A Chemiluminescence Method for the Determination of Ascorbic Acid based upon Its Reaction with Cerium(IV) in the Presence of $Ru(bipy)_3^{2+}$

  • A chemiluminescence (CL) method for the determination of ascorbic acid was proposed, which is based on the enhancement of the CL intensity of $Ru(bipy)_3^{2+}$ - Ce (IV) system with the addition of ascorbic acid. The CL intensity is proportional to the concentration of ascorbic acid in the range of $1{\times}10^{-10}\;to\;1{\times}10^{-5}M$ with a correlation coefficient of 0.9985. The detection limit is $3{\times}10^{-11}M$. The relative standard deviation (R.S.D.) of 10 determinations of 0.1 mM ascorbic acid was 1.75%. The proposed method has been successfully applied to the determination of ascorbic acid in the commercia] drinks. The possible mechanism of the CL reaction was studied.

Chemiluminescence Determination of Balofloxacin Based on Europium (III)-Sensitized KBrO3-Na2S2O4 Reaction in Micellar Medium

  • A novel chemiluminescence (CL) flow injection method for the determination of balofloxacin is described. The method is based on the weak CL signal arising from the reaction of $KBrO_3$ with $Na_2S_2O_4$ in acidic medium being significantly enhanced by balofloxacin in the presence of europium (III) ion and sodium dodecyl benzene sulfonate (SDBS). The experimental conditions that affected CL intensity were carefully optimized and the CL reaction mechanism was briefly discussed. Under the optimum conditions, the relative CL intensity was proportional to the concentration of balofloxacin in the range of $7.0{\times}10^{-11}$ to $3.0{\times}10^{-7}g\;mL^{-1}$. The detection limit was $2.7{\times}10^{-11}g\;mL^{-1}$ and the relative standard deviation was 2.1% for $7.0{\times}10^{-10}g\;mL^{-1}$ balofloxacin (n = 13). The proposed method was successfully applied to the determination of balofloxacin in pharmaceutical formulations and biological fluids.

  • A sensitive silver nanoparticles (Ag NPs) enhanced chemiluminescence (CL) method is reported for the determination of mandelic acid (MA). This method is based on the luminol-KIO4 system catalyzed by Ag NPs to produce CL spectra. Prepared Ag NPs were characterized by UV-visible spectra and TEM image. Under optimal condition, CL spectra of the system were responded linearly with the concentration of MA in the range of 2.5×10-9 to 2.0×10-8 mol L-1 (r=0.9989) with a detection limit of 1.2×10-10 mol L-1. The relative standard deviation of 1.0×10-7 mol L-1 MA was found 1.45 (n=9).

  • We have been studing sensitive non-enzymatic chemiluminescence (CL) imaging methods for the detection of DNA. For one of our methods, a unique chemical derivatization reagent. 3'. 4', 5'-trimethoxyphenylglyoxal (TMPG) was utilized. This reagent reacted specifically with guanine bases in nucleic acids to quickly produce a chemiluminescent derivative under mild reaction conditions. (omitted)

A Metal Enhanced Flow-Injection Chemiluminescence Method for the Rapid Determination of Norfloxacin in Pharmaceutical Formulations and Serum Sample

  • A simple and highly sensitive chemiluminescence method to determine norfloxacin (NFLX) has been proposed by measuring the chemiluminescence (CL) intensities using a flow injection (FI) system. The CL intensity of the luminol-$H_2O_2$ system is strongly enhanced by the addition of Cu (II) in alkaline condition. The CL intensity is substantially increased after the injection of NFLX into the luminol-$H_2O_2$-Cu (II) system. The enhancement effect is attributed to a catalytic effect of Cu (II) due to the interaction with NFLX which forms a complex with the catalyst. Under the optimal conditions, the sensitizing effect of the CL intensity is proportional to the concentration of NFLX in the range of $1.5{\times}10^{-9}-5.9{\times}10^{-7}molL^{-1}$ (r = 0.9994) with a detection limit ($3{\sigma}$) of $2.98{\times}10^{-10}molL^{-1}$. The proposed method had good reproducibility with the relative standard deviation (RSD, n = 5) of 1.6% for $1{\times}10^{-7}molL^{-1}$ of NFLX. The possible reaction mechanism of the CL reaction is also discussed. This method has been successfully applied for the determination of trace amount of NFLX in pharmaceutical preparations and serum samples.

은 나노입자를 이용한 화학발광법에 의한 L-alanine의 정량 (Determination of L-Alanine Using Silver Nanoparticles Chemiluminescence System)

  • A chemiluminescent method with silver nanoparticles for determination of L-alanine has been presented. The chemilumiscence intensity was further enhanced by silver nanoparticles in the luminol system by its catalytic role. The silver nanoparticles enhanced chemiluminescent method is applicable for the determination of an amino acid such as alanine. When alanine was introduced to the luminol system with silver nanoparticles, chemiluminescence intensity was reduced with the concentration of the added alanine. The effects of pH, concentrations of luminol, hydrogen peroxide and silver nanoparticles on the chemiluminescence intensity were investigated. The calibration curve for L-alanine was linear over the range from 6.60×10-8 M to 4.00×10-7 M, coefficient of correlation was 0.996 and detection limit was 3.5×10-9 M under the optimal conditions of 4.0×10-3 M, 4.0×10-2 M, 4.0×10-4 M, 12.8 for the concentration of luminol, H2O2, silver nanoparticles and pH, respectively.

A Generator of Gaseous Singlet Oxygen

  • In order to prevent side reactions due to free radical formation occurring in the reactions of singlet oxygen generated in solution phse, it was required that singlet oxygen is generated in gas phase from an apparatus of a solid-gas system. We have accomplished to construct an apparatus generating singlet oxygen in solid-gas system, which is composed of a flexible optical fibre tube connected by a dye-sensitizer probe containing rose bengal dye on polymer or inorganic material. Through the optical fibre tube visible light from a laser and an oxygen stream are passed into the sensitizer probe where singlet oxygen is generated. The determination of singlet oxygen was carried out by two methods. One involves the detection of the luminescence of singlet oxygen at 1268 mn and the other involves the chemiluminescence reaction of a dihydroisobenzofuran with singlet oxygen emitting luminescence at 456 nm.

Characterization of the ${\cdot}O_{2}^{-}$-Formation by Pyridine Nucleotide in Rat Hepatocytes

  • The detection with lucigenin under physiological conditions is selective for ${\cdot} O_{2}^{-}$, for it can be accepted that lucigenin indicates actual intramembranal $\cdot O_{2}^{-}- formation$. Lucigenin chemiluminescence (CL) was elicited from the plasma membrane (PM) only by addition of reduced pyridine nucleotide. NADPH was preferred to NADH in PM and hepatocytes. This specificity was masked by $NAD(P)^+$ inhibition. The half maximum rate of CL increase was obtained with 1.5 ${\mu}m$ NADH or 55 ${\mu}m$ NADPH in hepatocytes and 6 ${\mu}m$ NADH or 30 ${\mu}m$ NADPH in plasma membranes. Measurement of these NADPH values required the presence of a NADPH-regenerating system. With NADPH the maximal rate obtained was 10 fold higher than with NADH. NADPH and NADH could produce CL when having access from either side of the membrane. They seemed to react with the identical acceptor because NADH-induced CL was also inhibited by $NADP^+$. The characteristics of ${\cdot}O_{2}^{-}-formation$ produced by pyridine nucleotide will be discussed.

  • 정지흐름분석법을 이용하여 화학발광법으로 수용액 중의 Fe(II)와 Fe(III)을 분리 정량하는 방법에 대하여 연구하였다. 이 방법은 lucegenin과 $H_2O_2$ 혼합용액에 Fe(III)이온을 첨가하였을 때 화학발광의 세기가 증가하는 것을 기초로 하였다. KO H, $H_2O_2$ 및 Fe(III)의 가리움제로 이용한 citric acid의 농도와 시료의 주입속도가 화학발광의 세기에 미치는 영향을 조사하였다. 전체 철의 정량을 위하여 구한 [$H_2O_2$], [KOH] 및 흐름속도의 죄적조건은 각각 4.0M, 2.0M, 및 3.5mL/min 이었고, 이러한 최적조건 하에서 얻은 검정곡선에서 직선성이 성립하는 범위는 1.0${\times}10^{-6}$M에 서 1.0${\times}10^{-4}$M이었고, 상관계수는 0.996, 검출한계는 1.0${\times}10^{-7}$M이었다. Fe(III)이온의 정량을 위하여 구한 [$H_2O_2$], [KOH], [citric acid] 및 흐름속도의 최적조건은 각각 4.0M, 2.0M, 0.01M 및 3.5mL/min이었고 이러한 최적조건 하에서 얻은 검정곡선에서 직선성이 성립하는 범위는 1.0${\times}10^{-6}$M에서 1.0${\times}10^{-4}$M이었고, 상관관계수는 0.997, 검출한계는 5.0${\times}10^{-7}$M이었다.

Applications of Capillary Electrophoresis and Microchip Capillary Electrophoresis for Detection of Genetically Modified Organisms

  • In recent years, special concerns have been raised about the safety assessment of foods and food ingredients derived from genetically modified organisms (GMOs). A growing number of countries establish regulations and laws for GMOs in order to allow consumers an informed choice. In this case, a lot of methods have been developed for the detection of GMOs. However, the reproducibility among methods and laboratories is still a problem. Consequently, it is still in great demand for more effective methods. In comparison with the gel electrophoresis, the capillary electrophoresis (CE) technology has some unique advantages, such as high resolution efficiency and less time consumption. Therefore, some CE-based methods have been developed for the detection of GMOs in recent years. All kinds of CE detection methods, such as ultraviolet (UV), laser induced fluorescence (LIF), and chemiluminescence (CL) detection, have been used for GMOs detection. Microchip capillary electrophoresis (MCE) methods have also been used for GMOs detection and they have shown some unique advantages.
