  • PID 제어기는 구조가 간단하고 적용이 용이하다는 장점으로 인하여 널리 사용되고 있는 제어방식이다. 이러한 선형 PID 제어기는 시스템의 파라메터가 변화가 있거나 부하 특성이 비선형적으로 변화할 때에 적절한 이득과 성능을 얻기 어려워 고성능 제어 특성을 기대하기 어렵다. 본 연구에서는 세포성 면역 반응과 경사감소학습에 기초하여 비선형 PID 제어기를 설계하고, 설계된 제어기의 이득과 비선형 함수의 파라메터들을 실시간 적응적으로 학습할 수 있는 학습 알고리즘을 개발하고, 이를 제어시스템에 적용하였다. 제안된 비선형 PID 제어기는 비선형 직류 모터 시스템의 파라메터들이 변화하거나 주파수가 다른 추종 명령에 대하여, 적응적으로 이득을 변화 시키며 추종함을 보였다.

Cytokines Expression and Nitric Oxide Production under Induced Infection to Salmonella Typhimurium in Chicken Lines Divergently Selected for Cutaneous Hypersensitivity

  • In the present study, the impact of Salmonella Typhimurium on cell-mediated immunity (CMI) was investigated in 5 week-old immuno divergent broiler lines selected for the high and low response to phytohemagglutinin-P. The immune response was assessed in peripheral-blood mononuclear cells (PBMCs) induced with Salmonella Typhimurium at different time intervals (0 h, 0.5 h, 2 h, 4 h, 6 h, 12 h and 24 h). The differential mRNA expression patterns of IFN-${\gamma}$, IL-2 and iNOS were evaluated by quantitative real time PCR. In-vitro production of nitric oxide (NO) was also estimated in the culture supernatant and correlated with iNOS mRNA expression. Present study showed higher production of NO in the high cell-mediated line (HCMI) as compared to the low cell-mediated line (LCMI) upon stimulation with Salmonella Typhimurium. Correspondingly, higher mRNA expression of iNOS and IFN-${\gamma}$ were observed in high response birds (HCMI); but IL-2 was down regulated in this line compared to the low response birds (LCMI). Significantly (p<0.05) higher expression of iNOS, IFN-${\gamma}$ and higher production of NO in high line indicated that the selection for PHA-P response might be employed for increasing the immune competence against Salmonella Typhimurium in chicken flocks.

  • In a series of our screening for immunomodulating substances, we isolated prodigiosin from the culture broth qf Serratia marcescens B-1231. This compound inhibited the T cell-mediated immune responses such as concanavalin A-induced proliferation, mixed lymphocyte response, local graft versus host reaction and T-dependent antibody response at nontoxic concentrations. However. prodigiosin did not effect B cell-mediated immune functions such as lipopolysaccharide-induced proliferation and -activated polyclonal antibody production at the same concentrations. Prodigiosin did not cause death in vitro to lymphocytes at effective concentrations (<100 nM) and also did not show toxicity in vivo to lymphoid organs at effective dos-ages (10 and 30 mg/kg). The pharmacological potencies were comparable to the activities of well-known T-cell specific immunosuppressants such as cyclosporin A. In our continuing study, mechanism of action of PDG is investigated with respect to the effect of PDG on IL-2/IL-2R pathway and transcription factor.

The Role of Nrf2 in Cellular Innate Immune Response to Inflammatory Injury

  • Nuclear factor erythroid derived 2-related factor-2 (Nrf2) is a master transcription regulator of antioxidant and cytoprotective proteins that mediate cellular defense against oxidative and inflammatory stresses. Disruption of cellular stress response by Nrf2 deficiency causes enhanced susceptibility to infection and related inflammatory diseases as a consequence of exacerbated immune-mediated hypersensitivity and autoimmunity. The cellular defense capacity potentiated by Nrf2 activation appears to balance the population of $CD4^+$ and $CD8^+$ of lymph node cells for proper innate immune responses. Nrf2 can negatively regulate the activation of pro-inflammatory signaling molecules such as p38 MAPK, NF-${\kappa}B$, and AP-1. Nrf2 subsequently functions to inhibit the production of pro-inflammatory mediators including cytokines, chemokines, cell adhesion molecules, matrix metalloproteinases, COX-2 and iNOS. Although not clearly elucidated, the antioxidative function of genes targeted by Nrf2 may cooperatively regulate the innate immune response and also repress the expression of pro-inflammatory mediators.

소음인(少陰人) 십전대보탕(十全大補湯)이 면역반응(免疫反應)에 미치는 영향(影響) (An Experimental Study on the Effects of Shipjuntaepotang on Immune Response in Mice)

  • In order to investigate the effects of shipjuntaepotang (少陰人 十全大補湯) on immune response, the author performed this experimental study. Delayed type hypersensitivity (DTH) and rosette forming cells (RFC) for cell-mediated immune response, hemagglutinin (HA) titers, hemolysin (HL) titers, and carbon clearance for phagocytic function of MPS (mononuclear phagocyte system) were measured in ICR mice. The results were summarized as follows. 1. DTH in the experimental group was increased, as compared with the control group, with statistical significance. 2. RFC in the experimental group was increased, as compared with the control group, with statistical significance. 3. HA-titers were increased in the experimental group, as compared with the control group, with statistical significance. 4. HL-titers were increased in the experimental group, as compared with the control group, with statistical significance. 5. Carbon clearance was increased in the experimental group, as compared with the control group, with statistical significance. Through invivo experimental study in ICR mice, these findings suggest that shipjuntaepo-tang enhance both cellmediated and humoral immune responce.

Evaluation of Cytotoxicity for Immunity Rejection of US11, hDAF and FasL Transgene-Transfected Cells

  • Xenotransplantation is proposed as a solution to the problem of organ shortage. However, transplantation of xenogeneic organs induces an antigen-antibody reaction in ${\alpha}$-1,3-gal structure that are not present in humans and primates, and thus complement is also activated and organs die within minutes or hours. In this study, we used FasL gene, which is involved in the immune response of NK cell, and US11, which suppresses MHC Class I cell membrane surface expression, to inhibit cell mediated rejection in the interspecific immunity rejection, and also hDAF(CD55) was introduced to confirm the response to C3 complement. These genes were tranfeced into Korean native pig fetal fibroblasts using pCAGGS vector. And cytotoxicity of NK cell and human complement was confirmed in each cell line. The US11 inhibited the cytotoxicity of NK cell and, in addition, the simultaneous expression of US11 and Fas ligand showed excellent suppress to T-lymphocyte cytotoxicity, hDAF showed weak resistance to cytotoxicity of natural killer cell but not in CD8+ CTLs. Cytotoxicity study with human complement showed that hDAF was effective for reducing complement reaction. In this studies have demonstrated that each gene is effective in reducing immune rejection.

Immune Response and Plasma Alpha Tocopherol and Selenium Status of Male Buffalo (Bubalus bubalis) Calves Supplemented with Vitamin E and Selenium

  • An experiment was conducted using 20 male buffalo calves to study the effect of vitamin E and selenium supplementation on their immune response and plasma ${\alpha}$-tocopherol and selenium status. These buffalo calves (10-12 months old, average body weight $75.30{\pm}2.20 $ kg) were randomly allotted to four treatments on the basis of their body weights and were fed on wheat straw and concentrate mixture to meet their nutrient requirements of 500 g/d body weight gain. The buffalo calves were fed either a control diet (neither supplemented with Se nor VE) or diets supplemented with Se at 0.3 ppm (+Se), DL-alpha tocopheryl acetate at 300 IU (+VE), and both DL-alpha tocopheryl acetate at 300 IU and Se at 0.3 ppm (+Se+VE). These experimental diets were fed for 180 days. Blood samples were collected at day 0 and subsequently at 45 day intervals up to 180 days of experimental feeding to monitor plasma ${\alpha}$-tocopherol and Se concentrations. To assess humoral immune response, all calves were sensitized with formalin inactivated Pasteurella multocida antigen at 135 days of experimental feeding and blood was collected on 0, 7, 14, 21 and 28 days post vaccination (DPV) to measure antibody production using indirect ELISA. Cell mediated immune response of calves was assessed after 180 days of experimental feeding by in vivo delayed type hypersensitivity (DTH) reaction using phytohaemaglutinin-P (PHA-P) as a mitogen. Results revealed that feeding of VE and Se improved the plasma levels of these nutrients. Plasma levels of Se were affected by supplementation of both VE (p<0.001) and Se (p<0.001); however, no interaction ($Se{\times}VE$) was observed. Supplementation of Se improved the humoral immune response (p<0.008), whereas, VE showed a tendency towards improvement in cell mediated immune response (p<0.064). It was concluded that vitamin E and Se supplementation improved the status of these micronutrients and humoral immune response in buffalo calves.

Study of Cell-mediated Response in Mice by HPV16 L1 Virus-like Particles Expressed in Saccharomyces cerevisiae

  • The first vaccine against human papillomaviruses (HPV) formulated with HPV16 L1 virus-like particles (VLPs) produced in yeast was approved by the FDA in June 2006. Nevertheless, there have been few studies of the immunogenicity in mice of VLPs. In this study, we evaluated the cell-mediated immune response to VLPs produced in Saccharomyces cerevisiae. After immunization of mice with HPV16 L1 VLPs, we measured splenocytes proliferation and the levels of IFN$_{\gamma}$, IL2, IL4, and IL5. Splenocytes proliferation was significantly increased and a mixed Th1/Th2 response was indicated. IgG subtype immunoresponses were strongly induced and IgG1 titers were higher than those of IgG2a.

  • 본 논문은 생체시스템의 인공면역시스템을 이용한 Immunized PID 제어기를 구성하였으며, 상층부위에 제동시스템이 부착된 다층 건축물의 진동제어에 적용하였다. 인공면역시스템은 인체가 외부로부터 침투하는 바이러스, 병원균 등으로부터 보호하기 위하여 내부의 항체들이 이를 극복하기 위하여 행동하는 현상을 모델링한 것으로써 외란입력에 대한 적응성 및 강인한 응답특성을 가지고 있다. 본 논문에서 설계된 제어시스템은 항만의 건축물에 능동제어기로 적용하였으며, 바람이나 지진과 같은 환경적 부하로 인해 발생되는 건축물의 응답변위를 컴퓨터 시뮬레이션을 통해 나타내었다. 그리고, 기존의 진동제어에 널리 이용되는 제어기도 함께 적용하여 제안한 제어기 성능의 우수성을 입증하고자 함과 동시에 인공면역시스템을 응용한 제어기 설계에 대한 비젼도 제시하고자 한다.

Monitoring Cellular Immune Responses after Consumption of Selected Probiotics in Immunocompromised Mice

  • Probiotics are currently considered as one of tools to modulate immune responses under specific clinical conditions. The purpose of this study was to evaluate whether oral administration of three different probiotics (Lactiplantibacillus plantarum CJLP243, CJW55-10, and CJLP475) could evoke a cell-mediated immunity in immunodeficient mice. Before conducting in vivo experiments, we examined the in vitro potency of these probiotics for macrophage activation. After co-culture with these probiotics, bone marrow derived macrophages (BMDMs) produced significant amounts of proinflammatory cytokines including interleukin-6 (IL-6), IL-12, and tumor necrosis factor-α (TNF-α). Levels of inducible nitric oxide synthase (inos) and co-stimulatory molecules (CD80 and CD86) were also upregulated in BMDMs after treatment with some of these probiotics. To establish an immunocompromised animal model, we intraperitoneally injected mice with cyclophosphamide on day 0 and again on day 2. Starting day 3, we orally administered probiotics every day for the last 15 d. After sacrificing experimental mice on day 18, splenocytes were isolated and co-cultured with these probiotics for 3 d to measure levels of several cytokines and immune cell proliferation. Results clearly indicated that the consumption of all three probiotic strains promoted secretion of interferon-γ (IFN-γ), IL-1β, IL-6, IL-12, and TNF-α. NK cell cytotoxicity and proliferation of immune cells were also increased. Taken together, our data strongly suggest that consumption of some probiotics might induce cell-mediated immune responses in immunocompromised mice.
