Background: Artesunate, extracted from Artemisia annua, has been proven to have anti-cancer potential. Allicin, diallyl thiosulfinate, the main biologically active compound derived from garlic, is also of interest in cancer treatment research. This object of this report was to document synergistic effects of artesunate combined with allicin on osteosarcoma cell lines in vitro and in vivo. Methods: After treatment with artesunate and allicin at various concentrations, the viability of osteosarcoma cells was analyzed by MTT method, with assessment of invasion and motility, colony formation and apoptosis. Western Blotting was performed to determine the expression of caspase-3/9, and activity was also detected after drug treatment. Moreover, in a nude mouse model established with orthotopic xenograft tumors, tumor weight and volume were monitored after drug administration via the intraperitoneal (i.p.) route. Results: The viability of osteosarcoma cells in the combination group was significantly decreased in a concentration and time dependent manner; moreover, invasion, motility and colony formation ability were significantly suppressed and the apoptotic rate was significantly increased through caspase-3/9 expression and activity enhancement in the combination group. Furthermore, suppression of tumor growth was evident in vivo. Conclusion: Our results indicated that artesunate and allicin in combination exert synergistic effects on osteosarcoma cell proliferation and apoptosis.
Ikwi-tang (IW) is an oriental medicine that has been used for the treatment of general symptoms due to a stomach yin deficiency. The aim of this study is to investigate precisely the effect of IW on allergic rhinitis (AR). We well show the effects and the mechanism of IW in the ovalbumin-sensitized AR model. IW significantly decreased the number of nasal/ear rubs and the increment of IgE levels in the AR mice. The levels of interferon-${\gamma}$ were enhanced while the levels of interleukin (IL)-4 were reduced in the spleen tissue of the IW-administered AR mice. Expressions of IL-$1{\beta}$ and cyclooxygenase-2 were inhibited by IW-administration in the nasal mucosa tissues. Infiltration of eosinophils and mast cells was decreased in the IW-administered AR mice. Our results indicate that IW may attenuate the development of AR by the inhibition of caspase-1 activity.
Ethanol treatment during the brain growth spurt period has been known to induce the death of Purkinje cells. The underlying molecular mechanisms and the role of reactive oxygen species (ROS) in triggering ethanol-induced Purkinje cell death are, however, largely unresolved. We undertook TUNEL staining, western blotting assay and immunohistochemistry for the cleaved forms of caspase-3 and -9, with calbindin D28K double immunostaining to identify apoptotic Purkinje cells. The possibility of ROS-induced Purkinje cell death was immunohistochemically determined by using anti-8-hydroxy-2'deoxyguanosine (8-OHdG), a specific cellular marker for oxidative damage. The results show that Purkinje cell death of PD 5 rat cerebellum following ethanol administration is mediated by the activation of caspase-3 and -9. However, unexpectedly, TUNEL staining did not reveal any positive Purkinje cells while there were some TUNEL-positive cells in the internal and external granular layer. 8-OHdG was detected in the Purkinje cell layers at 8 h, peaked at 12-24 h, but not at 30 h post-ethanol treatment. No 8-0HdG immunoreactive cells were detected in the internal and external granular layer. The lobule specific 8-OHdG staining patterns following ethanol exposure are consistent with that of ethanol-induced Purkinje cell loss. Thus, we suggest that ethanol-induced Purkinje cell death may not occur by the classical apoptotic pathway and oxidative damage is involved in ethanol-induced Purkinje cell death in the developing cerebellum.

Minocycline is known to protect neurons from microglia-mediated cell death in many experimental models of brain diseases including ischemic stroke, Huntington's disease (HD), amyotrophic lateral sclerosis (ALS), traumatic brain injury, multiple sclerosis, and Parkinson's disease. Activation of caspase-2, 3, 8, and 9 was evident within 2-8 hr following oxidative insult with 0.5 mM hydrogen peroxide in PC12 cells. Minocycline significantly attenuated activation of these caspases up to 18 hr, resulting a significant increase in cell viability as assessed by MTT assay. (omitted)
전립선 암은 현대 남성에게 걸리기 쉬운 질환으로 매년 점점 증가하는 암 사망률 중 하나이다. 하지만 남성호르몬 비의존형 전립선 암 치료에 대한 효과가 거의 없어 이에 본 연구에서는 남성호르몬 비의존형 전립선 암세포인 PC-3 세포에서 감마선이 세포 성장 억제와 세포자멸사 기작에 대해 알아보고자 한다. 그리하여 본 연구에서는 5가지 방법으로 즉, 세포증식 억제, apoptotic cell의 형태학적 변화, DNA 분절 분석, AV/PI 염색, western blot 분석법을 사용하여 수행하였다. 본 연구의 결과로 감마선을 처리한 PC-3세포에서 형태학적(분절화) 변화와 DNA ladder가 관찰되었다. 또한 감마선을 처리한PC-3세포에서는 apoptosis와 관련된 Caspase3와 PARP cleavage가 유발되었고 Bax 단백질 증가도 보였다.
목 적 : Artemisinin은 인체에 대한 부작용이 적고 항말라리아 효능 뿐 아니라 강력한 항암효과가 있음이 알려져 있다. 저자들은 오까야마 약학대학에서 합성 된 350여개의 endoperoxide ring구조를 가진 artemisinin 유도체를 선별검사 하여 HL-60세포 주에 강력한 세포독성을 보여 준 c-127의 세포고사 유도 여부와 그 분자유전학적 기전을 알아보고자 하였다. 방 법 : HL-60세포는 RPMI 1640배지로 배양하였고 세포 생존율은 MTT분석으로 측정하였다. 세포고사 여부는 DNA추출과 전기영동법으로 확인하였으며 세포고사 유도 기전은 western blotting을 시행하여 알아보았다. 결 과 : C-127이 세포고사를 유도하여 HL-60세포의 세포 생존력을 농도 의존적으로 감소시켰다. C-127의 이런 항암효과의 분자 유전학적 기전으로는 caspase-8,3 활성화, Bcl-2 family인 Bid분절, JNK 인산화와 c-Jun 발현이 관여하였다. 결 론 : C-127이 HL-60 세포에서 세포고사를 유도하여 강력한 항암효과를 발현하였고, 그 기전으로 caspase cascade, Bcl-2 family, JNK인산화와 c-Jun활성화가 관여하였다.
PI3K/Akt 신호계는 세포 생존의 조절에 필수적인 경로로 대부분 암세포에서 활성이 증대되어 있다. 본 연구에서는 동충하초의 주요 생리활성 물질인 cordycepin에 의한 AGS 인체 위암 세포의 apoptosis 유도에 미치는 PI3K/Akt 신호계의 역할을 조사하였다. 본 연구의 결과에 의하면 cordycepin의 처리 농도의 증가에 따라 AGS 세포의 생존율은 억제되었으며, 이는 apoptosis 유도와 밀접한 관계가 있음을 핵의 형태적 변화와 flow cytometry 분석을 통하여 확인 하였다. 이러한 cordycepin의 apoptosis 유도 효과는 PI3K/Akt 신호계의 활성 저하와 연관성이 있었으며, 세포독성을 나타내지 않는 범위의 PI3K/Akt 신호계 저해제인 LY294002를 cordycepin과 동시 처리하였을 경우, cordycepin에 의한 apoptosis 유발을 더욱 증대시켰다. 그리고 cordycepin에 대한 LY294002의 apoptosis 유발 증대는 caspases (caspase-3, -8 및 -9)의 활성 증가와 poly(ADP-ribose) polymerase 단백질의 분해 증가를 촉진했다. 또한 cordycepin이 처리된 AGS 세포에서 LY294002는 apoptosis 유도에 관여하는 Bax의 발현을 증가시켰고 apoptosis 억제에 관여하는 Bcl-2의 발현은 감소시켰으며, 이는 미토콘드리아 기능 손상과 미토콘드리아에서 세포질로의 cytochrome c 유리를 증대시켰다. 따라서 PI3K/Akt 신호계의 활성 차단은 cordycepin의 항암 활성을 더욱 상승시켰으며, 이는 미토콘드리아 기능 손상과 caspase의 활성 증대를 통하여 이루어짐을 알 수 있었다.
오미자 종자에서 추출된 정유(Schisandrae Semen essential oil, SSeo)의 항암활성 및 작용 기전 해석을 위하여 U937 백혈병 세포를 대상으로 apoptosis 유도 여부를 조사하였다. SSeo 처리에 의한 U937 세포의 증식 억제는 apoptosis 유도와 연관성이 있음을 DAPI 염색을 통한 apoptotic body 출현의 증가, agarose gel 전기영도에 의한 DNA의 단편화 유도 및 flow cytometry 분석에 의한 Sub-G1기 세포 빈도의 증가로 확인하였다. SSeo 처리에 의한 apoptosis 유도에서 IAP family 단백질에 속하는 XIAP, cIAP-1 및 survivin의 발현 감소와 anti-apoptotic Bcl-2 단백질의 발현 저하, DR4 및 DR5의 발현 증가와 연관성이 있었다. SSeo 처리는 또한 Bid truncation, 미토콘드리아 기능 손상, caspases (-3, -8 and -9)의 활성화와 활성형 caspase-3의 기질 단백질인 PARP의 단편화를 동반하였다. 본 연구의 결과는 오미자 정유의 생화학적 항암기전 해석을 이해하고 향후 지속적인 연구를 위한 기초자료로서 활용될 수 있을 것으로 생각된다.
Objectives : The purpose of this research was to investigate the cytotoxic effect of Natural Killer(NK)-92 cell and Snake Venom, and to elucidate its mechanism on human lung carcinoma cell A549. Methods : In order to figure out whether Snake Venom enhances the cytotoxic effect of NK-92 cell in A549 cell, Cell Viability Assay was conducted. Also, in order to observe the changes of Caspase-3 and Caspase-8, both of which are proteinases that advance apoptosis, and the changes of TNRF and DR3, which are Death Receptors of the extrinsic pathway of apoptosis, Western Blot Analysis was conducted. By conducting RT-PCR analysis, we have tried to confirm Perforin, Granzyme B, and GADPH, all of which are cytotoxic-related proteins. Lastly, in order to observe the effect of Snake Venom on NO formation within human lung carcinoma cells, NO determination was conducted. Results : 1. After conducting Cell Viability Assay, Snake Venom enhanced the cytotoxic effect of NK-92 cell and inhibited the growth of A549. 2. Western Blot Analysis caused proteinases Caspase-3 and Caspase-8, which advance apoptosis, to increase in the combined treatment group, but not in treatment groups that focused only on either Snake Venom or NK-92 cell in A549 lung carcinoma cells. 3. Western Blot Analysis caused an expression of TNFR2 and DR3, both of which are Death Receptors of the apoptosis extrinsic pathway, in the combined treatment group, but not intreatment groups that focused only on either Snake Venom or NK-92 cell in A549 human lung carcinoma cells. 4. After conducting NO determination, NO formation within A549 cell showed no significant changes in both treatment groups that focused NK-92 cell and combined treatment group. 5. After conducting RT-PCR, the expression of Granzyme B and Perforin, which are cytotoxic-related proteins within A549 human lung carcinoma cells, showed growth in the combined treatment group, but not the treatment group that focused only on NK-92 cell. Conclusion : It has been indicated that, when it comes to the A549 cell, Snake Venom enhances the increase of Death Receptor expression and continuous apoptosis reaction, leading to the enhancement of the cancer cell cytotoxic effect of the NK-92 cell. It is expected that Snake Venom can be used with the NK-92 cell for further lung cancer treatment.
Oral squamous cell carcinoma is the most prevalent oral cancer, which is characterized by its high metastasis and recurrent rates and poor prognosis. Taxol is an anticancer agent which is microbial products extracted from jew tree. It combines with the tubulin and induces apoptosis by inhibiting mitosis of cell with microtubule stabilization. Recently, it was reported to be effective in various solid tumors, but only very slight effect has been seen in oral squamous cell carcinomas due to its cell-specific potencies. Cyclosporin A is used as immune suppressant and is being applied in anticancer therapy as its mechanism of induction of change of apoptotic process in various cells have been known. In this study, oral squamous cell carcinoma HN22 cell line was used for in vitro experiment and as for the experimental group taxol and cyclosporin A were applied alone and to observe the synergistic effect of apoptosis, Taxol and cyclosporin A were coadministered with different concentration of taxol for comparison. The results were obtained as follow: 1. There was no difference in Bcl-2, Bax, caspase 3, 8, 9 mRNA expression when cyclosprin A or taxol was applied alone to HN 22 cell line. 2. Caspase 3, 9 mRNA expression was prominently increased when cyclosprin A and taxol were applied together to cancer cell. 3. No significant difference was observed when cyclosporin A and taxol($1{\mu}g/ml$ and $3{\mu}g/ml$) were applied together to cancer cell line. 4. No significant difference was seen in Bcl-2, Bax, and caspase 8 mRNA expression in all the groups of in vitro experiments. 5. When cyclosporin A was applied alone in vivo study on the nude mice, histopathologi cal findings was similar to those of the control group. Oral squamous cell carcinoma induced by inoculation of HN 22 cell line was not reduced after treatment of cyclosporin A. 6. When taxol was applied alone, the islands of squamous cell carcinoma still remained, which meant insignificant healing effect. There was a lesser volume increase compared with the cyclosporin A alone. 7. When taxol and cyclosporin A were applied together, the connective tissue and calcification were seen in the histopathologic findings. Oral squamous cell carcinoma was decreased and cancer cell was disappeared. In observing the tumor mass change with time, there was a gradual decreased size and healing features. As the results of the in vitro experiment, it could conclud that only when the two agents are applied together, mitochondria-mediated apoptosis occurred by considerable increase of caspase 3, 9 mRNA expression, irrespectable of the concentration of taxol. In vivo experiment, there was a discrete synergistic effect when the two agents were applied together. But single use of cyclosporin A was not effective in this study. Based on the results of this experiment, if further clinical studies are done, taxol and cyclosporin A could be effectively used in treatment of oral squamous cell carcinomas.
