Expression of Chimeric Chicken-Yeast-Chicken H2B Histone Gene

  • A chicken H2B histone gene was cloned and expressed in Rat 3 cell line. Its messenger RNA level was about 10 times higher during S phase than during $G_1$ phase. A chimeric chicken-yeast-chicken H2B histone gene was made to change some of wobble sequences of chicken H2B gene. When the chimeric H2B gene was transfected into the Rat 3 cell line, it showed a pattern of expression similar to that of the original chicken H2B gene. At least in this gene, it was concluded that the wobble sequences were not required for the cell-cycle regulated pattern of expression.

Construction of a Transgenic Silkworm Carrying the Fibroin Gene of the Japanese Oak Silkworm, Antheraea yamamai

  • We describe the generation of transgenic silkworm that carrying the chimeric fibroin light chain (L-chain) gene. Previously, we have cloned the complete fibroin L-chain gene from the silkworm Baekok-Jam, Bombyx mori, and the complete fibroin gene from the oak silkworm, Antheraea yamamai. The 444 bp repetitive sequence of A. yamamai fibroin gene was inserted into the exon 6 of B. mori fibroin L-chain gene to produce chimeric fibroin L-chain gene. The chimeric fibroin L-chain gene was cloned into the polyhedrin gene site of Autographa californica nuclear polyhedrosis virus (AcNPV) to yield a recombinant baculovirus as a fibroin gene targeting vector, One-day-old fifth instar female silkworm larvae were injected with the recombinant baculovirus and then mated with normal male moths. Genomic DNA from their progenies was extracted and screened for the desired targeting event by using PCR and Southern blot analysis. The analysis showed that the chimeric fibroin gene had intergrated into the L-chain gene on the genome by homologous recombination and was transmitted through generations. The transgenic silkworm carrying the chimeric fibroin gene were approximately 43.2% in $F_2$ generation, and the silkworms synthesized the fusion protein in cocoons layer.

  • Fusion proteins resulting from chimeric sequences are excellent targets for therapeutic drug development. We developed a database of chimeric sequences by examining the genomic alignment of mRNA and EST sequences in the GenBank. We identified 688 chimeric mRNA and 20,998 chimeric EST sequences. Including EST sequences greatly expands the scope of chimeric sequences even though it inevitably accompanies many artifacts. Chimeric sequences are clustered according to the ECgene ID so that the user can easily find chimeric sequences related to a specific gene. Alignments of chimeric sequences are displayed as custom tracks in the UCSC genome browser. ChimerDB, available at http://genome.ewha.ac.kr/ECgene/ChimerDB/, should be a valuable resource for finding drug targets to treat cancers.

TT2 Embryonic Stem Cell 을 이용한 Chimeric Mouse 생산에 있어서 간단한 공배양방법 (Simple Methods for Production of Chimeric Mouse by Coculture with TT2 Embryonic Stem Cells)

  • 본 연구는 TT2 embryonic stem(ES) cell을 이용하여 chimeric mouse를 생산하는데 있어서 더욱 간편한 공배양방법 개발하기 위하여 수행되었다. 유전자 적중 생쥐의 개발은 유전자의 기능을 연구하는데 매우 중요한 수단으로 이용되고 있다. 이러한 생쥐의 개발에 있어서 chimeric mouse를 생산하는 과정은 ES cell의 종류의 차이는 있지만 주로 배반포기의 수정란에 ES cell을 주입하고 있다. 이 기술은 고가의 미세조작장치 뿐만 아니라 고도의 기술을 요하고 있다. 그러므로 본 연구에서는 TT2 ES cell를 8세포기 수정란과 공배양할 때의 필요로 하는 적절한 ES cell의 수를 검증함으로써 chimeric mouse의 생산 효율을 높일 수 있었다. 각각 0.5$\times$$10^{6}$, 1$\times$$10^{6}$과 2$\times$$10^{6}$$m\ell$의 ES cell을 8 세포기의 수정란과 공배양하였을때 0.5$\times$$10^{6}$과 1$\times$$10^{6}$$m\ell$에서 높은 배반포기로의 발달율을 나타내었다. 또한 가임신된 생쥐에 이들 배반포기를 이식한 결과 1$\times$$10^{6}$$m\ell$에서 높은 chimeric mouse 생산 효율을 나타내었다. 이러한 결과는 적절한 수의 ES cell과 수정란을 공배양함으로써 매우 간단하게 효율 좋은 chimeric mouse을 얻을 수 있음을 제시하고 있다.

고추의 sesquiterpene cyclase promoter-cinnamic acid 4-hydroxylase chimeric gene의 담배에서 발현 (Expression of Cinnamic Acid 4-Hydroxylase Chimeric Gene fused with Sesquiterpene Cyclase Promoter from Hot Pepper in Tobacco)

  • 고추에서 클로닝된 sesquiterpene cyclase (CASC) promoter에 cinnamic acid 4-hydroxylase (C4H)유전자를 클로닝하여 담배에 형질전환하였다. 형질전환체의 분석은 PCR, Southern blot분석으로 하였으며, CASC promoter의 작동여부를 조사하기 위하여 GUS histochemical assay를 하였다. 스트레스에 반응한다고 알려진 CASC promoter를 C4H gene과 fusion하여 C4H activity를 조사하여 스트레스에 반응하는 정도를 조사하였다. 최종적인 형질전환체로 확인된 개체는 평균 16.4% 정도였으며, 각 promoter별로 약간의 편차를 보였다. C4H activity 조사시 스트레스를 주지 않았을 때 대조구가 비슷한 양상을 나타내었다. 스트레스를 주었을 때 대조구와 비교하여 promoter 3번은 1.3배 activity가 높아졌으며 4번은 1.4배, 6은 2배, cyc 600은 1.1배 높아졌다. 이런 결과로 보아 이 promoter들은 UV에 반응하는 promoter이며 앞으로 stress에 저항하는 유전자의 적당한 promoter로, 타작물에 적용하여 stress저항성 작물의 육성 시 매우 유용하게 사용될 것으로 사료된다.

Chimeric RNAs as potential biomarkers for tumor diagnosis

  • Cancers claim millions of lives each year. Early detection that can enable a higher chance of cure is of paramount importance to cancer patients. However, diagnostic tools for many forms of tumors have been lacking. Over the last few years, studies of chimeric RNAs as biomarkers have emerged. Numerous reports using bioinformatics and screening methodologies have described more than 30,000 expressed sequence tags (EST) or cDNA sequences as putative chimeric RNAs. While cancer cells have been well known to contain fusion genes derived from chromosomal translocations, rearrangements or deletions, recent studies suggest that trans-splicing in cells may be another source of chimeric RNA production. Unlike cis-splicing, trans-splicing takes place between two pre-mRNA molecules, which are in most cases derived from two different genes, generating a chimeric non-co-linear RNA. It is possible that trans-splicing occurs in normal cells at high frequencies but the resulting chimeric RNAs exist only at low levels. However the levels of certain RNA chimeras may be elevated in cancers, leading to the formation of fusion genes. In light of the fact that chimeric RNAs have been shown to be overrepresented in various tumors, studies of the mechanisms that produce chimeric RNAs and identification of signature RNA chimeras as biomarkers present an opportunity for the development of diagnoses for early tumor detection.

형질전환 연초(Nicotiana tabacum L.)의 Mouse Adenosine Deaminase 유전자 발현 (Expression of Mouse Adenosine Deaminase Gene in Transgenic Tobacco (Nicotiana tabacum L.))

  • 동물유전자인 mouse adenosine deaminase (ADA)유전자가 안정적으로 연초 형질전환체에서 발현되었다. ADA cDNA 을 연초에서 강력히 발현시키기 위해서 35S/35S/AMV promoter을 부착시켰으며 식물세포에 도입을 위해서 binary vector인 pRD400을 이용하였다. 연초의 형질전환은 Tri- parental mating에 의해서 도입된 ADA 유전자 함유 binary vector와 disarmed Ti-plasmid을 함유하고 있는 Agrobacterium tmefacience MP9O을 사용하였다. 동시배양은 연초의 잎 disc을 이용해서 kanamycin이 첨가된 배지로부터 직접 shoots을 선발하여 형질전환체로 유도하였다. 형질전환 체에 ADA 유전자의 삽입여부는 PCR을 이용하였으며, 실험결과 ADA 유전자를 확인할 수 있었다. 또한 도입된 mouse ADA 유전자로부터 mRNA 및 단백질 합성여부를 각각 northern blot 및 immunoblot 분석한 결과 형질전환체 에서는 공히 확인되었다.

Protection against spring viremia carp virus (SVCV) by immunization with chimeric snakehead rhabdovirus expressing SVCV G protein

  • Spring viremia of carp virus (SVCV) poses a significant threat to numerous cyprinid fish species, particularly the common carp (Cyprinus carpio), often resulting in substantial mortalities. This study explores the potential use of a chimeric recombinant snakehead rhabdovirus carrying the SVCV G gene (rSHRV-Gsvcv) as a live vaccine against SVCV infection. Through virulence testing in zebrafish at different temperatures (15 ℃ and 20 ℃), no mortality was observed in groups infected with either rSHRV-wild or chimeric rSHRV-Gsvcv at both temperatures, whereas 100% mortality occurred in fish infected with wild-type SVCV. Subsequently, as no mortality was observed by rSHRV-Gsvcv, three independent experiments were conducted to determine the possible usage of chimeric rSHRV-Gsvcv as a vaccine candidate against SVCV infection. Fish were immunized with either rSHRV-Gsvcv or rSHRV-wild, and their survival rates against the SVCV challenge were compared with a control group injected with buffer alone at four weeks post-immunization. The results showed that chimeric rSHRV-Gsvcv induced significantly higher fish survival rates compared to rSHRV-wild and the control groups. These findings suggest that genetically engineered chimeric rSHRV-Gsvcv holds the potential for a prophylactic measure to protect fish against SVCV infection.

Expression of Chitinase Gene in Solanum tuberosum L.

  • In order to protect fungal diseases, leaf disc explants of Solanum tuberosum cultivar, Belchip, was infected with an Agrobacterium MP90 strain containing chimeric gene construct, consisting of antibiotic resistance and chitinase gene driven by the CaMV 35S promoter, for transformation. Regenerated multiple shoots were selected on a medium containing kanamycin and carbenicillin after exposure to Agrobacterium. The presence and integration of the npt II and chitinase gene were confirmed by polymerase chain reaction(PCR). Northern blot analysis indicated that the genes coding for the enzyme could be expressed in potato plants. The chitinase activity of transgenic potato plants was higher than the control potato.
