GENETIC POLYMORPHISMS OF THE GLUTATHIONE S-TRANSFERASE AND CYP1A1 GENES IN KOREAN ORAL SQUAMOUS CELL CARCINOMA (한국인 구강 편평세포암에서 Glutathione S-transferase와 CYP1A1 유전자의 다형성)

  • Many chemical compopunds are converted into reactive electrophilic metabolites by the oxidative(Phase I) enzymes, which are mainly cytochrome P-450 enzyme(CYPs). Phase II conjugating enzymes, such as glutathione S-transferase(GST), usually act as inactivation of enzymes. Genetic polymorphisms have been found to be associated with increased susceptibility to cancer of the lung, bladder, breast and colorectal. Many of the polymorphic genes of carcinogen metabolism show considerably different type of cancer among different ethnic groups as well as individuals within the same group. The aim of this study is (1) to establish the frequencies of genetic polymorphisms of GSTM1 and CYP1A1 in Korean oral squamous cell carcinoma(SCC), (2) to associate oral SCC with the risk of these genetic polymorphisms. The genetic polymorphisms of the GSTM1 and the CYP1A1 genes among 50 Korean oral SCC were analyzed using polymerase chain reaction(PCR). The results suggest that the homozygote and the mutant type of CYP1A1 MspI polymorphisms may be associated with genetic susceptibility to oral SCC in Korean. A combination of the GSTM1 null type with the homozygote(m1/m1), and the mutant(m2/m2) type of CYP1A1 MspI polymorphisms showed a relatively high risk of oral SCC in Korean. In the smoking group, the GSTM1 wild genotype may be the high risk factor of oral SCC in Korean. These data coincide with the hypothesis which states that different susceptibility to cancer of genetic polymorphisms exist among different ethnic group and different types of human cancer.

  • Biofilm has been described as a barrier, which produced by microorganisms to survive and protect themselves against various environments, like antibiotic agents. Staphylococcus spp. is a common cause of nosocomial and environmental infection. Thirty-six and thirty-five Staphylococci were isolated from animals and air, respectively. Based on the biofilm forming ability of the bacterium reported in our previous report, relationship between biofilm formation and antibiotic-resistance was investigated in this study. Regarding antibiotics susceptibility, cefazolin was the most effective agent to the bacteria. Strong biofilm-forming Staphylococcus spp. isolates might have a higher antibiotic resistance than weak biofilm isolates regardless of the presence of antibiotic resistance genes (p < 0.05). This result suggested that the chemical complexity of the biofilm might increase the antibiotic resistance due to the decrease of antibiotic diffusion into cells through the extensive matrix.

Experimental Studies on Comparison of Stress Corrosion Cracking Generation Due to Pipe Material Degradation in the Primary Stage of the Nuclear Power Plant (원전 1차 측 배관재질의 열화에 따른 응력부식균열 발생 비교 실험 연구)

  • In this report, stress corrosion cracking generation due to pipe material degradation in the primary stage of the nuclear power plant was investigated. Firstly, after artificially degrading the CF8A steel during 2, 4, and 6 months in actual temperature, $400^{\circ}C,$ assessed corrosion susceptibility of the degraded material following ASTM G5 standard. And next, the S.C.C. tests for the degraded material were conducted under the condition of $60^{\circ}C,$ 2wt.% H2BO3+Li70H solution, 0.8 oy. From the results, Corrosion rates linearly increased with degradation period and solution temperature increase. And both the raw material and the degraded materials were not failed in the S.C.C. test condition. In spite of long time test (about 3,900 hrs) under S.C.C. condition, surface pits or surface corrosion by the electro chemical reaction were not observed. And also, even though the nondestructive DCPD and ACPD methods were applied to on-line monitor the S.C.C. failure processes it was impossible because the surface pits and cracks were not generated.

Association of Cyclin D1 Variants with Head and Neck Cancer Susceptibility: Evidence from a Meta-analysis

  • Whether cyclin D1 (CCND1) gene variants increase susceptibility to head and neck cancer (HNC) is undetermined. Therefore, we performed the present meta-analysis to systematically assess any possible association between CCND1 variants (G870A and G1722C) and HNC risk. Seventeen studies for CCND1 G870A and three studies for CCND1 G1722C were included. Overall, CCND1 polymorphisms (G870A and G1722C) had no association with increased HNC risk (p>0.05). In the subgroup analysis by smoking status, significantly increased HNC risk was found among smokers under allele contrast, homozygous comparison and recessive models (p<0.05), smoking carriers of A allele and AA genotype appearing at elevated risk. In conclusion, while there was overall a lack of any association between CCND1 polymorphisms (G870A and G1722C) and HNC risk, smokers carrying the A allele and AA genotype of the CCND1 G870A polymorphism may be susceptible to HNC development.

Rapid Detection of Methicillin Resistant Staphylococcus aureus Based on Surface Enhanced Raman Scattering

  • Methicillin-resistant Staphylococcus aureus (MRSA) is one of the severe nosocomial infectious agents. The traditional diagnostic methods including biochemical test, antibiotic susceptibility test and PCR amplification are time consuming and require much work. The Surface enhanced Raman spectroscopy (SERS) biosensor is a rapid and powerful tool for analyzing the chemical composition within a single living cell. To identify the biochemical and genetic characterization of clinical MRSA, all isolates from patients were performed with VITEK2 gram positive (GP) bacterial identification and Antibiotic Susceptibility Testing (AST). Virulence genes of MRSA also were identified by DNA based PCR using specific primers. All isolates, which were placed on a gold coated nanochip, were analyzed by a confocal Raman microscopy system. All isolates were identified as S. aureus by biochemical tests. MRSA, which exhibited antibiotic resistance, demonstrated to be positive gene expression of both femA and mecA. Furthermore, Raman shift of S. aureus and MRSA (n=20) was perfectly distinguished by a confocal Raman microscopy system. This novel technique explained that a SERS based confocal Raman microscopy system can selectively isolate MRSA from non-MRSA. The study recommends the SERS technique as a rapid and sensitive method to detect antibiotic resistant S. aureus in a single cell level.

Application Evaluation of Asphalt mixtures using SDAR (Solvent DeAsphaltene Residue) (SDAR을 이용한 아스팔트 혼합물의 적용성 평가)

  • PURPOSES : The objective of this study is to evaluate the SDAR (solvent deasphaltene residue), which is obtained from the solvent deasphalting (SDA) process, as a pavement material. METHODS : The physical properties of the SDAR were evaluated based on its chemical composition, and asphalt mixtures with the SDAR were fabricated and used for the evaluation of mechanical properties. Firstly, the chemical composition of SARA (saturate, aromatic, resin and asphaltene) was analyzed using the TLC-FID (thin-layer chromatography-flame ionization detector). Moreover, the basic material properties of the asphalt binder with the SDAR were evaluated by the penetration test, softening point test, ductility test, and PG (performance grade) grade test. The rheological properties of the asphalt binder with the SDAR were evaluated by the dynamic shear modulus ($G^*$) obtained using the time-temperature superposition (TTS) principle. Secondly, the mechanical properties of the asphalt mixtures with the SDAR were evaluated. The compactibility was evaluated using the gyratory compacter. Moreover, the tensile strength ratio (TSR) was used for evaluating the moisture susceptibility of the asphalt mixtures (i.e., susceptibility to pothole damage). The dynamic modulus $E^*$, which is a fundamental property of the asphalt mixture, obtained at different temperatures and loading cycles, was used to evaluate the mechanical properties of the asphalt mixtures. RESULTS AND CONCLUSION : The SDAR shows stiffer and more brittle behavior than the conventional asphalt binder. As the application of the SDAR directly in the field may cause early failures, such as cracks on pavements, it should be applied with modifiers that can favorably modify the brittleness property of the SDAR. Therefore, if appropriate additives are applied on the SDAR, it can be used as a pavement material because of its low cost and strong resistance to rutting.

High $T_c$ Superconductors $LnBa_2Cu_3O_{7-\delta}$ from 2-Ethylhexanoate Precursors (2-Ethylhexanoate 화합물에서 제조한 고온초전도체 $LnBa_2Cu_3O_{7-\delta}$)

  • 2-Ethylhexanoate compounds which contained Y, Er, Yb, Ba, and Cu were prepared separately. These metallo-organic compounds were dissolved together in the common solvent and the solution was homogeneous. From these solution, high $T_c$ superconductors $YBa_2Cu_3O_{7-\delta}(Y_{123})$, $ErBa_2Cu_3O_{7-\delta}(Er_{123})$, and $YbBa_2Cu_3O_{7-\delta}(Yb_{123})$ were prepared. XRD analysis showed that these superconductors were not single phase and contained some impurity phases including $YBa_2Cu_4O_8(Y_{124})$. This was also confirmed from the magnetic susceptibility measurement. Generally, critical temperatures were higher than that of the superconductors prepared by ceramic method. Volume diamagnetic susceptibility showed little variation compared to the specimen made by ceramic method.

Synthesis, Physico-Chemical and Biological Properties of Complexes of Cobalt(II) Derived from Hydrazones of Isonicotinic Acid Hydrazide (Isonicotinic Acid Hydrazide의 Hydrazone으로부터 유도된 코발트(II) 착물의 합성, 물리-화학 및 생물학적 성질)

  • Hydrazones of isonicotinic acid hydrazide, viz., N-isonicotinamido-furfuralaldimine (INH-FFL), N-isonicotnamido-cinnamalidine (INH-CIN) and N-isonicotnamido-3',4',5'-trimethoxybenzaldimine (INH-TMB) were prepared by reacting isonicotinic acid hydrazide with respective aromatic aldehydes, i.e., furfural, cinnamaldehyde or 3,4,5-trimethoxy-benzaldehyde. A new series of fifteen complexes of cobalt(II) with these new hydrazones, INH-FFL, INH-CIN and INH-TMB, were synthesized by their reaction with cobalt(II) salts. The infrared spectral data reveal that hydrazone ligands behave as a bidentate ligand with N, O donor sequence towards the $Co^{2+}$ ion. The complexes were characterized on the basis of elemental analysis, magnetic susceptibility, conductivity, infrared and electronic spectral measurements. Analytical data reveal that the complexes have general composition [Co($L)_2X_2]\;and\;[Co(L)_3](ClO_4)_2$ where L= INH-FFL, INH-CIN or INH-TMB and X = $Cl^-,{NO_3}-,\;NCS^-\;or\;CH_3COO^-.$ The thermal behaviour of the complexes were studied using thermogravimetrictechnique. Electronic spectral results and magnetic susceptibility measurements are consistent with the adoption of a six-coordinate geometry for the cobalt(II) chelates. The antimicrobial properties of cobalt(II) complexes and few standard drugs have revealed that the complexes have very moderate antibacterial activities.

Syntheses, Spectral, Surface Morphological and Gamma Ray Irradiation Studies of Some Oxomolybdenum(V) and Dioxomolybdenum(VI) Complexes of an Azo Dye Derived from 4-aminoantipyrine

  • Syntheses of some novel oxomolybdenum(V) and dioxomolybdenum(VI) complexes with an azo dye methoxyphenolazoantipyrine (HL) derived from 4-aminoantipyrine and 2-methoxyphenol are reported. The complexes have been characterized by elemental analyses, molar conductance, magnetic susceptibility data, IR, UV-Vis, $^1H$ NMR, EPR and FAB mass spectral studies. The physicochemical studies and spectral data indicate that HL acts as a bidentate chelating ligand. The complexes have the general formulae [$MoO(HL)XCl_2$] and [$MoO_2(HL)XCl$],where X=Cl, NCS or $NO_3$. All the complexes are found to have distorted octahedral geometry. Structural and morphological characterization of the complexes [$MoO(HL)Cl_3$](1) and [$MoO_2(HL)Cl_2$](4) before and after gamma ray irradiation,was performed by X-ray diffraction and scanning electron microscopy( SEM).The ligand and the complexes were screened for their possible antimicrobial activities.

Effect of chemical composition on the weldability of quenched and tempered high strength steels (주질고장력강의 용접성에 미치는 화학조성의 영향)

  • In fabrication of various welded structures made of high strength steels, the occurence of hydrogen assisted cracking and embrittlement in HAZ is prime importance. The present work was carried out to clarify the effect of chemical compositions, especially B and/or Ti addition on the cold cracking susceptibility and HAZ embrittlement in low crabon equivalent steel. Tests results showed that the addtio of optimum boron content in steel with low Pem value i.e., below 0.20 % was the best way to improve the weldability as well as the mechanicla properties of $60kg/mm^2$ grade quenched and tempered high strength steels.
