Efficacy of a DNA Vaccine Carrying Eimeria maxima Gam56 Antigen Gene against Coccidiosis in Chickens

  • To control coccidiosis without using prophylactic medications, a DNA vaccine targeting the gametophyte antigen Gam56 from Eimeria maxima in chickens was constructed, and the immunogenicity and protective effects were evaluated. The ORF of Gam56 gene was cloned into an eukaryotic expression vector pcDNA3.1(zeo)+. Expression of Gam56 protein in COS-7 cells transfected with recombinant plasmid pcDNA-Gam56 was confirmed by indirect immunofluorescence assay. The DNA vaccine was injected intramuscularly to yellow feathered broilers of 1-week old at 3 dosages (25, 50, and $100{\mu}g/chick$). Injection was repeated once 1 week later. One week after the second injection, birds were challenged orally with $5{\times}10^4$ sporulated oocysts of E. maxima, then weighed and killed at day 8 post challenge. Blood samples were collected and examined for specific peripheral blood lymphocyte proliferation activity and serum antibody levels. Compared with control groups, the administration of pcDNA-Gam56 vaccine markedly increased the lymphocyte proliferation activity (P<0.05) at day 7 and 14 after the first immunization. The level of lymphocyte proliferation started to decrease on day 21 after the first immunization. A similar trend was seen in specific antibody levels. Among the 3 pcDNA-Gam56 immunized groups, the median dosage group displayed the highest lymphocyte proliferation and antibody levels (P<0.05). The median dosage group had the greatest relative body weight gain (89.7%), and the greatest oocyst shedding reduction (53.7%). These results indicate that median dosage of DNA vaccine had good immunogenicity and immune protection effects, and may be used in field applications for coccidiosis control.

Antiangiogenic Activity of the Lipophilic Antimicrobial Peptides from an Endophytic Bacterial Strain Isolated from Red Pepper Leaf

  • The induction of angiogenesis is a crucial step in tumor progression, and therefore, efficient inhibition of angiogenesis is considered a powerful strategy for the treatment of cancer. In the present study, we report that the lipophilic antimicrobial peptides from EML-CAP3, a new endophytic bacterial strain isolated from red pepper leaf (Capsicum annuum L.), exhibit potent antiangiogenic activity both in vitro and in vivo. The newly obtained antimicrobial peptides effectively inhibited the proliferation of human umbilical vein endothelial cells at subtoxic doses. Furthermore, the peptides suppressed the in vitro characteristics of angiogenesis such as endothelial cell invasion and tube formation stimulated by vascular endothelial growth factor, as well as neovascularization of the chorioallantoic membrane of growing chick embryos in vivo without showing cytotoxicity. Notably, the angiostatic peptides blocked tumor cell-induced angiogenesis by suppressing the expression levels of hypoxia-inducible $factor-1{\alpha}$ and its target gene, vascular endothelial growth factor (VEGF). To our knowledge, our findings demonstrate for the first time that the antimicrobial peptides from EML-CAP3 possess antiangiogenic potential and may thus be used for the treatment of hypervascularized tumors.

PROTEIN SPARING EFFECT AND AMINO ACID UTILIZATION IN BROILERS FED TWO TYPES OF LYSINE

  • A growth experiment was conducted to evaluate the nutritivie values of supplemental lysine and methionine in broiler chicks. Two types of L-lysine, liquid and powder type, and DL-methionine were added to the diets at different levels of dietary protein with two growth phases, 0-3 weeks and 4-6 weeks named starter and grower, respectively. Six hundred seventy two chicks were allotted in 14 treatments; 3 controls by dietary CP level (starter-grower) with CP 23-21%, CP 21-19% and CP 20-18, 8 groups of liquid and powder lysine supplementation of 0.1, 0.2, 0.3 and 0.4%, and 3 groups of lysine and methionine supplementation. Body weight, feed intake, and excreta were measured and analyzed to determine growth performance, amino acid digestibilities, and the quantity of excreted nitrogen in feces. Chicks fed CP 23-20 with 3,200 ME kcal showed significantly better growth performance than those fed CP 21-18 for 6 weeks. The supplementation of 0.2% of either type of lysine to CP 21-19 diet improved weight gain and feed efficiecy to the extent that CP 23-21 diet was fed. Physical type of lysine did not affect chick's growth and amino acid digestibilities of the diets. The level of CP in the diet significantly affected nitrogen excretion in feces. Supplementation of lysine and methionine to CP 21-18 diet reduced fecal nitrogen by 10% compared to CP 23-21 diet. It was confirmed that 0.2% of supplemental lysine to the broiler diet spared the dietary protein by 3%, and also reduced nitrogen excretion in feces by 10%.

  • Epoxy-methacrylate prepolymer was synthesized from methacrylic acid and a diglycidyl ether type low molecular weight epoxy resin, and the electron beam curing of the prepolymer was studied using styrene and polyethyleneglycol dimethacrylates as comonomers. The esterification was carried out quantitatively without any side reaction adding more than 250∼300ppm hydroquinone and less than $1{\%}$ (wt) triethanolamine by wt. of methacrylate acid, respectively. In the radiation curing under air atmosphere, the maximum gel fraction was observed at 10∼$30{\%}$ monomer contents. The rate of gel formation was decreased by the presence of unreacted epoxy group and acid in the prepolymer and increased with increasing degree of polymerization of polyethyleneglycol block in the dimethacrylates. Considerable oxygen effect in the curing, particularly when polyethyleneglycol dimethacrylate was used as a comonomer, was recognized through the differences of the product properties, though no remarkable differences were found in the rate of gel formation in air and nitrogen atmosphere.

A Study on the Cooking in 'The Ryuk-Jab-Rok' ("역잡록"의 조리가공에 대한 분석적 고찰)

  • This book is named 'The Ryuk-Jab-Rok' because it was written in the last page of 1820's almanac. This book is written purely in Korean and has not been published yet. This book comprises twenty eight items, among which there are eight items of vegetable preserving method, ten items of Jook preparation methods(a kind of cereal soup), nine items of Pyun-Myun methods(a kind of rice cakes and noodles) and Yak-Bab(a kind of spiced rice). In vegetable preserving method, the eggplant, the cucumber and a songi mushroom were preserved with the drying method., A radish, a sorojangii, and the root of white cabbage were used with the cold temperature preservation. A garlic was dried after salting. The sprouts of DooRub, which were coming in the hot room, were used. In the Jook preparations, there were five animal materials which were lamb, chick, crudian, oyster and abalone. In nine Pyun-Myun methods, Jap-Gua-Sil was illegible because the letters were not clear. Among eight items, the stick rice was used in four cases, the regular rice in two cases. The ground pine nuts, honey and the Chinese date were used most ofter. And the sesame salt and the chestnut were next. The analysis of the terms I this book revealed that 26 items were used for cooking processes. And it also showed us that there were seven kind of cutting procedure and eight kinds of heating procedure. The shapes and size of foods were revealed at only three places in all items. The one-chi(chi ; abut three cm) and three-Ja(Ja ; about thirty cm) which the terms represent the length were revealed twice and once respectively in this book. In the taste description, 'the good', the most common word, was used in seven times, and which was the most frequently introduce case. The measuring unit is hard to revive since the measurements were taken by the container, which were Jong-Ja, Sabal and Tang-gii, then in use. Fifteen kinds of containers and cookers were used for preparing foods. And all of them are now I use.

Effects of Vitamine and Mineral Supplements on the Performances of Broiler Breeder Hens (비타민과 미네랄 첨가가 육용종 암탉의 생산능력에 미치는 영향)

  • This work was designed to investigate the performances of broiler breeder hens upon the addition were vitamin and mineral supplements in feeds. Animals were divided into for feed groups such as control, T$_1$,T$_2$and T$_3$, and each treatment has four replications. All broiler breeder hens were housed individually, and were fed with identical iso-metaboilic energy and iso-protein, except for vitamins and minerals. Different sources and levels of vitamins and minerals were added to the basic feeds according to the treatments. At the end of the experiments, general laying rate(hen days), average egg weight, hatching egg laying rate(Hen day) every day. Hatching egg weight, sterility, embryo mortality, hatching rate and chick weight were examined. Although, early laying rate in all treatment groups were reduced during cold term management, general egg laying rate was increased only T$_3$group which was contanining vitamin C. However, similar results were obtained in the rates of general and hatching egg laying. General egg laying rate were somewhat higher of Control, T$_2$and T$_3$ groups than that of T$_1$group. In addition, the laying rates of general and hatching eggs slightly improved in the T$_3$group rather than those of Control group. Egg weight was reduced with the increasing of laying rate, showing the highest in the T$_3$group. The laying rate showed significantly lower value than those of any other groups. Egg weight was about 65g in T$_3$group. The results of hatching performance for production ability of Broiler breeder hens were fed experimental diet, although the rates of non-hatched and hatched eggs increased in order of Control, T$_1$T$_2$and T$_3$groups. However, the rate of embryo mortality was declined in order Control, T$_1$T$_2$and T$_3$groups.

Comparison of Broiler Performance according to Infection Rate of Chicken Mycoplasmosis in Broiler Breeders (육용종계 농장의 닭 마이코플라즈마병 감염율에 따른 육계 사육성적 비교)

  • Various factors negatively affect poultry production, including a multitude of avian pathogens. Among them, Mycoplasma gallisepticum (MG) and Mycoplasma synoviae (MS) cause world-wide economic losses in the poultry industry. Although respectively, MG and MS induce chronic respiratory disease and synovitis in poultry, these pathogens often manifest as clinically inapparent infections and many countries are therefore having difficulties effectively eradicating them. In this study, five broiler breeder farms in Korea were investigated for the presence of Mycoplasma infection from 2016 to 2018. Serum samples were collected for detection of antibodies to mycoplasma species using an enzyme-linked immunosorbent assay (ELISA) kit. Additionally, the production index and feed conversion rate of broilers were recorded. According to this investigation, broiler breeder farms in 2018 were serologically negative for Mycoplasma infection with broilers exhibiting a high production index and low feed conversion rate. Thus, eradicating mycoplasma in poultry farms is critical for improving chick quality and overall poultry production.

Verification of immunosuppression in chicks caused by Cryptosporidium baileyi infection using Brucella abortus strain 1119-3 (닭와포자충 감염닭에서 Brucella abortus의 이차감염에 대한 면역저하 현상의 입증)

  • Humoral immune response of young chicks to Brucella abortus strain 1119-3 inoculation was monitored to verify the degree of immunosuppression caused by infection with Cryptosporidium baileyi. Young chicks (2-day-old) were orally inoculated each with $2{\times}10^6$ oocysts of C. baileyi, and then injected intramuscularly with 0.3 $m\ell$ B. abortus strain 1119-3 containing $1{\times}10^9$ living organisms on day 14 postinoculation (PI). Serum samples were tested by plate agglutination test on day 17 PI onwards at an interval of 3-6 days over a period of 36 days. Infected chicks with the coccidium showed significantly lower antibody titers than those of uninfected controls (P<0.05). These findings document that C. baileyi infection in early life stage may predispose chicks easily to other potential poultry diseases.

Experimental infection of Campylobacter jejuni and Campylobacter coli in chicks (유추(幼雛)에 있어서 Campylobacter jejuni 및 Campylobacter coli의 실험적(實驗的) 감염(感染))

  • The present study was carried out to investigate the incidence of C jejuni and C coli in chicken. Also were examined the pathogenicity of the isolates in chick by experimental inoculation. Thermophilic Campylobacter were isolated from 34(45.9%) of the 74 specimens, and classified as 21.6% C jejuni, and 24.3% C coli. In the biotyping of 16 stranis of C jejuni isolates, 37.5% of the strains were grouped as biotype I, 62.5% as biotype II. In the case of 18 strains of C coli isolates, 49.9% of isolates were grouped as biotype I, 55.6% as biotype II. n oral inoculation with $10^4cfu$ of Campylobacter isolates into infant chicks(1 to 3 days-old), 17 days-old and 34 days-old chicks, 32.5% of the chicks developed diarrhea on day 1, 52.5% on day 3, 70.0% on day 5, and 27.5% on day 7, and the peak incidence of diarrhea was reached on day 5. The organisms were found to be discharged in feces one day afterwards. C jejuni and C coli strains were detected from the feces in 87.5% of the chicks on day 5. The organisms were multiplied from $10^4$ to $10^6cfu/gm$ in feces 5 to 7 days after inoculation. C jejuni and C coli recovered from 100% of the cecum, 64.3% of the duodenum, 50.0% of the spleen, 42.9% of the livers, and from 21.4% of gallbladders 7 days after inoculation.

Germ-line Transmission of Pseudotyped Retroviral Vector in Chicken

  • Using MLV (murine leukemia virus)-based retrovirus vectors encapsidated with VSV-G (vesicular stomatitis virus G glycoprotein), we tried to make transgenic chickens carrying the transferred genes in their chromosomes. Twenty one days after virus injection beneath the blastoderms of unincubated chicken embryos (stage Ⅹ, at laying), DNA isolated from the hatched chicks were analyzed by PCR with two sets of primers specific for EGFP (enhanced green fluorescence protein) gene or $Neo^R$ (E. coli neomycin resistant) gene. Among sixty-seven embryos injected with retrovirus, four of them were identified to carry the EGFP genes in their genomes. Remarkably, one transgenic chick showed presence of the retrovirus vector sequences in all organs differentiated from one of endoderm, mesoderm, and ectoderm. Expression of EGFP gene was not detected, however, the stable germ line transmission of transgene was verified in spermatozoa from the founder chicken and 50% of $F_1$ progenies.
