  • The pattern of sensory papillae, chaetotaxy, of the cercaria of Clenorchis sinensis was observed, The chaetotaxy was as follows; 5~6 Ci 1, 4~5 Ci 2, 5, ~6 Ci3 at 1st row, 4 Cii 1, 2 Cii2, 4 Cii3, 5~6 Cii4 at 2nd row, 3~4 Ciii 1, 2~3 Ciii 2 at 3rd row, and 2 Civl, 2~3civ2, 2~3 Civ 3, at 4th row, in cephalic region; 2 AiV, 1 AiD, 2 AiiV, 1 AiiD, 2 Aiiiv, 2 AiiiD, 1 AivV, 1 AivD, 1 PiiD, 1 PiiiD, in ventral(V) and dorsal(D) portions of body. Caudal region revealed 2-2-2-2 formula.

Studies on the Life History of Acanthoparyphium tyosenense Yamaguti, 1939 (Acanthoparyphium tyosenense Yamaguti, 1939의 생활사에 관한 연구)

  • Cercaria yamagutii Ito, 1957 (C. yamagutii) was found in Lunatia fortuni (L. fortuni) and Neverita didyma (N. didyma) collected from the tideland of Sim-po located at the estuary of the Mankyong River, Chonbuk. It was finally confirmed that the parasite is Acanthoparyphium tyosenense Yamaguti, 1939 (A. tyosenense) and its life history was clarified in this study. Mactra veneriformis (M. veneriformis) was artificially infected with C. yamagutii isolated from L. fortuni and N. didyma. It began to intrude into M. veneriformis through the inhalent canal. Five hours after infection, the tails of the cercaria began to be separated from the main body and the cercaria started to form cysts. Mature cysts were formed 340 hours (14 days) after infection. The cysts were $300\sim360{\mu}m$ in diameter and the encysted metacercarias were $790\sim800\times300\sim310{\mu}m$ in size. The metacercarias were administered orally to Larus crassiostris (L. crassiostris), and adult worms of $84.5\sim112.5\times55\sim65{\mu}m$ were found full of eggs with $2.20\~3.70$ mm long and $0.40\~0.59$ mm wide after 10 days. In a field study, it was observed that the infection rate of A. tyosenense is $99.5\%$ in M. venerifomis, $76.3\%$ Solen strictus (S. strictus), and $37\%$ Ruditapes philippinarum (R. philippinarum), No difference was found among different host sizes, It was concluded that the first intermediate hosts of A. tyosenense Yamaguti were L. fortuni, N. didyma, Tympanotonus microptera, Cerithidea (Cerithidea) largillierti, Cerithidea (Cerideopsilloa) cingulata, the second intermediate hosts M. venerifomis, S. strictus and R. philippinamn, and the final hosts L. crassiostris and Melanitta fusca stejnegeri.

Anthelmintic Natural Products against Clonorchis sinensis and the Analogues (간디스토마에 작용하는 천연물 및 그 유사체)

  • Among 230 species of herbal drugs screened, thirty one show the anthelmintic activities in vitro against Clonorchis sinensis, the chinese liver fluke. The active substances have been isolated and their structures identified. Some of the active substances and their derivatives have been synthesized, followed by anthelmintic activity tests. The extracts from the active drugs cause damages in organs of the adult worm in the hepatic duct of rabbit. Some fresh water fish, which function as bad hosts for the fluke, excrete defense substances against the cercaria of the fluke. The defense substances have been isolated from Cyprinus carpio and Cyprinus carpio nudus, followed by structural identification. The results are summarized as follows: l) The bark of Machilus thunbergii as well as the seed of Schizandra chinensis contain meso-dihydro-guaiaretic acid as the anthelmintic component. Among derivatives synthesized, 4-phenyl-1-((3, 4-dihydroxyphenyl)-, 4-phenyl-1-(3-hydroxy-4-methoxy phenyl)-and 4-phenyl-1-(4-hydroxy-3-methoxy phenyl)-2, 3-dimethyl butanes show considerable activities. Administration of the bark extract mainly damages the bladder of the adult worm. 2) The active substance from the roasted fruit of Prunus mume is 2-hydroxymethylfurfural. This substance is produced during the roasting process. Administration of the fruitextract causes a damage of the bladder of the adult worm. 3) The active substance from the root of Scutellaria baicalensis is 5.2'-dihydroxy-6, 7, 8, 6'-tetramethoxyflavone. 4) Beside alantolactone, a very strong anthelmintic component is contained in the root of Inula helenium. Administration of the root extract causes irreversible damage on the worm, affecting mainly the reproductive organs. 5) The cercaricidal substances from the epidermis of C. carpio and C. carpio nudus are ethyl linoleate and linoleic acid, respertively. 6) The cercaricidal substances from various kinds of fresh water fish have different $R_f$ values, implying that the defense substances are species-specific. Unexpectedly, the fish with good host function, for example Pseudorasbora parva, excrete the defense substances, too. The defense substances are possibly organ-specific in individual species; the organs essenstial for the existence of the species excrete the defense substances, allowing other parts to be invaded by the cercaria. 7) The cercaricidal fraction of Carassius carassius is detected only in the fish which have been collected during the summer time from May to September, Its secretion is not dependent on water temperature. Thus, it seems to be possible that the secretion of the defense substance would be stimulated through a contact between the fish and cercaria.

  • In order to judge appropriately the suitability as the second intermediate host of Clonorchis sinensis, the present experiments were proposed to survey the metacercaria of C. sinensis in the fish host, Cuztriculus eigenmanni, in the 6eld, and to observe, by experimental infection of the fish with Clonorchis cercaria, the penetrating ability of the cercaria, maturity, the process of degeneration and extinction, and infectivity of the metacercaria. The following is a brie summary of the leading facts gained through the experiments. 1. All the ten fish (mean body weight $13.4{\pm}3.439$) which were caught in Ok-ku irrigation reservoir were in acted with the metacercariae and the average number of the metacercariae found in the fish body/gram was 4.55. 2. The Bish was easily subject to invasion of the cercaria, and the examination of the cercariae after having the fish infected disclosed that 5:l hours after the infection, all of the cercariae formed their cysts in muscle and the metacercariae kept growing, that 7 days later the metacercariae were found folding their bodies twice, and that in 15 days the characteristic structure of the metacercariae was complete and made a vigorous rotary movement intermittently. 3. In the case of control fish, Pseudorasbora parka, both penetrating ability of the cercariae and the infective degree of the metacercariae to control host were stronger as compared with the 5sh host because that the control fish had higher suceptibility to C. sinensis. 4. Afterwards, the metacercariae in the fish host came to a state of maturity and beyond this stage some metacercariae started the Process of degeneration and extinction in as early as 30 days, and as more days elapsed, mortality of the metacercariae was not uniformly increased but difference of mortality appeared to be according to the individual of Ssh host. That is, in 390, 540 and 586 days, all of the metacercariae in the fish hosts became dead, meanwhile almost all of those found in the fish host of 596 days after the infection survived (mortality; 8, 51%) , Percentage of orally administered metacercariae developing to maturity in rabbit is 10.465 in the last case. 5. As the days were progressed the shapes of the metacercariae in the fish host tended to change from ellipsoid to almost round shapes, the length became shorter while the width was wider. The results shown above revealed that C. eigenmanni was, though it could not be proper host than p. larva. suitable as the second intermediate host in addition to our previous observation results.

  • The life history of Echinostoma cinetorchis was completed in the laboratory using Hippeutis cantors as the first and second intermediate host. The incubation for maturation of eggs and hatching of miracidia took 24 days at 28℃. On the 66th day after miracidial challenge, 16 snails were crushed and examined for the presence of E. cinetorchis larvae. The metacercariae were detected in all of the snails, and from three of them were found the rediae and cercariae. The morphological characteristics of the larvae and the experimentally obtained adults were compatible with that of E. cinetorchis.

  • A number of studies on the papillae of cercariae of trematodes reported that the papillar patterns (or chaetotaxy) of cercariae might be an excellent method to attain better understanding of the digenetic trematodes (Richard, 1971 ; Short and Cartrett, 1973; Bayssade-Dufour, 1979) . The present study was aimed to determine the number, distribution pattern and structure of the sensory papillae of Metagonimus yokogawai cercariae, and to elucidate the chaetotaxy of this digenetic trematode. M. yokogawai cercariae were pipetted from a vial in which infected snails (Semisulcospira libertina) had been kept for 3 hours. The snails were collected from an endemic area of M. yokogawai, Boseong river in west-southern part of Korea. Observations of papillae were based on light microscopy of those stained with silver nitrate, and on scanning electron microscopy The results are summarized as follows: 1, All papillae observed were uniciliated. 2. Cilia in anterior tip were shorter than the others in other portions. 3. The body papillae were arranged in essentially symmetrical patterns, Total number of the papillae was 126(63 pairs) in average; anterior tip 40(20 pairs), ventral 20(10 pairs), lateral 42(21 pairs), and caudal 8(4 pairs). 4. Pathology Survey of the Manila Clam Ruditapes philippinarum from Hwangdo Tidal Flat in Cheonsu Bay on the West Coast of Korea

  • Manila clam Ruditapes philippinarum is present at high rates of density in tidal flats in Cheonsu Bay on the west coast of Korea, where clams often exhibit mass mortalities in late summer. We monitored the pathologic condition of clams at Hwangdo tidal flat (HD) to understand the parasitic impacts on clam fitness. Manila clams were fully ripe in July and spawned during August and September, as the histology indicated. The histology revealed that clams in HD tidal flats were heavily infected by the protozoa parasite Perkinsus olseni, as the monthly prevalence ranged from 53% (September) to 93% (August). In addition, Manila clams were co-infected by the metazoan parasite Cercaria tapetis and Parvatrema duboisi with the prevalence of 0-33% and 0-14%, respectively. Massive hemocyte infiltration and subsequent inflammation were commonly observed from the gills of P. olseni infected clams. Clusters of P. olseni trophozoites and heavy hemocyte infiltration were also observed from the female gonad, suggesting that P. olseni interferes with host gonad maturation. The larval trematode occupied almost the entire host gonad, resulting in gonad castration. In addition, Metacercaria of P. duboisi were observed from the subsurface of the mantle. Ray's fluid thioglycollate medium assay (RFTM) indicated that clams collected in August and September contained approximately 4.0×106 P. olseni cells/g gills. Condition Index (CI) declined gradually from spring to early summer, and the decline in CI was interpreted as a consequence of the heavy parasitism, as the parasites drain the host's net energy to be used in somatic growth and gamete production.

  • As a series of studios on the suitability as a second intermediate host of Clcnorchis sinensis, the wormicidal effects of mucus from epidermis of Cyprinus carpio nudus were observed. The results obtained were sumlnarized as follows : On the silica gel thin layer chromatography with the greenisll yellow supernatant in acetone: benzene110:90 as carrier, seven spots were observed in iodine chamber. The spot of Rf. 0.225 value among them disclosed the strongest wormicidal effect on the cercaria, the encysted metacercaria, and the adult of C. sinensis. hfeanwhile, no seasonal variations were found in Rf. values and effect of the wormicidal fractions. The wormicidal materials in epidermal mucus of C. carpio nudes and Carassius carassius were compared in their effect, and the former was Inore effective killing substance than the latter. It seemed that C. carpio nodus could not be proper intermediate host of C. sinensis according to results of the present study, in addition to our previous observation results.
