Background: This study analyzed the physiological adaptations of a woody plant, Vitex rotundifolia, in Goraebul coastal sand dunes from May to September 2022. Environmental factors and physiological of plants growing under field and controlled (pot) conditions were compared. Results: Photosynthesis in plants growing in the coastal sand dunes and pots was the highest in June 2022 and July 2022, respectively. Chlorophyll fluorescence indicated the presence of stress in the coastal sand dune environment. The net photosynthesis rate (PN) and Y(II) were highest in June in the coastal sand dune environment and July in the pot environment. In August and September, Y(NPQ) increased in the plants in the coastal sand dune environment, showing their photoprotective mechanism. Chlorophyll a and b contents in the pot plant leaves were higher than those in the coastal sand dune plant leaves; however, chlorophyll-a/b ratio was higher in the coastal sand dune plant leaves than in the pot plant leaves, suggesting a relatively high photosynthetic efficiency. Carotenoid content in the coastal sand dune plant leaves was higher in August and September 2022 than that in the pot plant leaves. Leaf water and soluble carbohydrate contents of the coastal sand dune plant leaves decreased in September 2022, leading to rapid leaf abscission. Diurnal variations in photosynthesis and chlorophyll fluorescence in both environments showed peak activity at 12:00 hour; however, the coastal sand dune plants had lower growth rates and Y(II) than the pot plants. Plants in the coastal sand dunes had higher leaf water and ion contents, indicating that they adapted to water stress through osmotic adjustments. However, plants growing in the coastal sand dunes exhibited reduced photosynthetic activity and accelerated decline due to seasonal temperature decreases. These findings demonstrate the adaptation mechanisms of V. rotundifolia to water stress, poor soils, and high temperature conditions in coastal sand dunes. Conclusions: The observed variations indicate the responses of the V. rotundifolia to environmental stress, and may reveal its survival strategies and adaptation mechanisms to stress. The results provide insights into the ecophysiological characteristics of V. rotundifolia and a basis for the conservation and restoration of damaged coastal sand dunes.
Infection with rice blast fungus (Magnaporthe grisea) significantly reduced foliar net photosynthesis (A) of rice cultivars: Ilpoom, Hwasung, and Choochung in greenhouse experiments. By measuring the amount of diseased leaf area with a computer image analysis system, the relation between disease severity (DS) and net photosynthetic rate was curvilinearly correlated (r=0.679). Diseased leaves with 35% blast symptom can be predicted to have a 50% reduction of photosynthesis. The disease severity was linearly correlated (r=0.478) with total chlorophyll (chlorophyll a and chlorophyll b) per unit leaf area(TC). Light use efficiency was reduced by the fungal infection according to the light response curves. However, dark respiration (Rd) did not change after the fungal infection (p=0.526). Since the percent of reduction in photosynthesis greatly exceeded the percent of leaf area covered by blast lesions, loss of photosynthetic tissue on an area basis could not by itself account for the reduced photosynthesis. Quantitative photosynthetic reduction can be partially explained by decreasing TC, but cannot be explained by decreasing Rd. By photosynthesis (A)-internal CO$_2$ concentration (C$_i$ curve analysis, it was suggested that the fungal infection reduced ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco) activity, ribulose-1,5-bisphosphate (RuBP) regeneration, and inorganic phosphate regeneration. Thus, the reduction of photosynthesis by blast infection was associated with decreased TC and biochemical capacity, which comprises all carbon metabolism after CO$_2$ enters through the stomata.
Functional stay-green is a beneficial trait that may increase grain yield through the sustained photosynthetic competence during monocarpic senescence in cereal crops. The temporal changes of photosynthesis and related characteristics throughout the grain filling period of a stay-green japonica rice "SNU-SG1" was compared in growth chamber conditions with three high-yielding cultivars(HYVs) and their $F_1$ hybrids with SNU-SG1. SNU-SG1 exhibited a typical characteristic of functional stay-green in terms of chlorophyll degradation and photosynthetic competence during grain filling. According to the photosynthesis-light response curve measured at 10 and 35 d after heading for the flag leaf, SNU-SG1 exhibited higher initial light conversion efficiency and thus higher gross photosynthetic rate at light saturation compared to HYVs. Light saturation point was not different among genotypes, ranging from 1000 to 1500 ${\mu}mol$ photon $m^{-2}s^{-1}$. Net photosynthetic rate at light saturation($P_{max}$) of the upper four leaves in SNU-SG1 was much higher and sustained longer throughout grain-filling than HYVs and $F_1$ hybrids. The sustained high photosynthetic competence of SNU-SG1 during grain filling was ascribed to the longer maintenance of high mesophyll conductance that resulted from not only high chlorophyll content and its delayed degradation but also the slow degeneration of photosystem II(PS II) as judged by chlorophyll fluorescence($F_v/F_m$) of flag leaves. $F_1$ hybrids showed slow degeneration of photosystem II similar to the male parent SNU-SG1 while chlorophyll degradation pattern close to female parents, thus exhibiting a little higher $P_{max}$ than female parents. These results suggest that SNU-SG1 has a typical functional stay-green trait that can be utilized for increasing rice yield potential through the improved dry matter production during grain filling.
Pigments and thylakoid membrane proteins were investigated in wild type and PS I- mutants from Synechocystis sp. PCC6803 Comparing morphological features, B2 was less fluorescent than the other strains. The contents of chlorophyll a were propotional to the FNR activity in thylakoid membrane. The FNR activity of mutants was lower than that of wild type. In the result of pigments analysis, mutants had smaller cholophyll a than that of wild type. The major carotenoid was found to he $\beta$-caroene, but aeaxanthin was barely detected in thylakoid membrane of mutants. The polypeptide, 14.8kD was detected by electrophoresis in mutants. It was considered to be the modification of 15.4kD in wild type. Membrane polypeptides of 17.6 and 19.7kD were not detected in mutants. In the result of western blotting, subunit I was detected in all strains, but subunit II was barely detected in mutants. Subunit II was not detected in B2 at all. In view of the results so far achieved, the changes of contents of chlorophyll and zeaxanthin were affected by the defficiency or modification of functional domain in subunit I. Also the modification in subunit I affected the subunit II- binding site in PS I. As the result, efficiency of photosynthesis was decreased. Key words: Synechoystis sp. This study was conducted to determine the extent of drought resistance based on physiological responses of Calystegia soldanella under water deficit. In order to investigate the changes of plant growth, stomatal density, photosynthesis, chlorophyll fluorescence, the contents of chlorophyll and carotenoid, osmolality, total ion contents, the contents of carbohydrate and proline, C. soldanella was grown under well watered and drought stressed conditions for 12 days. In this study, water-deficit resulted in remarkable growth inhibition of C. soldanella. The effect of water-deficit on plant growth was associated with low osmotic potential of soil. On day 12 after drought treatment, dry weight, relative water contents, number and area of leaves and stem length were lower than those of control. The stomatal conductance and net photosynthetic rate were significantly reduced in water stressed plant to regulate inner water contents and $CO_2$ exchange through the stomatal pore. Chlorophyll fluorescence and chlorophyll contents were not different in comparison with the control, indicating that the efficiency of photosystem II was not affected by drought stress. This results could be explained that water-deficit in C. soldanella limits the photosynthetic rate and reduces the plant's ability to convert energy to biomass. A significant increase in total ion contents and osmolality was observed on day 7 and day 12. Accumulation of proline in leaves is associated with the osmotic adjustment in C. soldanella to soil water-deficit. Consequently, this increase in osmolality in water stressed plant can be a result in the increase of ion contents and proline.
The response of the freshwater microalga Chlorella vulgaris to mercuric ion ($Hg^{2+}$) stress was examined using chlorophyll a fluorescence image analysis and O-J-I-P analysis as a way to monitor the toxic effects of mercury on water ecosystems. The levels of photosynthetic pigments, such as chlorophyll a and b and carotenoids, decreased with increasing $Hg^{2+}$ concentration. The maximum photochemical efficiency of photosystem II(Fv/Fm) changed remarkably with increasing $Hg^{2+}$ concentration and treatment time. In particular, above $200{\mu}M\;Hg^{2+}$, considerable mercury toxicity was seen within 2 h. The chlorophyll a fluorescence transient O-J-I-P was also remarkably affected by $Hg^{2+}$; the fluorescence emission decreased considerably in steps J, I, and P with an increase in $Hg^{2+}$ concentration when treated for 4 h. Subsequently, the JIP-test parameters (Fm, Fv/Fo, RC/CS, TRo/CS, ETo/CS, ${\Phi}_{PO}$, ${\Psi}_O$ and ${\Phi}_{EO}$) decreased with increasing $Hg^{2+}$ concentration, while N, Sm, ABS/RC, DIo/RC and DIo/CS increased. Therefore, a useful biomarker for investigating mercury stress in water ecosystems, and the parameters Fm, ${\Phi}_{PO}$, ${\Psi}_O$, and RC/CS can be used to monitor the environmental stress in water ecosystems quantitatively.
Pathogen infection in plants induces complex responses ranging from gene expression to metabolic processes in infected plants. In spite of many studies on biotic stress-related changes in host plants, little is known about the metabolic and phenotypic responses of the host plants to Pseudomonas cichorii infection based on image-based analysis. To investigate alterations in tomato plants according to disease severity, we inoculated plants with different cell densities of P. cichorii using dipping and syringe infiltration methods. High-dose inocula (${\geq}10^6cfu/ml$) induced evident necrotic lesions within one day that corresponded to bacterial growth in the infected tissues. Among the chlorophyll fluorescence parameters analyzed, changes in quantum yield of PSII (${\Phi}PSII$) and non-photochemical quenching (NPQ) preceded the appearance of visible symptoms, but maximum quantum efficiency of PSII ($F_v/F_m$) was altered well after symptom development. Visible/near infrared and chlorophyll fluorescence hyperspectral images detected changes before symptom appearance at low-density inoculation. The results of this study indicate that the P. cichorii infection severity can be detected by chlorophyll fluorescence assay and hyperspectral images prior to the onset of visible symptoms, indicating the feasibility of early detection of diseases. However, to detect disease development by hyperspectral imaging, more detailed protocols and analyses are necessary. Taken together, change in chlorophyll fluorescence is a good parameter for early detection of P. cichorii infection in tomato plants. In addition, image-based visualization of infection severity before visual damage appearance will contribute to effective management of plant diseases.
To compare the vitality among cool-season turfgrasses under summer weather conditions and to obtain information to improve the management of turfgrasses in golf courses and sports fields., the chlorophyll fluorescence of three cool-season turfgrasses commonly planted on golf courses in the Jeju area was measured. The turfgrasses were perennial ryegrass (Lolium perenne L.), Kentucky bluegrass (Poa pratensis L.), and creeping bentgrass (Agrostis palustris Huds.). In perennial ryegrass and Kentucky bluegrass, the chlorophyll index was low in early summer and high in late summer. In creeping bentgrass, it remained low throughout the study. Fo tended to be low in the early summer and high in late summer in the three turfgrasses. However, the difference in Fo between late summer and early summer was markedly higher in perennial ryegrass than in Kentucky bluegrass or creeping bentgrass. Fm tended to be low in early summer and high in late summer, without obvious differences among the three turfgrasses. Fv/Fm, a measure of photochemical efficiency, was also low in early summer and high in late summer in the three turfgrasses. However, Fv/Fm in late summer was mostly higher in Kentucky bluegrass and creeping bentgrass than in perennial ryegrass, indicating that the former are more resistant to the high temperature and humidity of late summer. Furthermore, Kentucky bluegrass had a high chlorophyll index in late summer and would be most resistant to the harsh conditions of late summer.
버즘나무와 백합나무의 오존에 대한 생리 생화학적인 반응 특성을 비교하고, 단기간 동안의 노출에 따른 피해 및 내성 반응을 알아보고자 본 연구를 수행하였다. 두 수종 1년생 묘목을 100 ppb의 오존에 2, 4, 8 시간/일 동안 노출시키며 1주일간 광합성, 기공전도도, 오존흡입량을 측정하고 1주일 후 광화학 효율 및 탄소고정 효율, 엽록소 함량 및 항산화효소인 SOD의 활성을 측정하였다. 광합성과 기공전도도는 버즘나무의 경우 2시간 처리구는 감소하지 않았지만 오존 처리 시간이 길어질수록 감소하였으며, 백합나무는 버즘나무보다 민감하게 반응하여 2시간 처리구에서도 광합성과 기공전도도의 감소가 나타났으며, 오존 처리 시간이 길어질수록 더욱 많이 감소되었다 엽록소 형광반응을 이용한 광화학을 측정에서는 두 수종 모두에서 오존 처리 시간에 빠른 광화학효율의 차이가 크게 나타나지 않았지만, 탄소고정효율에 있어서는 오존 처리 시간이 길어질수록 매우 많이 감소하였으며, 백합나무의 감소 폭이 더 켰다. 엽록소 함량은 두 수종 모두 큰 변화가 나타나지 않았지만 SOD 활성에서는 수종간에 큰 차이가 나타났는데 버즘나무 2시간 처리구에서는 증가하였으며, 처리 시간이 증가함에 따라 감소하였지만 백합나무에 비하여 감소 폭이 적었다. 따라서 오존에 대해서 버즘나무보다 백합나무가 민감하게 반응하며 내성이 낮은 것으로 판단되었다.
