Verification of Practicality for the SSBL and Pinger Synchronizing Biotelemetry Method and System through the Tracking of Fish (어류의 행동추적에 의한 SSBL · 핑거동기 바이오텔레메터리 방식과 시스템의 실용성 검증)

  • The new biotelemetry method and system that the installation and the treatment of equipment is convenient and the instantaneously detailed position of the fish attached a pinger is able to track comparatively easily had been developed, an practicality of it were verified in the water tank and the small sea port through the tracking of fish. The biotelemetry method had been gotten the three dimensional locations of fish to the receiving transducer by combining of the super short base line (SSBL) method to detect the direction of pinger and the pinger synchronizing method to measure the range from receiving transducer to pinger. The receiving system had been designed to realize the high precision or wide detection range by application of the basic design method for receiving system of biotelemetry and the hydrophone array configuration. From tracking test of carp in the water tank, the migration course and the velocity of carp was investigated and the observed migration course was compared with measurement. The measured migration course of carp coincided with the observation in the main and the position of carp was able to track three dimensionally. The velocity of carp measured by the moving average method was 11.2cm/s. From tracking test of yellowtail in the small sea port, the migration course and the velocity of yellowtail was investigated at natural condition. The position of yellowtail was able to track three dimensionally and the velocity of it measured by moving average method was 43.9cm/s.

Studies on the Food Components of Triploid Carp Muscle 2. Lipid Components of Triploid Carp Muscle (3배체잉어 근육의 식품성분에 관한 연구 2. 3배체잉어 근육의 지방질 성분)

  • To evaluate the quality of lipids and their fatty acid compositions in diploid and triploid carps(Cyprinus carpio), we analyzed the compositions of neutral lipid, phospholipid and glycolipid, and fatty acid contents of each lipids in these carp muscles. Total lipid contents of diploid carps, nonspawning season and spawning season, and triploid carp were $2.5\~3.0\%,\;1.4\~1.9\%\;and\;2.4\~3.1\%$, respectively. Regardless of belly and dorsal muscles, diploid carp of nonspawning season and triploid one were higher in neutral lipid, but lower in glycolipid and phospholipid than those of diploid one of spawning season. The neutral lipid and phospholipid were mainly consisted of triglyceride($66.9\~86.1\%$), and phosphatidyl-choline($55.6\~62.9\%$), respectively. In fatty acid compositions of total lipid, triploid carp showed higher content in monoenes such as 16: 1, 18: 1, but lower in polyenes such as 22:6 than those of diploid carp. The major fatty acids in total lipid were 18:1, 16:9, 18:2, 18:3 and 16:1 in order. The fatty acid compositions of neutral lipids showed similar trend to those of total lipid, and the major fatty acids in phospholipid were 16:0, 18:1, 18:2 and 22:6, These fatty acid compositions of neutral lipid, glycolipid and phospholipid were slightly difference between dorsal and belly muscles in all the carps.

Physiological Responses of Common Carp (Cyprinus Capio) and Crucian Carp (Carassius Auratus) by Rapid Changes of Water Temperature (급격한 수온변화에 따른 잉어 (Cyprinus capio)와 붕어 (Carassius auratus)의 생리적 반응)

  • The blood and physiological changes of common carp (Cyprinus carpio) and crucian carp (Carassius auratus) were analyzed when the water temperature was rapidly increased from 20℃ (control) to 26 and 32℃. The water temperature reached 26℃ and 32℃ within 6 hours from the water temperature of 20℃, and the control was maintained at 20℃ for the duration of the experiment. From each experimental group, blood was collected every 3, 12, 24 and 48 hours after the water temperature rise, and the red blood cell count, hemoglobin, hematocrit, Na+, K+, Cl-, cortisol, glucose, aspartate amino transferase (AST) and alanine amino transferase (ALT) contents were analyzed. In the case of the crucian carp experimental group where the water temperature was raised to 32℃, the concentrations of plasma cortisol, glucose, AST and ALT increased 3 hours after the water temperature rise, and the concentrations did not decrease until 48 hours (P<0.05). Plasma cortisol, glucose, AST, and ALT in both C. carpio and C. auratus in the control group and the experimental group raised to 26℃ increased at 3 hours after the water temperature rise (P<0.05), and decreased to the value before the water temperature rise at 48 hours (P<0.05). The plasma Na+, K+ and Cl- concentrations of C. carpio and C. auratus in the experimental group where the water temperature was raised to 32℃ were higher than in the 20 and 26℃ experimental groups. The blood reaction of all experimental groups including the control group showed a common trend in both C. carpio and C. auratus increasing up to 12 hours after the water temperature rise and then decreasing at 48 hours after the water temperature rise. The results derived from this study would be useful for investigating the physiological response of fish stress in future. In addition, as fish mortality has recently occurred frequently in rivers and lakes, it is judged that it can be used as basic data.

Efficiency of Phototransduction Cascade in Carp Cones

  • In the vertebrate retina, rods mediate twilight vision and cones daylight vision. Rods have been purified easily from the retina, and thus the phototransduction mechanism in rods is now well documented. However, it has not been possible to purify cones in large quantities, and therefore, the knowledge on the mechanism in cones is limited. Here we report purification of carp (Cyprinus carpio) cones with a stepwise Percoll gradient. Using purified cells, we compared the phototransduction mechanism between rods and cones. The results showed that both transducin activation and phosphodiesterase activation are less effective, and visual pigment phosphorylation is faster in cones. These differences explain lower light-sensitivity and briefer photoresponse time course in cones.

  • Endocrine disruption in crucian carp (Carassius auratus) living in the branch of Han River were examined. Vitellogenin level in plasma was measured using ELISA system and aromatase mRNA level in brain was observed using RT-PCR technique. In all female fish, vitellogenin levels were in the range of 20∼40 $\mu\textrm{g}$/ml and aromatase mRNA expression could be detected on the agarose gel after RT-PCR. However, in case of males, vitellogenin level was elevated in only one fish, while vitellogenin was hardly detected in others. Aromatase was expressed in all males although the levels were relatively lower than the level in female fish. Testis-ova and any other histological changes of reproductive organ were not shown in both sexes.

  • Vitellogenin (Vtg) is found in the serum of both female and male fish that have been exposed to environmental endocrine disrupters or estrogen hormones, and is used as a biomarker for such contamination. In our current study antibodies raised against the purified flatfish Vtg were tested for their reactivity on immunoblots to flatfish and carp Vtg, and also to a Vtg protein fragment produced in E. coli. Polyclonal antibodies raised against purified flatfish Vtg reacted well with Vtg in the serum of flatfish and carp induced with $17{\beta}$-estradiol, but not with the Vtg protein fragment produced in E. coli.

  • A study was made to measure mortality and scoliosis of the fingerlings of common carp (Cyprinus carpio) which has been exposed to various concentrations of trichlorfon. Trichlorfon increased mortality of the fingerlings and it caused severe scoliosis. When exposed to 40 ppm trichlorfon for 120 hrs, $100\%$ mortality was observed. Scoliosis was observed when the fingerlings have been exposed 25 to 40 ppm trichlorfon.

  • Natural actomyosin was extracted from the dorsal muscle of carp (Cyprinus carpio), and some Physico-chemical properties were determined. The sedimentation constant ($s_{20},\;_\omega$) of the purified skeletal muscle actomyosin was about 24.76s and the ATP sensitivity 147-176. The specific viscosity was rapidly ascended with the increase of protein concentration as other vertebrate actomyosins. ATPase activating site under the effect of pH variation was showed on the both sides of acid and base resembling with the tendency of rabbit skeletal muscle actomyosin, and found at 6.0 on the acidic side and at 9.5 on the basic side.
