The concentrations of cadmium, metallothionein(MT), superoxide dismutase(SOD), and lactate dehydrogenase(LDH) were investigated in liver and kidney of rats which were fed the water containing 50 or 100ppm cadmium chloride with basal diet(group A), 5% horsetail diet(group, B), 5% mugwort diet(group C) and 5% champignon diet(group D) for weeks. Cadmium in liver decreased for the first 12 weeks of treatment, but thereafter increased, and was lower in experimental group B,C,D than in control group A. Cadmium in kidney increased linearly during the 16 weeks of treatment, and was lower in group B than in group A. MT in liver decreased for the first 12 weeks of treatment in group A, but increased linearly during the 16 weeks in group B,C,D, higher in group B than in group A. There were significantly higher accumulation of cadmium and MT in liver than in kidney in the beginning of cadmium treatment, but reversed in the ending of treatment. The SOD and LDH activities were not affected during the 16 weeks treatment, and there was no significant difference between groups. Histologic examination revealed moderate to severe hepatic and renal injury in group A compared to horsetail diet group B. These results indicate that the kidney is a major target organ of chronic cadmium poisoning, and suggest that Cd-induced hepatic injury, via release of Cd-MT, may play an important role in the nephrotoxicity. In addition, higher MT concentrations in liver and kidney in the group B constitute a plausible explanation of the protective effects of horsetail diet against the cadmium toxicity in relation to histologic findings.

Hemibarbus mylodon (Cypriniformes) is an endemic freshwater fish species in the Korean peninsula, for which urgent conservation efforts are needed. To understand their stress responses in relation to metal toxicity and thermal elevation, we performed a real-time RT-PCR-based expression assay of hepatic copper/zinc-superoxide dismutase (Cu/Zn-SOD), a key antioxidant enzyme, in response to experimental heavy metal exposure or heat treatment. The transcription of hepatic Cu/Zn-SOD was differentially modulated by acute exposure to Cu, cadmium (Cd), or Zn. Exposure to each metal at $5{\mu}M$ for 24 h revealed that Cu stimulated the mRNA expression of Cu/Zn-SOD to a greater extent than the other two heavy metals. The elevation in Cu/Zn-SOD transcripts in response to Cu exposure was dose-dependent (0.5 to $5{\mu}M$). Time course analysis of Cu/Zn-SOD expression in response to Cd exposure ($5{\mu}M$) revealed a transient pattern up to day 7. Exposure to thermal stress (an increase from 22 to $30^{\circ}C$ at a rate of $1^{\circ}C/h$ followed by $30^{\circ}C$ for 18 h) did not significantly alter SOD transcription, although heat shock protein 90 kDa (HSP90) transcription was positively correlated with an increase in temperature.

$Foxp3^+$$CD25^+CD4^+$ regulatory T (Treg) cells are crucial for the maintenance of immunological self-tolerance and are abundant in tumors. Most of these cells are chemo-attracted to tumor tissues and suppress anti-tumor responses inside the tumor. Currently, several cancer immunotherapies targeting Treg cells are being clinically tested. Cisplatin is one of the most potent chemotherapy drugs widely used for cancer treatment. While cisplatin is a powerful drug for the treatment of multiple cancers, there are obstacles that limit its use, such as renal dysfunction and the development of cisplatin-resistant cancer cells after its use. To minimize these barriers, combinatorial therapies of cisplatin with other drugs have been developed and have proven to be more effective to treat cancer. In the present study, we evaluated the effect of the combination therapy using methyl gallate with cisplatin in EL4 murine lymphoma bearing C57BL/6 mice. The combinatorial therapy of methyl gallate and cisplatin showed stronger anti-cancer effects than methyl gallate or cisplatin as single treatments. In Treg cell-depleted mice, however, the effect of methyl gallate vanished. It was found that methyl gallate treatment inhibited Treg cell migration into the tumor regardless of cisplatin treatment. Additionally, in both the normal and cisplatin-treated tumor-bearing mice, there was no renal toxicity attributed to methyl gallate treatment. These findings suggest that methyl gallate treatment could be useful as an adjuvant method accompanied with cisplatin therapy.
Particulate materials (PM) less than 10 ${\mu}m$ in diameter are of special interest in air pollution because they are respirable and responsible for the increasing mortality rate of lung cancer and cardiovascular diseases. These particles are often referred to as $PM_{10}$ and they are divided into a coarse fraction and a fine fraction which is also often referred to as $PM_{25}$. In this study, we monitored the TSP, $PM_{10},\;PM_{2.5}$ concentration of ambient air collected in northern part of Seoul in early spring and measured the concentration of heavy metals; Cr, Mn, Zn, As, Cd, and Pb. All the heavy metals were found in the collected particles and the concentrations were variable in the $PM_{10},\;and\;PM_{2.5}$ respectively. The detected concentration ranges were Cr: $ND\~2,889ng/m^3,\;Mn:2.4\~257.9ng/m^3,\;Zn:ND\~353.7ng/m^3,\; As:ND\~22.3ng/m^3,\;Cd:0.1\~2.9ng/m^3,\;and\;Pb:ND\~392.2ng/m^3$ in fine particles. Heavy metal toxicity of the particles were also tested in $H_9C_2$ cell line derived from rat cardiomyocytes. As for the results of health risk assessment calculated by unit risk of IRIS, heavy metals in ambient air of Seoul metropolitan area were found to be responsible for the increase of total excess cancer risk. Among them, chromium (hexavalent) was found to be the most risky in fine particles of ambient air collected in the northern part of Seoul in early spring.
Emission characteristics of air pollutants from three commercially operating cement kilns co-burning waste were investigated. The major heavy metals emitted were mercury (Hg), zinc (Zn), nickel (Ni), chromium (Cr), lead (Pb), cadmium (Cd), and arsenic (As) Removal efficiency of the bag filter was above 98.5% for heavy metals (except Hg), and above 60% for Hg. Higher fractions of heavy metals entering the bag filter were speciated to cement kiln dust. On average, 3.3% of the -heavy metals of medium and low toxicity (Pb, Ni, and Cr) entering the bag filter were released into the atmosphere. Among highly toxic heavy metals, 0.14% of Cd, 0.01% of As, and 40% of Hg entering the bag filter were released into the atmosphere. In passing through the bag filter, the proportion of oxidized Hg in all cases increased. Emission variations of hazardous air pollutants in cement kilns tested were related to raw materials, fuel, waste feed and operating conditions. Volatile organic compounds detected in gas emissions were toluene, acrylonitrile benzene, styrene, 1,3-butadiene, and methylene chloride. Although hazardous air pollutants in emissions from cement kilns co-burning waste were within the existing emission limit, efforts are required to minimize their levels.
In order to investigate the efficacy of BJBB on atopic dermatitis, various immune related factors were studied. The results and conclusions are as follows. Atopic dermatitis symptoms were improved in BJBB treated group and significant decrease in dermatitis index were observed in 13 weeks. ALT, AST and BUN, Cr levels were all with in the normal ranges in BJBB treated group, indicating no induced toxicity. BJBB treated group showed significant decrease in CD4+, CD11b+/Gr-1+ immune cell ratio in dorsal skin by 33% and 62% respectively. BJBB treated group showed significant decrease in the expression of IL-4, IL-5, IL-13 and histamine by 83%, 62%, 53% and 61% respectively. Also the group showed decrease in the transcription of IL-4, IL-5 and IL-13 mRNA in spleen by 41%, 52% and 50% respectively. BJBB treated group showed decrease in the expression of IgE by 57% respectively. The results above indicated that treatment of BJBB improved atopic dermatitis symptoms by immune modulation activity a clinical evidence. thus, BJBB has a potential use as a composition of medicinal plants for treatment against inflammation related disease.
Positive effect of multiple-PGPR (Plan Growth Promoting Rhizobacteria), isolated from heavy metal contaminated soil, on the germination of Barnyard grass (Echinochloa crus-galli var. frumentacea) was quantitatively estimated in 5 heavy metal (Cd, As, Ni, Cu, and Pb) contaminated liquid medium. The $EC_{50}$ value for respective heavy metal was estimated by TSK (Trimmed Speraman-Karber) model based on germination rate. The results showed overall increase in $EC_{50}$ with PGPR inoculation. The $EC_{50}$ value increased 1.4% from 96.0 mg/L (control) to 97.4 mg/L (PGPR-treated) in As contaminated medium. In Ni contaminated medium, the $EC_{50}$ value increased 31.9% from 148.0 mg/L (control) to 195.2 mg/L (PGPR-treated), while the $EC_{50}$ showed 4.8% increase from 63.4 mg/L (control) to 66.5 mg/L (PGPR-treated) in Cu medium. Overall seedling growth was stronger in the PGPR treated seeds than that in the control, but positive effect on seedling growth was not conspicuous. At effective concentration of 100 mg/L, the average seedling length of the PGPR treatment in As, Cd, Cu, and Ni medium, respectively, was 1.13, 0.14, 0.40, and 0.06 cm longer than that in the control. However, the increase of seedling growth was statistically insignificant (p < 0.05). These results suggest that inoculation of the isolated-PGPR exerts positive effects on seed germination by reducing heavy metal toxicity and can be an effective tool for application of phytoremediation on heavy metal contaminated soils.
Pollutants that are persistent, bioaccurnulative, and toxic have been linked to numerous adverse effects in human and animals, PBTs include heavy metals, polychlorinated biphenyls (PCBs), dioxins, polycyclic aromatic compounds (PACs) in addition to pesticides. This study focuses on toxic effects of the PBTs except pesticides on insects. Eight PBTs were selected from subgroups: three heavy metals (Pb, Hg, and Cd), two PCB mixtures (Aroclor mixtures 1 and 2), 2,3,7,8-tetrachlorodibenzo-p-dioxin, two monophenols (4-octylphenol and 4-nonylphenol), and tetrabutyltin, Beet armyworm, Spodoptera exigua, was used as test target insect species. Three physiological markers (metamorphosis, immune reaction, and follicle patency) were assessed in each exposure to different doses of the PCBs. Heat-shock proteins as molecular markers were also analyzed in response to the PCBs. All tested PBTs were toxic to metamorphosis from larvae to pupae when they were applied with diet. Two PCB mixtures were the most toxic compounds in this assay by giving significant toxicity at 0.005 ppm, while others had from 10 to 1000 ppm. Dioxin (0.1 ppb), tetrabutyltin (0.1 ppb), Pb (10 ppb), and Hg (0,01 ppb) were potent to inhibit immune reactions analyzed by inducing phenoloxidase activity and blocked phospholipase $A_2$ enzyme, Tetrabutyltin and dioxin significantly induced follicle cell patency, but their effects were lower than that of endogenous juvenile hormone, Dioxin, Pb, Hg, and Cd could induce the expression of heat shock proteins that were detected by immunoblotting against human HSP70 monoclonal antibody. HSP78 and HSP80 were upregulated in response to the PBTs. This expression was detected from the fat body and epidermis at as fast as 4h after injection. All these results clearly suggest that PBTs give significant ecotoxicity to insects that are valuable organisms in our environment.
