Photosystem I and II Reaction Centers in a New Type Oxygenic Photosynthesis of Acaryochloris marina Based on Chlorophyll d: Studies of Delayed Fluorescence and Triplet State ESR

  • A cyanobacteria-like organism Aaryochloris marina performs oxygenic photosynthesis with near far-red light by the use of chlorophyll d. Reaction center chlorophyll (Chl) of Photosystem (PS) II of A. marina was studied by analysis of millisecond-delayed fluorescence. Delayed fluorescence is emitted by Chi d indicating efficient energy transfer between antenna Chi d molecules and the unknown primary electron donor of PS II. P740 a reaction center Chl of PS I of A. marina is shown to give a dimer type cation, and triplet state with a D value of 245xlO$\^$-4/ cm$\^$-l/ in contrast to the 280-290 xlO$\^$-4/cm$\^$-l/ values of P700 suggesting triplet spins interacting at a 5% larger distance in P740 than in P700.

Developmental Changes in Photosynthetic Pigments and Chlorophyll Fluorescence in Etiolated Rice Seedlings During Greening

  • Developmental of photosynthetic pigments and changes in chlorophyll fluorescence of dark-grown rice seedlings were studied during greening. Light-illumination stimulated accumulations of total chlorophylls and carotenoids in leaves of etiolated seedlings, accompanied by a decrease in the ratio of chlorophyll a to chlorophyll b. When the composition of carotenoids was analyzed, violaxanthin level was shown to increase up to 24 h after the beginning of light illumination, followed by a subsequent decline. In contrast to this, zeaxanthin level increased consistently with progress of deetiolatin. The role of zeaxanthin is discussed in relation to chlorophyll fluorescence quenching. A study on chlorophyll fluorescence kinetics of the rice seedlings being deetiolated showed a time-dependent increase in Fv/Fm (yield of variable fluorescence/maximum yield of fluoresecnece) ratios, indicating that greening is responsible for the activation of photochemical reaction centers of the photosystem. When chlorophyll fluorescence quenching was examined, qNP (nonphotochemical quenching) and qE (energy-dependent quenching) exhibited a time-dependent decline with progress of greening. The presented results indicate that greening-induced development of the photosynthetic machinery is associated the conversion of the carotenoid violaxanthin to zeaxanthin, suggesting that zeaxanthin synthesized in the illuminated leaves may provide the protection from the damage when etiolated plants are exposed to light.

Chlorophyll a Fluorescence Parameters of Hulled and Hull-less Barley (Hordeum vulgare L.) DH Lines Inoculated with Fusarium culmorum

  • Barley worldwide is affected seriously by Fusarium seedling blight (FSB) and Fusarium head blight (FHB) diseases caused by the Fusarium species. The objective of this study was to facilitate the resistance of hulled and hull-less barley at different growth stages to F. culmorum according to direct parameters: disease rating (DR), fresh weight of leaves and roots, kernel weight per spike, kernel number per spike, plump kernels, and indirect parameters - chlorophyll a fluorescence (CF). Plate assay, greenhouse and field tests were performed on 30 spring barley doubled haploid (DH) lines and their parents infected with Fusarium culmorum. Direct parameters proved that hulled genotypes show less symptoms. Most studied chlorophyll a fluorescence (CF) parameters (apart from DIo/CS - amount of energy dissipated from PSII for laboratory test, TRo/CS - amount of excitation energy trapped in PSII reaction centers, ETo/CS - amount of energy used for electron transport and RC/CS - number of active reaction centres in the state of fully reduced PSII reaction center in field experiment) were significantly affected by F. culmorum infection. In all experiments, hulled genotypes had higher values of CF parameters compared to hull-less ones. Significant correlations were detected between direct and indirect parameters and also between various environments. It was revealed that ABS/CS, TRo/CS, and RC/CS have significant positive correlation in greenhouse test and field experiment. Significant correlations suggest the possibility of applying the CF parameters in selection of barley DH lines resistant to F. culmorum infection.

The Assessment of Photochemical Index of Nursery Seedlings of Cucumber and Tomato under Drought Stress (건조스트레스에 의한 오이와 토마토 공정육묘의 광화학적 지표 해석)

  • The purpose of this study is to analyze photochemical activity of nursery seedlings under drought stress, using chlorophyll fluorescence reaction analysis. Young nursery seedlings of tomato (Lycopersicon esculentum Mill.) and cucumber (Cucumis sativa L.), were grown under drought stress for 8 days. Analysis of chlorophyll fluorescence reaction (OJIP) and parameters, were performed to evaluate photochemical fluctuation in nursery seedlings under drought stress. Chlorophyll fluorescence reaction analysis showed maximal recorded fluorescence (P) decreased from the 5 day after treatment in tomato seedlings, while an amount of chlorophyll fluorescence increased at the J-I step. Thus, physiological activity was reduced. In cucumber seedlings, maximal recorded fluorescence (P) and maximal variable fluorescence ($F_V$) lowered from the 4 day after treatment, and chlorophyll fluorescence intensity of J-I step increased. Chlorophyll fluorescence parameter analysis showed electron transfer efficiency of PSII and PSI were significantly inhibited with decreasing $ET2_O/RC$ and $RE1_O/RC$ from the 5 day after treatment, in tomato seedlings and from the 4 day after treatment, in cucumber seedlings. $ET2_O/RC$ and $PI_{ABS}$ significantly changed. In conclusion, 6 indices such as $F_V/F_M$, $DI_O/RC$, $ET2_O/RC$, $RE1_O/RC$, $PI_{ABS}$ and $PI_{TOTAL}ABS$ were selected for determining drought stress in nursery seedlings. Drought stress factor index (DFI) was used to evaluate whether the crop was healthy or not, under drought stress. Cucumber seedlings were less resistant to drought stress than tomato seedlings, in the process of drought stress.

  • In order to determine the relationships between the lea(-burning disease and the light harvesting chlorophyll-protein (LHCP) complex in Panax ginseng C. A. Meyer, we investigated the chlorophyll-protein (CP) complex of the thylakoid membrane and its characteristics. In P. ginseng four Cp-complex bands determined by non-denaturing SDS-PAGE were identified CP I'(containing reaction center of photosystem I and LHCP I antennae), CP I (reaction center of photosystem I) LHCP II** (oligoform of LHCP II), and LHCP II (photosystem II antennae, CP 26 and CP 29) by Bassis and Dunahay's procedures. Under our experimental condition, the CP I band was only observed in P. ginseng and the band intensity of LHCP II** in P ginseng was higher than in spinach and soybean. There were differences in the absorption and fluorescence spectra and chlorophyll a/b ratio of the CP-complex bands between P. ginseng and other Plants. The Polypeptidr content of P. ginseng thylakoid was lower than in spinach and soybean thylakoid, and the Polypeptide profiles of P. ginseng was low band intensity, especially about 29-35 kD, 55 kD, and 60 kD, compared to spinach and soybean. The inhibitory effects of 2,5-dimethylfuran, specific singlet oxygen ($^1O_2$) quencher, showed that singlet oxygen destroyed 60% of chl.a, 90% of chl.b and 70% of carotenoid in bleaching P. ginseng with leaf-burning disease.

Sequestration of Orthophosphate by D(+)-Mannose Feeding Increases Nonphotochemical Quenchings in Chinese Cabbage Leaves (Mannose 처리된 배추 잎의 무기인산 감소에 따른 비광화학성 소산의 증가)

  • Limitation of photosynthesis in detached Chinese cabbage (Brassica campestris L.) leaves was induced by feeding of mannose (25 mM) for 12 h in the light, and changes in the basic thylakoid functions under this condition were investigated. The acid soluble phosphate contend and CO2 uptake rate was decreased by 66% and 67%, respectively. However, the starch content was increased by 24% compared to those of controls. From the fast induction curves of chlorophyll fluorescence, dark level fluorescence (Fo) slightly increased while intermediate plateau fluorescence level (FI) to peak level fluorescence (Fp) transient was significantly decreased with a slight decrease in the Fo-to-FI transient. This data means that reduction of secondary electron acceptor of PSII (QB) might be more severely inhibited than that of primary electron acceptor of PSII (QA) by decrease in phosphate level. The strong decline of (Fv)m//Fm ratio suggests that efficiency of excitation energy capture by PSII was decreased markedly. The quenching of Fo (qO), an indicator of state transition, was also occurred over the slow induction kinetics of chlorophyll fluorescence. From quenching analysis, fluorescence was dominantly quenched by nonphotochemical quenchings (qE+qT). These results showed that the capture and transfer efficiency of excitation energy to PSII reaction center in thylakoid was decreased with the decline of leaf phosphate level, and that the state transition was occurred during the induction of photosynthesis under these conditions.

Regulation of Chlorophyll-Protein Complex Formation and Assembly in Wheat Thylakoid Membrane

  • Lincomycin, an inhibitor of plastid protein synthesis, was found to block the synthesis of apoprotein P700 with a molecular mass of 72 kDa and the assembly of the Chl a-protein of PS I. Synthesis of the polypeptides of 48, 43.5, and 32 kDa of the PS II complex is also suppressed. This process is accompanied by the disappearance of the PS Two reaction center Chl a at 683 nm, and of the PS One reaction center Chl a at 690, 696, and 705 nm on the fourth derivative of the absorption spectra at 77K. Lincomycin does not affect the synthesis of LHC subunits. It increases the content of the two main Chl forms of LHC at 648 nm (Chl b) and 676 nm (Chl a). The low-temperature fluorescence ratio F736/F685 is also increased. However, the effect of cycloheximide (an inhibitor of cytoplasmic protein synthesis) leads to the reduction of polypeptides of the light-harvesting Chl a/b-protein complex in the range of 29.5-22 kDa. Under these conditions, the relative amount of Chl b and the F736/ F685 fluorescence ratio decrease significantly. This is obviously the result of blocking the LHC I and LHC II synthesis. At the same time rifampicin and actinomycin D (inhibitors which block transcription in chloroplast and nuclear genome, respectively) inessentially affect the characteristics of these complexes.

Photochemical Efficiency and psbA Gene Expression of Crinum Leaves under Natural Environmental Stress in Winter (겨울철 저온스트레스에 의한 문주란의 광합성효율과 psbA 유전자의 발현양상)

  • The change of chlorophyll fluorescence parameters, O-J-I-P transients and psbA gene expression were investigated in the leaves of Crinum asiaticum var. japonicum on the natural condition in winter, in order to elucidate physiological responses of photosystem II (PS II) activity to winter stresses. The photochemical efficiencies of PS II, Fv/Fm, were significantly low in winter, contrary to its high value in summer. The values of I -qN and I-qP were lower in midday than at dawn or night both in summer and winter, although their decrease in midday was less in winter than in summer. In the O-J-I-P transients, the fluorescence intensity of J, I, P-step decreased remarkably depending on temperature drop in winter. And the D I reaction center protein of PS II decreased in late winter more than in early winter, concomitantly with relatively high content of description products of psbA gene in midday. These results indicate that low temperature in winter causes irreversible damage to PS II and subsequently leads to cell death.

Effects of Salt in Soil Condition on Chlorophyll Fluorescence and Physiological Disorder in Panax ginseng C. A. Meyer (토양 염류 농도가 인삼 잎의 엽록소 형광반응 및 생리장해 발생에 미치는 영향)

  • Background : Excessively high concentration of sodium ion causednutrient deficiency and significantly decrease growth. This study was carried out to determine the limiting concentration range of sodium ion in the soil of ginseng field. Methods and Results : The growth of the ginseng cultivar Chunpoong reduced with increase in salinity, and the rate of growth reduction was higher in shoots than that of roots. Particularly, ginseng plants cultivated at high level of nitrate nitrogen or sodium may suffer delayed development and stunted growth. Chlorophyll damage occurred on the leaves of ginseng planted in relatively high levels (> $0.2cmol^+/kg$) of sodium ion, as determined by the fluorescence reaction. The incidence of physiological disorder in ginseng cultivated at 249 sites was correlated with the concentration of sodium ion in the soils. About 74% of ginseng fields in which physiological disorders occurred had concentrations of sodium ion in soil greater than $0.2cmol^+/kg$. In contrast, the concentration of sodium ions at 51 of 85 sites where no damage occurred was relatively ($0.05cmol^+/kg-0.15cmol^+/kg$). Conclusions : The concentration of sodium ion in soil of ginseng fields can be classified into three levels optimum (${\leq}0.15$), permissible allowance (0.15 - 0.2) and excessive (> 0.2).

Light-Dependent Chilling Injury on the Photosynthetic Activities of Cucumber Cotyledons (저온처리한 오이의 자엽에서 광합성 활성의 광의존성 저해)

  • The photosynthetic activities in relation to oxygen evolution rates, quantum yield, CO2 uptake rates and room temperature chlorophyll fluorescence were investigated in cotyledons of cucumber seedlings exposed to low temperature (at 4$^{\circ}C$) for 24 h. Light-chilling caused more inhibition on light-saturated maximum oxygen evolution rates, quantum yield, and CO2 uptake rates than dark-chilling did in the cucumber plant. Light-chilling induced more marked increase in Fo and decrease in (Fv)m/Fm than dark-chilling did in the room temperature chlorophyll induction kinetics. The above results affected by chilling in the light are considered to be associated with the partial damage of the reaction center of PS II and the decreased photosynthetic activities. There occurred a large decrease in qQ with little change in qNP in the light-chilling plant. When light- and dark-chilled plants were recovered at room temperature for 24 h and their chlorophyll fluorescences were induced with light doubling technique, light-chilled plants showed more smaller magnitude and rate of fluorescence relaxation than dark-chilled plants. These suggest that light-chilling might cause some alterations in transthylakoid pH formation, and that photosynthetic apparatus of cucumber cotyledons is more susceptible to light-chilling. In the fast fluorescence induction kinetics, FR was decreased by 60% in the light-chilled plants with reference to $25^{\circ}C$ light-grown plants, while the dark-chilled plants showed a decreased rate of only 20% with reference to $25^{\circ}C$ dark-treated plants for 24 h, indicating that cucumber seedling is very sensitive to chilling stress. So, it is certain that chilling injury to the photosynthetic apparatus is strongly dependent on the presence of light in cucumber seedlings.
