A New Selection System for Pepper Regeneration by Mannose

  • We report the development of a new selection system for the transformation of pepper plants by mannose. In order to achieve this, we first tested several factors related to regeneration conditions. Among the 30 inbred lines examined, line P9l5 was able to generate shoots at the highest rate from both cotyledons and hyporotyls in MS media. A dosage curve for optimizing the selection conditions was established by mixing mannose (range 0-50 g/L) and sucrose (range 0-30 g/L). The least selection pressure on shoot formation was created by a mixture of sucrose and mannose at 20 g/L and 15 g/L, respectively, and the threshold for ultimate tissue death was 50 g/L of mannose irrespective of the sucrose concentration. However, we found that mannose itself was not the sole inhibitor of pepper shoot development. High concentrations of sucrose (30 g/L) contributed additively to the inhibition of shoot formation at higher mannose concentrations. Genotype preference is a major factor that enhances regeneration ability in mannose media, as was observed in MS media. P9l5 and P410 line had high regeneration rates under mannose selection conditions in the presence of Agrobacterium infection. Different virulence levels of Agrobacterium strains did change the regeneration rates, probably due to interaction with the specificities of the inbred lines. Taken together, P9l5 offers the best pepper inbred line for transformation and we recommend a selection condition of 20 g/L of sucrose and 15 g/L or more of mannose up to 50 g/L in media.

  • This paper proposes an R&D project selection methodology for green technology centered on developing country-oriented technology commercialization. Eight selection criteria are derived from the R&BD logic model : technology needs of developing countries, effectiveness of green technology, technological potentials, domestic technological capability, commercialization feasibility, economic benefits, business feasibility, and spillover effects of developing countries. 21 qualitative and quantitative indicators are then defined for each criterion. The analytic hierarchy process is conducted to produce relative importance of evaluation indicators and to set final priority scores of R&D project candidates. The working of the proposed methodology is provided with the help of a case study example of Green Technology Center. The proposed methodology is expected to be effectively utilized for policy practices of R&D project selection in the field of green technology.

  • The human genome has evolved as a consequence of evolutionary forces, such as natural selection.   • This paper proposes a selection procedure of geospatial data for location guidance map generation system. The selection procedure requires some targets appointed by users as input data and outputs generation. The procedure is embedded in a prototype of object-oriented GIS. We show sample maps generated by the system.

The Relationships among Selection Attribute, Trust, Experiential Value, and Recommendation for Sport Center Consumers (스포츠센터 이용객들의 레스토랑선택속성이 신뢰, 경험가치, 그리고 추천의도에 미치는 영향)

  • This study was performed to verify the relationships among selection attribute, restaurant trust, experiential value, and recommendation focusing on sport center consumers. The data were collected from visitors who registered more than three months in the sport center in South Korea. Total 500 survey was distributed and 330 participants were used for further statistical analysis. SPSS 23.0 and AMOS 21.0 for Windows were used for statistical analysis. Five factors of selection attribute (menu, interior, exterior, staff, convenience) were extracted, and measured by using 15 questions. According to the results of this study, interior, exterior, and staff factors have positive effects on restaurant trust, and interior and menu were significant predictors of the experiential value. In addition, present study confirmed the theoretical relationship among trust, experiential value, and recommend intention as perceived by sport center visitors. Although there are many studies which demonstrated the relationships among selection attribute and other outcome variables, little research explained the relationships among the variables from sport center consumers. Therefore, this study will contribute to provide meaningful results and some practical implications for both academia and the related foodservice industry.

  • As port functions change to act as an economic catalyst and take on a central position in industries engaged in international trade, port hinterland has become a significant component in international shipping. The success of port hinterland as a strategic base of logistic activities critically depends on location selection factor for international distribution center that links elements of global supply chain management. By examining multi-measurement items empirically, this paper analyzed location selection factor for international logistics distribution center in port hinterland, and evaluated Busan new port hinterland from the user's perspective. Employing exploratory factor analysis, the results revealed that the model structured around five factors incorporating geo-location and accessibility, availability, political supports, cost factors, and quality of business environment is valid and reliable in the context of the location selection factors for logistics distribution center in the context of port hinterland operations. The evaluation of Busan new port hinterland provides useful insights for strategic improvement to accommodate the users' expectation. Further, the model offers both a descriptive and diagnostic strategic management tool for port hinterland development and operations, to guide future improvement.

Estimation of Genetic Improvement Based on the Breeding Values of Growth Traits of the Abalone Haliotis discus hannai (육종가 기반 북방전복(Haliotis discus hannai) 성장형질의 유전적 개량량 추정)

  • This study was conducted to estimate the genetic improvements by selection criteria using the genetic parameters and breeding values for population of abalone Haliotis discus hannai. Genetic parameters and breeding values were estimated using all measurement data of growth traits (shell length, shell width and total weight) at 18 and 30 months old after artificial fertilization for 3,029 individuals produced in April 2014. Growth traits all exhibited moderate heritability (0.253-0.354). So it is considered that family selection will be more advantageous than individual selection. It was found that a higher genetic improvement could be expected when selecting the top 10% based on the breeding values of total weight rather than other traits. In particular, a higher genetic improvement could be expected when selecting the top 10% at 30 months old than 18 months old after artificial fertilization. This seems to be because the selection differential and heritability were higher at the 30 months old. Therefore, by estimating genetic parameters and breeding values of a population for production of the next generations by stage of growth, if they are used properly in selection and mating according to the improvement direction, it is considered that more breeding effects can be expected.

  • A fully differential RC calibrator for accurate cut-off frequency of a programmable channel selection filter is proposed. The proposed RC calibrator consists of an RC timer, clock generator, synchronous counter, digital comparator, and control block. To verify the proposed RC calibrator, a six-order Chebyshev programmable low-pass filter with adjustable 3 dB cut-off frequency, which is controlled by the proposed RC calibrator, was implemented in a $0.18-{\mu}m$ CMOS technology. The channel selection filter with the proposed RC calibrator draws 1.8 mA from a 1.8 V supply voltage and the measured 3 dB cut-off frequencies of the channel selection LPF is controlled accurately by the RC calibrator.

  • Many Firms consider the application of a cross-docking system to reduce inventory and lead-time. However, most studies mainly concentrate on the design of a cross-docking system. This study presents the method that selects the cross-docking center under the existing logistics network. Describing the operation environment to apply the cross-docking system, the selection criteria of the cross-docking center, and the main constraints of transportation planning under the environment of multi-level logistics network, we define the selection problem of the cross-docking center applied to a logistics field. We also define the simulation model that can analyze variously the cross-docking volume and develop the selection methodology of the cross-docking center. The simulation model presents the algorithm and influence factors of the cross-docking system, the decision criteria of the system, policy parameter, and input data. In addition, this study analyzes the effect of increasing the number of simultaneous receiving and shipping docks, and the efficiency of the overnight transportation and cross-docking by evaluating each scenario after simulating the scenarios with the practical data of the logistics field.

Optimized Serological Isolation of Lung-Cancer-associated Antigens from a Yeast Surface-expressed cDNA Library

  • The technique of serological analysis of antigens by recombinant cDNA expression library (SEREX) uses autologous patient sera as a screening probe to isolate tumor-associated antigens for various tumor types. Isolation of tumor-associated antigens that are specifically reactive with patient sera, but not with normal sera, is important to avoid false-positive and autoimmunogenic antigens for the cancer immunotherapy. Here, we describe a selection methodology to isolate patient sera-specific antigens from a yeast surface-expressed cDNA library constructed from 15 patient lung tissues with non-small cell lung cancer (NSCLC). Several rounds of positive selection using patient sera alone as a screening probe isolated clones exhibiting comparable reactivity with both patient and normal sera. However, the combination of negative selection with allogeneic normal sera to remove antigens reactive with normal sera and subsequent positive selection with patient sera efficiently enriched patient sera-specific antigens. Using the selection methodology described here, we isolated 3 known and 5 unknown proteins, which have not been isolated previously, but and potentially associated with NSCLC.
