Analysis of cellular fatty acid methyl esters (FAMEs) for the identification of leuconostoc strains isolated from kimchi

  • The cellular fatty acid methyl esters (FAMEs) analysis data obtained for clusters defined at a Euclidian distance of 17.5, in the classification of lactic acid bacteria isolated from kimchi, described by Lee et al. (4), was used for the identification of 79 Leuconostoc isolates. The test strains were isolated using a selective isolation medium specific for the genus Leuconostoc. These strains were then characterized according to their fatty acid profiles. The results show that all seventy nine test strains were identified to the known Leuconostoc clusters B, C, and D. Cluster B had the highest relative amount of the saturated fatty acid 16 : 0. The saturated fatty acid 16 : 0 and summed feature 9 were found as a major components in cluster C, which had a higher level of summed feature 9 than cluster B. Cluster D is characterized by the highest relative amount of the unsaturated fatty acid 18 : 1 w9c. It is suggested that FAMEs analysis can be successfully applied in the identification of lactic acid bacteria isolated from kimchi.

Physiological Responses of Oxygen-Tolerant Anaerobic Bifidobacterium longum under Oxygen

  • In order to investigate what kind of response anaerobic bifidobacteria has on oxygen stress, five oxygen-tolerant bifidobacteria were isolated from human fecal samples. All were temporarily identified as Bifidobacterium longum through an analysis of carbohydrate utilization patterns and cellular fatty acid profiles. In the presence of oxygen, the lag phase became extended and the cell growth was suppressed. Bifidobacterial cell was able to remove dissolved oxygen in an early stage of growth and to overcome oxygen stress to a certain extent. The cell became long n size and showed a rough surface containing many nodes which were derived from abnormal or incomplete cell division. Cellular fatty acid profiled changed remarkably under a partially aerobic condition, so that the carbon chain of cellular fatty acid became short. All the dimethyl acetals originated from plasmalogen were reduced, any cyclopropane fatty acid, 9, 10-methyleneoctadecanoic acid ($C_{19:0}cyc9,10$), was increased remarkably. Oxygen stress induced a 5.5 kD protein in B. longum JI 1 of the oxygen-teolerant bifidobacteria, that was named Osp protein, and its N-terminal amino acid sequence was as follows: unknown amino acid-Thr-Gly-Val-Arg-Phe-Ser-Asp-Asp-Glu. Therefore, the oxygen-tolerant bifidobacteria seemed to defend against oxygen stress byincreasing the content of short fatty acid and cyclopropane fatty acid, and induction of an oxygen stress protein, but not the plasmalogen.

Chemotaxonomic Classification of Marine Bacteria on the Basis of Fatty Acid Compositions

  • The cellular fatty acids of 47 marine bacteria representing the genus Alteromonas, Arthrobacter, Bacillus, Micrococcus, Pseudomonas, Shewanella, Staphylococcus and Stenotrophomonas were determined by a gasliquid chromatographic analysis. Sixty-eight different fatty acids with 10 to 20 carbon atoms were detected in marine bacteria. Of the eight genus examined, 14:0, 16:0 and i17:0 were detected in all, while i14:0, a15:0, i16:0, and 15:0 were found in most of all. There were significant differences in the fatty acid patterns between Gram positive and Gram negative bacteria. Bacteria of Gram positive genus showed relatively high contents of the branched type fatty acids, while the major fatty acids in Gram negative were unsaturated and straight forms. Phylogenetic relationships between marine bacteria defined by the cellular fatty acid patterns represented obvious differences between Gram positive and Gram negative genera, even in respective genus. Therefore, the bacterial classification and identification can be accomplished more easily and rapidly based on the cellular fatty acid profiles than the conventional methods.

Comparison of Cellular Fatty Acid Composition and Genotypic Analysis of Bifidobacterium longum MK-G7 with Commercial Bifidobacteria Strains

  • This study was conducted to compare the cellular fatty acid composition and genotypic analysis of Bifidobacterium longum MK-G7 originated from Koreans with other commercial type strains of bifidobacteria. The cellular fatty acid of Bif. longum MK-G7 was shown to be composed of $C_{160FAME},C_{181\;c18DMA},C_{18.1\;CIS9\; FAME},C_{14.0FAME},C_{19\;0cye9,10 DMA},Feature7(C_{17.2 FAME), and Feature 10(C_{181\; Cll/t9/t6 FAME}$. Bif. longum MK-G7 showed 99.9% homology and the highest relatedness with Bif. longum ATCC 15707 type strain. Both Bif. longum MK-G7 and Bif. longum ATCC 15707 showed 153 bp products on RAPD (randomly amplified polymorphic DNA) analysis, however, they showed quite different band patterns on PFGE (pulsed-field gel electrophoresis) analysis. Consequently, our present study showed that Bif. longum MK-G7 was different from any commercial type strains of Bif. longum tested.

Effects of three different formulae of Gamisoyosan on lipid accumulation induced by oleic acid in HepG2 cells

  • Background: Gamisoyosan (GSS) is an herbal formula which has been used to treat women's diseases for several hundred years in Korea. GSS is one of the three most common prescriptions among women and is used to treat menopausal symptoms. Fatty liver disease is also common in postmenopausal women and can precede more severe diseases, such as steatohepatitis. The present study compared the effects of GSS on fatty liver using three different formulae, Dongui-Bogam (KIOM A), Korean Pharmacopeia (KIOM B) and Korean National Health Insurance (KIOM C). Methods: In oleic acid-induced HepG2 fatty liver cells, cellular lipid accumulation, triglycerides and total cholesterol were measured after treatment with three GSS formulae and simvastatin as a positive control. To investigate the phytoestrogen activity of GSS, MCF-7 cells were treated with GSS, and hormone levels were quantified. Also, qualitative analysis was performed with UPLC. Results: All types of GSS decreased cellular lipid accumulation. KIOM A was slightly less effective than the other two GSS formulae. KIOM B and KIOM C decreased cellular triglycerides more effective than simvastatin, but KIOM A did not affect cellular triglycerides. Cellular total cholesterol was decreased by all GSS and simvastatin. GSS showed phytoestrogen activity in MCF-7 cells. From the UPLC analysis data, geniposide, paeoniflorin and glycyrrhizin were detected form three GSS formulae. Conclusion: These results suggest that all GSS formulae have a beneficial effect on fatty liver disease during menopause and that differences of formula have no effect on the efficacy of the prescription.

Lipid analysis of streptomycetes isolated form volcanic soil

  • The cellular fatty acids and quinones of streptomycetes isolated from volcanic soils were analysed. The strains contained fatty acids of 14 to 17 carbon chains, and 12-methyltetradecanoic acid and 14 methylpentadecanoic acid were dominant in most strains. The total profiles consisted of 74% branched fatty acid family, 16.8% linear family and 8.2% unsaturated family. The largest cluster of grey spore meases defined by numerical classification was separated from the remainders in the principal component analysis, but the other clusters were overlapped with one another. In the analysis of respiratory quinones, all of the strains contained either the menaquinone of 9 isoprene units with 6 hydrogenations of 8 hydrogenations as the major species. The distribution of menaquinones among the clusters could provide an important key in the chemotaxonomy of streptomycetes.

지방산분석을 이용한 Pseudomonas aeruginosa의 동정 (Identification of Pseudomonas aeruginosa Using Fatty Acid Analysis)

  • Cellular fatty acid composition of 17 strains of the Pseudomonas aeruginosa was determined by gas-liquid chromatography isolated from environmental and clinical sample in a C university hospital. Straight-chain saturated acid of C16:0 and straight-chain unsaturated acid of C18:1 with a double bond were commonly found in all the strains tested. The presence of 12:0 3OH (3-10%), 16:0 (18-28%), and 18:1w7c (17-37%) showed the characteristics of the species in the Pseudomonas. Bacterial fatty acid composition was considered to be useful for the study of interrelation and for rapid identification of the bacteria.

  • Candida속 효모 27종이 표준 균주를 대상으로 세포내 지방산 조성을 기체 크로마토그래피로 분석하여, 함유 지방산의 종류와 함량비를 근거로 27종이 균주를 3개의 분류군으로 나누었다. 이들 지방산 분석에 의한 분류군을 Candida속의 다른 분류학적 결과들인 coenzyme Q 분자종의 종류, DNA 염기함량 분석, ITS 부위의 제한효소 절단 다형질의 양상, 리보좀 소단위 RNA 유전자의 염기배열 보고와 비교 분석하였다. 그 결과 같은 분류군에 위치하는 균주들 사이의 연관 관계가 거의 일치하였다. 다라서 세포내 지방산 분석은 Candida속 규준들을 유전적 동질성을 갖는 균주들로 분류하는 수단으로 사용될 수 있으며, 유전적으로 매우 다양한 균주들의 집단으로 알려진 Candida속에 관한 분류적 고찰에 일차적으로 사용될 수 있는 유용한 수단임을 알 수 있었다.

Physicochemical Characters of Ultra Violet Ray Resistant Deinococcus sp. Isolated from Air Dust

  • Among a few number of UV-resistant isolated form various environmental sources (10), we made a comparative physio-chemoanalytical study on one of spherical bacteria isolated from air dust, presumably Deinococcus sp. (CM strain 29) with an UV resistant bacterium, Deinococcus radiophilus ATCC 27603 as the reference strain. Our isolate of UV resistant coccus, Deinococcus sp. CM 29 and D. radiophilus ATCC 27603 showed more than 75% matching coefficient in metabolic activity of various substrates. The most predominant cellular fatty acid of both strains was palmitoleic acid (C 16 :1, cis 9), but the detail fatty acid profiles were slightly dissimilar to each other. Cell-bound arange pigment seemed to be an identical chemicals on spectrophotometric analysis. L-ornithine was detected as cell-wall amino acid in both strains. Galactose was detected as cell-wall sugar in D. radiophilus ATCC 27603, whereas glucose in Deinococcus sp. CM 29. G-C molar ratio of both strains was comparable, 63-65%.

Dudleya brittonii extract promotes survival rate and M2-like metabolic change in porcine 3D4/31 alveolar macrophages

  • Objective: Although alveolar macrophages play a key role in the respiratory immunity of livestock, studies on the mechanism of differentiation and survival of alveolar macrophages are lacking. Therefore, we undertook to investigate changes in the lipid metabolism and survival rate, using 3D4/31 macrophages and Dudleya brittonii which has been used as a traditional asthma treatment. Methods: 3D4/31 macrophages were used as the in vitro porcine alveolar macrophages model. The cells were activated by exposure to phorbol 12-myristate 13-acetate (PMA). Dudleya brittonii extraction was performed with distilled water. For evaluating the cell survival rate, we performed the water-soluble tetrazolium salt cell viability assay and growth curve analysis. To confirm cell death, cell cycle and intracellular reactive oxygen species (ROS) levels were measured using flow cytometric analysis by applying fluorescence dye dichlorofluorescein diacetate and propidium iodide. Furthermore, we also evaluated cellular lipid accumulation with oil red O staining, and fatty acid synthesis related genes expression levels using quantitative polymerase chain reaction (qPCR) with SYBR green dye. Glycolysis, fatty acid oxidation, and tricarboxylic acid (TCA) cycle related gene expression levels were measured using qPCR after exposure to Dudleya brittonii extract (DB) for 12 h. Results: The ROS production and cell death were induced by PMA treatment, and exposure to DB reduced the PMA induced downregulation of cell survival. The PMA and DB treatments upregulated the lipid accumulation, with corresponding increase in the acetyl-CoA carboxylase alpha, fatty acid synthase mRNA expressions. DB-PMA co-treatment reduced the glycolysis genes expression, but increased the expressions of fatty acid oxidation and TCA cycle genes. Conclusion: This study provides new insights and directions for further research relating to the immunity of porcine respiratory system, by employing a model based on alveolar macrophages and natural materials.
