Development of Carrot Medium Suitable for Conidia Production of Venturia nashicola

  • The causal fungus of pear scab, Venturia nashicola, grows slowly and rarely produces conidia on artificial media in the laboratory, but it produced conidia on the Cheongah medium containing Cheongah powder. V. nashicola grew too slow to produce conidia until 15 days after cultivation but produced conidia with $4{\time}10^4$ conidia/plate 30 days after cultivation on the Cheongah medium containing 1% Cheongah powder. V. nashicola showed a peak production of conidia with $4.5{\times}10^5$ conidia/plate 60 days after cultivation on the carrot medium containing 2% carrot powder, one of the constituents of Cheongah powder. The carrot medium is considered to be the best medium to obtain conidia of V. nashicola in the laboratory until now. This is the first report on the development of a suitable medium for conidia production of V. nashicola, as far as we know.

  • This study was carried out to obtain basic information for possibility of oral vaccine in carrot using Agrobacteruim -mediated transformation system. The epitope region of porcine epidemic diarrhea virus (PEDV) spike gene which is classified as a member of the Coronaviridae and causes an acute enteritis in pigs was successfully expressed in carrot (Daucus carota) using the Agrobacterium-mediated transformation system. Hypocotyl segments of in vitro germinated plantlets were infected with Agrobacteriun tumefaciens LBA 4404 harboring PEDV spike gene. Embryogenic callus (EC) was induced on MS selection medium with 1 mg/L 2,4-D, 50 mg/L kanamycin and 300 mg/L cefotaxime after 45 days of culture. Subcultured ECs on MS selection medium without 2,4-D were converted to somatic embryos (SE) of various stage; globular, heart and torpedo stage. Putative transgenic embryos were selected on MS medium with 50 mg/L kanamycin and 300 mg/L cefotaxime. Regenerated plantlets from transformed SE were induced on MS medium containing 50 mg/L kanamycin after 30 days of culture. Genomic PCR confirmed the integration of PEDV spike gene into nuclear genome of carrot and northern blot analysis demonstrated the expression of PEDV spike gene in transgenic carrot.

  • Effects of the special media on the mycelium growth in Agaricus campestris has been studied. The results are summarized as follows; 1. The mycelium growth of Agaricus campestris were considerably Stimulated on the Carrot (Dancus carota L.) basal medium which was added 4ml. of carrot extract, Cucumber(Cucumis sativus L.) basal medium added 3ml of cucumber extract, and Radish (Rhaphanus sativus L.) basal medium added 3ml. of radish extract during the culture for 144 hours. 2. The mycelium of Agaricus campestris on the media which was added the several kinds of vegetable extracts shows a lot of growth for 144 hours. The orders are as follows; Carrot basal medium(4ml/100ml)>Cucumber basal medium (3ml/100ml)>Radish basal medium (3ml/100ml)>Lettuce basal medium (2ml/100ml)>Cabbage basal medium (2ml/100ml). However, the Lettuce (Lactuca scariota L.) basal medium and the Cabbage (Brassica chinensis L.) basal medium among these five media are no significant differences.

Effects of Ascorbate on Somatic Embryogenesis in Carrot Cell Cultures (당근 세포배양으로부터 체세포배 발생에 미치는 아스콜빈산의 효과)

  • This study was conducted to elucidate the effects of ascorbic acid and dehydroascorbic acid on somatic embryogenesis from the cultured cells of carrot. Ascorbic acid in culture medium merely stimulated the proliferation of non-embryogenic cells but dehydroascorbic acid in medium induced embryogenic cells from non-embryogenic cells accompanying the inhibition of cell proliferation. Ascorbic acid in medium inhibited somatic embryogenesis from embryogenic cells while dehydroascorbic acid in medium enhanced somatic embryogenesis from the cells as well as non-embryogenic cells. This enhancement was limited to globular embryos and the maturation to cotyledonary embryos was inhibited by dehydroascorbic acid treatment. From the above results it is suggested that carrot callus cultures on medium containing dehydroascorbic acid could quickly induce embryogenic cells. In addition after brief culture of embryogenic cells on development medium containing dehydroascorbic there by acid the subculture of the cells to MS basal medium resulted in the high frequency production of somatic embryos.

Effect of carbon sources on somatic embryogenesis and cotyledon number variations in carrot (Daucus carota L.)

  • In order to investigate the effect of carbon sources on somatic embryogenesis and cotyledon number variations in carrot, embryogenic callus were cultured in the medium supplemented with various concentrations of sucroseor glucose, and with combination of 2% sucrose and various concentrations of mannitol or sorbitol. The maximum number of somatic embryos formed per flask (1,555.70) was obtained in the medium supplemented with 2% sucrose rather than glucose alone or a combination of mannitol or sorbitol and 2% sucrose, and the number of somatic embryos was decreased with the increasing of mannitol or sorbitol concentration. The frequencies of somatic embryos with two cotyledons were 35.14% for sucrose, 19.88% for glucose, 32.55% for mannitol + 2% sucrose, and 38.59% for sorbitol + 2% sucrose, respectively, and the frequencies of abnormal somatic embryos having 3 or more cotyledons were 64.86% for sucrose, 80.12% for glucose, 67.44% for mannitol + 2% sucrose, and 61.41% for sorbitol + 2% sucrose, respectively. Particularly, the frequency of somatic embryos with two cotyledons (59.16%) was the highest in the 2% sucrose medium compared to the frequency of abnormal somatic embryogenesis with three or more cotyledons, and the frequency gradually decreased with increasing concentration of glucose, mannitol or sorbitol. According to these results, it was found that the ratio of abnormal somatic embryo was higher than the normal somatic embryo in carrot, and was shown that somatic embryogenesis and the cotyledon number was affected by the concentrations of sucrose, glucose as carbon source, and mannitol and sorbitol as osmotic agents in culture medium.

The Effects of Various Hormones on Growth of Carrot Tissue Culture (당근 배양세포의 생육에 미치는 수종 Hormone의 영향)

  • Experiments were conducted to investigate the effects of several hormones on the growth of suspension culture of carrot (Daucus carota L.) cells, where changes in pH and the amount of $NH_4-Nin$ the medium were observed with regard to growth. A treatment with 2,4-D at $10^{-5}M$ resulted in a highest rate of growth; the hormone at this concentration caused an increase in dry weight by about 40 to 50% over the control, measured at a stationary phase. It was thus indicated that 2,4-D at $10^{-5}M$ provided the optimal condition for the suspension culture. Changes in pH of the medium were found to be affected by hormonal treatments during the first 2-3 days following the inoculation, after which the pattern of pH changes in hormone enriched media paralleled that of the control. Subsequent changes of $NH_4-N and NO_3-N$ from the medium by the cells, and also by growth of the cells. The uptake of $NH_4-N$ by the cells did not appear to be influenced by hormonal treatments. At a stationary phase, a considerable amout of $NO_3-N$ played a more important role than $NH_4-N$ in the growth of the carrot cell suspension culture.

Effects of Cell Size, Density, Conditioned Media and pH on Carrot (Daucus carota L.) Cell Embryogenesis (당근(Daucus carota L.) 세포 배양시 세포의 크기, 밀도, Conditioned 배지 및 pH가 배발생에 미치는 영향)

  • The effects of sizes and densities of cells cultured, conditioned medium, and media pH on the somatic embryogenesis of carrot (Daucus carota L.) were examined. A large number of globular embryoids was formed after 4 days in cell culture, and later globular embryoids developed into heart and torpedo shape. High cell density resulted in higher number and better growth of embryos, especially on conditioned medium than Murashige-Skoog medium. The fresh weight and number of embryoids formed increased with the decrease in cell size. The significant reduction in fresh weight and number of embryoids was obtained when culturing cells with diameter of over 90 ${\mu}{\textrm}{m}$. Dry weight and number of embryoids were markedly reduced with medium pH of 4 or 7, but promoted with pH 6.0.

Rapid Detection of Growth factors of intestinal Lactic Acid Bacteria (장내유산균 증식인자의 신속한 검색)

  • The growth of Bifidobacterium and Lactobacillus isolated from human interstinal bacteria were induced by water extract and U-step extract of soybean and carrot and the pH of these bacteria-cultured media were decreased. The increasing growth rates of these bacteria are related to the decrease of the pH of these bacteria-cultured media. When human intestinal flora as starter were inoculated into the medium containing water extract of soybean and carrot. the growth of lactic bacteria were also induced and the pH of the media were decreased. By measuring the pH of the media which were inoculated and cultured intestinal bacteria as a starter, it is possible to determine whether the food are the growth factors of intestinal lactic acid bacteria or not. By this method, the food which decreased pH of the medium were soybean, turnip, carrot. leek, garic, dropwork, wonnwood and onion. 'These foods may induce lactic acid bacteria in human in1estlne.

Studies on Growth and Differentiation of Suspension-Cultured Carrot Cells I. Alterations in Peroxidase Activity, Polyamine Content and Ethylene Production during Somatic Embryogenesis (당근 현탁 배양세포의 생장과 분화에 관한 연구 I. 배형성 과정에서 Peroxidase 활성, Polyamine 함량 및 Ethylene 성성의 변화)

  • Changes of peroxidase activity, polyamine content and ethylene production during somatic embryogenesis in suspension-cultured carrot (Daucus carota L.) cells were investigated. As compared with nonembyrogenic cells and their medium, embryogenic cells and their medium were characterized by higher levels of peroxidase at all times of culture period. Peroxidase in embryogenic cells showed higher oxidation activity of IAA than in nonembryogenic cells at the torpedo stage, but the IAA oxidation activity of peroxidase released into embryogenic medium was lower than that of peroxidase released into nonembryogenic medium. Peroxidase patterns of embryogenic and nonembryogenic cells showed three cathodic bands, and one anodic band, while peroxidase patterns released into embryogenic and nonembryogenic media did not show any anodic bands and the isoelectric points of cathodic peroxidase were pH 7.7, 7.5 and 6.6. Compared with nonembryogenic cells, polyamine content in embryogenic cells was increased by 15% at the torpedo stage, but polyamine ratio was constant, and ethylene production was extremely low at all times of culture period. Therefore, it is suggested that the peroxidase in embryogenic cells is correlated with embryogenesis by regulating hormone ratios through IAA oxidation, while the peroxidase isozyme patterns may be used as a biochemical marker of embryogenesis. The increase of polyamine content and the decrease of ethylene production suggest an interaction between polyamine and ethlyene during embryogenesis.

Optimization of SOD Biosynthesis by Controlling Sucrose Concentration in the Culture of Carrot Hairy Root

  • In order to optimize the carrot hairy root culture for SOD production, a fed-batch culture of hairy roots was performed in a bioreactor. Maximum SOD activity was obtained when the hairy roots were transferred to the MS medium containing 110 g/1 concentration of sucrose. By controlling the sucrose concentration (70 g/1 sucrose for growth and 110 g/1 sucrose far production, respectively) In a two-stage fed-batch culture, 29 g/1 of the hairy roots was obtained based on the final dry mass. The volumetrically determined SOD activity and productivity in the fed-batch culture were about 6 times higher than those from the flask culture containing sucrose at 30 g/1 concentration.
