It was investigated the morphological changes of differentiating epidermal cells in chick embryos. Ectodermal cells at 3 days after incubation were cuboidal, their nuclei were large, and rough endoplasmic reticulum and mitochondria were distributed in the cytoplasm. At 5 days after incubation, there were periderm and one basal layer in epidermis. The cells of basal layer were columnar, their nuclei were round, and rough endoplasmic reticulum and free ribosomes were developed. Also, peridermal cells were flat, chromatins were partially condensed and glycogen particles were abundant. No periderm showed and cells of basal layer formed intermediate layer at 9 days after incubation. Basal cells of intermediate layer were cuboidal, neighboring cells were anchored by desmosomes and tonofibrils and free ribosomes were evenly scattered. At 15 days after incubation, stratum corneum and stratum germinativum were distinguished. In cells of stratum germinativum, tonofibrils, free ribosomes and desmosomes were well developed. And then, the shedding of stratum corneum were showed at 17 days after incubation and stratum corneum were well developed after hatching.
Primordial germ cells (PGCs) were manipulated as part of the system to produce transgenic chickens. PGCs were isolated from the germinal crescent of developmental stage 6 to 8 donor emhryos of the Korean Native Ogol Chickens (KNOC). These PGCs were transfected with plasmid DNA containing the lac Z gene by liposome mediated transfection methods. The lac Z gene was transfected and expressed in the PGCs. These transfected PGCs were injected into the germinal crescent of White Leghorn embryos (stage 6 to 8). The injected transfected PGCs migrated via the circulatory system into the future gonad and expression observed in the gonads of 3 day old chick. Of the 47 embryos and 3 day old chickens, one positive PGCs gonad from sacrificed young chickens was detected by appearance of blue cells. Plasmid DNA with the foreign gene was incorporated into the population of germ cells in the gonad. These results demonstrate that PGCs can he transfected and then transferred for colonization into the gonad, and show the potential to ultimately manipulate the genetic material of the chicken gernline.
To understand the role of protein kinase C (PKC) in the regulation of chondrogenesis, we examined proteins which are phosphorylated by PKC. Stage 23/24 chick embryo wing mesenchymes were micromass-cultured to induce chondrogenesis and cell extracts were phosphorylated in a condition that activates PKC. Several proteins including 63 and 66 kDa proteins were phosphorylated. The 66 kDa protein was phosphorylated only in the presence of phorbol 12-myristate 13-acetate (PMA) and phosphatidylserine CPS), and the phosphorylation was almost completely diminished by bisindolylmaleimide, a PKC inhibitor. In addition, partially purified PKC increased the phosphorylation of the 66 kDa protein. Treatment of cultures with lysophosphatidylcholine (LPC) promoted chondrogenesis and phosphorylation of 66 kDa protein, while PMA and thymeleatoxin inhibited both of the two events. Our results suggest that the 66 kDa protein is a putative substrate of PKC, and phosphorylation of the 66 kDa protein, probably by $PKC\alpha$ is required for chondrogenesis.
A trial involving 200 day-old 288 Shaver chicks in a randomized experimental design tested the effects of using diets based on local feed materials (LF). Commercial imported counterpart feeds (CF) served as control diets. Birds were raised from 0-6 w on Chick Starter feed, from 7-17 w on Pullet Developer feed and from 18 to 72 w on Layer feed. The results obtained indicated that there were no significant differences in the performance of birds fed LF and CF diets during the Chick Starter and Pullet Developer phases. During the Laying phase, there were also no differences in laying percentage, (66 vs 65) and in egg size (62 vs 63 g) between LF and CF diets. Feed cost was lower on LF diet had a better egg yolk colour score, (4 vs 1) than those fed on CF diet. Feed cost was lower on LF than CF diets and the feed cost for producing eggs was approximately 50 percent lower on LF compared to CF (P$ 0.06 vs P$ 0.12). It was concluded that the use of locally available feed resources produces comparable performance to that obtained using commercial imported feeds. Secondly, using local materials markedly reduces feed cost and cost of producing eggs.

Effects of polyunsaturated fatty acid (PUFA) and vitamin E (VE) supplementation in the diet of breeder hens on the egg quality and hatchability, lipid peroxides of the egg yolk, and development of the newly-hatched offspring chicks were investigated. A total of 800 Avian 48, 28 wk-old broiler breeders were assigned randomly to 4 groups with 4 replicates of 45 females and 5 males. Each group was fed one of the following four diets with different oil sources and levels of VE: corn oil (CO), fish oil (FO), CO+VE and FO+VE. The results showed that: i) Addition of FO in the breeder diet reduced the whole egg weight, yolk weight, albumen weight, yolk color score and neonatal offspring chick body weight without affecting the hatchability as compared to the CO treatment. ii) Addition of VE efficiently reduced the lipid peroxides of egg yolk from hens fed diets containing FO. iii) VE in the breeder diet significantly promoted the development of liver and heart of the chick offspring.
To investigate the effects of mercuric chloride ($HgCl_2$) on the differentiation in the cerebral neuron of chick embryo 7 days, the ultrastructural changes in nerve cells injected with a various doses of mercuric chloride were observed with transmission electron microscope. The enzyme activity of the some dehydrogenases, and adenosine triphophate (ATP) were also analyzed. The results obtained are as follows; The ultrastructural changes in 1.0mg-injected group, the nuclear envelope were irregular, and the RER, Golgi complexes and mitochondria were not well developed. In 2.0mg-injected group, the nuclear envelope were partly destroyed or detached, and mitochondria were decreased in number and their cristae were destroyed, too. The RER and Golgi complexes were less developed than those of the normal groups. In general, the activities of dehydrogenases were declined by increasing the dose of mercuric chloride. Lactate dehydrogenase (LDH) activity fatted to below 85% of the normal group in 1.0mg-injected group, and 69% in 2.0mg-injected group. Malate dehydrogenase (MDH) activity was decreased greatly to 76% in 2.0mg-injected group. Succinate dehydrogenase (SDH) activity fatted to 85% in 1.0mg-injected group, and 74% in 2.0mg-injected group. ATP content in 1.0mg-injected group was almost near to the normal level, but it was increased significantly in 2.0mg-injected group.
This study was undertaken to obtain more detailed knowledge of the oxygen consumption rate of chick embryos, of the relationship between the increasing rate of embryo weight and that of oxygen consumption, of the daily increase of oxygen consumption by that of embryo weight, and of the metabolic rate of the White Leghorn eggs. The results obtained are summarized as follows. 1. The average oxygen consumption rate of the chicken embryo at the 3rd day of incubation is 0.8ml/h, STPD. It is strongly suggested that this value can used as a physiological criterion to classify the fertilized and unfertilized eggs, on the other hand oxygen consumption rate of the fertilized eggs reaches a plateau between the 18th and 20th days. 2. There exists a parallel relationship between the increasing rate of embryo weight and that of oxygen consumption rate during the incubation period. 3. There are not exist a parallel relationship between the daily increase of embryo weight and that of oxygen consumption during the whole incubation period. 4. The metabolic rate of chicken embryo(ml/h/g) is highest in the early stage of incubation and it started to decrease sharply until the 8th day follow by slow decrease thereafter.
Embryonic and postembryonic chicken lenses have been analyzed morphologically to investigate the differentiation of the lens fibers by light and electron microscopes. Morphogenesis of the chick lens was initiated as lens epithelial cells were proliferated and proceeded to elongate the cells characteristically at posterior side, by which the disintegrations of nuclei were accompanied during the early developmental stages. Primary and secondary lens fibers were identified at the late developmental stages, while interconnections between neigh-boring cells well developed and denucleation commenced. On day of hatching, the chicken lens fibers contained few cell organelles within the cytoplasm and showed the homogeneity of cytoplasmic appearance. On day 10 of hatching, the lens were fully differentiated; fiber cells, in which most cell organelles except polysomes were disappeared, showed a slender and elongated prismatic shape. At that stage gap junctions were particularly developed or cytoplasmic ridges are closely interlocked between adjoining cells. In conclusion, differentiation of chick lens involves the division of epithelial cells, the elongation into fiber cells, the loss of cell organelles and the increase of gap junction.
Ion-exchange polymeric stationary phases presenting amino acid and amino groups were prepared by the surface grafting of glycidyl methacrylate onto a silica gel surface and subsequent amination. Three kinds of amino acids-L-arginine (Arg), D-lysine (Lys), and D-histine (His)-were used in this study. An ion-exchange polymeric stationary phase presenting ethylene diamine (EDA) was also prepared by surface graft polymerization. Separation of the model proteins bovine serum albumin (BSA), chick egg albumin (CEA), and hemoglobin (Hb) was performed using the amino acid- and amine-derived columns. In separating the CEA/BSA mixture, the resolution time of BSA was longer than that of CEA when using the EDA column, whereas the resolution time of BSA was shorter than that of CEA when using the Arg, Lys, and His columns. In the separation of the Hb/BSA mixture, the resolution time of BSA was longer than that of Hb in the EDA column, whereas the resolution time of BSA was shorter than that of Hb in the amino acid columns (D-Lys, L-Arg, and D-His).
A Potent inhibitor of trypsin and other various serine proteases including chymotrypsin, elastase, kallikrein, plasmin and subtilisin, has been purified to homogeneity from chick skeletal muscle by convendonal chromatographic procedures. The Inhibitor has an apparent molecular weight of 66, 000 dalton as determined by gel filtration. When the purified inhibitor was electrophoresed in the presence of sodium dodecyl sulfate, there appeared rwo protein bands having molecular weights of 66, 000 and 64, 000 dalton. The 64, 000 dalton protein seems to be the product of 66, 000 dalton protein by a lin'ited proteolysis during the purification procedure or in viuo. Thus, it seems to consist of a single polypeptide. The inhibitor appeared to be glycoprotein and have an isoelectric point of 7.4. It contains relatively large amount (8.33 mole%) of cysteine residues.
