Manufacture and Physicochemical Properties of Chitosan Oligosaccharide/A2 β-Casein Nano-Delivery System Entrapped with Resveratrol

  • The purposes of this research were to form chitosan oligosaccharide (CSO)/A2 ${\beta}$-casein nano-delivery systems (NDSs) and to investigate the effects of production variables, such as CSO concentration levels (0.1%, 0.2%, and 0.3%, w/v) and manufacturing temperature ($5^{\circ}C$, $20^{\circ}C$, and $35^{\circ}C$), on the production and physicochemical characteristics of CSO/A2 ${\beta}$-casein NDSs to carry resveratrol. The morphological characteristics of CSO/A2 ${\beta}$-casein NDSs were assessed by the use of transmission electron microscopy (TEM) and particle size analyzer. High-performance liquid chromatography (HPLC) was applied to determine the entrapment efficiency (EE) of resveratrol. In the TEM images, globular-shaped particles with a diameter from 126 to 266 nm were examined implying that NDSs was successfully formed. As CSO concentration level was increased, the size and zeta-potential values of NDSs were significantly (p<0.05) increased. An increase in manufacturing temperature from $5^{\circ}C$ to $35^{\circ}C$ resulted in a significant (p<0.05) increase in the size and polydispersity index of NDSs. Over 85% of resveratrol was favorably entrapped in CSO/A2 ${\beta}$-casein NDSs. The entrapment efficiency (EE) of resveratrol was significantly (p<0.05) enhanced with an increase in manufacturing temperature while CSO concentration level did not significantly affect EE of resveratrol. There were no significant (p<0.05) changes observed in the size and polydispersity index of NDSs during heat treatments and storage in model milk and yogurt indicating that CSO/A2 ${\beta}$-casein NDSs exhibited excellent physical stability. In conclusion, the CSO concentration level and manufacturing temperature were the crucial determinants affecting the physicochemical characteristics of CSO/A2 ${\beta}$-casein NDSs containing resveratrol.

Effects of Dietary Casein, Soy, and Methionine-Supplemented Soy on Serum Lipids Level in Rats

  • The objective of the current study was to determine the influences of dietary proteins and methionine on plasma lipid concentrations. Thirty growing male Sprague-Dawley rats were fed diets similar in all respects except that dietary protein was from either casein, soy protein isolate, or soy protein isolate supplemented with L-methionine (0.24 %). The animals were fed experimental diets ad libitum for nine weeks. Plasma total-cholesterol concentrations were unaffected by the protein source or methionine supplementation. Plasma triglyceride concentrations were lower in rats of methionine supplemented soy protein diets (76 mg/dL) than in the rats fed casein or soy diet (120 mg/dL, 109 mg/dL, respectively). These results indicate that soy protein reduces plasma triglycerides relative to casein in rats fed cholesterol free diets, and that methionine-supplemented soy diets decrease plasma triglyceride concentrations more than soy protein alone.

  • Casein was chemically modified with acetic, succinic, and maleic anhydride and changes in functional pro-perties were evaluated as affected by the degree of modification. Chemical modification resulted in casein with unique functional properties depending upon the type of anhydrid used and the degree of modification. It was possible to control heat coagulation, calcium precipitability, forming and emulsion capacity and stability. At pH 4.5 heat coagulation was 0% in the case 74.1% acetylated casein; on the contrary, succinylation and maleyation resulted in highly heat sensitive protein. Foaming properties were improved markedly by suc-cinylation and maleylation at pH 4.5. However, emulsifying properties were enhanced only by maleylation.

  • The effect of lysine or methionine addition on the lipid oxidation and protein properties in casein-fish oil model system during the storage of 21 days at 37$^{\circ}C$ was studied. The peroxide and TBA values were increased markedly and the amino acids addition to the system caused to reduce the extent of lipid oxidation in comparison with that of casein-fish oil system which was a control group. Significant changes in fatty acid composition of each group were observed. Polyenoic acid contents were drastically decreased during the storage in the groups with and without the amino acids additions, . And also significant losses of several amino acids were occurred with the reduction of solubility and digestibility of casein during the storage. However no different effects were observed in both additionsof lysine and methionine to the system.

  • Effects of dietary protein sources and their amino acid compositions on the liver and plasma cholesterol levels in growing Single Comb white Leghorn male chicks were studied. A diet containing isolated soy protein (21% cp) was supplemented with 0.5% DL-methionine and 0.3% L-glycine. and another diet containing casein(21% cp) was supplemented with 1.5% L-arginine HCl, 0.4% DL-methionine. and 1.0% L-glycine. Plasma cholesterol level was markedly lower in groups force-fed the diets containing either soy protein or casein supplemented with amino acids compared to those found in birds fed corresponding diets without amino acids supplementation. The cholesterol lowering effect of the casein diet. when balanced with various supplemental amino acids appeared to be due to arginine instead of methionine or glycine. It is likely that amino acid balance rather than the composition of a specific amino acid is one of the major factors determining the effect of dietary protein sources on the blood cholesterol levels in chicks.

Influence of the Combination of Casein and Isolated Soyprotein with or without Methionine Supplementation on the Growth, Metabolism, and Body Composition of Growing Rats (카제인과 대두단백질(大豆蛋白質)의 혼합(混合) 및 Methionine보충(補充)이 흰쥐의 성장(成長), 체내(體內) 대사(代謝) 및 체조성(體組成)에 미치는 영향)

  • This experiment was conducted to study effects of different sources of protein, and of the different combination ratios of the two protein supplemented or unsupplemented with methionine on the growth and body metabolism of the growing male rats. Casein and isolated soyprotein (ISP) were combined at different ratios of 100:0, 80:20, 50:50, 20:80 and 0:100 to supply the constant levels of dietary energy and protein of 3,600kcal/kg and 20%, respectively. Each combination was supplemented with either 0 or 0.3% methionine. Results obtained in these experiments were summarized as follows: The body weight and weight gain in the ISP diet were the lowest, but were increased with the increase of ratios of casein to ISP. The body weight gain in the ISP diet supplemented with methionine was similar to that in the combination diets of casein and ISP. The combination diets of casein and ISP supplemented with methionine had the higher weight gain than the casein diet. The food intake in the casein diet was the highest, and was increased with the increase of ratios of casein to ISP, and was lowered with methionine supplementation. The food intake in the ISP diet was the lowest and was not increased with methionine supplementation to the ISP diet. The food efficiency ratio of the combination diets of casein and ISP was improved compared to those of either the casein or the ISP diets. The food efficiency ratio was improved in the diet of either casein of ISP supplemented with methionine, but was not improved in the combination diets of casein and ISP with methionine supplementation. The gross energy intake had direct relation with the food intake of rats and the relation was increased either in the casein diet of with the increase of the ratios of casein to ISP. The energy efficiency and protein efficiency ratios were improved in the diet of either casein of ISP by the supplementation of methionine, and the effect of methionine supplementation of the energy efficiency and protein efficiency atio was increased with the increase of the ratios of ISP to casein. The nitrogen intake and urinary nitrogen excretion of the casein diet were the highest and those of the ISP diet were the lowest. They were increased with the increase of ratios of casein to ISP. The nitrogen balance and retention were not significantly among the treatments. The gross energy intake and fecal and urinary energy were the highest in the casein diet but the digestible and metabolizable energy and the digestibility and metabolizability of energy were not significantly different among the treatments. The body fat content of rats was increased by supplementation of methionine but the body protein content was decreased (r=-0.65, p<0.01). The body fat content of rats was negatively correlated with body moisture content (r=-0.83, p<0.01). The liver weight was highly correlated with the carcass weight (r=0.79, p<0.01), and was increased by the methionine supplementation and by the increase of the ratios of casein to ISP.

  • The present study was designed to observe the effects of soyprotein and casein with or without cholesterol on serum and liver lipids in male rats. The 6 experimental groups were as fellows ; SF ; soyprotein, cholesterol-free diet. SC ; soyprotein, 0.5% cholesterol added diet. CF ; casein, cholesterol-free diet CC ; casein,0.5% cholesterol added diet. SCF ; protein mixture of soyprotein and casein(1 : 1), cholesterol-free diet SCC ; mixed protein, 0.5% cholesterol added diet. The hypocholesterolemic and hypotriglyceridemic effects of soyprotein were observed at 3 weeks, but these effects disappeared at 6 weeks. The hypocholesterolemic effect of soyprotein was more obvious when the 0.5% cholesterol was supplemented in the diets. The serum free cholesterol level was not affected by the dietary protein source or the dietary cholesterol, therefore, the difference in serum total cholesterol among groups seems due to the difference in cholesterol esters. There was a tendency of a higher percentage of HDL in soyprotein groups compared to casein groups at 1 week, however, this tendency disappeared with time. The liver cholesterol and triglyceride contents were not differ among cholesterol-free diet groups, however, with addition of cholesterol, those of soyprotein groups were significantly lower than casein groups. The higher serum arginine/lysine ratio of soyprotein groups may offer the part of explanation of its hypocholesterolemic effect.

  • A feeding experiment was conducted to determine the nutritional value of various dietary protein sources without a vitamin premix supplementation in juvenile black rockfish, Sebastes schlegeli. Diets containing similar quantities of lipid and carbohydrate with 50% crude protein from fish meal (FM)/soybean meal (SBM)/corn gluten meal (CGM), FM only, casein/gelatin, high FM/low casein and low FM/high casein were fed to rockfish along with a commercial rockfish diet. The diet containing only 65.6% FM and the commercial rockfish diet produced significantly greater weight gain (WG) and feed efficiency (FE) than all other semipurified diets (SPDs); the economical diet with 52.8% FM, 10% SBM and 5% CGM produced lower WG than the diet containing FM and commercial rockfish diet, but FE and survival were similar both diets. Intermediate responses were observed for rockfish fed the SPD containing 32.8% FM and 27.4% casein, while SPDs containing 10% FM and 41.8% casein and 38.5% casein and 10% gelatin yielded the lowest WG and FE. Some differences in tissue indices and body composition of rockfish were induced by the diets containing various protein sources and levels without the vitamin premix supplementation.

  • The nomenclature of genetic variants of casein which is major protein in milk have had a lot of confusion, but now have established. Genetic variants of ${\alpha}_{s1}-CN,\;{\alpha}_{s2}-CN,\;{\beta}-CN,\;{\kappa}-CN$ have reported 8 variants(A, B, C, D, E, F, G, H), 4 variants(A, B, C, D), 13 variants ($A_1,\;A_2,\;A_3,\;A_4$, B, C, D, E, F, G, $H_1,\;H_2$, I), 11 variants(A, B, C, E, $F_1,\;F_2,\;G_1,\;G_2$, H, I, J), respectively. Their data detailed have introduced in several web sites including www.uniprot.org. The studies on genetic variants of casein from Korean native cattle have been reported only ${\beta}-casein\;A_4$ but still not established the protein sequence. The classification and distinct nomenclature of genetic variants of bovine casein were required because the development of milk science and technology have been focused in the region that have to studied biochemically such as functional foods, EMC and GMO et al.
