  • The effect of chitosan treatment on the dyeing and dye fastness, and mechanical properties of hair was investigated in this study when the carotinoid dyestuffs extracted from African Marigold(Tagetes erecta L.) were applied to the hair. The sequences of the chitosan treatment were changed in dyeing and mordanting procedures, i.e., pre-treatment, mid-treatment, and post-treatment. While the effect of chitosan application on the color shade change was not significant, the pre-treatment of the chitosan increased the dye uptake. Discoloration and fading was observed in the lightfastness test when chitosan was mid-treated. Alkali perspiration and acid perspiration fastness test results showed that chitosan post-treatment gave lower tendency. Washing fastness results showed that chitosan post-treatment and mid-treatment gave lower values, which indicates that chitosan deters the direct formation of insoluble complex among fiber-dyestuff-mordant. In the mechanical characteristics results, however, initial modulus and breaking strength increased significantly in the post-treatment and pre-treatment of chitosan.

The Ultrastructure of the Cutaneous Pigment Cells in the Frog, Rana nigromaculata Hallowell, during Hibernating Phases (동면기 개구리 (Rana nigromaculata) 피부색소세포의 미세구조)

  • The authors observed the ultrastructure of the pigment cells of the frog, Rana nigromaculata Hallowell, during the hibernation. The specimens from the skin were fixed in 2.5% glutaraldehyde-paraform-aldehyde fixative in phosphate buffer at pH 7.2 prior to fixation in 2% osimium tetroxide, dehydrated in graded ethanol and acetone, embedded in Epon 812 mixture, and sectioned with LKB-ultramicrotome. the ultrathin sections were contrasted with uranyl acetate and lead citrate and observed with a JEOL-100B electron microscope. The results were as follows. In hibernating phase, pigment cells of the frog were consisted of the three kinds of chromatophores (xanthophore, iridophore and melanophore) in their dorsal skin. The traits of these cells were as follows. 1. Xanthophores A. Xanthophores were filled with pterinosomes and carotenoid vesicles. Many ribosomes, a few mitochondria and glycogen particles were dispersed in the cytoplasm. B. Pterinosomes were spherical or ellipsoidal in shape. They were divided into 6 types (type I, type II, type III, type IV, type V, type VI pterinosomes) by the their inner structure and especially, type I, type II, type III pterinosomes were well developed.

The Effect of $SO_2$ on the Degradation of Chlorophyll in Green perilla (Perilla frutescens Suwon No. 8) (($SO_2$ 에 의한 들깨(수원(水原) 8호(號))의 엽록체분해(葉綠體分解)에 대하여)

  • The effects of light, active oxygen, inorganic $SO_3\;^{-2}\;ion$, $SO_2$ fumigation duration, $SO_2$ concentration on the degradation of photosynthetic pigments in Green perilla (Perilla frutescens Suwon No. 8) fumigated by $SO_2$ were investigated inside a phytotron. The results were as follows: 1. With the increase of the $SO_2$ dosage, visible injury and the degradation of chlorophyll increased. 2. The degradation of chlorophyll b was less than that of chlorophyll a and the carotinoid was more easily degratated than chlorophyll b. 3. The degradation of chlorophyll by $SO_2$ fumigation was induced directly by $SO_2$, itself. 4. Light is necessary to the degradation of chlorophyll. $SO_2$ and $O_2^-$ made in the photolysis process of the water molecule were the major components in the degradation of chlorophyll.

Use of Lycopene, an Antioxidant Carotinoid, in Laying Hens for Egg Yolk Pigmentation

  • The possibility of lycopene affecting egg yolk pigmentation was studied with lycopene diets containing 0, 4, 8, and $12{\mu}g/g$ meal, respectively. The addition of lycopene above $4{\mu}g/g$ meal significantly improved yolk color after four days of supplementation. The transfer of lycopene into egg yolk was confirmed by thin layer chromatography, and high-performance liquid chromatography-mass spectrometry (HPLC-MS). The deposition rate of lycopene into egg yolk was approximately 2%, which was quantitatively determined using a HPLC with a UV detector. The result indicates that lycopene is a good candidate for egg yolk pigmentation and for making functional eggs.

  • Color(and appearance)is one of the most important visual attributes in food. The favorite color of food is depend on the nation and its culture. Unfortunately the word on color ash superseded appearance as the description of the total visual perception of food. Fine Structural Approach of Granular Gland Regeneration after Skin Injury in Bombina orientalis (Bombina orientalis 피부손상 후 과립선 재생에 관한 미세구조적 연구)

  • Granular gland regeneration in the toad after dorsal skin wound histologically was examined using scanning and transmission electron microscopy. After cutaneous wounds were induced by excision, animals were maintained in special cages for up to 20 days. In transmission electron microscopy (TEM), newly formed granular gland, though poorly developed, was seen on 4 day after injury. Epithelial cells moved toward apical region of newly formed gland. The cells had smooth surface and were not connected to other cells by desmosomes. Mitochondria rich cell (MRC) possessing long cytoplasmic processes formed a gland cavity and hemidesmosomes were found under the cell processes. Basal cavity of newly formed gland consisted of MRC, pro-granular producing cells (pGPC), and granular producing cell (GPC). Moreover it was observed that xanthophores moved to the base of the epithelial tissue on 10 day after the injury. These cells contained numerous pterinosomes and carotenoid vesicles. Immature pterinosomes were large and carotenoid vesicles were moderately electron dense. On 13 day after the injury, xanthophores contained abundant carotinoid vesicles and lammelated pterinosomes. Iridophores were also observed adjacent the developing xanthophores on 16 day post-injury. These observations indicated that regeneration of granular gland from glandular precursor cells during wound healing and subsequent expansion of the glandular cells might be dependent on maturation and proliferation of these newly formed cells.

Isolation of Lipid High-yielding Chlorella vulgaris Mutants by UV Irradiation (자외선 조사에 의한 지질 고생산성 Chlorella vulgaris 변이주 분리)

  • Chlorella vulgaris, a genus of single-cell green algae, is considered to be a very essential resource for the higher value-added business including functional food and biodiesel, due to its high contents of protein, carbohydrate and lipid. In this study, ultraviolet rays were irradiated in order to induce the mutation of C. vulgaris. After inducing the mutation, UV1-20 mutant, high in lipid was selected and its cell growth rate, dry weight, pigment content and lipid content were measured. The growth rate of the UV1-20 mutant was increased almost 1.5 times than the wild type, but pigment contents of chlorophyll and carotinoid were decreased. In addition, the lipid content of UV1-20 was increased 1.8 times than the wild type. Therefore, C. vulgaris mutant, isolated in this study, is considered to have sufficient potential to be used as a material for the higher value-added business.

  • Climate change has occurred frequently, and it lead to a severe decrease in marketability and productivity. Onion has various functional elements related to antioxidant, antibacterial, anticancer and immune enhancement, the productivity is closely affected by climate conditions. In this study, to environmentally-friendly produce onions even though drought condition, effect of Bacillus aryabhattai H19-1 on the yield, marketability, and functional component of onions was evaluated in a field. When strain H19-1 was drenched, onion production and marketability significantly increased, and the contents of ascorbic acid and carotenoid of harvested onion bulbs also increased. Therefore, B. aryabhattai H19-1 is expected to be effective in enhancing onion productivity by mitigating drought stress.

Dyeing Properties of Yerba Mate Tea on the Fabrics (마테차를 이용한 직물의 염색성)

  • The yerba mate that grows in the subtropical forests of South America is an evergreen tree within the Aquifoliaceae family. The yerba mate (Ilex paraguariensis) tea is habitually used as a drink in Argentina, Paraguay, Uruguay, and Southern Brazil. A wild mate tree needs about 25 years to fully grow to 15 a height of meters; however, they only grow to a height of 3-5 meters when cultivated. The leaves are 7-11 cm long and 3-5cm wide with a serrated margin. Yerba mate tea-based beverages are made from the leaves and stems of the mate tree. It is known that they contain a rich content of antioxidants and polyphenol, vitamins, amino acids, minerals and colorants. Dyeing properties of a mate plant in the silk, wool, linen and cotton fabrics were investigated. Constituents of colorant of mate tea are chlorophyll and carotinoid; in addition, it has more minerals such as (Fe, Ca, Mn, Mg, Na, K, Zn, and Cu) than green tea. Mate tea colorants showed good affinity to silk and wool fabrics. It was found that the optimum condition for dyeing was 30 minutes for dyeing time, 7 for pH of dyebath, and at a dyeing temperature of $80^{\circ}C$. The maximum absorbance wavelength (${\lambda}_{max}$) of silk and wool fabrics dyed with mate tea were at 420-440nm; however, that of linen and cotton fabrics were at 400nm. K/S values of fabrics dyed under optimum conditions were 1.979 for silk fabrics, 1.541 for wool fabrics, 0.551 for linens and 0.465 for cotton fabrics. Munsell hue values of dyed fabrics measured 3.1Y-6.4GY for silk, 1.4Y-8.3Y for wool, 5.5Y-3.7GY for linen, and 5.3Y-1.3GY for cotton. All dyed fabrics showed hues in-between greenish yellow and greenyellow. Colorfastness to rubbing, perspiration and dry-cleaning of dyed fabrics with mate tea were very good; however, the grades of colorfastness to washing of dyed linen and cotton fabrics were poor, and colorfastness to light were poor.
