DEVELOPMENT OF POLYETHOXYLATED RETINAMIDE AS AN ANTI-AGING AGENT

  • A novel retinol derivative, polyethoxylated retinamide(Medimin A) was synthesized, as an anti-aging agent. Collagen synthesis, skin permeation, stability, and toxicity of Medimin A were evaluated and compared with those of retinol and retinyl palmitate. In vitro collagen synthesis was evaluated by quantitative assay of $[^3H]-proline$ incorporation into collagenase sensitive protein in fibroblast cultures. For in vitro skin permeation experiments, Franz diffusion cells(effective diffusion area: 1,766 $cm^2$) and the excised skin of female hairless mouse aged 8 weeks were used, The stabilities of retinoids were evaluated at two different temperature($25^{\circ}C\;and\;40^{\circ}C$) and under UV in solubilized state and in O/W emulsion. To estimate the safety, acute oral toxicity, acute dermal toxicity, primary skin irritation, acute eye irritation and human patch test were performed. The effect of Medimin A on collagen synthesis was similar to that of retinol. The skin permeability of Medimin A was higher than those of retinol and retinyl palmitate. The Medimin A was more stable than retinol and retinyl palmitate. Medimin A was nontoxic in various toxicological tests. These results suggest that Medimin A would be a good anti-aging agent for enhancing bioavailability and stability.

환경성질환과 환경유해인자의 연관성을 규명하기 위한 독성 연구 고찰 (A Systematic Review of Toxicological Studies to Identify the Association between Environmental Diseases and Environmental Factors)

  • Background: The occurrence of environmental disease is known to be associated with chronic exposure to toxic chemicals, including waterborne contaminants, air/indoor pollutants, asbestos, ingredients in humidifier disinfectants, etc. Objectives: In this study, we reviewed toxicological studies related to environmental disease as defined by the Environmental Health Act in Korea and toxic chemicals. We also suggested a direction for future toxicological research necessary for the prevention and management of environmental disease. Methods: Trends in previous studies related to environmental disease were investigated through PubMed and Web of Science. A detailed review was provided on toxicological studies related to the humidifier disinfectants. We identified adverse outcome pathways (AOPs) that can be linked to the induction of environmental diseases, and proposed a chemical screening system that uses AOP, chemical toxicity big data, and deep learning models to select chemicals that induce environmental disease. Results: Research on chemical toxicity is increasing every year, but there is a limitation to revealing a clear causal relationship between exposure to chemicals and the occurrence of environmental disease. It is necessary to develop various exposure- and effect-biomarkers related to disease occurrence and to conduct toxicokinetic studies. A novel chemical screening system that uses AOP and chemical toxicity big data could be useful for selecting chemicals that cause environmental diseases. Conclusions: From a toxicological point of view, developing AOP related to environmental diseases and a deep learning-based chemical screening system will contribute to the prevention of environmental diseases in advance.

알루미늄 가공 공장 배출 방류수의 독성 원인물질 탐색 (Application of Toxicity Identification Evaluation Procedures for Toxic Effluents from the Aluminum Rolling Industry)

  • Objectives: The objective of this study is to identify toxicants causing acute toxicity in effluents from the aluminum rolling industry that violate the discharge limits in Korea. Methods: Whole effluent toxicity tests (WET) were conducted on effluent discharged from the aluminum rolling industry following the US EPA WET test methods. We collected effluent samples three times and evaluated acute toxicity by using Daphnia magna. We employed toxicity identification evaluation (TIE) procedures to identify toxicants causing toxicity in the effluent. Results: No specific chemical groups were identified in the seven different manipulations applied to the of wastewater effluent samples showing 1.3 toxic units (TU) according to the TIE phase I procedures. Water quality parameters for water hardness, electric conductivity and heavy metals (Mn) were 4,322 mg/l as $CaCO_3$, 11.39 mS/cm, and $5,551{\mu}g/l$, respectively. Considering water hardness and reference toxicity, high concentrations of Mn can be disqualified from the causative toxicants. Consequently, high ionic concentrations of $Na^+$(1,648 mg/l), $Ca^{2+}$(1,048 mg/l), $Mg^{2+}$(1,428 mg/l) and $SO_4{^{2-}}$(7,472 mg/l) were identified to be causative toxicants. Water hardness and electric conductivity exceed the $EC_{50}$ value obtained by biological toxicity tests using Daphnia magna. Conclusion: According to TIE procedures, high salt concentration is determined to be a major toxicant in the effluent of agro-industrial wastewater treatment plants receiving wastewater from the aluminum rolling industry.

농약의 노출시간에 따른 급성어독성의 변화 (The changes of acute toxicity of pesticides depending on the exposure time to killifish, Oryzias latipes.)

  • 담수산 어류인 송사리(Oryzias latipes)에 대한 6개 농약의 급성독성을 7일동안 시간의 경과에 따라 측정, toxicity curve를 작성하여 각 농약의 시간의 경과에 따른 급성독성 발현의 현상을 평가하고 48시간 및 96시간 LC50값과 lethal threshold 농도와의 관계를 분석하여 얻은 결과는 다음과 같다. 농약의 toxicity curve를 중심으로 시간의 경과에 따른 급성독성값의 변화를 보면 butachlor가 120시간째, fenobucarb가 48시간째, chlorothalonil이 48시간째 lethalthreshold 농도를 나타내어, 각각 120시간, 48시간만에 급성독성현상이 더이상 시간의 경과에 따라 발현되지 않으나, 나머지 3개의 농약인 alachlor, diazinon, iprobenfos는 노출 7일 이후에도 계속 급성독성현상을 나타냄을 알 수 있었다. 이로써 6개의 농약에 대한 lethal threshold 농도는 butachlor 0.53 mg/l(0.49-0.57 mg/1), alfachlor 1.15 mg/1>, fenobucarb 10.3 mg/1(9.3-11.4 mg/1), diazinon 6.70mg/1>, chlorothalonil 0.085 mg/1(0.082-0.087 mg/1), iprobenfos 3.42 mg/1>이었다. 결국 노출시간 96시간이내에 독성현상이 발현되어 더이상 치사율이 증가하지 않은 농약은 6개중 2종류뿐이며 나머지 4개중 3개에서는 노출후 7일이 경과하여도 계속하여 독성이 발현됨을 알 수 있었다.

Toxicoinformatics: The Master Key for Toxicogenomics

  • The current vision of toxicogenomics is the development of methods or platforms to predict toxicity of un characterized chemicals by using '-omics' information in pre-clinical stage. Because each chemical has different ADME (absorption, distribution, mechanism, excretion) and experimental animals have lots of variation, precise prediction of chemical's toxicity based on '-omics' information and toxicity data of known chemicals is very difficult problem. So, the importance of bioinformatics is more emphasized on toxicogenomics than other functional genomics studies because these problems can not be solved only with experiments. Thus, toxicoinformatics covers all information-based analytical methods from gene expression (bioinformatics) to chemical structures (cheminformatics) and it also deals with the integration of wide range of experimental data for further extensive analyses. In this review, the overall strategy to toxicoinformatics is discussed.

시너(thinner)의 물리화학적 특성과 랫트의 생식기에 미치는 영향 연구 (A Study on thinner's Physicochemical property and its effect on genital organ of rat)

  • The aim of this study is to confirm the physicochemical property and hazard of thinner (012), which is a diluent of enamel paint used for floor coating for waterproofing and oil painting for the outer wall. The literatures of physicochemical property and hazard of thinner were surveyed and its physicochemical property were evaluated. And then, the inhalation toxicity of thinner affecting the central nervous system and reproductive organs in rats were examined by subchronic (6 h./day. 5 days/ week for 13 weeks) inhalation test. 1) According to the 13-week subchronic inhalation test, there were no significant changes in clinical test and body weight. However, a significant evidence of toxicity was observed in the hematological test and organ weight such as heart, kidney, liver and brain (p<0.01) in the 200 ppm and 1,000 ppm exposure groups in a dose response manner. In the histopathology analysis, there were no significant evidence of toxicity. Therefore, thinner was not classified as an organ targeted toxic agent. In case of Harmfulness, it could be classified as a chronic toxic agent 3($500 ppm/4hr, rat). 2) The reproductive toxicity such as extension of the period of estrous cycle, reduction of serum estradiol concentration and increase of frequency of the abnormal sperm was observed in the 1,000 ppm exposed animals. 3) The result of the physicochemical property of the test material showed that the specific gravity was 0.793, boiling point $155.8^{\circ}C$, steam pressure 2.1 kPa, ignition point $34.5^{\circ}C$, and spontaneous ignition point $280^{\circ}C$. The endothermic and exothermic values were 371.4 J/g and 159.1 J/g. respectively. The explosion limit was 214 mg/l. These data showed that thinner could be classified as an explosion agent level 1.2 and ignitive liquid agent 3 ($23-60^{\circ}C$) according to the notification No. 2008-1 of the Labor Ministry, "Classifying Standard of Chemical Materials."

Assessment of quantitative structure-activity relationship of toxicity prediction models for Korean chemical substance control legislation

  • Objectives For successful adoption of legislation controlling registration and assessment of chemical substances, it is important to obtain sufficient toxicological experimental evidence and other related information. It is also essential to obtain a sufficient number of predicted risk and toxicity results. Particularly, methods used in predicting toxicities of chemical substances during acquisition of required data, ultimately become an economic method for future dealings with new substances. Although the need for such methods is gradually increasing, the-required information about reliability and applicability range has not been systematically provided. Methods There are various representative environmental and human toxicity models based on quantitative structure-activity relationships (QSAR). Here, we secured the 10 representative QSAR-based prediction models and its information that can make predictions about substances that are expected to be regulated. We used models that predict and confirm usability of the information expected to be collected and submitted according to the legislation. After collecting and evaluating each predictive model and relevant data, we prepared methods quantifying the scientific validity and reliability, which are essential conditions for using predictive models. Results We calculated predicted values for the models. Furthermore, we deduced and compared adequacies of the models using the Alternative non-testing method assessed for Registration, Evaluation, Authorization, and Restriction of Chemicals Substances scoring system, and deduced the applicability domains for each model. Additionally, we calculated and compared inclusion rates of substances expected to be regulated, to confirm the applicability. Conclusions We evaluated and compared the data, adequacy, and applicability of our selected QSAR-based toxicity prediction models, and included them in a database. Based on this data, we aimed to construct a system that can be used with predicted toxicity results. Furthermore, by presenting the suitability of individual predicted results, we aimed to provide a foundation that could be used in actual assessments and regulations.

염색폐수의 수질독성시험을 이용한 한국의 수질배출허용기준 평가연구 (Assessment of Korean Water Quality Standards for Effluent Discharged from the Dye Industry Based on Acute Aquatic Toxicity Tests Using Microbes and Macroinvertebrates)

  • Acute aquatic toxicity of effluents discharged from five dyeing plants in Gyeong-gi province were evaluated to assess whether the current Korean water quality standards(KWQS) could protect aquatic life. Chemical analyses of all parameters regulated under KWQS, except for E-coli, were also carried out to determine regulation compliance of the samples. All the effluent samples were satisfied with KWQS except for the color in only one sample. In acute Daphnia magna toxicity tests, significant mortality was observed in one of five samples and EC50 was 12.1%(95% confidence interval 9.1-16.2), which was in compliance with KWQS. The result of the Microtox assay indicated that acute microbial toxicity existed in effluents from three out of five plants, two of which were in compliance with KWQS. The agreement between regulation compliance of chemical concentrations of effluent and observed toxicity from various biological toxicity tests was very poor to fair (kappa = 0.194~0.250). The data presented suggest that exposure to dyeing wastewater which were in compliance with Korean water quality standards may not be safe to aquatic biota, and multiple tropical levels should be considered in aquatic toxicity monitoring of dyeing industry.

다양한 시험생물종을 이용한 산업폐수 생태독성 평가 및 원인물질 탐색 (Ecotoxicity Test of Wastewater by a Battery of Bioassay and Toxicity Identification Evaluation)

  • Toxicity identification and quantification are important factors to evaluate the effect of industrial effluent on the aquatic environment. In order to measure the potential and real toxicity of mixed chemicals in the effluents, the biological method (i.e., WET test) should be used as well as chemical analysis method. In this study, we conducted WET test for various kinds of industrial effluents using aquatic organisms such as water flea (Daphnia magna), algae (Pseudokirchneriella subcapitata), fish (Oryzias latipes, Danio rerio), and microorganism (Vibrio fisheri). In addition, we carried out chemical analysis and TIE (Toxicity Identification Evaluation) for effluents in order to identify the substances causing toxicity. Among the 30 kinds of wastewater, S13 showed the highest eco-toxicity and $Ca^{2+}$ and $Cl^-$ ion were suspected as major compounds causing toxicity for aquatic organisms. In order to confirm these suspected compounds, various confirmation procedures need to be carried out.
