동면중인 박쥐의 시상하부에 대한 전자현미경적 연구 I. 신경세포의 미세구조 (An Electron Microscopic Study on the Hypothalamus of the Hibernating Bat I. Fine Structure of the Nerve Cell)

  • The posterior hypothalamus of the hibernating greater horseshoe bats (Rhinolophus ferrumequinum korai Kuroda) were observed with an electron microscope. The posterior hypothalamus is known to be closely related to the reflex responses activated by cold, and the following observations were obtained in the cellular type of nerve cells: there are three types of neurons in the posterior hypothalamus. 1. The first type of neuron was the largest, ovoid or conical in shape, the nucleus was elliptic and the nuclear envelope had many deep invaginations. The cell organelles were well developed, in particular there was an abundance of variously shaped mitochondria, and the Golgi complex and the polysomes were observed in the cytoplasm. 2. The second type of neuron was moderate in size, ovoid or elliptic in shape, the nucleus was located nearer to the plasma membrane and the nuclear envelope had. a few invaginations. The cytoplasm was rich in amount compared with that of the third type of neuron, and the cell organelles, especially the rough endoplasmic reticulum were well developed. Also lipofuscin pigments were observed. 3. The third type of neuron was the smallest in size and round in shape. The nucleus and the nucleolus were observed in the central portion of the cell body and the nuclear envelope had a few invaginations. The cytoplasm was small compared with those of the first and second types, but the rough endoplasmic reticulum, the mitechondria and the polysomes were relatively well developed. The cytoplasm was characterized by the presence of membrane-bound small bodies with a single membrane containing a fine particular substance around the rough endoplasmic reticulum and the Golgi complexes.

마산만 식물플랑크톤의 단기적 변화양상 : 1. 동태 (Short Term variability of the Phytoplankton Populations in Masan Bay: I. Dynamics)

  • 마산만은 부영영화의 영향으로 적조와 무산소층이 빈발하는 해역으로 알려져 있 다. 이 해역의 빠른 turnover rate를 고려할 때 보다 짧은 관측 간격을 필요로 하므 르, 1988년 여름 장마 직후 16일간 일일관측을 실시하였다. 비교적 짧은 조사기간에도 불구하고 식물플랑크톤의 양 및 조성에 큰 변화가 있었다. 세포수와 클로로필 농도는 각각 70, 10배의 변화폭을 보였다. 식물플랑크톤 현존량의 첫 번째 최고점에서는 규조 류인 Skeletonema costatum이 압도적 우점종으로 나타났고, 이는 약 1주일 후의 두 번 째 최고점에서 Prorocentrum minimum으로 우점종의 천이가 일어났다. 이러한 변화의 시간 규모로 볼 때 마산만과 같은 복잡한 연안환경에서는 적어도 1주 간격의 조사가 고려되어야 할 것이다. 성층은 표층에서 높은 성장을 가능하게 했던 반면, 저층에서 표층으로의 규산염 공급을 방해하여 규조류의 지속적인 성장을 제한한 것으로 보인다. 표층에서 잇따라 일어났던 규산염 및 규조류 양의 두 번째 최고점은 조류 (tidal current)에 의한 저층으로부터 불연속면을 거쳐 일어나는 규산염의 주기적 (flux)를 시사한다.

A Proteomics Based Approach Reveals Differential Regulation of Visceral Adipose Tissue Proteins between Metabolically Healthy and Unhealthy Obese Patients

  • Obesity and the metabolic disorders that constitute metabolic syndrome are a primary cause of morbidity and mortality in the world. Nonetheless, the changes in the proteins and the underlying molecular pathways involved in the relevant pathogenesis are poorly understood. In this study a proteomic analysis of the visceral adipose tissue isolated from metabolically healthy and unhealthy obese patients was used to identify presence of altered pathway(s) leading to metabolic dysfunction. Samples were obtained from 18 obese patients undergoing bariatric surgery and were subdivided into two groups based on the presence or absence of comorbidities as defined by the International Diabetes Federation. Two dimensional difference in-gel electrophoresis coupled with matrix-assisted laser desorption/ionization time-of-flight mass spectrometry was carried out. A total of 28 proteins were identified with a statistically significant difference in abundance and a 1.5-fold change (ANOVA, $p{\leq}0.05$) between the groups. 11 proteins showed increased abundance while 17 proteins were decreased in the metabolically unhealthy obese compared to the healthy obese. The differentially expressed proteins belonged broadly to three functional categories: (i) protein and lipid metabolism (ii) cytoskeleton and (iii) regulation of other metabolic processes. Network analysis by Ingenuity pathway analysis identified the $NF{\kappa}B$, IRK/MAPK and PKC as the nodes with the highest connections within the connectivity map. The top network pathway identified in our protein data set related to cellular movement, hematological system development and function, and immune cell trafficking. The VAT proteome between the two groups differed substantially between the groups which could potentially be the reason for metabolic dysfunction.

Anti-inflammatory Effects of Various Mushrooms in LPS-stimulated RAW264.7 Cells

  • Mushrooms have been widely cultivated and consumed as foods and herbal medicines owing to their various biological properties. However, few studies have evaluated the anti-inflammatory effects of mushrooms. Here, we investigated the effects of mushroom extracts (MEs) on lipopolysaccharide (LPS)-induced inflammation in macrophages (RAW264.7 cells). First, we extracted MEs with either water or ethanol. Using LPS-treated RAW264.7 cells, we measured cell proliferation and NO production. Gene expression of tumor necrosis $factor-{\alpha}$ ($TNF-{\alpha}$), interleukin (IL)-6 (IL-6), and $IL-1{\beta}$ was assessed by RT-PCR, and protein abundance of inducible NO synthase (iNOS) and cyclooxygenase-2 (COX-2) and phosphorylation of p65 were determined by immunoblotting. MEs prepared using both water and ethanol inhibited LPS-induced inflammation in RAW264.7 cells. Nitric oxide (NO) levels induced by LPS were reduced by treatment with MEs. Isaria japonica Yasuda water extracts and Umbilicaria esculenta (Miyoshi) Minks ethanol extracts significantly decreased the mRNA expression of inflammation-related cytokine genes including $TNF-{\alpha}$, IL-6, and $IL-1{\beta}$. Similarly, the protein abundance of iNOS and COX-2 was also decreased. The phosphorylation of p65, a subunit of nuclear $factor-{\kappa}B$ was at least partly suppressed by MEs. This study suggests that mushrooms could be included in the diet to prevent and treat macrophage-related chronic immune diseases.

Identification of Histone Deacetylase 2 as a Functional Gene for Skeletal Muscle Development in Chickens

  • A previous genome-wide association study (GWAS) exposed histone deacetylase 2 (HDAC2) as a possible candidate gene for breast muscle weight in chickens. The present research has examined the possible role of HDAC2 in skeletal muscle development in chickens. Gene expression was measured by quantitative polymerase chain reaction in breast and thigh muscles during both embryonic (four ages) and post-hatch (five ages) development and in cultures of primary myoblasts during both proliferation and differentiation. The expression of HDAC2 increased significantly across embryonic days (ED) in breast (ED 14, 16, 18, and 21) and thigh (ED 14 and 18, and ED 14 and 21) muscles suggesting that it possibly plays a role in myoblast hyperplasia in both breast and thigh muscles. Transcript abundance of HDAC2 identified significantly higher in fast growing muscle than slow growing in chickens at d 90 of age. Expression of HDAC2 during myoblast proliferation in vitro declined between 24 h and 48 h when expression of the marker gene paired box 7 (PAX7) increased and cell numbers increased throughout 72 h of culture. During induced differentiation of myoblasts to myotubes, the abundance of HDAC2 and the marker gene myogenic differentiation 1 (MYOD1), both increased significantly. Taken together, it is suggested that HDAC2 is most likely involved in a suppressive fashion in myoblast proliferation and may play a positive role in myoblast differentiation. The present results confirm the suggestion that HDAC2 is a functional gene for pre-hatch and post-hatch (fast growing muscle) development of chicken skeletal muscle.

Thiazolidinedione 유도체(TD49) 물질을 이용한 적조생물 Akashiwo sanguinea의 제어 (Algicidal Effects of a Newly Developed Thiazolidinedione Derivative, TD49, on Dinoflagellate Akashiwo sanguinea)

  • To evaluate the algicidal impact of a newly developed algicide thiazolidinedione derivative, TD49, on dinophyceae Akashiwo sanguinea in aquatic ecosystems, tentative culture experiments for the target species were conducted in small (SS), middle (MS), and large scale (LS) culture vessels. When TD49 was introduced at the final concentration of $2{\mu}M$ in SS and MS, as well as $1{\mu}M$ in LS, the abundance of A. sanguinea decreased significantly in all the treatments. On the other hand, total phytoplankton abundance, except A. sanguinea in the TD49 treatments, gradually increased with culture time, which implies that a cell destruction of A. sanguinea by TD49 is a major cause of the population growth by other phytoplankton species. Also, A. sanguinea was easily destroyed, which was likely to be a source of extracellular substances. In particular, a pH decrease was significant in the treatments than in the control, which indicates that the water in the treatments has been acidified, due to an increase in the heterotrophic metabolisms of bacteria and degradation of A. sanguinea cells. Our results indicate that the TD49 substance is the potential agents for the control of A. sanguinea in the enclosed and eutrophic water bodies.

가열 및 저장조건에 따른 호박즙의 이화학적 성분변화 (Changes in Physicochemical Components of Stewed Pumpkin Juice Heated and Stored under Different Conditions)

  • This study was designed to determine the most suitable manufacturing conditions of stewed pumpkin juice(SPJ) in terms of nutrition and taste. Well ripened pumpkin was heated for 3, 4, 5, 6 and 7 hours in a pressure cooker and then extracted and packed in retort pouches. These were kept at 4$^{\circ}C$ and 28$^{\circ}C$ for 30 days, respectively and changes of the physicochemical components of SPJ were investigated. The free amino acid compositions of pumpkin and SPJ occupied asparagine, aspartic acid, glutamine, arginine and serine in increasing order of abundance. Except SPJ heated for 3 hours, SPJ stored at 28$^{\circ}C$ had the most abundant free amino acids, followed by SPJ stored at 4$^{\circ}C$, and SPJ not stored. The fatty acid composition of pumpkin occupied behenic acid, erucic acid, palmitic acid, linoleic acid, heneicosanoic aicd, in order of abundance. When SPJ was heated over 3 hours, palmitic acid and linolenic acid decreased greatly. SPJ stored at 4$^{\circ}C$ and 28$^{\circ}C$ increased contents of saturated fatty acids such as behenic acid, palmitic acid than SPJ not stored. As SPJ was heated for a long time, carotenoid was mostly destroyed and the Hunter "L", "a" and "b" values of SPJ decreased. Among the mineral constituents of SPJ, Na and Ca were dominantly occupied. It was found that the range of the total visible cell count was 3~4CFU/ml from the SPJ which was heated for 3 hours and it increased by 1$\times$109CFU/ml when the SPJ was stored at 28$^{\circ}C$. It seems that the contamination by microorganism have occurred at packing process. In sensory evaluation, the SPJ which was heated for 5 hours had the highest scores in overall preference, sweet smell and sweetness.ce, sweet smell and sweetness.

Transient Receptor Potential Cation Channel V1 (TRPV1) Is Degraded by Starvation- and Glucocorticoid-Mediated Autophagy

  • A mammalian cell renovates itself by autophagy, a process through which cellular components are recycled to produce energy and maintain homeostasis. Recently, the abundance of gap junction proteins was shown to be regulated by autophagy during starvation conditions, suggesting that transmembrane proteins are also regulated by autophagy. Transient receptor potential vanilloid type 1 (TRPV1), an ion channel localized to the plasma membrane and endoplasmic reticulum (ER), is a sensory transducer that is activated by a wide variety of exogenous and endogenous physical and chemical stimuli. Intriguingly, the abundance of cellular TRPV1 can change dynamically under pathological conditions. However, the mechanisms by which the protein levels of TRPV1 are regulated have not yet been explored. Therefore, we investigated the mechanisms of TRPV1 recycling using HeLa cells constitutively expressing TRPV1. Endogenous TRPV1 was degraded in starvation conditions; this degradation was blocked by chloroquine (CLQ), 3MA, or downregulation of Atg7. Interestingly, a glucocorticoid (cortisol) was capable of inducing autophagy in HeLa cells. Cortisol increased cellular conversion of LC3-I to LC-3II, leading autophagy and resulting in TRPV1 degradation, which was similarly inhibited by treatment with CLQ, 3MA, or downregulation of Atg7. Furthermore, cortisol treatment induced the colocalization of GFP-LC3 with endogenous TRPV1. Cumulatively, these observations provide evidence that degradation of TRPV1 is mediated by autophagy, and that this pathway can be enhanced by cortisol.

헴철이 풍부한 영양원이 혐기성 세균의 생장과 생존에 미치는 영향: 락토바실러스 가세리 모델연구 (Effect of Heme-rich Nutrient on Anaerobic Bacterial Growth and Survival: A Model Study on Lactobacillus gasseri)

  • Lactic acid bacteria (LAB), belonging to the Firmicutes phylum, lack heme biosynthesis and, thus, are characterized as fermentative and catalase-negative organisms. To verify the hypothesis that heme-rich-nutrients might compensate the heme-biosynthesis incapability of non-respiratory LAB in animal gut, a heme-rich-nutrient was fed to a dog and its fecal microbiome was analyzed. Firmicutes abundance in the feces from the heme-rich-nutrient-fed dog was 99%, compared to 92% in the control dog. To clarify the reason of increased Firmicutes abundance in the feces from the heme-rich-nutrient-fed dog, Lacobacillus gasseri were used as model anerobic LAB to study a purified heme (hemin). The anaerobic growth of L. gasseri in the medium with 25 µM hemin supplementation was faster than that in the medium without hemin, while the growth in the 50 µM hemin-supplemented medium did not vary. Cellular activities of the cytochrome bd complex were 1.55 ± 0.19, 2.11 ± 0.14, and 2.20 ± 0.08 U/gcell in the cells from 0, 25, and 50 µM hemin-supplemented medium, while intracellular ATP concentrations were 7.90 ± 1.12, 11.95 ± 0.68, and 12.56 ± 0.58 µmolATP/gcell, respectively. The ROS-scavenging activities of the L. gasseri cytosol from 25 µM and 50 µM hemin-supplemented medium were 68% and 82% greater than those of the cytosol from no hemin supplemented-medium, respectively. These findings indicate that external hemin could compensate the heme-biosynthesis incapability of L. gasseri by increasing the cytosolic ROS-scavenging and extra ATP generation, possibly through increasing the electron transfer. Increase in the number of anaerobic bacteria in heme-rich-nutrient-fed animal gut is discussed based on the results.

Cheonggukjang Fermented with Bacillus subtilis SCGB574 Ameliorates High Fat Diet-Deteriorated Large Intestinal Health in Rat Model

  • Cheonggukjang is a traditional fermented food in Korea, which is known to exert beneficial effects on health. In this study, we evaluated the effects of cheonggukjang fermented by Bacillus subtilis SCGB 574 (B574) on high fat diet (HFD)-deteriorated large intestinal health. Rats were fed with HFD or HFD supplemented with 10.1% cheonggukjang (B574). Fecal microbiota was analyzed based on 16S rRNA gene sequences, and the fecal and serum metabolome were measured using GC-MS. Our results showed that SCGB574 intake significantly reduced body weight, restored tight junction components, and ameliorated inflammatory cell infiltration. SCGB574 also shifted gut microbiota by increasing the abundance of short chain fatty acid producers such as Alistipes and Flintibacter, although it decreased the abundance of Lactobacillus. Serum and fecal metabolome analyses showed significantly different metabolic profiles between the groups. The top five metabolites increased by SCGB574 were i) arginine biosynthesis, ii) alanine, aspartate, and glutamate metabolism; iii) starch and sucrose metabolism; iv) neomycin, kanamycin, and gentamicin biosynthesis; and v) galactose metabolism. These results showed that cheonggukjang fermented by SCGB574 ameliorates adverse effects of HFD through improving intestinal health.
