Inhibitory Effect of Ginkgolide B on Platelet Aggregation in a cAMP- and cGMP-dependent Manner by Activated MMP-9

  • Extracts from the leaves of the Ginkgo biloba are becoming increasingly popular as a treatment that is claimed to reduce atherosclerosis, coronary artery disease, and thrombosis. In this study, the effect of ginkgolide B (GB) from Ginkgo biloba leaves in collagen (10 ${\mu}g/ml$)-stimulated platelet aggregation was investigated. It has been known that human platelets release matrix metallo-proteinase-9 (MMP-9), and that it significantly inhibited platelet aggregation stimulated by collagen. Zymographic analysis confirmed that pro-MMP-9 (92-kDa) was activated by GB to form an MMP-9 (86-kDa) on gelatinolytic activities. And then, activated MMP-9 by GB dose-dependently inhibited platelet aggregation, intracellular $Ca^{2+}$ mobilization, and thromboxane $A_2$ ($TXA_2$) formation in collagen-stimulated platelets. Activated MMP-9 by GB directly affects down-regulations of cyclooxygenase-1 (COX-1) or $TXA_2$ synthase in a cell free system. In addition, activated MMP-9 significantly increased the formation of cyclic adenosine monophosphate (cAMP) and cyclic guanosine monophosphate (cGMP), which have the anti-platelet function in resting and collagen-stimulated platelets. Therefore, we suggest that activated MMP-9 by GB may increase the intracellular cAMP and cGMP production, inhibit the intracellular $Ca^{2+}$ mobilization and $TXA_2$ production, thereby leading to inhibition of platelet aggregation. These results strongly indicate that activated MMP-9 is a potent inhibitor of collagen-stimulated platelet aggregation. It may act a crucial role as a negative regulator during platelet activation.

The Effect of Pulsatile Versus Nonpulsatile Blood Flow on Viscoelasticity and Red Blood Cell Aggregation in Extracorporeal Circulation

  • Background: Extracorporeal circulation (ECC) can induce alterations in blood viscoelasticity and cause red blood cell (RBC) aggregation. In this study, the authors evaluated the effects of pump flow pulsatility on blood viscoelasticity and RBC aggregation. Methods: Mongrel dogs were randomly assigned to two groups: a nonpulsatile pump group (n=6) or a pulsatile pump group (n=6). After ECC was started at a pump flow rate of 80 mL/kg/min, cardiac fibrillation was induced. Blood sampling was performed before and at 1, 2, and 3 hours after ECC commencement. To eliminate bias induced by hematocrit and plasma, all blood samples were adjusted to a hematocrit of 45% using baseline plasma. Blood viscoelasticity, plasma viscosity, hematocrit, arterial blood gas analysis, central venous $O_2$ saturation, and lactate were measured. Results: The blood viscosity and aggregation index decreased abruptly 1 hour after ECC and then remained low during ECC in both groups, but blood elasticity did not change during ECC. Blood viscosity, blood elasticity, plasma viscosity, and the aggregation index were not significantly different in the groups at any time. Hematocrit decreased abruptly 1 hour after ECC in both groups due to dilution by the priming solution used. Conclusion: After ECC, blood viscoelasticity and RBC aggregation were not different in the pulsatile and nonpulsatile groups in the adult dog model. Furthermore, pulsatile flow did not have a more harmful effect on blood viscoelasticity or RBC aggregation than nonpulsatile flow.

발생계배 연골세포의 분화기구에 대한 연구 I. 세포응집과 분화와의 관계 (Studies on the Differentiation of Chondrogenic Cells in Developing Chick Embryo I. Cellular Aggregation and Chondrogenesis)

  • 연골세포 분화기구 연구의 기초단계로서 미세 세포배양법을 정립하였으며, 세포의 응집정도와 연골분화의 관계를 조사하기 위하여 계배 limb bod 간충직세포를 여러가지 농도로 micromass 배양하면서 세포농도에 따른 세포증식정도와 proteoglycan에 결합된 alcian blue의 양 및 [35 S] sulfate의 sulfate proteohlycan에 표지되는 속도를 측정하고, 면역조직화학법을 이용하여 type II collegen의 발현을 관찰하였다. 각 농도별로 배양한 Hambruger-Hamilton stage 23/24 간충직세포 중 5. $\times$ 106 cells/ml 이상의 농도로 배양한 세포는 연골세포로 분화하였으나, 저농도로 배양한 세포는 분화하지 않았다. 반면에 stage 18/19 간충직세포는 분화단계 중에서 동일세포들끼리 응집되는 단계이며 stage 23/24 간충직세포 이 단계를 지나 분화능을 갖는 세포응축의 단계인 것으로 생각된다. 본 연구 결과 세포응집 및 분화능은 간충직세포의 발생시기에 따라 다르며, 분화능을 갖기 위해서는 세포응집이 선행조건이고, 그 적정 미세 배양 농도는 5-10 $\times$ 106 cells/ml임을 알았다. 한편, hyaluronidase의 활성은 stage 23/24세포 배양 전 과정에서 비교적 일정한 것으로 나타나 이 시기의 세포분화에는 별로 중요하지 않는 것으로 보인다.

The Effect of Vibration on the Hemorheological Characteristics of Non-aggregated Blood

  • The present study investigates the hemorheological characteristics of blood flow with applying vibration to a non-aggregating red blood cell suspension. In order to obtain the non-aggregating RBC suspension, blood samples were treated with vibration at a specified condition, which viscosities were taken before and after the treatment, respectively. The viscosity of the blood samples after treatment was higher than before treatment. These treated blood samples were forced to flow through a capillary tube that was vibrated perpendicularly to the direction of the flow. The experimental results showed that vibration caused a reduction of the flow resistance of the non-aggregated blood. The reduction of the flow resistance was strongly dependent on both frequency and amplitude of vibration. These results show potential in treating various diseases in the microcirculation associated with blood cell aggregation.

The inhibitory mechanism of crude saponin fraction from Korean Red Ginseng in collagen-induced platelet aggregation

  • Background: Korean Red Ginseng has been used as a traditional oriental medicine to treat illness and to promote health for several thousand years in Eastern Asia. It is widely accepted that ginseng saponins, ginsenosides, are the major active ingredients responsible for Korean Red Ginseng's therapeutic activity against many kinds of illness. Although the crude saponin fraction (CSF) displayed antiplatelet activity, the molecular mechanism of its action remains to be elucidated. Methods: The platelet aggregation was induced by collagen, the ligand of integrin ${\alpha}_{II}{\beta}_I$ and glycoprotein VI. The crude saponin's effects on granule secretion [e.g., calcium ion mobilization and adenosine triphosphate (ATP) release] were determined. The activation of mitogen-activated protein kinases (MAPKs), including extracellular signal-regulated protein kinase 1/2 (ERK1/2), c-Jun N-terminal kinases (JNKs), and p38 MAPK, and phosphoinositide 3-kinase (PI3K)/Akt was analyzed by immunoblotting. In addition, the activation of integrin ${\alpha}_{II}b{\beta}_{III}$ was examined by fluorocytometry. Results: CSF strongly inhibited collagen-induced platelet aggregation and ATP release in a concentration-dependent manner. It also markedly suppressed $[Ca^{2+}]_i$ mobilization in collagen-stimulated platelets. Immunoblotting assay revealed that CSF significantly suppressed ERK1/2, p38, JNK, PI3K, Akt, and mitogen-activated protein kinase kinase 1/2 phosphorylation. In addition, our fraction strongly inhibited the fibrinogen binding to integrin ${\alpha}_{IIb}{\beta}_3$. Conclusion: Our present data suggest that CSF may have a strong antiplatelet property and it can be considered as a candidate with therapeutic potential for the treatment of cardiovascular disorders involving abnormal platelet function.

Feasibility of Coculture Method for Production of Chimeric Mice Using J1 Embryonic Stem Cells

  • The demand for the production of gene-defective mice from embryonic stem (ES) cells is increasing to clarify decisive gene function in vivo. Although blastocyst injection is widely used to generate ES cell-mediated knockout mice, coculture method has been alternatively used because of several advantages, such as low cost and simple procedure. Thus, this experiment was designed to demonstrate the feasibility of the coculture method using J1 ES cells, which are known to be efficient for blastocyst injection. Eight-cell embryos were harvested from 2.5 days post-coitum (dpc), denuded with acid tyrode's solution, and transferred onto trypsinized J1 ES cells. Aggregation was carried out following two typical methods, which are simple coculture method and aggregation in groove prepared by aggregation needle. Successfully aggregated-embryos were developed to blastocysts for 24 h and transferred into uterus of pseudo-pregnant foster mother. Chimeric offspring was judged by coat pigmentation. In this study, we could obtain chimeric mice from all the two aggregation methods, but the chimera production efficiencies in coculture using groove were three times higher at least than those in the other group. In conclusion, these observations suggest that coculture method should be available for production of knockout mice from J1 ES cells. Presently, the germ-line transmission rates of the chimeras produced from the two methods are under investigation.

Spatio-temporal Load Forecasting Considering Aggregation Features of Electricity Cells and Uncertainties in Input Variables

  • Spatio-temporal load forecasting (STLF) is a foundation for building the prediction-based power map, which could be a useful tool for the visualization and tendency assessment of urban energy application. Constructing one point-forecasting model for each electricity cell in the geographic space is possible; however, it is unadvisable and insufficient, considering the aggregation features of electricity cells and uncertainties in input variables. This paper presents a new STLF method, with a data-driven framework consisting of 3 subroutines: multi-level clustering of cells considering their aggregation features, load regression for each category of cells based on SLS-SVRNs (sparse least squares support vector regression networks), and interval forecasting of spatio-temporal load with sampled blind number. Take some area in Pudong, Shanghai as the region of study. Results of multi-level clustering show that electricity cells in the same category are clustered in geographic space to some extent, which reveals the spatial aggregation feature of cells. For cellular load regression, a comparison has been made with 3 other forecasting methods, indicating the higher accuracy of the proposed method in point-forecasting of spatio-temporal load. Furthermore, results of interval load forecasting demonstrate that the proposed prediction-interval construction method can effectively convey the uncertainties in input variables.

  • Ginseng has been widely studied due to its various therapeutic properties on various diseases such as cardiovascular disease (CVD). Cardiovascular disease has been canonically known to be caused by high levels of low-density lipoproteins (LDL) in the bloodstream, in addition to the impaired vasodilatory effects of cholesterol. However, current research on CVD has revealed a cascade of mechanisms involving a series of events that contribute to the progression of CVD. Although this has been elucidated and summarized in previous studies the detailed correlation between platelet aggregation and innate immunity that plays an important role in CVD progression has not been thoroughly summarized. Furthermore, immune cell subtypes also contribute to the progression of plaque formation in the subendothelial layer. Thrombus formation and the coagulation cascade also have a vital role in the progression of atherosclerosis. Hence, in this mini review we aim to elucidate, summarize, and propose the potent therapeutic effect of ginseng on CVD, mainly on platelet aggregation, plaque formation, and thrombus formation.

Effects of taurine on plasma and liver lipids, erythrocyte ouabain sensitive Na efflux and platelet aggregation in Sprague Dawley rats

  • The effects of taurine on plasma and liver cholesterol, erythrocyte ouabain sensitive Na efflux and platelet aggregation were examined in Sprague Dawley rats fed control or 0.5% cholesterol with 0.2% cholate diet. Plasma and liver levels of total cholesterol were increased significantly (p<0.05) in rats fed cholesterol diet compared to the control, and taurine significantly decreased the elevated plasma level of cholesterol in rats fed cholesterol diet (p<0.05). HDL-cholesterol was decreased in groups fed the cholesterol diet regardless of taurine supplementation and the difference between groups with and without cholesterol was significant (p<0.01). Plasma triglyceride was decreased and liver triglyceride was increased both significantly (p<0.05) in rats fed cholesterol compared to the control. Plasma and liver triglyceride in rats fed taurine was decreased significantly compared to the control (p<0.05). Intracellular Na tended to be lower in rats fed cholesterol or taurine and higher in rats fed cholesterol plus taurine compared to the control. Na efflux through Na-K ATPase and the passive leak of Na was somewhat reduced in rats fed cholesterol or taurine and was augmented in rats fed cholesterol plus taurine compared to the control, which showed a similar trend to the intracellular Na. Taurine supplementation caused a suppression of Na efflux in groups fed control diet and restored the suppressed Na efflux in groups fed cholesterol. Platelet aggregation was significantly decreased in the group fed taurine compared to the control (p<0.05) and the group fed cholesterol plus taurine was also a little lower in aggregation than the group fed cholesterol. Microscopic examination showed that taurine prevented fatty liver in rats fed cholesterol diet. Taurine known for stimulating Na-K ATPase in some cell types rather decreased erythrocyte ouabain sensitive Na-K ATPase in the present study. Taurine had hypolipidemic and hypocholesterolemic effects and inhibited platelet aggregation which may be favorable for prevention of cardiovascular diseases.
