Effects of Simulated Acid Rain on the Chlorophyll Contents in the Needles of Pinus koraiensis and Ligustrum obtusifolium Seedlings (인공산성우(人工酸性雨)가 잣나무 및 쥐똥나무 유묘(幼苗)의 엽내(葉內) 엽록소(葉綠素) 함량(含量)에 미치는 영향(影響))

  • With the purpose of pursuing the increase and decrease of chlorophyll a and chlorophyll b caused by application of the simulated acid rain treatment on Pinus koraiensis seedlings and Ligustrum obtusifolium cuttings, the experimental design of randomized block arrangement with three replications was implemented in the experimental field of Yesan National Agricultural Junior College, during the growing season of 1985. Pinus koraiensis seeds stratified in cool and moist condition were sown on pots, and in case of Ligustrum obtusifolium, C1/1 cuttings were potted for experimental use in the early spring. The regime of artificial acid rain, based upon precipitation frequency and density, was simulated from the learning of climatological data averaged from 30 years records. The spray of acid water containing pH values of 4.0 and 2.0 was initiated from the 1st of May and ended on the 31st of August. As control, ground water was also treated at the same time. To analyse the chlorophyll content, those leaves looking representative and unaffected by other harmful agents were sampled on the 18th of September, and UV-visible spectrophotometer was used. With decrease in pH values of acid rain, the content of chlorophyll a and chlorophyll b decreased in both species. The decrease in chlorophyll a could be confirmed through statistical significance, but not in chlorophyll b. And when we discussed the chlorophyll decrease index which was explained in detail in the paper, an attention might be given to similarly decreasing values in both chlorophyll a and chlorophyll b, according as pH levels of acid rain decreased. The ratios of chlorophyll a to chlorophyll b in both species were not affected by different pH leaves of acid rain.

Varietal Difference in Relationship between SPAD Value and Chlorophyll and Nitrogen Concentration in Rice Leaf (벼 잎의 엽록소 및 질소함량과 엽록도 관계의 품종적 차이)

  • This experiment was conducted to find out the varietal differences in the relationships of chlorophyll and nitrogen concentration with leaf greenness value (SPAD value). We used two rice varieties with different leaf color type: Hwaseongbyeo has pale green leaves, and Gwanganbyeo has dark green leaves. SPAD value was correlated with chlorophyll a concentration significantly in both varieties, but not with chlorophyll b in Hwaseongbyeo. The correlation coefficients between SPAD value and nitrogen concentration of rice flag leaves were highly significant. However, the regression analysis between SPAD value and nitrogen concentration of rice flag leaves showed different relation among varieties. The slope of the regression line in Gwanganbyeo was steeper than that in Hwaseongbyeo. In conclusion, SPAD value can be applied to estimate the chlorophyll a concentration without consideration of varietal differences. Chlorophyll b and nitrogen concentration in the leaf were regressed to the SPAD values differently with varieties. This implies that the SPAD value can not be used directly to estimate chlorophyll b and nitrogen concentration without consideration of varietal differences.

A study on the change up chlorophyll due to the fermentation of Kim chi (열무김치 숙성에 따른 chloropyll 변화에 관한 연구)

  • This investigation was attempted to determine changes in chlorophyll. Pheophytin content PH and titratable acidity during young redish Kim chi fermentation. Young redishes divied in to leaf and vein and fermented in dark room for 35 days at 10$^{\circ}C$ after pickling each group. Samples were collected at 5 days interval for the chemical analysis. The following results were obtained. 1. Chlorophyll content in young redish was decreased by decreasing PH during fermentation periods. Chlorophyll content decreased from 11.60mg/g to 7.62mg/g in leaf, from 0.48mg/g to 0.33mg/g in vein respectively during 35 days fermentation periods. In contrast to chlorophyll. pheophytin content increased as chlorophyll content decreased during the period. The ratio of chlorophyll a and b content in young redish about 3 : 1 in leaf and vein and decreased nearly the same ratio during fermentation periods. In case of pheophytin a and b it was increased without any relation with the ratio chlorophyll a and b. In generally, the optimum PH for eating was known 4.3 to 5.0. Young redish Kim chi was reached the optimun PH at 15 to 25 days of fermentation at 10$^{\circ}C$. At this periods. Chlorophyll content varied from 11.32mg/g to 9.30mg/g for leaf and from 0.43mg/g to 3.37mg/g for vein, respectively. 2. Titratable acidity(as lactic acid) was increased to 0.54% in leaf at 20 days fermentation period during Kim chi fermentation. and nearly unchanged until 30days. At the end of fermentation period the titratable acidity was about 0.30%. Titratable acidity was increased as decreasing pH. and the correlation coefficients was -0.92 in leaf and -0.77 in Vien. 3. Correlation between titratable acidity and chlorophll, pheophytin content was not significant statistically.

The Protective Effect of Chlorophyll a Against Oxidative Stress and Inflammatory Processes in LPS-stimulated Macrophages

  • This study was designed to investigate the suppressive effect of chlorophyll a on nitric oxide (NO) production and intracellular oxidative stress. In addition, chlorophyll a regulation of nuclear factor (NF) ${\kappa}B$ activation and inducible NO synthase (iNOS) expression were explored as potential mechanisms of NO suppression in a lipopolysaccharide (LPS)-stimulated macrophage cell line. RAW 264.7 murine macrophages were preincubated with various concentrations ($0-10\;{\mu}g/ mL$) of chlorophyll a and stimulated with LPS to induce oxidative stress and inflammatory response. Treatment with chlorophyll a reduced the accumulation of thiobarbituric acid-reactive substances (TBARS), enhancing glutathione level and the activities of antioxidative enzymes including superoxide dismutase, catalase, glutathione peroxidase (GSH-px), and glutathione reductase in LPS-stimulated macrophages compared to LPS-only treated cells. NO production was significantly suppressed in a dose-dependent manner (p<0.05) with an $IC_{50}$ of $12.8\;{\mu}g/mL$. Treatment with chlorophyll a suppressed the levels of iNOS protein and its mRNA expression. The specific DNA binding activities of NFkB on nuclear extracts from chlorophyll a treated cells were significantly suppressed in a dose-dependent manner with an $IC_{50}$ of $10.7\;{\mu}g/mL$. Chlorophyll a ameliorates NO production and iNOS expression through the down-regulation of NFkB activity, which may be mediated by attenuated oxidative stress in RAW 264.7 macrophages.

  • On the east slope of the front ranges of the Colorado Rockies, USA, three species of Populus are distributed at different elevations from the Great Plains to the timberline. At elevations 1, 720m (P. sargentii), 2, 335m (P.angustifolia) and 3, 190m (P.tremuloides) chlorophyll cotents of leaves of thses dioecious trees were determined. Total chlorophyll content was in the range of 0.86-1.56mg/g leaf, increasing with elevation as a second-degree polynomial, and the peak content was estimated to be reached at about 2, 800m. Total chlorophyll content was greater in female than in male trees in the two higher elevation species, but not in P. sargentii, the low elevation species. Chlorophyll b content was greater than chlorophyll a content in the same two higher elevation species. The increase of chlorophyll b than to increase of chlorophyll a content. These phenomena appear to be the result of adaptation of these species to different environmental conditional at different elevation. Sequential distribution of plant populations on environmental gradients in such a way that variation in given plant characters paralell the environmental gradient must occur not only in ecotypes of a species, but also in different but closely related species.

  • Study on effectiveness of Bradyrhizobium japonicum on soybean [Glycine max (L.) Merr.], local cultivar 'Sathiya' were carried out in the garden soil of Trib-huvan University. Different parameters like nodulation, chlorophyll content in fresh leaves and growth of plant in inoculated and uninoculated plant was studied. Pot experiment was conducted in the green house to evaluate the effectiveness of B. japonicum on soybean. It was observed that B. japonicum inoculation increased the number of nodules, shoot length of plant and total chlorophyll content in fresh leaves of soybean plant. However, root length was decreased in all inoculated plants.

HPLC Analysis of Biomass and Community Composition of Microphytobenthos in the Saemankeum Tidal flat, West Coast of Korea (한국 서해 새만금 갯벌에서 저서미세조류의 생체량과 군집조성에 대한 HPLC 분석)

  • Biomass and community composition of microphytobenthos in the Saemankeum tidal flat were studied by HPLC analysis of the photosynthetic pigments from November 2001 to November 2002. The environmental factors of sediment were also investigated to examine the relationship between microphytobenthos biomass and sedimentary environments. The detected photosynthetic pigments of microphytobenthos were chlorophyll a, b, c, fucoxanthin, 19'-hexanoyloxyfucoxanthin, violaxanthin, diadinoxanthin, alloxanthin, diatoxanthin, zeaxanthin+lutein, peridinin and beta-carotene. Pheophytin a, the degradation product of chlorophyll a, was also detected. The results of pigmen analysis suggest the presence of diatom (fucoxanthin), euglenophytes (chlorophyll b), chlorophytes (chlorophyll b + lutein), cyanobacteria (zeaxanthin), cryptophytes (alloxanthin), chrysophytes (fucoxanthin + violaxanthin), prymnesiophytes (19'-hexanoyloxyfucoxanthin) and dinoflagellates (peridinin). Chlorophyll a concentration in the top 0.5 cm of sediment was in the range of $0.24\;mg{\cdot}m\^{-2}\;-32.11\;mg{\cdot}m\^{-2}$ in the study area. The increase of chlorophyll a concentration in the spring indicates the occurrence of a microphytobenthic bloom. In the summer, there was a sharp decrease of the chlorophyll a concentration which was probably due to high grazing activity by macrobenthos. The annual mean chlorophyll a concentration in the study area was low compared to that in most of other tidal flat areas probably due to active resuspension of microphytobenthos and high grazing activity by macrobenthos. There was no clear relationship between microphytobenthos biomass and sedimentary environments because of a large variety of physical, chemical and biological factors, Pigment analysis indicated that while diatoms were dominated in the microphytobenthic community of the Geojon tidal flat, euglenophytes and/or chlorophytes coexisted with diatoms in the Mangyung River tidal flat.

  • Leaf colors of rice can be used to identify stress level due to its adaptation to environmental change. For most leaves green-related colors are sourced from chlorophyll a and b. For most leaves green-related colors are consisted of chlorophyll a and b. Chlorophyll concentration is normally measured using a spectrophotometer in laboratory. In some remote observation fields, it is impossible to collect the leaves, preserve them, and bring them to laboratory to measure their chlorophyll content. The measurement of chlorophyll content is observed through its color. Using CIE chromaticity diagram leaf color information in RGB is transformed into wavelength (in nm). Pheophytin contents were also analyzed in 95% ethanol extracts. In the process of leaf development of rice young seedling, both pigments were compared. Leaf samples from different rice seedling bed is taken, their colors and RGB values are recorded using Photoshop Image Analysis. SPAD-502 values were also measured. The chlorophyll and Pheophytin contents were fully estimated by ${\rightthreetimes}_{avg}$ on CIE chromaticity diagram.

  • This study was conducted to investigate the changes of chlorophyll contents and chlorophyll fluorescence in barley(Hordeum vulgare L.) 7 day old seedling treated with 0.2M, 0.4M, 0.6M, 0.8M, and 1.0M NaCI concentration containing Hepes buffer(pH 7.5). Barley was affected by NaCI treatment. The chlorophyll a, b and carotenoid of barley decreased with an increase in NaCI concentration. However, chlorophyll a, b and carotenoid of barley were not greatly influenced by o.8M and 1.0M NaCl. Fv, Fv/Fm and qP were gradually decreased by higher concentration of NaCI. qP, qNP, qR and qE were gradually decreased by 6hr. During barley chloroplast was development NaCI affected chlorophyll synthesis than photosynthetic activity. Whereas barley seedling leaves were more influenced photosynthetic activity than chlorophyll contents by NaCI.

Effect of Light Quality on the Growth of Panax ginseng in a Phytotron With Natural Light (자연광기상실에서 인삼생육에 대한 광질의 영향)

  • Ginseng plants in the 2nd year were grown under the shade (90-95%) of colored cellophane films at $15^{\circ}C$ in a natural light phytotron. Relative root grown (final/initial) was in decreasing order of blue, green, white, yellow and red. The growth of aerial part, especially stem length was poorest in blue light with relative root growth negatively correlated with stem length. In this relation each color showed specific domain. Stem length showed positive linear correlation with leaf area per plant in each and among light though it was not significant in red light. The content and ratio of chlorophyll a and b in leaf were in the decreasing order of White, Blue, Green, Red and Yellow. There is positive correlation between chlorophyll a and chlorophyll b, and between total chlorophyll and chl:a/b ratio. Blue color shade is expected to be beneficial for ginseng production.
