Pseudomonas aeruginosa BYK-2의 균체고정화법을 이용한 생물유화제의 생산

  • The optimal conditions and properties for the immobilization of marine bacterium Pseudomonas aeruginosa BYK-2 have been determined. For the high productioon of biosurfactant, Na-alginate, PVA, modified PVA were used as a carrier. The optimal emulsifying activity on immobilized Pseudomonas aeruginosa BYK-2 showed 1036Unit (about 2.2g/L biosurfactant) in Basal salt medium(B.S.M.) at $25^{\circ}C$, 100rpm. Ca-alginate was selected the optimal bead among PVA, modified PVA and Ca-alginate. The optimal cell load in alginate bead was 10 gCWW/100g carrier. As the results of incubation of immobilized 5g Ca-alginate bead (conditions; 3% alginate, bead diameter: 2.3mm, 10% cell load) in 50m1 production medium, The emulsifying activity of 1407Unit, about 3.0g/L biosurfactant was obtained from immobilized cell after cultivation of 92hr at $25^{\circ}C$, 100rpm.

  • Arthrobacter simplex (ATCC 6946) was cultured, induced and immobilized in acrylamide polymer. The characteristics of the immobilized whole-cell enayme were studied using hydrocortisone as the substrate. The enzyme activity was increased during the incubation of the gel particle in 0.5% peptone media. The ennzyme reaction kinetics of the Δ'-dehydrogenase (3-oxosteroid Δ'-oxydo reductase, E. C. 1.3.99.4) foliowed the Michaelis-Menten type. Km and Vm values were different significantly after immobilization of the cell. The optimum pH and temperature were changed, too. Nitrogen sources such as casitone, peptone or tryptone were good media for the enzyme reaction. And there was no need to add cofactors of the enzyme in the pre-sence of energy sources used in the test. The effect of metal ions on the enzyme activity was insignificant. Organic solvents were used increase the substrate concentration and there was no optimum solvent concentration depending on the substrate concentration.

  • Stability of bioluminescence was investigated with Photobacterium phosphoreum immobilized on the strontium alginate in order to develope continuous real time monitoring of pollutants. The stability of bioluminescence emission was improved by prolonged aging time. The aging time of ${\geq}40$ min and the cell concentration of ${\leq}0.6\;of\;OD_660$ were selected for the immobilization of P. phosphoreum to give linearity between cell concentrations and bioluminescence intensity. In sensitivity tests using phenol, it was found that this compound quenched bioluminescence proportional to the concentration without lowering of cell growth. The lower value for maximum quenching ($q_s$) and higher dissociation constant ($K_s$) were observed with strontium-alginate immobilized cells compared to free cells. The response of bioluminescence to toxicants was evaluated with the immobilized luminescent bacteria. The sensitivity of the immobilized cells was found to be good in response to toxicants, 4-nitrophenol, salicylate and cadmium, when evaluated with a specific rate of bioluminescence quenching.

Cell Surface Display of Poly(3-hydroxybutyrate) Depolymerase and its Application

  • We have expressed extracellular poly(3-hydroxybutyrate) (PHB) depolymerase of Ralstonia pickettii T1 on the Escherichia coli surface using Pseudomonas OprF protein as a fusion partner by C-terminal deletion-fusion strategy. Surface display of depolymerase was confirmed by flow cytometry, immunofluorescence microscopy and whole cell hydrolase activity. For the application, depolymerase was used as an immobilized catalyst of enantioselective hydrolysis reaction for the first time. After 48 h, (R)-methyl mandelate was completely hydrolyzed, and (S)-mandelic acid was produced with over 99% enantiomeric excess. Our findings suggest that surface displayed depolymerase on E. coli can be used as an enantioselective biocatalyst.

  • Immobilization of anchorage-dependent animal cells was investigated using macroporous gelatin microcarriers developed in our laboratory. For the observation of the distribution of cells in macroporous beads, Vero-6 cells and CHO cells were cuttured and their distribution in macroporous beads was observed using a confocal microscope. In results, the final concentration of Vero-6 cells and CHO cells on macroporous beads was 2-3 times higher than that on commercial solid microcarriers (Cytodex-3). Also, macroporous microcarriers could hold cells in their macropores. Consequently, the pores protected cells against hydrodynamic shear. Based on Kolmogorov eddy length scale, the smaller eddies (80 $\mu $m) showed the detrimental effect on cells in macroporous beads as compared with 160 $\mu $m of eddies in conventional solid microcarriers.

  • Platinum is a transition metal that is very resistant to corrosion. It is used as catalyst for converting methyl alcohol to formaldehyde, as catalytic converter in cars, for hydrocracking of heavy oils, in Fuel Cell devices etc. Moreover, Platinum compounds are important ingredient for cancer chemotherapy drugs. The nano forms of Platinum due to its unique physico-chemical properties that are not found in its bulk counterpart, has been found to be of great importance in electronics, optoelectronics, enzyme immobilization etc. The stability of Platinum nanoparticles has supported its use for the development of efficient and durable proton exchange membrane Fuel Cells. The present review concentrates on the use of Platinum conjugated with various metal or compounds, to fabricate nanocomposites, to enhance the efficiency of Platinum nanoparticles. The recent advances in the synthesis methods of different Platinum-based nanocomposites and their applications in Fuel Cell, sensors, bioimaging, light emitting diode, dye sensitized solar cell, hydrogen generation and in biosystems has also been discussed.

  • Choristoneura fumiferana (Cf-2Cl) insect cells were cultured and immobilized by using cellulosesulfate (NaCS) and polydiallyldimethylammoniumchloride (PDADMAC). A concentration of CfMNPV (Choristoneura fumiferana multiple-mucleopolyhedrovirus) and a Cf-2Cl cell density in the microspheres have been achieved at the densities of $1.57\times10^{10}$ PIBs/ml and $7.5\times10^7$ cells/ml, respectively. Additionally, MTT-test was used to measure the viable cell density in the microspheres, and the confidence of MIT-test was investigated before and after baculovirus infection in the immobilized cell culture.

Stabile Fermentation of Citric Acid Using Immobilized Saccharomycopsis lipolytica

  • The effects of media composition on citric acid fermentation using surface immobilized Saccharomycopsis lipolytica were studied. The use of the standard medium for these organisms resulted in rapid decrease of citric acid production and a transformation of immobilized cell morphologies from a yeast-type to a mycelium-type. When the standard medium was enriched with vitamins, trace minerals, a growth factor and ammonium to form a Vigorous Stationary Phase (VSP) fermentation type medium, relatively stable citric acid production (10 mg/lㆍh) was obtained. Using the VSP type medium, the surface immobilized cells also retained their yeast-type form.
