CELL CULTURE STUDIES OF MAREK'S DISEASE ETIOLOGICAL AGENT (조직배양(組織培養)에 의한 Marek 병(病) 병원체(病原體)의 연구(硏究))

  • Throughout the studies the following experimental results were obtained and are summarized: 1. Multiplication of agents in primary cell cultures of both GF classical and CR-64 acute strain of Marek's disease infected chicken kidneys was accompanied by the formation of distinct transformed cell foci. This characteristic nature of cell transformation was passaged regularly by addition of dispersed cell from infected cultures to normal chicken kidney cell cultures, and also transferred was the nature of cell transformation to normal chick-embryo liver and neuroglial cell cultures. No cytopathic changes were noticed in inoculated chick-embryo fibroblast cultures. 2. The same cytopathic effects were noticed in normal kidney cell monolayers after the inoculation of whole blood and huffy coat cells derived from both forms of Marek's disease infected chickens. In these cases, however, the number of transformed cell foci appearing was far less than that of uninoculated monolayers prepared directly from the kidneys of Marek's disease infected chickens. 3. The change in cell culture IS regarded as a specific cell transformation focus induced by an oncogenic virus rather than it plaque in slowly progressing cytopathic effect by non-oncogenic viruses, and it is quite similar to RSV focus in chick-embryo fibroblasts in many respects. 4. The infective agent (cell transformable) were extremely cell-associated and could not be separated in an infective state from cells under the experimental conditions. 5. The focus assay of these agents was valid as shown by the high degree of linear correlation (r=0.97 and 0.99) between the relative infected cell concentration (in inoculum) and the transformed cell foci counted. 6. No differences were observed between the GF classical strain and the CR-64 acute strain of Marek's disease as far as cell culture behavior. 7. Characterization of the isolates by physical and chemical treatments, development of internuclear inclusions in Infected cells, and nucleic acid typing by differential stainings and cytochemical treatments indicated that the natures of these cell transformation agents closely resemble to those described fer the group B herpes viruses. 8. Susceptible chicks inoculated with infected kidney tissue culture cells developed specific lesions of Marek's disease, and in a case of prolonged observation after inoculation (5 weeks) the birds developed clinical symptoms and gross lesions of Marek's disease. Kidney cell cultures prepared from those inoculated birds and sacrificed showed a superior recovery of cell transformation property by formation of distinct foci. 9. Electron microscopic study of infected kidney culture cells (GF agent) by negative staining technique revealed virus particles furnishing the properties of herpes viruses. The particle was measured about $100m{\mu}$ and, so far, no herpes virus envelop has been seen from these preparations. 10. No relationship of both isolates to avian leukosis/sarcoma group viruses and PPLO was observed.

Studies on Calcium Availability in Various Sources by Chicken (닭에 대(對)한 칼슘 공급원별(供給源別) 효율(?率)에 관(關)한 연구(硏究))

  • The calcium balance study was carried out to determine the availability of calcium in different sources for chicks and laying hens. The sources of calcium were calcium carbonate (CC), dicalcium phosphate-dihydrate (DCPH), and dicalcium phosphate-anhydride (DCPA) for chicks and calcium carbonate (CC) and oyster shell (OS) for laying hens. The radioisotope dilution method was employed to measure the endogenous excreta calcium during the period of balance study following preliminary feeding. A. Experimental results with chicks: No significant difference was found among feed consumption of chicks fed diets containing different sources of calcium. Body weight gain of chicks was dependent upon the source of calcium. The gain decreased in the order of DCPH, DCPA and CC (P<0.01). The feed conversion efficiency in chicks fed DCPH was better than those in chicks fed CC or DCPA. The average tibia ash contents for chicks fed different sources of calcium were similar. The DCPH was superior to CC or DCPA regarding the calcium content in tibia ash. There were no significant differences among the average calcium contents in plasma trichloracetic acid filtrate in chicks irrespective of calcium sources. The mean apparent retention of calcium by chicks fed DCPH, CC and DCPA were 65.9, 64.0 and 59.9% respectively. The calcium to phosphorus ratios in tibia ash and plasma trichloracetic acid filtrate for chicks fed different sources of calcium were similar. The chicks fed DCPH showed the partition of endogenous excreta calcium in total excreta calcium as 35.6% which was higher than 31.0 or 31.4% for chicks fed CC or DCPA. The endogenous excreta calcium per day per chick in group fed DCPH, DCPA or CC were 17.2, 16.1 and 14.6mg respectively. The true retained calcium per day per chick in group fed DCPH were 109.9 mg which was higher than those observed with CC or DCPA group (P<0.01). The true retention of calcium by the birds fed diets containing DCPH, CC or DCPA were 78.1, 75.1 or 72.6% respectively. B. Experimental results with laying hens: The feed consumption, egg production and feed converion efficiency of laying hens fed diets containing different sources of calcium were similar. Calcium concentration in plasma trichloracetic acid filtrate in laying birds fed CC was equivalent to the value obtained by feeding OS. The apparent calcium retention by laying birds fed CC was 61.6% and it was significantly more than that of hens fed OS of 51.6% (P<0.05). The partition of endogenous excreta calcium in total excreta calcium of laying hens fed CC was 23.5% and this was higher than that of birds fed OS of 15.6%. The laying hens fed CC showed 310 mg of endogenous excreta calcium per day per bird while birds fed OS showed 261mg. The true retention of calcium by layers fed CC was 70.7% against 59.2% for birds fed OS (P<0.05).

The Effects of Live Yeast(Saaccharomyces cerevisiae) Supplementation on the Performance of Laying Hens (활성효모 첨가가 산란계의 생산성에 미치는 영향)

  • In order to study the effects of supplementation of live yeast(Saccharomyces cerevisiae) on the performance of laying hens, five experiments were conducted. Two experiment were conducted during summer period, one with 37 wk old Dekalb-Delta strain laying birds(Exp. 1) and the other one with 100 wk old molted Nick Chick Brown laying birds(Esp.2) . In each experiment, 240 birds were divided into 12 groups of 20 birds each and randomly distributed. Each of the two experimental diets(Control 71 and 0.05% live yeast supplemented 72) was fed to 6 groups for 4 wks in Exp.1 and 3 wks in Exp.2. Three experiments were conducted during winter period, Exp.2 with 54 wk old Hy-Line strain laying birds, Exp.4 with 52 wk old Hy-Line strain laying birds, and Exp.5 with 36 wk old broiler breeder(Indian River strain). In each experiment, 540 birds were divided into 18 groups of 30 birds each and randomly distributed. Each of the 3 experimental diets(Control:$T_1$0.05% live yeast supplemented:$T_2$ and 0.1% live yeast supplemented : T$_3$) was fed to 9 groups for 6 wks in Exp.3, 9 wks in Exp.4 and 4 wks in Exp.5. In Exp.4, Latin Square design was employed to determine the effects of switching feeds at 3 wk intervals. All hens were housed in cages of commercial farm and experimental diets were made with commercial layer feeds. In Experiment 1, egg production was significantly(P<0.05) higher in $T_2$. Feed intake was significantly (P<higher in 72 at 1st wk but 4 wk average was not significantly different. Feed efficiency was significantly(P<0.01) better in 72 at End wk but 4 wk average was not significantly different. Other parameters, such as weight, soft egg production, cracked egg production and mortality were not significantly different. In Experiment 2, egg production was significantly(P<0.05) higher in $T_2$. Feed efficiency was significantly (P<0.05 and P<0.01) better at End wk and 3rd wk but 3 wk average was not significantly different. Soft egg production was significantly(P<0.05) higher in 72. Other parameters were not significantly different. In Experiment 3, egg productions were significantly(P<0.05) different among treatments : $T_3$ was higher than $T_1$ and $T_2$ was higher than $T_1$. Egg weight of $T_1$ and $T_2$was significantly(P<0.05) heavier than $T_3$. Feed intake of $T_2$ and $T_3$ was significantly(P<0.05) higher than $T_1$ at 6th wk but overall average was not significantly different. Soft egg production were significantly(P<0.01) different among treatments:$T_1$ was higher than $T_3$ was higher than $T_2$. Feed efficiency cracked e99 Production and mortality were not signifcantly different. In Experiment 4, egg production tended to increase as the level of live yeast supplementation increased but they were not statistically different. In Experiment 5, egg production of broiler breeders of $T_3$ was significantly(P<0.01) higher than $T_1$. Feed intake of $T_3$ was significantly(P<0.05) greater than $T_1$ and $T_2$ at 3rd wk but overall average was not significantly different. Fertility and hatchability tended to be higher in the supplemented groups than in the control.

Recovery rate, growth and development of Heterophyopsis continua in experimental chicks (병아리에서 Heterophuopsis continua의 충체 회수율 및 성장 발육)

  • The growth and developmental pattern of H. continua was observed after experimental infection of their metacercariae to chicks. The recovery rate of worms from the chicks at 1 to 28 days post-infection(PI) was 12.8% in average. The rate remained fairly high for early 4 days of infection but decreased thereafter rapidly till 28 days PI. Most of the nukes, 91.9%, were recovered from the ileum of the chicks. In metacercariae, genital organs such as the ovary, testes, seminal vesicle, seminal receptacle and genital sucker were recognizable. At one day PI Mehlis'gland appeared, and at 2 days follicular vitellaria were observed. At 3 days PI, eggs were formed in the uterine tubule and increased in number as the worm grew old. The worms reached $2,990{\;}{\mu\textrm{m}}$ in length and $525{\;}{\mu\textrm{m}}$ in width at 28 days PI. Genital organs developed rapidly in early stages of infection but slowly thereafter to 28 days Pl, whereas non-genital organs developed steadily through the infection period. It was proved by this experiment that chicks should be a moderately suitable final host of H. continua.

Effect of Cecectomy on Nitrogen Utilization ]Rate and Uric Acid Excretion in Growing Chicks (성장중인 닭에 있어서 맹장절제수술이 질소의 이용률과 요산의 배설량에 미치는 영향)

  • A 2 ${\times}$ 2 factorial experiment was conducted to determine the effect of cecectomy and urea supplementation on nitrogen utilization In growing chicks. Birds were either cecectomized or shamoperated and fed a 19% protein diet with or without 1% urea supplementation. All chicks were provided ad libitum access to feed and water throughout the six-day experimental period. In the cecectomized chicks, fred efficiency was significantly decreased when fed a 19% protein diet supplemented with 1% urea (P<0.05). When chicks were fed a 19% protein diet, cecectomy significantly shortened gastrointestinal passage time (GPT) of food (P<0.05). However, the GPT was significantly lengthened in chicks fed a 19% protein diet supplemented with urea (P<0.05). Regardless of diets, cecectomized chicks increased the moisture contents of the excreta (P<0.05) and feeding a 1% dietary urea also significantly increased (P<0.05). Cecectomy significantly decreased uric acid N excretion by about 30 mg per 100 g body weight per day (P<0.05) and uric acid N/ total N excretion when fed both diets (P<0.05). The present study suggests that cecectomy improves nitrogen utilization rate by decreasing uric acid excretion in growing chicks. These results are in good agreement with those obtained previously in a cecectomy study of adult chickens.

A Study on the Correlation of Cecal Anaerobic Bacterial Counts with Cecal Length in Growing Chicks (성장중인 닭에 있어서 맹장내의 혐기성 미생물수와 맹장 길이와의 관계에 관한 연구)

  • A study was conducted to investigate the correlation of cecal anaerobic bacterial counts with the cecal length in growing chicks. Half of 160 chicks of five weeks old were housed in cages and the remainings were kept with goats in free-range and allowed to have free access to goat's feed and faeces. All the experimental birds were fed ad libitum on a commercial chick formula diet during the period of 5 to 12 weeks of age. Body weight was larger in birds reared in the cage than in those reared on the field at 8 weeks of age (P<0.05), but vice versa at the end of experiment (P<0.05). The cecal length was longer in field-rearing than in cage-rearing at 6, 9, 10 and 11 weeks of age (P<0.05). The cecal length was correlated with body weight gain in both rearing groups (r=0.816 f3r cage-rearing, r=0.816 for field-rearing). The cecal anaerobic bacterial counts were significantly higher in field- rearing than in cage-rearing at 6, 9, 10 and 11 weeks of age (P<0.05). The cecal length was highly correlated with cecal anaerobic bacterial counts in cage- (r=0.9549) and field-(r=0.9866) rearing. It is concluded that the correlation of increase cecal length with increased cecal anaerobic bacterial counts ted a libelous of goat's faeces in growing chicks.

Effects of Dietary Levels of Glycine, Threonine and Protein on Threonine Efficiency and Threonine Dehydrogenase Activity in Hepatic Mitochondria of Chicks

  • This study was carried out to evaluate the relationship between threonine (Thr) efficiency and Thr dehydrogenase (TDG) activity as an indicator of Thr oxidation on chicks fed with levels of diets (CP [17.5% and 21.5%] and Thr [3.8 and 4.7 g/100 g CP]; glycine [Gly][0.64% and 0.98%] and true digestible Thr [dThr] [0.45% and 0.60%]). Calculation of the Thr efficiency was based on N-balance data and an exponential N-utilization model, and TDG activity was determined as accumulation of aminoacetone and Gly during incubation of hepatic mitochondria. This study found that in the liver of chicks who received a diet containing up to 0.79% Thr (4.7 g Thr/100 g of CP) in the 17.5% CP diet, no significant (p>0.05) effect on TDG activity was observed. However, significantly (p = 0.014) increased TDG activity was observed with a diet containing 21.5% CP (4.7 g Thr/100 g of CP) and the efficiency of Thr utilization showed a significant (p = 0.001) decrease, indicating the end of the Thr limiting range. No significant (p>0.05) effect on the total TDG activity and accumulation of Gly was observed with addition of Gly to a diet containing 0.45% dThr. In addition, addition of Gly to a diet containing 0.60% dThr also did not result in a change in accumulation of Gly. Due to an increase in accumulation of aminoacetone, an elevated effect on total TDG activity was also observed. No significant (p>0.05) reduction in the efficiency of Thr utilization was observed after addition of Gly at the level of 0.45% dThr. However, significantly (p<0.001) reduced efficiency of Thr utilization was observed after addition of Gly at the level of 0.60% dThr. Collectively, we found that TDG was stimulated not only by addition of Thr and protein to the diet, but also by addition of Gly, and efficiency of Thr utilization was favorably affected by addition of Gly at the level near to the optimal Thr concentration. In addition, no metabolic requirement of Gly through the TDG pathway was observed with almost the same accumulation of Gly and a slight increase in TDG activity by addition of Gly. Thus, our findings suggest that determination of TDG activity and parameter of efficiency of Thr utilization may be useful for evaluation of dietary Thr level.

The effect of protease on growth performance, nutrient digestibility, and expression of growth-related genes and amino acid transporters in broilers

  • During the course of this trial, our team assessed the influence of protease upon the growth performance, the nutrient digestibility, and the expression of growth-related genes and amino acid transporters within the liver, muscle, and small intestines of broilers. During the first step, our team allocated 600 broilers into four dietary treatments for a period of 35 days in order to measure the growth performance and nutrient digestibility of the broilers selected. The separate treatments contained 10 replicates (15 birds per replicate). The treatments were composed of: 1) CON, basal diet; 2) T1, basal diet + 0.03% protease; 3) T2, basal diet + 0.06% protease; and 4) T3, basal diet + 0.09% protease. Next, the broiler chick sample tissue was harvested from the CON and T3 groups in order to conduct gene expression analysis following the feeding trials the broilers underwent. Our team discovered that the broilers fed protease diets possessed increased body weight and an average daily gain, but conversely, had lower feed conversion ratios when their dietary protease levels increased from 0% to 0.09% (p < 0.05). Additionally, significant linear improvements were identified among the nutrient digestibility of dry matter, crude protein, energy, and amino acids within broilers supplied with protease diets when contrasted and compared with broilers supplied with the basal diet (p < 0.05). In addition, the gene expression of the genes IGF1, IGF2, GH, and LEP in the liver, and the genes MYOD1 and MYOG in the breast muscles, was significantly increased after broilers were fed with a protease diet as compared to broilers that subsisted on a basal diet (p < 0.05). Protease supplementation also raised the expression levels within these amino acid transporters: SCL6A19, SLC7A1, SLC7A7, SLC7A2, SLC7A6, SLC7A9, and SLC15A1, located in the small intestine, when compared to the basal diet (p < 0.05). Our results suggest that protease supplementation in their diet improved the growth performance of broilers via an increase in the expression growth-related genes within broiler liver and muscle tissue. In addition, protease supplementation enhanced broiler digestibility via the upregulation of amino acid transporter expression within the small intestine.

Biological Characteristics and Mass Rearing System for Cadra cautella (Walker) as a Substitute Diet for Natural Enemies (천적의 대체먹이로서 줄알락명나방(Cadra cauteiia)의 생물적 특성과 대량사육)

  • Biological characteristics of the almond moth, Cadra cautella, of which eggs will be substitute diets for Orius spp. and Trichogramma spp., were investigated and a mass-rearing system for the moth was developed. At 25$^{\circ}C$, egg, larval, and pupal period was 4.2, 29.8 and 8.3 days, respectively, and adult longevity was 5.8 days for female and 4.8 days for male. Total number of eggs at 20, 25 and 30$^{\circ}C$ was 128.9,207.9 and 139.9, respectively. The moth could be successfully reared with all food substrates tested, of which rice bran (50%)+chick feed (50%) assumed to be proper for massrearing in view of cost. Eggs could be stocked at 9 C for 7 days, representing 82% hatchability. In the rearing cage (16${\times}$24${\times}$9cm) used, 1,000 eggs was better for initial level of inoculation, showing relatively high emergence rate and adult weight. Mass-rearing procedures were explained in detail.

Development of Infectious Bronchitis Virus (IBV) ELISA Kit for Detection of Antibodies against Nephropathogenic IBV Vaccine (국내회사와 다국적기업 제조 ELISA 키트의 전염성 기관지염 백신에 따른 항체 검출능 비교)

  • Infectious bronchitis virus (IBV) is an economically important disease in the poultry industry worldwide. This disease commonly manifests respiratory signs, poor egg quality, and decline in egg production. Since IBV is a RNA virus, the emergence of new variant strains is continuously reported and the immunization of susceptible chickens with only one antigenic type of the virus has been shown to induce partial or no protection against other unrelated types. Therefore, it is difficult to diagnose IBV due to variants serotypes. In this study, we collected serum from various ages of Broiler GP (Grandparent) to Layer CC (Commercial chick) and performed detectability comparison test between domestic company and multinational company manufacturing ELISA kit. Results of this experiment suggest that domestic company manufacturing ELISA kit is more sensitive to infectious bronchitis antibody than that of the multinational company. Our findings also suggest that antibody's change trends after infectious bronchitis vaccination. Thus, the use of appropriate kit for domestic situations is important.
