In Vivo H-1 MR Spectroscopy of Intracranial solid Tumors (두개강내 고형성 종양의 H-1 자기공명분광법)

  • Purpose: To assess the ability of in vivo H-1 MRS to determine the degree of malignancy and to characterize the histopathologic type of intracranial solid tumors. Materials and Methods: In vivo H-1 MR spectra of the pathologically-proven 81 intracranial soild tumors (low-grade glioma 17 cases, high-grade glioma 31 cases, lymphoma 9 cases, meningioma 8 cases, central neurocytoma 4 cases, medulloblastoma 3 cases, PNET 3 cases, metastasis 2 cases, others 4 cases) were analyzed. H-1 MR spectroscopy was performed on a 1.5T MR unit using PRESS sequence with a TR of 2000ms, a TE of 270 or 135ms and a voxel size of $2{\times}2{\times}2cm^3$ for all spectra. N-acetyl aspartate (NAA)/Creatine complex(Cr), Choline complex (Cho)/Cr, and lactate (Lac)/Cr ratios were measured on the peak heights of each resonance and compared among the different tumors. Results: All intracranial solid tumors demonstrated decreased NAA, elevated Cho and lactate, and variable Cr levels. All tumors showed increased Cho/Cr and Lac/Cr, whereas NAA/Cr level was decreased. Mean Cho/Cr and Lac/Cr ratios were significantly higher in high-grade gliomas than in low-grade gliomas. However, NAA/Cr ratio showed no significant difference between low-grade and high-grade gliomas. Very high Cho peaks were seen in lymphomas, meningiomas, medulloblastomas, and neurocytomas in addition to high-grade gliomas. Conclusion: H-1 MRS may be useful in differentiating between low-grade and high-grade gliomas, however cannot characterize the histologic types or subtypes of tumors.

Ultraviolet Radiation-Induced Apoptosis is Inversely Correlated with the Expression Level of Poly(ADP-ribose) Polymerase

  • The present study was conducted to elucidate whether the expression level of poly(ADP-ribose) polymerase (PARP) is related to the ultraviolet radiation (UV)-induced apoptosis. After treatment of the mammalian cell lines HeLa S3 and Chinese hamster ovary (CHO) with 50 J/m2 UV, induction of apoptosis was determined by several means during 24 h post-incubation. Incidence of apoptosis was much lower in CHO than HeLa S3 cells based on the percentage of apoptotic cells in terms of morphological changes in nucleus or direct counting of viable cells and qualitative or quantitative DNA fragmentation. Interestingly, when the expression level of PARP was measured by western blotting, the amounts of PARP that was retained at each time point inversely correlated with the incidences of apoptosis in these cells. Concomitant with generation of the 85 kDa fragment, 116 kDa PARP disappeared in HeLa S3 within 6 h after UV treatment, whereas a fair amounts of 116 kDa band was still retained in CHO cells at 36 h post-incubation. This inverse relationship was also observed in the adaptive response system, in which cells weve treated with a high dose of UV after pretreatment with a low dose. As expected, typical adaptive responses appeared in CHO cells but not in HeLa cells, showing greater cell viability and lesser DNA fragmentation. During the adaptive response in CHO cells, PARP was expressed at much higher level compared to the single, high dose-treated cells. Interestingly, even though PARP was induced at 6 h post-incubation In both cell types, its expression was more prominent in CHO cells. Thus, our data indicate that the retained level of intact PARP against UV damage inversely correlates with incidence of apoptosis in mammalian cells, and also suggest that a machinery to protect the PARP degradation against UV damage exists in CHO but not in HeLa S3 cells.

A Research on Housing Plan Competitions Promoted by Daily Cho-Sun Company in Japanese Colonial Period (1939년 조선일보 주최 조선주택설계도안 현상모집에 관한 고찰)

  • This study examines the details and characteristics of prizewinning plans of Cho-Sun housing plan competition promoted by Daily Cho-Sun Company during Japanese colonial period. The main features of Cho-Sun housing plan competition were that the competition held with an application of popular newspaper readers and a judging committee was composed of only Korean architects, especially Park, Gil-Ryong as the president of the board of examiners. Cho-Sun housing plan competition put ideological factors for the improvement of living as like advancing to the cultured(modem) life, respecting for family life, respecting for privacy of family members and so on. First of all, the competition suggested the direction of the improvement of dwelling spaces which focused on making harmonious home life. This study found common characteristics of prizewinning plans of Cho-Sun housing plan competition as the result. First, Cho-Sun housing plan competition introduced a centralized house plan as a model of the improvement of small-sized houses. Second, architectural orders of western or Japanese style mingled with ones of korean traditional style in prizewinning plans as entrance hall, western style reception room(parlour), maid's room, bathroom for example. And a manner of seating using chairs was brought into the style of living which would be assumed in the prizewinning plans. Third, traditional way of composing dwelling spaces was maintained, receiving the characteristics of dwelling spaces of japanese style simultaneously, which central corridor was put at the heart of house for improving arrangement of rooms and flow lines. Fourth, lavatory and bathroom were placed inside of the house.

Characteristics of Recombinant CHO Cell Growth and Erythropoietin Production in Serum-Containing Media and Serum-Free Media (혈청배지와 무혈청배지에서의 재조합 CHO 세포 성장과 Erythropoietin 생산)

  • We have investigated the characteristics of recombinant CHO cell growth and erythropoletin(EPO) production at different concentrations of serum and inoculation density. Cell growth and EPO production were increased with the increase of serum concentration and inoculation density. Enhancement of CHO cell growth and EPO production by medium exchange using serum-free medium at the growth phase of cells was studied. It was found that the exchange of culture medium with serum-free medium was favorable for growth of cells and production of EPO. The maximum number of cell and concentration of EPO obtained by exchanging culture medium were $6.2{\times}105cells/$\textrm{cm}^2$ and 7,470units/m1, respectively, compared to $2.1{\times}105cells/\textrm{cm}^2$ and 2,380units/m1 in serum-containing medium without medium exchange. It was observed that CHO cell growth was correlated with EPO production in serum-free media.

  • The study is on the beauty of the line in the Cho-sun Costume. In the aspect of the Costume Aesthetics and Study of Clothing Design, I try to find the beauty of the line in the external form and the immanent beauty of the external line in the Cho-sun Costume. The results of the study is that : 1) The beauty of the line external form have ① the functional beauty of the line ② the beauty of drape line ③ combination of the line of internal and external structure. ④ the beauty of wearing with the Chi-ma. ⑤ the beauty of sharp line in the ornaments 2) The immanent beauty of the line in the Cho-sun Costume : There are a natural beauty found by the reason of nature, temperate beauty, simple beauty in the Cho-sun Costume.

Proteome Analysis of Recombinant CHO Cells Under Hyperosmotic Stress

  • Under hyperosmotic stress, rCHO cells display decreased specific growth rate $({\mu})$ and increased specific antibody productivity $(q_{Ab})$. The effects of hyperosmotic stress on batch culture cellular dynamics are not well understood. To this end, we conducted a proteome profile of rCHO cells, using 2D-gel, MALDI-TOF-MS and MS/MS. As a result, the proteome profile of rCHO cells could be established using 41 identified proteins. Based on this proteome profile of rCHO cells, we have found at least 8 differently expressed spots at hyperosmotic osmolality (450 mOsm/kg). Among these spots, two metabolic enzymes were found to be up-regulated (pyruvate kinase and GAPDH), while down-regulated protein was identified as tubulin. It shows that hyperosmotic stress can alter metabolic state, by up-regulated activities of two glycolysis enzymes, which could lead to activate the generation of metabolic energy. Tubulin expression was down-regulated, suggesting a reduction of cell division. Finally, the increased conversion energy could leads to improve overall productivity.

Characterization of Soluble Type Human Folate Receptor on Folate Binding and Transport (Soluble type의 human folate receptor의 folate 결합과 수송에 관한 특성)

  • We have cloned a soluble type human folate receptor(hFR type${\gamma}$) from human thymus cDNA library using the PCR amplification technique. To examine whether hFR type${\gamma}$ has a folate transport activity, CHO cells were transfected with the pcDNAhFR${\gamma}$ expression plasmid, and the stable cell line CHO/hFR${\gamma}$ expressing a high level of the hFR type${\gamma}$ was identified by northern and western blot analysis. The CHO/hFR${\gamma}$ cells produced a [$H^3$]folic acid binding protein in the culture medium. However, we couldn't detect any cell surface [$H^3$] folic acid binding and transport activities. The growth of the CHO/hFR${\gamma}$ cells was more rapidly inhibited than the wild type CHO cells in the low concentration folic acid media. These observations indicate that although soluble type human folate receptor can bind [$H^3$]folate, it does not involve in folate transport.

  • In this paper, noble electrical equivalent circuit ultrasonic motor is applied by model of parallel one from mechanical equation of piezoelectric device and characteristics estimation is done by equivalent converter. Uitrasonic motor is driven by oscillation circuit of two-phase high frequency inverter. Operating frequency of inverter is described by tracking control method of resonance frequency, according to change of load, temperature etc.

  • Korean folk medicine 'Sin Seong Cho' (神聖草)has been used to cure common cold, neuralgia and cancer of the stomach in Korea. The botanical origin of the crude drug has been no pharmacognostical confirmation on it. To clarify the botanical origin of 'Sin Seong Cho', the morphological and anatomical characteristics of the rhizomes of Davallia mariesii Moore and Neocheiropteris ensata (Thunb.) Ching growing in Korea were examined. As the result, it was made clear that 'Sin Seong Cho' from Korea was derived from the rhizomes of Davallia mariesii, rarely from the rhizomes of Neocheiropteris ensata.
