Growth and Nutrient Utilization in Buffalo Calves Fed Ammoniated Wheat Straw Supplemented with Sodium Sulphate

  • Ten male buffalo calves (aged 6-8 months, average body wt. $88.5{\pm}0.5kg$) were divided into two groups of five animals in each. All the animals were fed on urea-ammoniated wheat straw (4% urea, 50% moisture) along with concentrate mixture (50:50 on DM basis). In addition animals in group II were given sodium sulphate to see the effect of sulphur on the utilization of nitrogen added through urea-ammoniation. This feeding practice continued for a period of 120 d, during which fortnightly body weights were taken to assess their growth rate. A metabolism trial was conducted after 90 days of feeding to know the digestibility of nutrients and their balance. Results revealed no significant difference in the intake of DM and other nutrients in two groups. The digestibility of DM, OM, EE, NDF, ADF and cellulose was alike in animals fed ammoniated straw and ammoniated straw+sodium sulphate supplemented group, whereas the digestibilities of CP and hemicellulose was significantly (p<0.01) more in group I and II respectively. There was no significant difference in intake of nitrogen, calcium and phosphorus in 2 groups. Similarly, the balance of these 3 nutrients was positive and statistically alike in two groups. Intake and excretion of sulphur through faeces and urine was significantly (p<0.01) more in group II than in group I. Inspite of higher excretion of sulphur through faeces and urine in group II, the sulphur balance was significantly (p<0.05) more in group II than in group I, probably due to significantly (p<0.01) higher intake of sulphur in this group. There was no significant difference in total body weight gain or average daily gain between two groups, indicating that addition of sodium sulphate did not have any positive effect on these parameters. Similarly the intake of DM, DCP and TDN were also alike in two groups. The DCP and TDN values of the two diets were 8.0, 60.4 and 6.8, 56.6% respectively. Feeding cost/unit gain was alike in both the groups.

Thermotropic Liquid Crystalline and Photochemical Phase Transition Behavior of Octa[8-{4-(4'-cyanophenylazo)phenoxy}]octyl and Octa[8-{4-(4'-cyanophenylazo) phenoxycarbonyl}]heptanoated Disaccharides (옥타[8-{4-(4'-시아노페닐아조)펜옥시}]옥틸 그리고 옥타[8-{4-(4'-시아노페닐아조) 펜옥시카보닐}]헵타노화 이당류의 열방성 액정과 광화학적 상전이 거동)

  • Octa[8-{4-(4'-cyanophenylazo)phenoxy}]octyl and octa[8-{4-(4'-cyanophenylazo)phenoxycarbonyl}]heptanoated disaccharide derivatives were synthesized by reacting cellobiose, maltose, and lactose with 1-{4-(4'-cyanophenylazo) phenoxy}octylbromide or 1-{4-(4'-cyanophenylazo)phenoxycarbonyl}]heptanoyl chloride, and their thermotropic liquid crystalline and photochemical phase transition behavior were investigated. All the {(cyanophenylazo)phenoxy} octyl disaccharide ethers (CADETs) formed monotropic nematic (N) phases, whereas all the {(cyanophenylazo) phenoxycarbonyl}heptanoated disaccharide esters (CADESs) exhibited enantiotropic N phases. Compared with CADETs, CADESs showed higher isotropic (I)-to-N phase transition temperatures. Photoirradiation of the disaccharide derivatives in a glass cell or in a cell with a rubbed polyimide (PI) alignment layer at a N phase resulted in disappearance of the N phase due to trans-cis photoisomerization of azobenzene, and the initial N phase was recovered when the irradiated sample was kept in the dark because of cis-trans thermal isomerization and reorientation of trans-azobenzenes. The rates of the photochemical N-I and the thermal I-N phase transition of disaccharide derivatives in a cell with a rubbed PI alignment layer were faster than those in a glass cell, and were significantly different from those observed for the monomesogenic compounds containing cyanoazobenzene and the 4-{4'-(cyanophenylazo)phenoxy}octyl glucose and cellulose ethers. The results were discussed in terms of difference in cooperative motion of azobenzene groups due to the flexibility of the main chain, the number of mesogenic units per repeating units, and the distance between the azobenzene groups.

New High Recovery Membrane Modules for Desalination

  • Desalination by reverse osmosis (RO), which first entered commercial use in the 1970s, was initially mainly used for treating brackish water. Technological progress led to the development of an RO membrane enabling single-pass seawater desalination. Toyobo succeeded in developing a single-pass seawater desalination RO module composed of hollow fiber type membranes made of cellulose triacetate in 1978, and then in 1979 began production of the first commercially available double-element module. This double-element module has many advantages suitable for seawater desalination. It has high chlorine tolerance and high salt rejection, derived from the properties of the membrane material, and it is highly resistant to fouling and scaling matters due to the unique flow pattern and fiber bundle configuration. These advantages help to explain why the Toyobo double-element module has been used so successfully at the many seawater desalination plants around the world. Since the 1980s, large plants capable of desalinating several tens of thousands of cubic meters a day have sprung up around the Mediterranean and In the Middle East. The Jeddah RO Phase I Plant, which has a capacity of 56, 800m$^3$/day, went into operation in 1989. In 1994, the same sized Phase II Plant came on stream, giving the plant a huge total capacity of 113, 600m$^3$/day. The plant constructor Mitsubishi Heavy Industries, Ltd. (MHI), and the RO membrane manufacturer Toyobo Co., Ltd. In 1998, the world's largest RO seawater desalination plant in operation, which has a capacity of 128, 000m$^3$/day and is run by Saudi Arabia's Saline Water Conversion Corporation (SWCC), went into operation at Yanbu. RO seawater desalination technology has thus already reached the stage of full-scale commercial use. In order to encourage its wider use, however, RO desalination needs to be made more economical by lowering construction and water treatment costs. Toyobo has therefore developed a new economical RO desalination system by a recovery ratio of 60% using a high-pressure module with a high product flow rate. In 2000, Toyobo high recovery membrane module was selected for the largest seawater desalination plant in Japan, which has a capacity of 50, 000m$^3$/day.

  • WeI have studied the process for synthesizing porous silica with a specific surface area of minimum $800m^2/g$ by adding surfactant [Poly Etylene Glycol(PEG) and Hydroxy Propyl Cellulose(HPC)] to the sol-gel reaction between sodium silicate and hydrochloric acid. NaCl, the by-product of the sol-gel reaction, was water cleaned and removed; when 200 ml of water was used to clean 50 g of silica gel, NaCl remaining in the silica gel was reduced to maximum 0.81wt%. The appropriate level of surfactant for silica gel synthesizing proved to be below 5%. As a result of the experiment, for the silica synthesized by adding surfactant of HPC(2.5%)+PEG(2.5%), the surfactant area was $860m^2/g$ and grain size was $20-50{\mu}m$. From this study, we have concluded that it is of industrial significance that specific surface area is improved and silica of a regular grain size is obtained just by adding surfactant in the gel process or drying process of silica.

  • 자원재활용과 원가절감를 위해 고지재활용율이 크게 상승하고 있으며, 이와 아울러 고 지원료로부터 고품질의 제품을 생산할 수 있는 기술에 대한 관심이 증대되고 있는 실 정이다. 저급 고지로부터 고품질의 탈묵펄프를 얻기 위해서는 고지재활용공정의 핵심 공정인 부유부상 공정의 효율화가 가장 중요하다고 믿어진다. 이를 위해서는 부유부상 공정에 가장 큰 영향을 미치는 고형입자의 표면 화학적 특성에 따른 공정의 분리효율 평가와 이에 근거한 공정해석과 개선이 요청된다. 이러한 부유부상 공정을 표면화학적 측면에서 구명하기 위해 마이크로 크리스탈린 셀룰로오스(Microcrystalline cellulose: M MCC)를 모댈 물질로 사용하여 표면화학적 특성은 다르나 입자의 크기와 형태는 동일 한 시료를 준비하였다. 친수성의 표면 특성을 나타내는 MCC의 표면 특성을 바꾸기 위 하여 AKD(alkyl ketene dimer)로 처리비율을 달리하여 솔벤트 사이징 처리를 실시하 였다. 이렇게 준비된 MCC의 표면화학적 특성을 IGC를 이용하여 평가하였다 .. IGC는 G GC를 응용한 표면분석 기술로 고체의 물리화학적 특성과 흡착성을 분석하기 위해서 사용되며 이로부터 흡착제의 표면특성을 평가할 수 있다. 본 실험에서는 AKD로 소수화 정도가 다르도록 소수화시킨 MCC 시료를 이용하여 I IGC 칼럼을 준비하고 n-알칸과 몇가지 극성 용매률 이용하여 이들의 칼럼 내 체류시 간을 측정함으로써 흡착특성을 평가하고 이로부터 흡착현상을 열역학적으로 분석하였다. IGC 분석 시에는 칼럼의 온도를 $30^{\circ}C$, $35^{\circ}C$, $40^{\circ}C$의 3수준으로 변화시켰다. 측정결 과로부터 MCC 표면의 흡착자유에너지와 엔탈피, 엔트로피의 변화량을 평가하였으며, 또한 MCC 표면의 극성에너지와 산염기적 성질을 평가하였다. 실험결과 MCC의 소수화도에 따른 열역학적 흡착현상의 차이가 명백하였다. 이는 소 수화 수준에 따라 소수성 및 친수성 물질의 흡착성이 변화된다는 것을 보여준다. 따라서 탈묵 시 진행되는 기포에의 부착현상을 평가할 수 있는 기초자료로 활용될 수 있다 고 사료되었다.

Characteristics of Biodegradation under Composting and Vermiculite Condition (폐 MDF 퇴비화 사용을 위한 생분해 환경 특성에 관한 연구 (I))

  • This study is performed that furniture and interior materials of MDF's (Medium Density Fiberboard) biodegradation properties, and the goal of this study is investigation of possibility of waste-MDF's composting after landfilling. To investigate biodegradation, this study was performed according to KS M ISO 14855-1, and there were two different soil conditions including a compost condition and an activated vermiculite condition as artificial soil. This experiment was tested for 40 days. The measurement of carbon-dioxide generation was processed every 24 hour in 1-2 week, and every 48 hour after 3 week. In the same days, MDF showed 24.4% of biodegradation in compost condition, and 6.2% in activated vermiculite. Also, the reference material of TLC (thin-layer chromatography) grade cellulose showed 26.4%, 11.4% in compost and activated vermiculite respectively. The dilution plate method was performed for biological analysis in the study. This experiment was used for investigation of inoculum's (Bacillus licheniformis) activity. As the result of bioassay, compost has more other germs include inoculum than activated vermiculite in the first week. Especially in the 2nd week, the reference material under the compost condition showed the most germ's activity, and also the biodegradation was the highest. Consequentially, compost condition was able to reduce a performing period of biodegradation testing than activated vermiculite. However, activated vermiculite could be stabilizing errors between repetition.

Purification and Properties of Pectate Lyase Produced by Recombinant Strain -Containing pelK Gene from Alkalitolerant Bacillus sp. YA- 14 (알칼리내성 Bacillus sp. YA-14 유래의 Pectate Lyase 유전자를 함유한 재조합균주로부터 효소의 정제 및 특성)

  • Pectate lyase produced by recombinant strain containing pectate lyase gene from alkalitolerant Bacillus sp. YA-14 was succesively purified with 257.6 purification folds and a 10.2% yields by the affinity method, eM-cellulose column chromatography followed by gel filtration on Sephadex G-I00 column. The optimal pH and temperature for pectate lyase activity were 10.0 and $60^{\circ}C$, respectively. The enzyme was stable between pH 4.0 and 10.0, and up to $50^{\circ}C$. The molecular weight of this enzyme was estimated to be 43,000 daltons by SDS-PAGE. Amino acid analysis showed that the enzyme contained more polar and basic amino acids, especially serine, glycine and tyrosine, than that of various pectate lyase from other strains. The N-terminal amino acid sequence was Ala-Asp-Leu-Gly-His-Gln-Thr.

Controlled Release of Three Nutrients from Dual-layered Coated Compact Pellets (이중 코팅된 압착 펠렛으로부터 3종 영양소의 방출 제어)

  • The purposes of this study were to prepare dual-layered coated compact pellets containing three nutrients Glucose, Chromium picolinate, Vitamin C) for rumen bypass. The core compact pellets were prepared by an extrusionspheronization method and then double layered coated with pH independent EC (ethyl cellulose) and pH-dependent polymers ($Eudragit^{(R)}$ E100) using a fluid-bed spray coater. Depending on the coating levels of EC and $Eudragit^{(R)}$ E100, release profiles were variable in simulated rumen (pH 6.8) and abomasums (pH 2.0) fluid using USP apparatus I (basket method). When compact pellets were coated with EC (about 10% level in inner layer) and then $Eudragit^{(R)}$ E100 (20% level in outer layer) in a dual-layered manner, rumen-bypass delivery resisting rumen fluid followed by release in abomasums fluid could possible. The friability was also satisfactory based on chewing behavior of ruminants. The dual-layered coated compact pellets showed smooth surface and distinct inner/outer layers using scanning electron microscopy (SEM). The current rumen bypass delivery system can be also applicable to deliver other nutrients in ruminants.

Study of Macrophage Activation and Structural Characteristics of Purified Polysaccharide from the Fruiting Body of Cordyceps militaris

  • Cordyceps militaris, an entomopathogenic fungus belonging to the class Ascomycetes, has been reported to have beneficial biological activities such as hypoglycemic, anti-inflammatory, antitumor, antimetastatic, hypolipidemic, immunomodulatory, and antioxidant effects. In this study, the crude water-soluble polysaccharide CMP, which was obtained from the fruiting body of C. militaris by hot water extraction and ethanol precipitation, was fractionated by DEAE-cellulose and Sepharose CL-6B column chromatographies. This process resulted in three polysaccharide fractions, termed CMP Fr I, CMP Fr II, and CMP Fr III. Of these fractions, CMP Fr II, with an average molecular mass of 127 kDa, was able to upregulate effectively the phenotypic functions of macrophages such as NO production and cytokine expression. The chemical property of the stimulatory polysaccharide, CMP Fr II, was determined based on its monosaccharide composition, which consisted of glucose (56.4%), galactose (26.4%), and mannose (17.2%). Its structural characteristics were investigated by a combination of chemical and instrumental analyses, including methylation, reductive cleavage, acetylation, Fourier transform infrared spectroscopy (FTIR), and gas chromatography-mass spectrometry (GCMS). Results indicated that CMP Fr II consisted of the (1${\rightarrow}$4) or (1${\rightarrow}$2) linked glucopyranosyl or galactopyranosyl residue with a (1${\rightarrow}$2) or (1${\rightarrow}$6) linked mannopyranosyl, glucopyranosyl, or galactopyranosyl residue as a side chain. The configuration of the ${\beta}$-linkage and random coil conformation of CMP Fr II were confirmed using a Fungi-Fluor kit and Congo red reagent, respectively.

  • The dried ground needles (2.0 kg) of Chamaecyparis pisifera (Sieb. et Zucc.) Endlicher were extracted with acetone-$H_2O$ (7:3, v/v), concentrated, and fractionated with a series of n-hexane, methylene chloride, ethyl acetate and water on a separation funnel. Each fraction was freeze dried, then a portion of ethyl acetate soluble powder was chromatographed on a Sephadex LH-20 column using a series of aqueous methanol and ethanol-n-hexane mixture as eluents. The isolated compounds were identified by cellulose TLC, $^1H$-, $^{13}C$-NMR, COSY, HETCOR, FAB and EI-MS. (+)-catechin, taxifolin-3-O-${\beta}$-D-xylopyrano-side, quercetin-3-O-${\alpha}$-L-rhamnopyranoside were isolated from the ethyl acetate soluble fraction of Chamaecypairs pisifera needle. Antioxidative tests on the isolated compounds indicated that all of the compounds showed similar values to ${\alpha}$-tocopherol and BHT as controls.
