Antiobesity Effect of Recombinant Human Caseinomacropeptide tide in Sprague-Dawley Rat

  • Caseinomacropeptide (CMP) is a glycopeptide of 64 amino acid residues derived from the C-terminal of mammalian milk K-casein. Recently, human CMP (hCMP) was produced from the recombinant yeast Saccharomyces cerevisiae. In this study, the antiobesity activity of the recombinant hCMP (rhCMP) was investigated in vivo using Sprague-Dawley rats. The rhCMP did not affect the rats fed with a normal fat diet (fat content, $5.0\%$), but decreased the body weight gain of the rats fed with a high fat diet (fat content, $20\%$) by up to $19\%$. Autopsies revealed that the weights of the liver, kidney and adipose tissues decreased when the rats were given the rhCMP, which also reduced the lipid concentrations in the plasma and liver, but enhanced the fecal excretion of lipids. These results suggest that rhCMP prevent the accumulation of lipid by stimulating its fecal excretion, so could be used as a food supplement to alleviate the obesity problem caused by a high fat diet.

An Immunohistochemical study on the endocrine cells in the gastrointestinal tract of the snakehead, Ophicephalus argus (가물치 위장관 내분비세포에 관한 면역조직학적 관찰)

  • The regional distribution and relative frequency of occurrence of endocrine cells in nine segments of the gastrointestinal(GI) tract of snakehead(Ophicephalus argus) were investigated by immunohistochemical methods using specific antisera against 5- hydroxyptrytamine(5-HT), somatostatin, gastrin/cholecystokinin(GAS/CCK), glucagon, bovine chromogranin, porcine chromogranin and insulin. Four types of immunoreactive cells for 5-HT, somatostatin, GAS/CCK and glucagon were observed in the GI tract. These cells were generally appeared in the mucosal epithelia or located at the interface of the mucosal epithelial layer and intestinal glandular region. 5-HT-immunoreactive(IR) cells were found in segment II, III, IV, V and VI, and the most numerous in segment IV. Somatostatin-IR cells were found in segment II, III, IV and V, and the most numerous in segment III. GAS/CCK-IR cells in segment VI, VII and glucagon-IR cells in segment III, IV, V were detected but a few in these segments. No bovine chromogranin-, porcine chromogranin- and insulin-IR cells were detected throughout the GI tract of the snakehead.

Response of appetite-related genes in relation to the rearing water temperature in red spotted grouper (Epinephelus akaara)

  • Growth of fish is controlled by various environmental factors, including water temperature (WT). WT is also a major factor that affects the eating behavior of fish. In this study, we studied the relationship between WT and expression profiles of factors involved in appetite regulation and digestion in red spotted grouper. Fish were divided into three groups with 30 fish per group and reared for 2 weeks in a recirculating system at three different WT (natural WT, 20 ℃, and 25 ℃). After 2 weeks, they were sacrificed immediately after food intake (+ 0 h), 3 h after (+ 3 h), 6 h after (+ 6 h), and 3 h before being fed the next day (- 3 h). Fish samples were obtained for analysis of mRNA expression levels of cholecystokinin (CCK), leptin A (LepA), leptin B (LepB), and neuropeptide Y (NPY) in the brain, liver, and intestine. Expression of CCK and NPY mRNA was significantly higher at 25 ℃ compared to that at natural WT and 20 ℃, but no difference was observed for leptin. This suggests that the red spotted grouper shows characteristics typical of subtropical fish and has higher levels of feeding and digestion when reared at 25 ℃ water temperature condition.

  • The gastrointestinal tract of three Percida, Lateolabrax japonicus, Epinephelus septemfasciatus and Mugil cephalus, was investigated immunocytochemically for the occurrence of somatostatin-. seotonin-, gastrin-, pancreatic polypeptide(PP)-, cholecystokinin-8(CCK-8)- and glucagon-immunoreactive cells. In Lateolabrax japonicus and Epinephelus septemfasciatus, five endorcrine cell types, such as somatostatin-, serotonin-, gastrin-, PP- and CCk-8-immunoreactive cells were demonstrated. In Mugil cephalus, however, six endocrine cell types, such as somatostatin-, serotnin-gastrin-, PP-, CCK-8- and glucagon-immunoreactive cells were detected. Somatostatin- and serotonin-immunoreactive cells were detected in the gastric mucosa of all species. Glucagon-immunoreactive cells were found only in the gastric mucoas of Mugil cephalus. In the pyloric caeca, PP-and CCK-8-immnuoreactive cells fo all species. gastrin-immunoreactive cells of Epinephelus septemfasciatus and Mugil cephalus, and serotonin-immunoreactive cells of Epinephelus septemfasciatus were demonstrated. In the intestinal mucosa of all species, gastrin-, PP- and CCK-8-immunoreactive cells were detected, and in the intestinal mucosa of Epinephelus septemfasciatus serotonin-immunoreactive cells were also detected. The frequency of these immunoreactive cells differs from each portion of the gastrointestinal tract of all species.

  • Objectives : Sasang Constitutional Medicine is based on the diversity of human beings and medically developing a variation of responses to diseases and medicines. This diversity is categorized into four concerning morphology, physiology, pathology, and pharmacology. It is supposed that sasang constitutional medicine is related the genetic diversity of individuals. Single nucleotide polymorphism is the basic tool to research genetic polymorphism as a landmark of genomes. Each constitution has different processes of pathophysiology and metabolisms to herb medications. In clinical research, the stroke incidence is significantly different by constitution. Methods : We researched whether the polymorphic expression of CCK (rs=2241997) depends on sasang constitution. The [c/t] polymorphism site of promotor region of CCK gene on 3p22-p21.3 was investigated. Results : The allele frequency of [c/t] polymorphism of CCK promotor region was different in constitution groups compared to the average allele frequency of SNP DB. The allele frequencies of Soeumin and Soyangin groups were (c:0.70/t:0.30). and (c:0.71/t:0.29), that of Taeumin group was (c:0.57/t:0.43) and of Taeyangin group was (c:1.00/t:0.00) Conclusions : It was regarded the [c/t] polymorphism of CCK promotor region is available to classify the constitution. However, it is necessary to research about CCK gene polymorphism and more constitution population groups. It is also necessary to research the more functional gene's polymorphism and sasang constitution.

  • The regional distribution and relative frequency of endocrine cells in the alimentary tract of the snake, Rhabdophis tigrinus tigrinus, were investigated by immunohistochemical method using 7 antisera. Chromogranin (Cg)-, glucagon-, somatostatin-, gastrin/cholecystokinin (Gas/CCK)-, serotonin-, bovine pancreatic polypeptide (BPP)-immunoreactive cells were identified in this study. Cg-immunoreactive cells were detected throughout the alimentary tract including the esophagus, with predominant frequency in the pylorus. Numerous immunoreactive cells were observed from the esophagus to the pylorus but a few cells were detected in the large intestine. Glucagon-immunoreactive cells were observed from the proximal portions to the distal portions of the small intestine. They were increased to the middle portions but thereafter decreased, and no cells were found in the terminal portions. Somatostatin-immunoreactive cells were restricted to the small intestine and these cells were decreased toward to distal portions of the small intestine. Gas/CCK-immunoreactive cells were detected in the pylorus and small intestine. They were most predominant in the pylorus and the proximal portions of the small intestine but thereafter decreased toward to the distal regions. Serotonin-immunoreactive cells were observed throughout the alimentary tract. They were most predominant in the pylorus and proximal portions of the small intestine but a few cells were observed in the large intestine. BPP-immunoreactive cells were restricted to the distal portions of the small intestine with rare frequency. No bombesin-immunoreactive cells were found in this study.

  • The pepsinogen secretion was stimulated by the cholecystokinin, caerulein, isoproterenol, and carbachol, respectively. But it was increased slightly and returned to control level by the combiantions of total saponin with each above the agents. Even though the atropine had the inhibition effect, the pepsinogen secretion was recovered to normal level from depressed condition by the combination of the atropine with total saponin. Propranolol showed the same pattern as atropine, too. On the other hand, the pepsinogen secretion was stimulated by the DBcAMP alone, but decreased to control level by the combination with the total saponin. In the case of DBcGMP, the pepsinogen secretion was decreased by itself, but stimulated the above control level by the combination with total saponin. Histamine alone had little effect on the pepsinogen secretion, but when combinated with total saponin, the pepsinogen secretion was increased. Serotonin alone and with total saponin, had no effect respectively, From the above results, the total saponin may have the normalization action stimulating or decreasing the pepsinogen secretion to the control level.

Histological and immunohistochemical effects of Jengjengamiyjintang on the duodenal ulcer induced by HCl-aspirin (정전가미이진탕이 HCl-aspirin으로 유발된 십이지장 궤양에 미치는 조직학적 및 면역조직화학적 연구)

  • In order to study the effects of Jengjengamiyjintang on the duodenal ulcer induced by HCl-aspirin in rats, the changes of histological profiles, goblet cells(PAS-positive cells), and the distribution and frequency of cholecystokinin(CCK)-8 and serotonin-producing gastro-entero-endocrine cells were observed after oral administration of Jengjengamiyjintang. Histologically, very severe injury to duodenal epithelium were observed in control groups and these injuries were increased with time intervals. But in the Jengjengamiyjintang administrated groups, no gross lesion of ulcer were demonstrated and histologically minor injury to the mucosal epithelium were observed. PAS-positive cells were increased in the Jengjengamiyjintang administrated groups compared to that of control groups. Severe degranulation of CCK-8- and serotonin-immunoreactive cells were observed in control groups but these phenomenon was seldom in the Jengjengamiyjintang administrated groups. Serotonin-immunoreactive cells were significantly decreased in control groups but increased in Jengjengamiyjintang administrated groups compared with control groups. According to these result, it is suggested that Jengjengamiyjintang would accelerat the healing of the duodenal ulcer but the functional mechanisms were unknown.

Effect of Phenoxybenzamine on the Contraction of the Isolated Esophagus Strip Induced by CCK-PZ (Cholecystokinin-pancreozymin의 식도절편 수축반응에 미치는 phenoxybenzamine의 영향)

  • In this study, the effects of phenoxybenzamine and related drugs on the action of CCK-PZ and caerulein were examined in isolated gall bladder of guinea pig and higher esophagus strip of fowl. The strips were placed in a bath containing Locke-Ringer solution maintained at $38^{\circ}C$. Oxygen was continuously bubbled through the solution. The contractile response was measured isometrically by a force displacement transducer connected to polygraph. In isolated gall bladder preparation caerulein produced contractile response of CCK-PZ type, but the relative potency on a weight basis was 30 times stronger than CCK-PZ. The response of caerulein or CCK-PZ was not blocked by cholinergic blocking agent and both alpha and beta adrenergic blockades, however, the response of caerulein or CCK-PZ was exceptionally blocked by phenoxybenzamine. In isolated esophagus strip CCK-PZ with high concentration produced marked contraction which was not modified by atropine and other blocking agents, whereas the response was blocked by phenoxybenzamine. These results lead to the conclusion that phenoxybenzamine inherently inhibits the contractile response of CCK-PZ and caerulein on esophagus and other smooth muscle.

Immunohistochemistry of Endocrine Cells in the Alimentary Tract of the Tree Frog, Hyla arborea japonica

  • The regional distribution and relative frequencies of endocrine cells were studied immunogistochemically (PAP methods) in the alimentary tract of the tree frog, Hyla areorea japonica, using specific antisera against serotonin, somatostatin, bovine Sp-1/chromogranin (BCG), cholecystokinin (CCK)-8, vasoactive intestinal polypeptide (VIP), gastrin, bombesin, secretin and pancreatic polypeptide (PP). Six kinds of endocrine cells were identified in this study, These immunoreactive cells were located in the gastric glands of stomach regions and in the basal portion of the epithelium of the intestinal tract or esophagus with variable frequencies. They were spherical or spindle-shaped. Serotonin-immunoreactive cells were observed in the whole alimentary tract including the esophagus. Somatostatin-immunoreactive cells were also detected throughout the alimentary tract except the rectum. CCK-8-immunoreactive cells were observed from the pylorus to ileum. Vip-immunoreactive cells were restricted to the rectum. Bombesin-immunoreactive cells were restricted to the fundic gastric regions and gastrin-immunoreactive cells were restricted to the pylorus. However, no BCG-, secretin and PP-immunoreactive cells were demonstrated in this study. In conclusion, the regional distribution and relative frequency of the endocrine cells in the alimentary tract of the tree frog were similar to other anuran species, but some differences which may be caused by feeding habits and species specification were also observed.
