Ultrastructural Studies of the Brain Tumors (뇌종양세포의 미세형태학적 연구)

  • To investigate ultrastructural characteristics of cancer cells of the nervous system, 25 cases; i.e. astrocytoma(9), oligodendroglioma(1), medulloblastoma(1), meningioma(5), pinealoma(2) and pituitary adenomas(7). The common findings were marked irregularity of nuclear membrane with pronounced infoldings, clumping of heterochromatin along inner nuclear membrane, enlargement of nucleolus, and frequent observations of nuclear bodies and nuclear inclusions. But these findings are also the signs that can be observed in hyperactive cells. Thus, ultrastructural characteristics of cancerous nucleus are the great variability of nuclear size, shape and composition. but none of them appear to be specific. Among cytoplasmic organelles, massive fibrils are characteristic of astrocytoma and meningiomas, cytoplasmic protofibrils such as glial process and microvesicles in oligodendroglioma, secretory granules are characteristic in pituitary adenomas, and fine filamentous fibrils and desmosomes are characteristic of fibroblastic type of meningioma. Intercellular relationships and cell membrane specialization are important features in the differential diagnosis of various undifferentiated tumors. The frequent resolution of difficult diagnosis problems by electron microscopy outweighs the disadvantages of this technique, such as the expense and time required.

  • According to the enlargement of production facilities and structures, the requirements of high-capacity load cells are increased for monitoring the process conditions in many fields. Generally, however, the accuracy of the column-type high-capacity load cells is not enough due to the geometric nonlinearity. It is supposed to result from the fact that the whole spring element is under high-level stress for the uniform strain field. In this paper, a new shape of spring element is developed which utilizes the stress concentration. As a design criterion, an object function which quantifies the degree of nonlinearity is defined and optimized by use of response surface modeling. As a result, the weight of the spring element is reduced shout 50% in comparison to the conventional shape. The bonding positions of stain gages are found. which show theoretically zero geometrical nonlinearity, while the ratio of overload protection is reduced from 130% to 125% Also it is shown that the response surface method is very efficient in the optimization approach by use of FEM.

Ultrastructural Alterations of Rabbit Liver after Overdose of Nitrate (질산염과잉투여(窒酸鹽過剩投與)에 의(依)한 간장(肝臟)의 변화(變化)에 관한 전자현미경적연구(電子顯微鏡的硏究))

  • In order to know the morphological changes of liver in nitrate poisoning, the ultrastructural studies were carried out on the rabbit liver after potassium nitrate was administered orally at lethal dose, in single treatment, as acute case and at two different levels. 1.0 and 0.5g/kg of body weight daily for 43 and 60 days as chronic case, respectively, The results were summarized as followings: 1. In the hepatic cells of acute case, mitochondria were swollen, disappearance of cristae and variable in shape. Dilatation of rough endoplasmic reticulum and vacuoles containing degenerated cell organells were observed. Glyogen particles were decreased in number. Degenerated Kupffer cells were often seen in acute case. 2. In the hepatic cells of chronic case, there were increase of smooth endoplasmic reticulum, marked enlargement of rough endcplasmic reticulum, detachment of membrane bound ribosome and some rough endoplasmic reticulum changed into smooth endoplasmic reticulum. Secondary lysosome, abundant glycogen paricles and myelin-figure structures were also observed in the cytoplasm of the hepatic cells. The endothelial cells were proliferated in the area of the necrotic cells.

Neuroendocrine Tumor of Unknown Primary Accompanied with Stomach Adenocarcinoma

  • A 67 year old male at a regular checkup underwent esophagogastroduodenoscopy. On performing esophagogastroduodenoscopy, a lesion about 1.2 cm depressed was noted at the gastric angle. The pathology of the biopsy specimen revealed a well-differentiated adenocarcinoma. On performing an abdominal computed tomography (CT) scan & positron emission tomography-computed tomography (PET-CT) scan, no definite evidence of gastric wall thickening or mass lesion was found. However, lymph node enlargement was found in the left gastric and prepancreatic spaces. This patient underwent laparoscopic assisted distal gastrectomy and D2 lymph node dissection. On final examination, it was found out that the tumor had invaded the mucosal layer. The lymph node was a metastasized large cell neuroendocrine carcinoma with an unknown primary site. The patient refused chemotherapy. He opted to undergo a close followup. At the postoperative month 27, he had a focal hypermetabolic lesion in the left lobe of the liver that suggested metastasis on PET-CT scan. He refused to undergo an operation. He underwent a radiofrequency ablation.

  • Necrotizing lymphadenitis, or Kikuchi's disease, a newly recognized disease of unknown origin, occurred usually in young woman and mostly resolved spontaneously without treatment within a few months. Clinically, characteristic symptoms and sings were local lymph node enlargement, sometimes accompanied by tenderness, fever, weightless, leukopenia, and elevated erythrocyte sedimentation rate, and so necrotizing lymphadenitis can be confused with malignant lymphoma and tuberculosis. The histologic features of necrotizing lymphadenitis are distinctive. : lymph node biopsy reveals areas with frank cellular necrosis, karyorrhexis, and absence of plasma cell. We present nineteen cases of necrotizing lymphadenitis during from March 1990 to January 1993 and discuss their exact diagnosis and proper treatment.

Chronic Subdural Hematoma in the Aged, Trauma or Degeneration?

  • Chronic subdural hematomas (CSHs) are generally regarded to be a traumatic lesion. It was regarded as a stroke in 17th century, an inflammatory disease in 19th century. From 20th century, it became a traumatic lesion. CSH frequently occur after a trauma, however, it cannot occur when there is no enough subdural space even after a severe head injury. CSH may occur without trauma, when there is sufficient subdural space. The author tried to investigate trends in the causation of CSH. By a review of literature, the author suggested a different view on the causation of CSH. CSH usually originated from either a subdural hygroma or an acute subdural hematoma. Development of CSH starts from the separation of the dural border cell (DBC) layer, which induces proliferation of DBCs with production of neomembrane. Capillaries will follow along the neomembrane. Hemorrhage would occur into the subdural fluid either by tearing of bridge veins or repeated microhemorrhage from the neomembrane. That is the mechanism of hematoma enlargement. Trauma or bleeding tendency may precipitate development of CSH, however, it cannot lead CSH, if there is no sufficient subdural space. The key determinant for development of CSH is a sufficient subdural space, in other words, brain atrophy. The most common and universal cause of brain atrophy is the aging. Modifying Virchow's description, CSH is sometimes traumatic, but most often caused by degeneration of the brain. Now, it is reasonable that degeneration of brain might play pivotal role in development of CSH in the aged persons.

Effects of Hyeongbangpaedok-san (HBPDS) on Allergic Contact Dermatitis (ACD) induced by DNFB in Mice (형방패독산(荊防敗毒散)이 DNFB로 유발된 생쥐의 알레르기접촉피부염에 미치는 영향)

  • Objectives The present study was carried out to investigate effects of HBPDS on allergic contact dermatitis (ACD) induced by 2,4-Dinitro-1-fluorobenzene (DNFB) in mice. Methods In this experiment, effects of HBPDS on body weights, skin thicknesses, skin weights, histopathological changes, clinical aspects, erythema index, melanin index, production levels of cytokines in ACD mice were investigated. In addition, effects on proliferation rates, release of ${\beta}$-hexosaminidase and histamine were also investigated in vitro. Results & Conclusions 1) HBPDS inhibited enlargement of skin thickness and weight significantly (P < 0.05). 2) HBPDS treatment prevented spongiosis, edema and immune cell infiltrations. 3) Erythema, desquamation and keratosis were diminished by oral administration of HBPDS. 4) Production levels of TNF-alpha and IFN-gamma in serum were decreased by HBPDS treatment in vivo. 5) More than 200 ${\mu}g/ml$ of HBPDS treatment decreased ${\beta}$-hexosaminidase release and more than 400 ${\mu}g/ml$ of HBPDS treatment also decreased histamine release in vitro.

  • Canine T-zone lymphoma (TZL) is a mature T-cell lymphoma in dogs. The diagnosis and sub-classification are impossible without biopsy or immunophenotyping by flow cytometry. An 11-year-old, spayed, female Golden Retriever presented with lymph node enlargement. Clinical examination was consistent with canine multicentric lymphoma. However, immunophenotyping revealed positive for CD3, CD4, CD5, CD8, CD21, TCRαβ, and MHCII but negative for CD34, CD45, CD79a, and TCRγδ. Histopathology revealed lymphocytes expanding to the cortex-preserving architecture and thinning of the nodal capsule, and CD3 positive but PAX-5 negative. Owing to the indolent nature of TZL, careful monitoring approach without clinical intervention was utilized.

LSD1-S112A exacerbates the pathogenesis of CSE/LPS-induced chronic obstructive pulmonary disease in mice

  • Lysine-specific demethylase 1 (LSD1) is an epigenetic regulator that modulates the chromatin status, contributing to gene activation or repression. The post-translational modification of LSD1 is critical for the regulation of many of its biological processes. Phosphorylation of serine 112 of LSD1 by protein kinase C alpha (PKCα) is crucial for regulating inflammation, but its physiological significance is not fully understood. This study aimed to investigate the role of Lsd1-S112A, a phosphorylation defective mutant, in the cigarette smoke extract/LPS-induced chronic obstructive pulmonary disease (COPD) model using Lsd1SA/SA mice and to explore the potential mechanism underpinning the development of COPD. We found that Lsd1SA/SA mice exhibited increased susceptibility to CSE/LPS-induced COPD, including high inflammatory cell influx into the bronchoalveolar lavage fluid and airspace enlargement. Additionally, the high gene expression associated with the inflammatory response and oxidative stress was observed in cells and mice containing Lsd1-S112A. Similar results were obtained from the mouse embryonic fibroblasts exposed to a PKCα inhibitor, Go6976. Thus, the lack of LSD1 phosphorylation exacerbates CSE/LPS-induced COPD by elevating inflammation and oxidative stress.

p38 MAPK Inhibitor NJK14047 Suppresses CDNB-Induced Atopic Dermatitis-Like Symptoms in BALB/c Mice

  • Atopic dermatitis (AD) is a chronic inflammatory skin disorder. Suppression of MAPKs and NF-κB is implicated as a vital mechanism of action of several traditional Chinese medicines for AD therapy. Although overexpression of MAPK mRNA in the skin tissue has been shown in the AD model, the roles of each MAPK in AD pathogenesis have rarely been studied. This study examined the effect of NJK14047, an inhibitor of p38 MAPKs, on AD-like skin lesions induced in BALB/c mice by sensitization and challenges with 1-chloro-2,4-dinitrobenzene (CDNB) on dorsal skin and ears, respectively. After induction of AD, NJK14047 (2.5 mg/kg) or dexamethasone (10 mg/kg) was administrated for 3 weeks via intraperitoneal injection. Following its administration, NJK14047 suppressed CDNB-induced AD-like symptoms such as skin hypertrophy and suppressed mast cell infiltration into the skin lesions. It also reduced CDNB-induced increase in TH2 cytokine (IL-13) and TH1 cytokines (interferon-γ and IL-12A) levels but did not decrease serum IgE level. Furthermore, NJK14047 blocked CDNB-induced lymph node enlargement. These results suggest that NJK14047, a p38 MAPK inhibitor, might be an optimal therapeutic option with unique modes of action for AD treatment.
