High-Yield Production of Xylitol from Xylose by a Xylitol Dehydrogenase Defective Mutant of Pichia stipitis

  • This study was carried out in order to investigate the characteristics of xylitol fermentation by a xylitol dehydrogenase defective mutant PXM-4 of P stipitis CBS 5776 and to determime optimum conditions for the high yield ofxylitol production from xylose. Gluconic acid was selected as a co substrate for the xylitol fermentation, since gluconic acid neither blocked xylose transport nor repressed xylose reductase expression. An increase of gluconic acid concentration reduced the rates of xylitol production and cell growth by decreasing medium pH, and the optimal concentration of gluconic acid was determined to be 20 gll with approximately 100% xylitol conversion yield. A fed-batch cell culture resulted in a 44.8 g/l xylitol concentration with 100% yield, based on the amount of xylose consumed.

  • The investigations of specific growth rate, specific alcohol production rate, cell yield, alcohol yield of K. marxianus LG were performed according to the sugar concentrations, 50, 80, 110, 190, and 250g/l of extracted solution of Jerusalem Artichoke. The functipnal relationship between specific growth rate, specific alcohol production rate, and alcohol concentrations were devoted study to. In case of low concentration of alcohol, the fuctions were linear relationships. But in the region of high concentration of alcohol, they expressed the exponential relationships. The growth rate of K. marxianus was prohibited at higher than 50g/l of alcohol concentrations regardless of concentration of residual sugar. Cell and alcohol yield showed the maximum values around 25g/l of alcohol concentraton without being related to initial sugar concentrations.

  • A hyperthermophilic archaeon, $Thermococcus$ $onnurineus$ NA1 was studied to investigate its fermentation characteristics using various carbon sources including formate, maltose and carbon monoxide during the anaerobic batch cultivation at $80^{\circ}C$. Formate was the best carbon source for cell growth and hydrogen production among others. In the batch culture on formate, it was found that the cell concentration increased exponentially by 12 hrs of culture, after which the cell growth and formate consumption was retarded. Hydrogen production was continued more than 24 hrs although the cell growth was ceased at 18 hrs. Hydrogen production rate was directly correlated with the cell growth and formate degradation up to 18 hrs, and the average hydrogen production yield was 1.05 mole-$H_2$/mole-formate. Cell growth and hydrogen production were optimized at the initial pH 6-7, while inhibited at the initial pH lower than 5 and higher than 9.

  • A two-stage fed-batch fermentation was carried out to increase xylitol productivity by Candida tropicalis. The first stage for cell growth was performed in the pH-stat and continuous fed-batch modes. The higher cell growth and lower ethanol production obtained in the fed-batch mode where the growth medium was fed when pH of culture broth increased over 5.7. And also the effect of oxygen transfer on xylitol production was investigated by changing agitation speed under 0.5 vvm of aeration. The maximum xylitol productivity and yield were obtained at 500 rpm of agitation.

  • The green alga, Dunaliella salina under fed-batch cultivation produced 51.12 mg of hydrocarbon per liter with maintaining 0.313 (g dry wt/l). About 20% of hydrocarbon production yield based on dry biomass was obtained from both batch and fed-batch processes. Optimum culture conditions of light intensity, pH and salt concentration were obtained as 0.0080 (kJ/$cm^2$/h), 8.0 and 1.4 (g of NaCl/l), respectively by response surface analysis. The production of hydrocarbons in D. salina was closely correlated to cell growth. Fed-batch cultivation produced more hydrocarbons and maintained better cell growth than a batch process.

  • This experiment was undertaken with the purpose of investigating effect of gibberellin on the growth and electron microscopic ultrastructure of hop(Humulus lupulus var. Hallertau) shoot apex. The results are as follows. 1) The growth of stem in the plots treated with GA(10, 25, and 50ppm) is more predominant than control. The leaves and cones of treated plots turn to pale green. 10ppm plot of GA treatment is the most effective in growth and yield than the others. 2) Under the GA-treated condition, it is believed that the nucleoplasmic index (NP) are higher than the control, and so nucleus, nucleolus, mitochondria, primary cell wall, middle lamella, and etc. tends to be larger than the control. 3) Mitochondria contained in the cell of shoot apex treated with concentration of 10ppm GA was seemed to increases in number and be larger in size than the control.

Selection of Protoplasts-Derived Cell Lines for High Yields Indole Alkaloids from Suspension Cultures of Vinca (Catharanthus roseus) (일일초(Catharanthus roseus) 현탁배양으로부터 원형질체유래 Indole Alkaloid 고생산성 세포주 선발)

  • To produce economically important indole alkaloids by cell cultures, we selected protoplastsderived clones (protoclones) of vinca (Catharanthus roseus) for high yields of catharanthine and ajmalicine. Protoplasts were enzymatically isolated from suspension-cultured cells. The highest plating efficiency (1%) was obtained when protoplasts were plated at a density of 1$\times$105 protoplasts/ml in a culture medium solidified with 0.4% Seaplaque agarose. The growth rates of 40 protoclones subcultured on a solid medium varied over a wide range. Protoclone VPC-6, which had the highest growth rate, was observed to produce relatively high yields of catharanthine and ajmalicine when cultured in a liquid medium. Although the original cell line did not produce catharanthine at a detectable level by HPLC, protoclone VPC-10 produced it at a level of 5.9$\mu\textrm{g}$/g fresh weight of cells for 10 days of culture. Under the same conditions, protoclone VPC-15 produced ajmalicine at a level of 133.6$\mu\textrm{g}$/g, of which productivity was improved about ,3 times than that of the original cell line. The results indicate that differences in the growth rate and indole alkaloid yield among the protoclones reflect the somaclonal variation in suspnsion-cultured cells.

Effect of Ammonium Phosphate Concentration on the Growth of Recombinant E. coli (재조합 대장균의 세포성장에 대한 인산암모늄 농도의 영향)

  • The growth of recombinant E. coli and formation of the by-products were investigated. Ammonium phosphate is known to affect the cell growth as well as the enzyme formation. When initial ammonium phosphate concentration was 0.5g/L, cell mass was 4.1g/L. By adding tryptone to the medium, acetic acid formation increased while lactic acid formation decreased. In cultivating recombinant E. coli, lactic acid and acetic acid turned out to be important by-products which affected cell yield and growth rate. Initial ammonium phosphate and tryptone concentration were optimized in our research and can be applied for other culture of recombinant E. coli.

  • Hydrogen production and its dynamics were investigated in the continuous anaerobic digestion process using Clostridium beijerinckii Donker 1926. In this work, glucose was used as a substrate and hydraulic retention times (HRT) were 0.5, 0.25 or 0.125 day. The removal efficiency of carbohydrate was over 99% under all of HRT conditions. As HRT was shorter, COD removal efficiency became lower while hydrogen content in the total gas and hydrogen production rate became higher. The cell growth yield and hydrogen production yield were 0.27 g-VSS/g-glucose and 0.26 L/g-glucose, respectively, at the steady state. It is expected that the microorganism is able to produce hydrogen when used in the wastewater treatment containing carbohydrate such as glucose. Also, the results in this study could be applied to the actual hydrogen gas production, a promising alternative energy.

Comparison of Immuno-stimulatory Activities by Purification Process of Schizandra chinensis Baillon Fruits

  • Two compounds from Gomisin N and Gomisin A were isolated from the fruits of Schizandra chinensis Baillon. The highest extraction yield as 21.36% was observed in the ethanol extract, compared to the yield obtained form the water extract. The extraction yields of the single compounds were measured to be 0.13 and 0.014 Gomisin A and Gomisin N, respectively. Approximately, 90% of the growth of human stomach adenocarcinoma cancer cells was inhibited after adding 1.0 g/l of the ethanol extract. The growth of the human normal lung cell was limited to 24% after adding the ethanol extract. The water extract lowered the specific secretion of $TNF-\acute{a}$ and IL-6 from T cells, $10.3{\times}10^{-4}\;pg/cell\;and\;12.1{\times}10^{-4}\;pg/cell$, respectively, compared to the ethanol extracts. On the other hand, a treatment with the ethanol extract increased the specific secretion of $TNF-\acute{a}$ and IL-6 from human T cells, to $11{\times}10^{-4}\;pg/cell\;and\;14.3{\times}10^{-4}\;pg/cell$, respectively. The crude ethanol extract had the highest effect on the differentiation of human promyelocytic leukemia cells compared to the other extracts and Gomisin A and N. In general, the biological activities of the extracts gradually decreased as the purification process proceeded, which suggests that higher immunostimulatory activities can be maintained by adding the crude extracts of the fruits rather than by adding a single compound.
