation of hyperthyroidism, but the levels of them are useful for interpretation of hypothyroidism patients. Our results of serum protein electrophoresis suggest that the concentration of serum protein electrophoresis fractions can be useful to understand the thyroid disease.
