전처리가 오배자 추출물에 의한 여름용 인견직물의 염색 및 기능성 향상에 미치는 영향 (Effect of Pretreatment on Dyeability and Functionalities of Summer Rayon fabrics Finished by Gallnut Extract)

  • Viscose rayon filament fabrics have been called 'artificial silk' and beloved as summer clothing materials for a long time in Korea. This is because the viscose rayon filament fabrics reveals glossy surface and cool touch feeling compared to other conventional fabrics composed of staple fibers. Therefore, we tried to prepare the higher value added viscose rayon filament fabrics for summer textile products. In this study, we applied gallnut extract to the viscose rayon filament fabric to develop summer fabrics with natural color and multi-functions such as antibacterial and antioxidant properties. This process also pursue eco-friendly and multi-functional fabric finishing from the natural material "gallnut". In addition, various pre-treatment with cationizer, chitosan, or chito-oligomer was applied to the finishing process to improve the finishing efficacy and durability. Consequently, it was found that the active component of gallnut extract was successfully incorporated to the viscose rayon filament fabric through a pad-dry-cure process. And, the treated viscose rayon filament fabrics showed excellent antibacterial and antioxidant properties. Therefore, it was expected that the rayon filament fabrics treated by gallnut extract could be used as effective summer fabrics preventing the growth of bacteria and skin ageing as well as providing cool touch feeling. However, the pre-treatments were not that meaningful on the functionalities but effective on coloring.

Membrane Filtration에 의한 약주의 저장성 증진 (Improvement of Shelf-life of Yakju by Membrane Filtration)

  • 막여과 약주의 저장 중 품질변화를 관찰하기 위하여 여러 가지 종류의 여과막으로 여과한 약주를 $25^{\circ}C$에서 50일간 저장하면서 약주의 pH, 적정산도, 탁도 및 총균수, 젖산균수, 효모수를 측정하였다. 비살균 약주는 저장 중 적정산도, 탁도, 생균수가 증가한 반면 pH는 감소하였다. 막여과 약주의 경우 적정산도와 탁도의 변화가 관찰되지 않았으며 약주내 미생물들이 관찰되지 않아 높은 저장성을 보여주었다. 키토산을 0.1% (w/v) 첨가하였을 경우에는 생균수의 급격한 감소현상을 나타내어 높은 살균력을 지니고 있음을 알 수 있었다.

면역기능 증강 신물질에 대한 마우스의 면역학적 및 혈액학적 변화 (Changes of immunostimulatory effects by Immu-Forte on mice)

  • Immu-Forte composed of chitosan, ${\beta}-glucan$, manno-oligosaccharide and pangamic acid was evaluated for its effectiveness as a nonspecific immunostimulator in mice. The effects of Immu-Forte were determined by analysis of cytokines using ELISA and phenotype of leukocyte subpopulations using monoclonal antibodies specific to mouse leukocyte differentiation antigens and flow cytometry. All T cells, all B cells, CD4 T cells, CD8 T cells, macrophages, IL-2, IL-4, IL-12 and IFN-r in Immu-Forte A-treated group increased in 1 months posttreatment and were significantly higher (p < 0.05) than that of control at 1 months posttreatment. All T cells, all B cells, CD4 T cells, CD8 T cells, macrophages and IL-2 in Immu-Forte EX-treated low and middle dose groups increased in 1 months posttreatment and were significantly higher (p < 0.05) than that of control at 1 months posttreatment. In the Immu-Forte soybean-treated group, NK cells and IL-4 were significantly higher in middle dose-treated group, and IL-2, IL-4 and IFN-r were significantly higher in low dose-treated group. In the Immu-Forte F-treated group, all T cells, all B cells, CD4 T cells, CD8 T cells, macrophages, NK cells, IL-2, IL-4, IL-12 and IFN-r in high dose-treated group and all T cells, all B cells, CD4 T cells, CD8 T cells, macrophages, IL-2, IL-4, IL-12 and IFN-r in middle dose-treated group and NK cells, IL-2, IL-4, IL-12 and IFN-r in low dose-treated group were significantly higher (p < 0.05) than that of control at 1 months posttreatment. In conclusion, this study has demonstrated that Immu-Forte had an immunostimulatory effect on mice through proliferation and activation of mouse immune cells.

부추첨가 김치의 발효과정 중 chitinase 활성과 조직감 (Chitinase Activity and Textural Property of Leek Added Kimchi During Fermentation)

  • 김치재료들의 chitinase 활성을 측정하고, 부추의 첨가량을 달리한 김치를 제조하여 15$^{\circ}C$에서 9일간의 숙성과정 중 김치의 chitinase 활성과 조직감의 변화를 측정한 결과는 다음과 같았다. 김치의 주요 재료들은 쪽파(특히 잎부분)>마늘>부추>대파(특히 잎부분) 순으로 chitinase 활성을 나타내었다. 김치숙성 중의 chitinase 활성은 김치액이 김치조직 보다 월등히 높게 나타났으며 각 김치시료간의 효소활성 차이도 김치액에서 더 뚜렷하게 나타났다. 김치액에서의 chitinase 활성은 숙성이 진행됨에 따라 점차적으로 감소한 반면, 김치조직에서의 chitinase 활성은 숙성 3일 또는 5일까지 증가한 후 감소하였다. 그러나 김치조직과 김치액 모두에서 대조구보다 부추첨가구가 더 높은 chitinase 활성을 나타내었으며 부추첨가구에서는 부추첨가량이 증가할수록 높은 chitinase 활성을 보였다. 숙성이 진행됨에 따라 모든 시료구에서는 김치조직의 침투관통력은 감소하였으며 부추첨가량에 따른 차이는 미미하였으나 대조구가 숙성 전 기간동안 가장 낮은 침투관통력을 보였다. 이상의 결과를 종합해보면 부추첨가는 김치 숙성 중 부추의 chitinase 활성에 의해 새우젓 등의 키틴질을 분해하여 chitosan을 형성하므로서 김치의 조직을 향상시키는 것으로 생각된다.

SPCE에 HRP 효소가 고정화된 바이오센서의 전기화학적 특성에 관한 연구 (A Study on the Electrochemical Characteristics of Biosensor with HRP Enzyme Immobilized on SPCE)

  • 과산화수소를 이용한 펜톤(Fenton)산화법은 수처리 및 토양 복원분야에서 활용되는 친환경 산화방법이다. 이 방법으로 오염물질을 제거할 때, 오염물의 농도에 따라 과산화수소의 농도를 적절하게 조절하는 것이 상당히 중요하다. 이에 본 연구에서는 HRP (horseradish peroxidase) 효소를 이용한 전기화학적 바이오센서를 제조하고 효소의 활성과 과산화수소의 검출 특성에 대한 연구를 수행하였다. SPCE (Screen Printed Carbon Electrode)의 작업 전극 표면에 키토산과 AuNP를 이용하여 HRP를 전착하였다. 이 후, 전위주사법(CV)과 전기화학적 임피던스 분광법(EIS)을 이용하여 효소의 고정화를 확인하였다. 또한 시간대전류법(CA)과 UV 분광법으로부터 HRP 효소의 활성을 확인하였다. 본 연구에서 제조한 바이오센서를 PBS 전해질에 담그고 과산화수소를 적정하여 CA 분석으로부터 전극에서 발생하는 전류를 측정하였다. 발생 전류는 과산화수소의 농도에 대하여 선형적으로 증가하였으며, 전류로부터 과산화수소의 농도를 예측할 수 있는 검정곡선을 도출하였다.

저장고 형태에 따른 온주밀감의 저장 중 품질변화 (Quality Changes of Satsuma Mandarin during Storage by Storage Warehouse)

  • 저장고 형태에 따른 온주밀감의 저장 중 품질특성을 검토하였다. 항균제로서 베프란(iminoctadime-triacetate) 2,000배 희석액과 키토산 1.5%에 0.5% CaCl$_2$를 혼합한 용액에 감귤을 충분히 침지 하였다. 풍건시킨 후 26L인 플라스틱 컨테이너에 감귤을 12kg 정도씩 담아 3$0^{\circ}C$에서 24시간동안 저장전처리를 하고, 상온저장과 내부온도 4$^{\circ}C$ 상대습도 87%를 기준으로 저장하였다. 저장고내의 온습도 편차가 적은 저장고에서 가장 낮은 부패율이 나타내어, 감귤저장 중 온도관리가 중요함을 알 수 있었다. 감귤 저장 중 Penicillium italicum, Monilia candida Alternaria citri, Mucor hiemalis, Phomopsis citri, Botrytis cinerea, Phoma citricarpa, Glomererella cingulata, Penicillium digitatum 등이 부패미생물로서 동정되었다. 중량 감소, 껍질과 과육의 수분 함량은 저장 중 서서히 감소하였다. 산 함량의 감소는 저장 4개월 동안 상온저장에서 24%에 비하여, 저온저장에서는 15~18%로 저장효과가 컸다. 총당 함량은 저장 3개월 이후부터 감소 폭이 많았으며, 비타민 C 는 저장 60일 후부터 감소가 많아 장기간 저장으로 감귤 품질이 떨어짐을 알 수 있었다.

Tumoral Accumulation of Long-Circulating, Self-Assembled Nanoparticles and Its Visualization by Gamma Scintigraphy

  • The enhanced permeability and retention (EPR) effect is used extensively for the passive targeting of many macromolecular drugs for tumors. Indeed, the EPR concept has been a gold standard in polymeric anticancer drug delivery systems. This study investigated the tumoral distribution of self-assembled nanoparticles based on the EPR effect using fluorescein and radio-labeled nanoparticles. Self-assembled nanoparticles were prepared from amphiphilic chitosan derivatives, and their tissue distribution was examined in tumor-bearing mice. The size of the nanoparticles was controlled to be 330 run, which is a size suited for opening between the defective endothelial cells in tumors. The long-circulating polymer nanoparticles were allowed to gradually accumulate in the tumors for 11 days. The amount of nanoparticles accumulated in the tumors was remarkably augmented from 3.4%ID/g tissue at 1 day to 25.9%ID/g tissue at 11 days after i.v. administration. The self-assembled nanoparticles were sustained at a high level throughout the 14 day experimental period, indicating their long systemic retention in the blood circulation. The ${\gamma}$-images provided clear evidence of selective tumor localization of the $^{131}I$-labeled nanoparticles. Confocal microscopy revealed the fluorescein-labeled nanoparticles to be preferentially localized in the perivascular regions, suggesting their extravasation to the tumors through the hyperpermeable angiogenic tumor vasculature. This highly selective tumoral accumulation of nanoparticles was attributed to the leakiness of the blood vessels in the tumors and their long residence time in the blood circulation.

Effect of Skim Milk-Alginate Beads on Survival Rate of Bifidobacteria

  • In this study, an attempt was made to increase the survival rate of bifidobacteria entrapped in alginate in the gastrointestinal tract, and to investigate the potential industrial applications, for example lyophilized capsules and yogurt. First, the protective effect of various food additives on bifidobacterial survivability was determined after exposure to simulated gastric juices and bile salts. The additives used in this study were skim milk (SM), polydextrose (PD), soy fiber (SF), yeast extract (YE), chitosan (CS), $\kappa$-carageenan ($\kappa$-C) and whey, which were added at 0.6% concentration (w/v) to 3% alginate-bifidobacterial solution. In the simulated gastric juices and bile salts, the protective effect of 0.6% skim milk-3% alginate (SM-A) beads on the survival rate of bifidobacteria proved to be higher than the other additives. Second, the hydrogen ion permeation was detected through SM-A vessel without bifidobacterial cells at different SM concentrations (0.2%, 0.4%, 0.6%, 0.8%, and 1.0%). There were no differences in terms of the pH decrease in SM-A vessels at 0.6%, 0.8%, and 1.0% (w/v) SM concentrations. The survival rate of bifidobacteria in SM-A beads would appear to be related to the SM buffering capacity against hydrogen ions and its tendency to reduce the pore size of bead. In this experiment, the survival rate of bifidobacteria entrapped in beads containing 0.6% SM showed the highest viability after exposure to simulated gastric juices for 3h, thereby indicating that 0.6% SM is the optimum concentration fir 3% alginate bead preparation. Third, the effect of SM-A beads on the freeze-drying and yogurt storage for 10 days was investigated. SM-A beads were found to be more efficient for freeze drying and yogurt storage than untrapped cells and the alginate bead. Consequently, the survival rate of bifidobacteria entrapped in SM-A beads was increased in simulated gastric juices, bile salts and probiotic products, such as lyophilized capsules and yogurt, SM-A beads can be expected to produce high value probiotic products.

에탄올 내에서 $\alpha$-Chymotrypsin에 의한 Acetyltyrosine의 에스테르화 반응 (The Esterification of Acetyltyrosine by $\alpha$-Chymotrypsin in EtOH/Water Mixture)

  • Ac-Tyr-OH의 에스테르화 반응은 water/water­m miscible organic solvent를 이용하여 단일상계 (one phase system) 조건 하에서 ${\alpha}$-chymotrypsin의 촉 매작용으로 진행하였으며, 그 결과는 다음과 같다. Ac- Tyr-OH(1OmM)이 함유된 에탄올(lOml)[ 4 % %(v/v) buffer 함유] 용액에서 ${\alpha}$-chymotrypsin의 촉매작용에 의한 에스테르화 반응에 영향을 마치는 흡착물질 chitin, chitosan, Sephadex G-25 및 G-50, CM 및 DEAE-Sephadex, CM 및 DEAE cellulose 중 chitin을 첨가하였을 경우, 에스테르화 수율이 62%로 가장 높았다. 그리고 에스테르화 수 율에 마치는 최적반응조건은 chitin/${\alpha}$-chymotry psm의 비가 20(w/w), 온도 $35^{\circ}C$, pH 8.0 빛 유기 용매에 첨가된 수용액의 농도는 2%(v/v)였다. 또한, water/water-miscible organic solvent system 에서 chitin의 첨가는 효소의 안정성에 기여하는 것으로 나타났다. 최적반응조건 하에서 에스테르화 반응의 수율, Km 및 Vmax 값은 각각 93%, 3.093mM 및 1.088mM/mg/hr이었다.

Single-Protein Molecular Interactions on Polymer-Modified Glass Substrates for Nanoarray Chip Application Using Dual-Color TIRFM

  • The immobilization of proteins and their molecular interactions on various polymer-modified glass substrates [i.e. 3-aminopropyltriethoxysilane (APTS), 3-glycidoxypropyltrimethoxysilane (GPTS), poly (ethylene glycol) diacrylate (PEG-DA), chitosan (CHI), glutaraldehyde (GA), 3-(trichlorosilyl)propyl methacrylate (TPM), 3'-mercaptopropyltrimethoxysilane (MPTMS), glycidyl methacrylate (GMA) and poly-l-lysine (PL).] for potential applications in a nanoarray protein chip at the single-molecule level was evaluated using prismtype dual-color total internal reflection fluorescence microscopy (dual-color TIRFM). A dual-color TIRF microscope, which contained two individual laser beams and a single high-sensitivity camera, was used for the rapid and simultaneous dual-color detection of the interactions and colocalization of different proteins labeled with different fluorescent dyes such as Alexa Fluor® 488, Qdot® 525 and Alexa Fluor® 633. Most of the polymer-modified glass substrates showed good stability and a relative high signal-to-noise (S/N) ratio over a 40-day period after making the substrates. The GPTS/CHI/GA-modified glass substrate showed a 13.5-56.3% higher relative S/N ratio than the other substrates. 1% Top-Block in 10 mM phosphate buffered saline (pH 7.4) showed a 99.2% increase in the blocking effect of non-specific adsorption. These results show that dual-color TIRFM is a powerful methodology for detecting proteins at the single-molecule level with potential applications in nanoarray chips or nano-biosensors.
