Purification Properties and Treatment Conditions of Cellulase for Papermaking Processes (제지 공정 적용을 위한 Cellulase의 정제 특성 및 처리 조건 선정에 관한 연구)

  • The main protein peak was observed in fraction No.9 and 109 when cellulase seperation was conducted by use of DEAE-Sephadex. The protein obtained from fraction No.9 has the characteristics of Cx component and that from fraction No.109 characteristics similiar to $C_1$ component. The effective reaction condition of the ensyme used was $40^{\circ}C$ in temperature. pH 5.0 and 90 minutes in treatment time. For the case of $C_1$ pH 5.5 in temperature range of $30^{\circ}C 50^^{\circ}C$, 4.0 5.5 in pH, and over 30 minutes of treatment time, the reaction was in the range of 80% of the maximum. Affinity of enzyme increased as freeness, increased, and this effect was more visible in fiber than in fines.

  • Cotton and cellulosic other fabrics, such as rayon, polynosic, and linen were treated with liquid ammonia, and then were treated with cellulase after or before dyeing, as well as in the presence of dye. Dyeing was carried out with C. I. Direct Blue 1 at 5$0^{\circ}C$, for 6hr in the case of rayon, and 24hr in the case of cotton, polynosic, and linen. The optimum condition of cellulase was at 55$^{\circ}C$, pH 4~5. Weight loss of fabrics were increased by the liquid ammonia treatment and it was predominant when they were treated with cellulase alone and in the presence of dye. Changes of color strength of the cotton, polynosic, and linen were increased by liquid ammonia treatment compare with untreatment. Especially, in the presence of dye, K/S value of the liquid ammonia-treated fabrics were nearly the smae as untreated.

Real-time Monitoring of Glucose Using a Differential Refractive Index Detector (시차 굴절률 측정에 의한 실시간 글루코오스 모니터링)

  • A differential refractive index detector was used for monitoring glucose contents in textile processing solutions. The sensitivity of the device was high enough to measure 0.05% aqueous glucose solution that could not be measured by normal refractive index measurement. The device was set to monitor glucose concentration continuously in real time by measuring differential refractive index and calibrated by standard glucose solutions in a range of 0.1 to 1.0%. The possibility of industrial application of the device was demonstrated by real-time monitoring of glucose concentration in textile processing solutions such as desizing bath and cellulase treatment bath. Both of solutions contained glucose as a major degraded product. The device would be able to control the weight loss of cellulosic fiber during cellulase treatment since the amount of degraded products in a processing bath is proportional to its weight loss.

  • Effects of ultraviolet stress and elicitors, cellulase and jasmonic acid (JA), for the production of capsidiol, sesquiterpenoid phytoalexin, in seedlings and suspension cultures of pepper (Capsicum annum L. cv, Soobicho) were examined. Extracellular capsidiol in the medium of suspension cultures was absent from control cells, but accumulated in the elicitor treated cells with 0.05 $\mu\textrm{g}$/mL of cellulase or 0.1 $\mu\textrm{g}$/mL JA. Elicited cells gradually decreased their viability and eventually died within 48 hours of elicitor treatment by the toxicity of capsidiol accumulated in the culture medium. Capsidiol production in the leaves of pepper seedlings was markedly increased by the treatment of ultraviolet stress and reached maximum level at 48 hours of irradiation. Infiltration of elicitors, 0.05 $\mu\textrm{g}$/mL cellulase or 1.0 $\mu\textrm{g}$/mL JA, to the surface of leaf or fruit, stimulated the elicitation of the cells which resulted in the production of capsidiol and expansion of pathogene-like lesion around the elicitor treated region.

Extract Characteristics of Old Pumpkin on Enzyme Treatment (효소제 처리에 따른 늙은 호박의 추출 특성)

  • In the present study, we investigated the quality characteristic of old pumpkin extract treated with enzymes. As a results, all the groups treated with pectinase were better in quality characteristic than control group and the group treated with 0.15%(w/w) pectinase was specially great. All the groups treated with simultaneous pectinase and cellulase were higher in the extraction rate than the groups treated with pectinase or cellulase. The experimental groups were divided into non-treated control(I) and three treatment groups(II- IV) for optimum condition of enzyme treatment. The II and IV groups were treated with 0.15%(w/w) pectinase and 0.15%(w/w) cellulase, respectively, and the ill group was treated with both 0.15%(w/w) petinase and 0.05%(w/w) cellulase. Yield for old pumpkin extract of the III group (86.94%) was higher than that of other groups, but there were no significant difference among the groups in soluble solide content and pH of the extract. Reducing sugar and total sugar contents in the ill group were 2.81% and 4.60%, respectively. Total carotene content in the II group (5.36 mg%) was higher than other groups. Old pumpkin extracts in all the groups showed nitrite-scavenging ability to pH 1.2, 3.0 and 4.0. Total free amino acid content in the III group (176.7 mg%) was higher than other groups. Citrulline contents in the II and III groups were detected 1.66 and 1.41 mg%, respectively but the contents in other groups were not detected.

Effect of Sizing Agent on the Enzymatic Finishing of Tencel Fabric by Cellulase (셀룰라아제에 의한 텐셀직물의 가공에 있어서 호제의 영향)

  • The effect of pasting agent on the defibrillation of Tencel fabric was investigated. It was evaluated by the weigth loss of fabric when the fabric was treated with cellulase containing various kinds of pasting agents. The surface appearance of Tencel fabric was checked by SEM. Under the treatment condition without pasting agent, the weight loss of fabric was high at pH 5.0 and $60^\circ{C}$. This means that the cellulase activity was high at this condition. By increasing the concentration of carboxymethyl cellulose(CMC), the weight loss of fabric was decreased monotonously. This tendency was not appeared in other pasting agents. CMC is synthesized by the reaction of chloroacetic acid and cellulose. The glucose units may be remained after the reaction. So, it was considered that the degradation of glucose unit in Tencel was decreased, because cellulase had to attack both Tencel and CMC.

  • Since cellulose is the only organic material that is annually replenishable in very large quantities, we must explore ways to utilize it as a source of energy, food and chemicals. For the utilization of this resource, it is first enzymatic hydrolyzed to glucose, then the glucose can be used as a food, converted single cell protein by microorganism, fermented to clean burning fuel and other chemicals. Cellulolytic enzyme, cellulase, consists of two or three major components, $C_1-cellulase$, $C_x-cellulase$ and ${\beta}-glucosidase$. $C_x-cellulase$ are fairly common but $C_1-cellulase$ are quite rare. Trichoderma viride is the best source of active cellulose, especially $C_1-enzyme$. Saccharification rate of cellulose in greatly influenced by the degree of crystallinity and extent of lignification. But by the pretreatment the substrate with cellulose swelling agent, delignifying reagent and physical treatment, the degree of saccharification is enhanced. Thus, glucose syrups of 2 to 10% concentration are realized from milled newspaper. The enzymatic hydrolysis of such energy rich material, such as cellulose, to glucose is technically feasible and practically achievable on a very large scale.

  • An attempt was made to increase the yield of extraction of ginkgoflavonglycosides from leaves of Ginkgo biloba by treatments of with Cellulase C and macerating enzymes. The yield of dried extract and its contents of ginkgoflavonols, when treated only with cellulase C, were analyzed to be 1. 99% and 0. 38%, respectively. The contents of ginkgoflavonglycosides in the dried extracts were calculated to be 25. 28%. By the treatment with a mixture of three enzymes, cellulase C: cellulase NC and macerosin (1 : 1 : 2), the yield of the dried extract, ginkgoflavonols as well as their glycosides were determined to be 2. 48%, 0. 48% and 24. 16%, respectively.

  • The soil of Kwangneung area(Kyeunggi-Do) was inoculated directly into wheat-bran-media and after $3{\sim}4$ days of incubation, a Penicillium species whose cellulase activity was 1011u/g was isolated. With the treatment of mutagenic agents an improved strain(cellulase activity: 1303u/g) was obtained. This strain was screened again by mono-spore isolation method. Finally a strain C8-14 (cellulase activity: 2351u/g) which had lesser spores than the wild strain was obtained.

  • A Trichoderma sp. SO-571 producing cellulase was isolated from soil, and a pilot-scale cultivation and separation of cellulase were conducted. The cellulase activity was about 14.5 unit/ml after 112 hr of cultivation in a 301 fermenter containing 3.0% cellulose, 4.0% soybean powder, 3.0% wheat bran, 0.5% ($NH_4$)$_2$$SO_4$0.2% urea, 1.0% CSL, 0.5% $KH_2$PO$_4$, and 0.2% Tween 80. The cellulase was purified over 4.6 folds in three steps with 47.86% yield. The optimum pH of cellulase was pH 5.0 and optimum temperature was $60^{\circ}C$. To investigate the effect of the cellulase-treated cellulosic fabric, the weight loss was compared. The weight loss of denim treated with cellulase from Trichoderma sp.SO-571 was 2.9% and that with Celluclast 1.5L was 2.2%. In tencel treatement with enzyme, cellulase showed 0.7% higer weight loss than that with Celluclast 1.5L.
