Effect of Holding Time of Broiler at Slaughter House on Color, PSE, Appearance of Chicken Meat (육계의 계류 시간에 따른 닭고기의 육색, PSE 발생 및 외관 특성)

  • This studies were conducted to investigate the effect of holding time of broiler at slaughter house on chicken meat quality. Short holding time ($1{\sim}2hrs $) increased chicken meat quality compared to long holding time ($14{\sim}15 hrs$), which resulted in 14% higher grade $1^+$ chicken. In portioned meat, short holding time ($1{\sim}2 hrs$) resulted in 15% higher incidence of grade $1^+$ chicken breast. Also, long holding time occurred higher PSE incidence of chicken breast. pH value of the chicken meat with short holding time was similar to that with long holding time, and that with medium holding time showed the lowest. $L^*$ (lightness) value of the chicken meat was lower with medium holding time than with short holding time, but that in muscle and skin showed increasing tendency with long holding time. Holding time showed no effect on $a^*$ (redness) of the chicken meat, but longer holding time decreased $b^*$ (yellowness) value. This indicated that long holding time adversely affected chicken meat quality.

  • 근세포의 분화에 있어서의 근특이 단백질의 합성 순서를 구명하기 위하여 계배 근세포를 2$\\sim$9일간 배양하면서 단백질합성야상을 SDS-polyacrylamide 겔전기 영동법, 등전점초점2차원 전기영동법 및 방사자기법으로 분석하였다. Actin은 분화의 초기부터 활발히 합성되어 그 양이 다량으로 축적되나, myosin은 배양 3일째부터 대량 합성되기 시작하였다. Myosin의 대량합성시기는 배양 근원세포가 융합을 활발히 일으키는 시기와 거의 같았다. Myoglobin은 분화초기부터 서서히 합성축적되기 시작하여 배양 5일에서 최대치에 달하였다. Creatine phosphokinase는 배양 3일만에, 그리고 glyceraldehyde dehydrogenase는 6일만에 전기영동상에 검출되었다. Tropomyosin $\\alpha$와 $\\beta$, 그리고 troponin C는 분화초기부터 비교적 다량 합성되고 있었다. 젖산탈수소효소의 활성은 배양 2$\\sim$5일 사이에서 급격히 증가하고 이후 거의 변화가 없었다. 이 효소의 동위효소 조성은 초기 근원세포에서는 $H_4$와 $H_3M$형이 많으나 분화가 진행됨에 따라 $HM_3 와 M_4$형이 서서히 출현하였다. 그리고 배양 5일만에 5종의 동위효소가 모두 검출되었다.

  • Drastic alterations in myogenesis could be induced by ultraviolet irradiation of the myogenic cells derived from 12 day old chick embryo skeletal muscle. The effects of irradiation on various aspects, including cell division, transformation to myotubes, and morphology of myoblasts and myotubes, were examined. Irradiated cells were smaller in size, and only few cells transformed resulting in smaller size of myotubes with a narrow width. Both the inhibiting actions to cell division and to fusion were more striking when irradiated at earlier stages after plating. As well, cell division and fusion were inhibited more effectively with increasing UV dose and excessive amount caused cell death. A lowering cell density was thought to account for the decrease in myogenesis and possible reasons for the decrease in the capacity for fusion were discussed in view of the results presented in this report and of the findings from other laboratories.

Physicochemical and histopathological parameters of broilers with dorsal cranial myopathy

  • Objective: This study aimed to investigate the effect of dorsal cranial myopathy (DCM) on chicken meat quality. Methods: Sixty-six Ross 308 AP broilers, 47 days old, of both sexes, weighing about 3.51 kg, were slaughtered according to standard industrial practices, and evaluated for meat color, pH, chemical composition, collagen content, fatty acid profile, and histopathological parameters. Comparisons between normal and DCM-affected meat were performed using Student's t-test at the 5% significance level. Results: Histological analysis of muscle tissues affected by DCM showed myofiber degeneration, proliferation of inflammatory cells, fibroplasia, and necrosis with fibrosis. DCM samples had lower protein content and higher moisture, ash, insoluble collagen, total collagen, and pH. DCM-affected meat was redder and more yellowish. There were no differences in lipid or soluble collagen contents between groups. DCM-affected meat had higher percentages of arachidonic acid (C20:4n-6) and eicosapentaenoic acid (C20:5n-3). Conclusion: This study revealed that DCM-affected meat exhibits considerable changes in quality parameters.

  • This survey was carried out to determine five sulfonamide(sulfamerazine, sulfamethazine, sulfadimethoxine, sulfamonomethoxine, sulfaquinoxaline) residues in beef, pork, chicken and swine kidney. For this survey, 30 samples of beef, 15 samples of chicken, 10 samples of pork and 10 samples of swine kidney were collected in Chonnam from June, 1992 to June, 1993, and were analyzed by HPLC. The recoveries of sulfamerazine, sulfamethazine, sulfamonomethoxine, sulfadimethoxine, and sulfaquinoxaline in spiked samples between $0.25{\sim}1.00$ ppm were 71.7%, 80.3%, 71.6%, 70.9%, 68.4%, respectively. None of 65 samples which were examined exceeded 0.1 ppm. Of 15 chicken muscle samples, 2 samples exceeded 0.05 ppm in sulfamerazine (0.077 ppm) and sulfamethazine (0.075 ppm), respectively. Of 10 swine kidney samples, 1 sample exceeded 0.05 ppm in sulfadimethoxine (0.052 ppm). And sulfanilamide concentration of swine kidney were higher than pork.

Effects of Carcass Processing Method and Curing Condition on Quality Characteristics of Ground Chicken Breasts (분쇄 계육 가슴살의 도체가공방법과 염지조건이 품질특성에 미치는 영향)

  • This study was carried out to investigate the effect of hot boning and curing condition on the quality characteristics of ground chicken breast. Treatments were cured by four conditions follows; control (general curing method), T1 (after hot-boning and then immediately cured), T2 (after hot-boning and immediately cured, then frozen), and T3 (after hot-boning, immediately frozen, refrigerated and then cured). The pH of chicken breast in the state of pre-rigor was 6.22. The pH of cold storage or freezing chicken breast meat respectively were 5.70 or 5.61. The pH of T1 and T2 treatments were significantly higher than those of control and T3 treatment (p<0.05). After stored for 1 wk, the pH value of T1 treatment had a higher value than those of other treatments. T1 treatment had the highest water holding capacity and the lowest cooking loss among all treatments, regardless of the cooking methods. The reduction in diameter for T1 and T2 treatments was lower than those of control and T3 treatment (p<0.05). T1 treatment had the lowest fat loss and moisture loss among all treatments, and the emulsifying capacity of T1 treatment was the highest. The protein solubility of T1 treatment was significantly lower than that of T3 treatment.

  • This experiment was carried out to investigate the minerals content and post-mortem biochemical change of broiler leg and breast muscle during hot and cold storage period. The leg and breast muscle samples were stored at $2^{\circ}C$and $30^{\circ}C$ for 32 hours, and analyzed 0, 4, 8, 32 hours after slaughter, the results obtained as follows (1) Minerals composition were different between breast and leg muscle. (2) Total nitrogen content changed according to the storage period and temperature in the breast (P<0.005) and the leg (P<0.005) (3) The Content of phosphate (P<0.005), magnesium (P<0.005), sodium (P<0.025) in breast muscle was higher than in the leg mucle, and did not change according to the storage period and temperature. (4) Calcium and iron contents were not different between breast and leg muscle but significantly changed according to the storage period and temperature. (5) Sodium content was not different among the various factors, and pH was different between leg and breast (P<0.005) muscle, and ultimate pH was appeared at 8 hours of slaughter, but 20 hours of slaughter in hot stored breast muscle. (6) pH decline in cold stored breast muscle was faster than in the hot stored. (7) Penetration value was higher in the breast muscle than in the leg muscle (P<0.005) (8) Regormortis were not occurred in cold stored breast muscle and hot stored leg muscle, but occurred in cold stored leg muscle and hot stored breast muscle at 4 hours after slaughter.

Effect of Maternal Passive Autoimmunization against Myostatin on Growth Performance in Chickens

  • Myostatin is a negative regulator of skeletal muscle growth and a loss of functional myostatin protein increases muscle hypertrophy and hyperplasia in cattle. The present study was conducted to investigate whether maternal passive immunization against myostatin would improve growth performance in chickens. A complete broiler myostatin cDNA was cloned and it was expressed into two transcripts as 1,128 bp and 985 bp by alternative splicing. A conjugated mature myostatin (350 bp) was used to induce autoimmunization and maternal passively immunized chickens was used for the experiment. It was confirmed that there was a maternal passive immunization against myostatin at zero weeks of age, but its effect was reduced by 6 weeks of age. The auto-immunized groups showed smaller body weights than those of control group during the growing period and the difference was getting bigger with time until 6 weeks of age. These results suggest that passive autoimmunization against myostatin used in this study is not potent enough to stimulate growth performance in chickens.

Effects of Dietary Fish Oil, Vitamin E and C Supplementation on DHA Deposition and Shelf-Life in Broiler Chickens (어유, 비타민 E 및 C의 급여가 닭고기의 DHA 축적 및 저장성에 미치는 영향)

  • This study was compared the effect of shelf-life and DHA accumulation in chicken meat from broilers fed experimental diets for two weeks($21{\sim}35$ days) of growers. Two hundred-ten male Ross broilers, 1 day of age, were randomly allocated to seven treatment groups. Experimental diets were assigned to each of the seven groups: control diet containing tallow, T1 with 1.00% fish oil, T2 with 2.00% fish oil, T3 with 2.00% fish oil, 200 ppm vitamin E and 200 ppm vitamin C, T4 with 2.00% fish oil and 200 ppm vitamin C, T5 with 2.00% fish oil and 200 ppm vitamin E, and T6 with 3.00% fish oil. The levels of DHA in chicken meat was the highest in T6, and T1 in breast muscle and thigh muscle with skin was higher than that of $T2{\sim}T5,\;T2{\sim}T5$ in raw chicken meat and wing with skin was about two-fold higher than that of T1. The contents of DHA in chicken meat according to storage days were significantly reduced to 42.30%, 49.38% and 48.51% in T1, T2 and T6, respectively, and this decrease was higher than that of T3, T4 and T5 (p<0.05). Particularly, the rate of reduction of DHA was the lowest in the T3 and T5, which were the lowest in TBARS(thiobarbituric acid reactive substances). TBARS increased in the order of T6, T2 and T1, but reduced in the order of T3, T5 and T4 according to storage days, and there was a significant difference among the treatment groups (p<0.05).

Effect of Dietary Root Powder of Angelicae gigantis on Growth Performance, Organ Weight and Serum Components in Broiler Chicken (당귀근부 첨가사료가 육계의 생산성과 장기발육 및 혈액성장에 미치는 영향)

  • The root powder of Angelicae gigantis Radix (RPAGR) is known to he beneficial for liver and blood circulation. In a 6-wk broiler feeding trial, the RPAGR was added at levels of 0, 0.2, 0.5, and 1.0%, respectively. The RPAGR was found to be palatable as birds consumed moreof the test diets than the control(0% RPAGR) diet. The BW gain of 1.0% RPAGR group was higher than the control group. The feed efficiency(F/G) was improved significantly(P<0.01) by feeding 1.0% RPAGR. The RPAGR also caused beneficial effects in various carcass characteristics, i.e., carcass weight, dressing percentage, breast muscle, and thigh meat production. There was no consistent trend in various organ weights by the addition of RPAGR. Also, no consistent trend was found in abdominal fat weight. The serum cholesterol level tended to decrease as the BW increased. No consistent trend was detected in serum triglyceride contents.
