Highly Selective Synthesis of β-Amino Carbonyl Compounds over ZSM-5-SO3H under Solvent-free Conditions

  • ZSM-5-$SO_3H$ efficiently catalyzed the one-pot three-component Mannich reaction of aldehydes, anilines, and ketones. ${\beta}$-Aminocarbonyl compounds were obtained in reasonable yields and excellent stereoselectivities when the reaction was carried out at room temperature under solvent-free conditions. Simple experimental conditions and product isolation procedure makes this protocol potential for the development of clean and environment-friendly strategy for the synthesis of ${\beta}$-amino-ketones. The catalyst was recovered and reused for subsequent runs.

Salsolinol, a catechol neurotoxin, induces oxidative modification of cytochrome c

  • Methyl-6,7-dihydroxy-1,2,3,4-tetrahydroisoquinoline (salsolinol), an endogenous neurotoxin, is known to perform a role in the pathogenesis of Parkinson's disease (PD). In this study, we evaluated oxidative modification of cytochrome c occurring after incubation with salsolinol. When cytochrome c was incubated with salsolinol, protein aggregation increased in a dose-dependent manner. The formation of carbonyl compounds and the release of iron were obtained in salsolinol-treated cytochrome c. Salsolinol also led to the release of iron from cytochrome c. Reactive oxygen species (ROS) scavengers and iron specific chelator inhibited the salsolinol-mediated cytochrome c modification and carbonyl compound formation. It is suggested that oxidative damage of cytochrome c by salsolinol might induce the increase of iron content in cells, subsequently leading to the deleterious condition which was observed. This mechanism may, in part, provide an explanation for the deterioration of organs under neurodegenerative disorders such as PD.

S$_H$2 Reaction on Silicon-Carbon Bond in the Photoreactions of 2, 3-Benzo-1, 1-diphenyl(or dimethyl)-1-sila-2-cyclobutene with Carbonyl Compounds

  • The photoreaction of 2, 3-benzo-1, 1-diphenyl (or dimethyl)-1-sila-2-cyclobutene (9 or 10) with an aldehyde or ketone results in 1:1 cycloadduct of [4 + 2] type. In the reactions of 2, 3-benzo-1, 1-dimethyl-1-sila-2-cyclobutene (10) with acetone and butanone, another 1:1 adducts (13) were also formed, respectively. The following facts indicate that the formation of adduct involves an attack of a triplet carbonyl compound on the silicon of the benzosilacyclobutene, an $S_H2$ process. (1) Even when the reaction of 9 with acetophenone was carried out under conditions such that more than 99% of incident light was absorbed only by acetophenone using the filter solution of aq. cupric sulfate, the same adduct was still formed. (2) When the reaction of 9 with acetone was carried out under oxygen atmosphere, only trace amount of adduct was formed.

On the Oxidation of Rice Lipid Fractions Extracted from the Whole Grain by Immersion (용매침지(溶媒浸漬)에 의하여 추출(抽出)된 쌀알의 유지획분(油脂劃分)의 산화(酸化)에 대(對)하여)

  • Oxidative changes in the rice lipids under accelerated condition were studied by measuring the changes in weight gain, total carbonyl compound, malonaldehyde, and fatty acid composition. Rice lipids were prepared by extraction with either n-hexane or ethanol from polished rice grain and purified by Folch's method. The lipid preparations were either incubated in dark at $40^{\circ}C$ or irradiated with ultra-violet light for a period of 55 days. Weight gain by oxygen absorbed sharply increased within 3 days in the rice lipids under UV light irradiation. However, with the rice lipids at $40^{\circ}C$ incubation a moderate increase in weight was observed only after 45 days storage. Their induction periods were one day (hexane extracted, under UV light), 2 days (ethanol extracted, under UV light), 30 days (hexane extracted, at $40^{\circ}C$), and 40 days (ethanol extracted, at $40^{\circ}C$) respectively. Oxidative rancid odor appeared at the end of the induction period. Total carbonyl compound and malonaldehyde markedly increased within 7 days, and decreased in the rice lipids under ultra violet light irradiation, while at $40^{\circ}C$ incubation they were continued to increase slowly through out the storage. The hexane extracted lipid were less stable than ethanol extracted lipid on the basis of oxygen absorption, malonaldehyde and other carbonyl compound formation. With the hexane extracted lipid during 55 days incubation at $40^{\circ}C$, the contents of linoleic and linolenic acids decreased, while the oleic, stearic, and palmitic acids increased.

A Novel Bromoindole Alkaloid from a Korean Colonial Tunicate Didemnum sp.

  • Chemical investigation on a colonial marine tunicate, Didemnum sp. led to the isolation of a series of indole alkaloids including a new (1) and two known metabolites (2-3). Based on the spectroscopic analysis including 1D and 2D NMR along with MS spectra, the structure of 1 (16-epi-18-acetyl herdmanine D) was elucidated as a new amino acid derivative. The absolute configuration of 1 was determined by comparison of specific rotation with the known compound. The structures of compounds 2 and 3 were also identified as bromoindole containing compounds N-(6-bromo-1H-indole-3-carbonyl)-L-arginine and (6-bromo-^1H-indol-3-yl) oxoacetamide, respectively, based on $^1H$ and $^{13}C$ NMR data, MS data and specific rotation value. Their pharmacological potentials as antibacterial agents and FXR antagonists were investigated, but no significant activity was found. However, the structural similarity of compound 1 to compound 4 suggested the anti-inflammatory potential of compound 1.

  • This paper describes a racemic and stereoselective synthetic route for a novel cyclohexenyl carbocyclic adenine analogue. The required stereochemistry of the target compound was controlled using a stereoselective glycolate Claisen rearrangement followed by α-chelated carbonyl addition. The introduction of 6-chloropurine was achieved using Mitsunobu conditions, and further modifications of the corresponding heterocycle gave the target cyclohexenyl nucleoside.

  • For metabolic study of pinusolide, a naturally occurring platelet activating factor antagonist, a synthetic method for preparation of radiolabeled pinusolide was studied. Pinusolide was first oxidized with $OsO_4/NaIO_4 $to 17-nor-8-oxo compound (2), which was subsequently converted to pinusolide by treatment with the Lombardo reagent $(Zn/CH_2Br_2/TiCI_4)$. The Wittily reaction was unsuccessful in the latter carbonyl methylenation of 2.

Effect of an Extra Chloro Substituent on Photochemistry of o-Alkylphenacyl Chloride

  • The title compound, ${\alpha},{\alpha}$-dichloro-o-methylacetophenone, was prepared and its photochemical behavior was investigated. Addition of an extra chlorine at alpha position to the carbonyl showed many different features from photochemical reactivities of mono chloro analogue, 2,5-dimethylphenacyl chloride. In benzene, a rearrangement product with a formal 1,5-Cl shift and a reduction product were formed beside indanone. In methanol, solvolysis and cyclization of a common dienol intermediate occurred at comparable reaction rates.

  • It is known that hydantoin derivative can be synthesized according to the method of Bucherer-Bergs, which employs the interaction of the carbonyl compounds with potassium cyanide and ammonium carbonate in dilute alcohol solution. In a converting study of .alpha.-dietylaminoacetophenone with KCN and ($NH_{4})_{2}CO_{3}$ to hydantoin, the attempted 5-diethylaminomethyl-5-phenylhydantoin was not formed. In this reaction diethylaminomethly group was unexpectedly eliminated and 5-phenylhydantoin was obtained, instead of the anticipated compound.
