Ultrastructure and Dehydrogenase activity on the Differentiation of the Cerebral Nerve Cell in the Chick Embryo (1) (계배 대뇌의 신경세포 분화에 따른 탈수소효소 활성 및 미세구조 (1))

  • To investigate the changes during the differentiation of the cerebral neurons of chick embryo of tne embryogenic day (ED) 7 and 8, the ultrastructural changes in the cerebral neurons, the activity of dehydronases (LDH, MDH and SDH), protein expression profile and adenosine triphosphate concentration were analyzed. In ED 7 chick embryos, relatively large nucleus, centrally located nucleolus, evenly spread chromatin over nucleoplasm, and prominent nuclear envelope were observed. Oval-shaped mitochondria with well-developed cristae were present over entire cytoplasm. In ED 8 chick embryos, evenly spread chromatin over nucleoplasm, and prominent nuclear envelope were observed. In the cytoplasm, well-developed rough endoplasmic reticulum and Golgi complex were observed. In ED 7 chick embryos and ED 8 chick embryos, 31 polypeptide bands and 34 polypeptide bands were observed, respectively. The activities of dehydrogenases were lower in ED 7 chick embryos than in ED 8 chick embryos. LDH activity was 8.16 (ED 7) and 9.28 (ED 8), MDH activity was 7.98 (ED 7) and 10.10 (ED 8), and SDH activity was 5.49 (ED 7) and 7.14 (ED 8) respectively. The ATP concentration remained unchanged over ED 7 and 8.

  • A classical technique to study somitic cell fate is to employ the cross-transplantation of quail somites into a chick host. The densely stained nucleoli of the quail cells makes it possible to assess the fate of the donor quail cells in the chick host. Classical somite transplantation techniques have been hampered by the necessity of a small opening in the chick eggshell, difficulty in hatching the offspring and interspecies post-hatch graft rejection. With the advent of transgenic chicken technology, it is now possible to use embryos from transgenic chickens expressing reporter genes in somite cross-transplantation techniques to remove any possibility of interspecies graft rejection. This report describes using a surrogate eggshell system in conjunction with transgenic chick:chick somitic cell cross-transplantation to generate viable chimeric embryos and offspring. Greater than 40% of manipulated embryos survive past 10 days of incubation, and ~80% of embryos successfully cultured past 10 days of incubation hatched to produce viable offspring.

Characterization of the Pathogenesis Mechanism after Pseudomonas aeruginosa Infection through Food Consumption Using Chick Embryo Model

  • This study introduced a chick embryos’ infection model to elucidate the pathogenesis mechanism of Pseudomonas aeruginosa, which causes serious diseases in human after ingestion of P. aeruginosa-contaminated animal originated foods. The embryonic chick model is able to give a rapid and relatively inexpensive method to assess bacterial pathogenicity compared to embryos of other vertebrates. Embryos were infected with P. aeruginosa and elastase-deficient P. aeruginosa. After infection with P. aeruginosa cells, total bacterial cell numbers and gelatinase activities in the embryos were compared. Thereafter, precartilage condensation and chondrogenesis were assessed by peanut agglutinin (PNA) binding on day 3 and by Alcian blue staining for sulfated proteoglycans on day 5, respectively. P. aeruginosa significantly increased in embryos, resulting in abnormal limb development, whereas P. aeruginosa defective in elastase activity partly impaired proliferation. In addition, P. aeruginosa-infected chick embryos significantly stimulated the production of matrix metalloproteinases. Several analyses showed that elevated proteases suppressed the proliferation and survival of chondrogenic cells. The results show that this infection model was a useful assay to determine the virulence mechanism of P. aeruginosa in human after intake of microbiologically contaminated foods.

Chiari Malformation with Surgically Induced Open Neural Tube Defect in Late Chick Embryos : Characterization by Magnetic Resonance Imaging and Histopathological Analysis

  • Objective : Chiari II malformation (CM II) is still the main cause of severe morbidity and mortality in children with open neural tube defects (ONTDs). The goal of this study was to validate a CM II model in late-stage chick embryos with surgically induced ONTDs. Methods : To make the chick embryo model of ONTD, their neural tubes were opened for a length of 5-6 somites at the thoracic level in Hamburger and Hamilton stage 18 chick embryos (n=150). They were reincubated in ovo up to a total age of 17-21 days. A total of 19 embryos survived and were assigned to either the postoperative day (POD) 14-15 group (n=6) or the POD 17-18 group (n=13). Magnetic resonance imaging (MRI) and histopathologic findings of embryo heads with spinal ONTDs were compared with age-matched normal chick embryos. Results : The chick embryos with ONTDs demonstrated definite and constant structural changes, such as downward displacement of the cerebellum to just above the foramen magnum and narrow and small cerebrospinal fluid spaces in the crowded small posterior fossa. These morphologic features were more prominent in the POD 17-18 group than in the POD 14-15 group. Conclusion : This is the first description of CM II with spinal ONTD in a late-stage chick embryo model with MRI and histopathological analysis. The morphological changes of the posterior fossa in this study mimic those of CM II associated with spinal ONTD in humans. This model will facilitate investigation of the pathogenesis of CM II.

Effects of Radioactive Phosphorus (32P) on the Growth of Chick Embryo and Young Chick (방사성(放射性) 인(燐) (32P)이 계태아(鷄胎兒) 및 초생추발육(初生雛發育)에 미치는 영향(影響))

  • This experiment was performed to study the effects of radioactive phosphorus($^{32}p$) on the growth of chick embryo and young chick. Radioactive phosphorus($^{32}p$) was administered into the yolk sac of chick embryo in doses of 2 uci/gm and 1 uci/gm and was administered intraperitoneally to the young chick in doses of 1 uci/gm and 0.5 uci/gm. The chick embryos were sacrificed on 6th, 7th, 8th, 9th and 10th day and the chicks were sacrificed on 1st, 3rd, 6th and 10th day after the administration and the liver, kidney, spleen, brain, eye ball and femur were weighed to observe the effects of growth inhibition on them. The results obtained were as follows. 1. A marked growth inhibitory effect was found on 8th, 9th and 10th day after the administration of $^{32}p$ in chick embryos and the same effect was found on 6 th and 10 th day after the administration in chicks. 2. The growth of the liver, kidney, spleen and femur was inhibited markedly but the brain and eye ball were not affected in chick embryos and chicks.

Effects of Actinomycin D and Puromycin on Early Chick Embryos (초기 계배에 대한 Actinomycin D와 Puromycin의 영향)

  • Chick embryos received a single injection of actinomycin D($0.1{\mu}g,\;0.05{\mu}g\;or\;0.1{\mu}g$) or puromycin($10.0{\mu}g,\;30.0{\mu}g\;or\;50.0{\mu}g$) into the yolk sac of Arbor acres chick embryos either prior to incubation or at certain periods of time (48, 96 and 144 hours) after incubation. After 10days of incubation, surviving embryos were investigated morphologically and biochemically. Embryos treated with actinomycin D or puromycin showed a high mortality when they were exposed prior to incubation and at 48 hours after incubation. Electron micrographs of chondrocytes in tarso-metatarsal of antibiotics (actinomycin D or puromycin) treated embryos showed the destruction of cytoplasm and nuclei when they were exposed prior to incubation. Endoplasmic reticulum was expanded and mitochondria were damaged in chondrocytes of surving embryos treated with low doses at 48 hours, 96 hours or 144 hours after incubation. The activities of enzymes such as lactate dehydrogenase, malate dehydrogenase and succinate dehydrogenase in embryos treated with actinomycin D or puromycin were much less than those of the saline treated group. Also, the amounts of DNA, RNA and protein were greatly decreased.

  • A teratogenicity test of 'folpet' was carried out in the developing chick embryos to investigate and validate the safety of rural environmental hazardous materials. Folpet was administered to chick embryos' yolk sac at a rate of 0.1mg and 0.01mg per SPF eggs at 96 hours of incubation. The morphological changes were examined. Fertility ratio of SPF eggs used was 94.9%. Hatching rate of untreated control group was 74.4% and the group dosed with 100ul of corn oil into the yolk sac was 70.0%. The $LD_{50}$ of folpet was 0.663mg/100ul/egg. After hatching, mean body weight, body length, claw length and beak length of high and low dose administered groups were not significantly different from untreated and vehicle control group. There was no abnormal appearence in all the groups. Therefore it seems that, within the doses applied, folpet dose not induce potential teratogenicity in the developing chick embryos.

  • To obtain the genetic information of Korean native livestock, the karyotyping of Korean native chick were performed by high-resolution banding technique. The chromosomes were prepared from lymphocyte culture and early embryos with 200 Korean native chick which have been raised at National Livestock Research Institute. There were no significant difference between Korean native chick and Leghorn in the number of chromosomes and chromosomal morphological pattern. Using high resolution banding technique, the yield of G-bands of prophase is much greater than that can be obtained by International System for Standardzed Avian Karyotypes(ISSAK, 1999). The G-band landmarks of Korean native chick were similar to those of ISSAK and Leghorn except some macrochromosomes. chromosome Z and 3 had C-band variants with heteromorphic patterns on distal and centromeric site. The proportion of constitutive heterochromatin, the heterochromatin ratio of Korean native chick was significantly more than that of Leghorn in all chromosomes.
