Protective Effect of Korean Red Ginseng against Oxidative Damage by Carbon Tetrachloride in Rat (흰 쥐에서 사염화탄소에 의한 산화적 손상에 대한 홍삼의 방어효과)

  • To investigate the protective effect of Korean red ginseng (RG) against oxidative damage, rats were intoxicated by carbon tetrachloride and liver tissues and blood were taken and analyzed histopathologically and biochemically. Light microscopy of the liver showed that RG prevented the necrosis of hepatocytes remarkably and reduced the change of fat. RG increased the capability for serum to suppress oxygen radical in the generating system. It is suggested that RG enhanced the antioxidative potential of the body against $CCl_4$, which would prevent the necrosis of hepatocytes in vivo.

  • The protective effects of acteoside, a phenylethanoid glycoside, on carbon tetrachloride-induced hepatotoxicity and the possible mechanisms involved in this protection were investigated in mice. Pretreatment with acteoside prior to the administration of carbon tetrachloride significantly prevented the increased serum enzymatic activities of alanine and aspartate aminotransferase in a dose-dependent manner.   • This study was performed to evaluate the effect of liver damage on toluene metabolism in rats pretreated with carbon tetachloride. Liver damage in rats was induced by administration of 0.1ml of carbon tetrachloride per 100g of body wight intraperitoneally every day for four weeks except the last day before sacrifice. One day before sacrifice, toluene was administered to the animals instead of carbon tetrachloride. Rats were sacrificed at the 1st, the 2nd, the 3rd and the 4th week after the first administration of carbon tetachloride. Based on the histopathological findings, liver weight and serum alanine aminotransferase, the $CCl_4$-preteated group was found to have gradual severe liver damage. Especially the degree of liver injury became increasingly severe throughout the whole course of the experiment. The contnts of hippuric acid in urine lower in the all groups pretreated with $CCl_4$than that of the control. The contents of hepatic cytochrome P450(CYP), benzylalcohol dehydrogenase and benzaldehyde dehydrogenase activities were decreased in $CCl_4$-pretreated rats than those of the control. The $CCl_4$treated animals showed the gradual decreased activities of these enzyme as injection times elapsed. Km values of the benzylalcohol dehydrogenase in pooled liver samples from $CCl_4$-pretreated or control groups were similar. On the other hand, Vmax values of the $CCl_4$-pretreated group was lower than of the control. Therefore, it can be concluded that reduction of the toluene metabolism in damaged rat liver induced with $CCl_4$was due to the inhibition of CYP content, bezylalcohol and benzaldehyde dehydrogenase activities which related with toluene metabolic enzyme system.

  • Ionic dissociation rates of $\alpha$-chlorobenzyl ethyl ether in each solvent of toluene-$d_8$ and carbon tetrachloride were measured by the method of dynamic NMR spectroscopy. The spin system of these 1H NMR spectra was $AB_3$. The theoretical spectrum was calculated by computer simulation of dynamic NMR spectra, which agreed very well with observed spectra. From this computer simulation, the ionic dissociation rate constant k was obtained, and by Eyring plot with it, slope and intercept length was gained, from which kinetic parameters were calculated.The easiness of ionic dissociation depended upon solvent polarity. Activation enthalpy was 4.7 kcal/mole in toluene-$d_8$, 10.7 kcal/mole in carbon tetrachloride, and activation entropy was -35. 8 e.u. in toluene-$d_8$, -14.4 e.u. in carbon tetrachloride. It was understood that though the ${\Delta}H^{neq}$ value was small, this ionic dissociation had an easier procession in nonpolar solvents with increasing temperatures. Considering that the ionic dissociation could be thought as the first step of $S_N1$ mechanism, attention might be paid to the results that the value of ${\Delta}S^{neq}$ had a large negative value in comparison with a small ${\Delta}H^{neq}$.

  • The purpose of this study is to investigate the effects of bowontang on serum reactions of $CCl_4$- treated mouse. In this study, the experimental mouse were divided into three groups, normal, control, and sample. Under the same condition, the normal group was fed basal diet and water, the control group was injected with carbon tetrachloride ($CCl_4$, 0.6ml/kg) and fed a basal diet for 2 weeks, and the sample group was injected with carbon tetrachloride ($CCl_4$, 0.6ml/kg) and fed the bowontang extract (900mg/kg) for 2 weeks. It was shown that scavenging effect on DPPH radical was depended on concentration of the bowontang. And activities of GOT, GPT and ALP in blood serum of the sample group were significantly decreased as compared with the control group, respectively. Also, activities of LPO, GSH and catalase in the liver were significantly changed in experimental mice. In the liver of the sample groups, the activity of LPO was decreased as compared with control group, while activities of GSH and Catalase were significantly increased.

  • Alcoholic extract of the leaves of Andrographis paniculata Ness (=AAP) was obtained by cold maceration. A dose of 300 mg/kg $(1/6of\;LD_{50})$ of the extract was selected to study hepatoprotective action against carbon tetrachloride-induced liver damage. The extract was found to be effective in preventing liver damage which was evident by morphological, biochemical and functional parameters.

  • Hepatoprotective activity of different extracts of Citrullus colocynthis L. sch. (roots) (cucurbitaceae) was investigated in albino rats by inducing hepatotoxicity with carbon tetrachloride. The alcoholic extract of Citrullus colocynthis sch. 100 mg/kg b.w. has been shown to posses significant hepatoprotective effect by lowering the serum level of transaminases (GPT & GOT), alkaline phosphate (ALP) and bilirubin (P < 0.05 to P < 0.001).

  • The effects of ursodeoxycholic acid (UDCA) were studied on the hepatotoxicity induced by several hepatotoxicants such as carbonte trachloride, thioacetamide and 1-naphthylisothiocyanate in ICR male mice. UDCA (50 mg/kg, 100 mg/kg) decreased the elevated serum bilirubin in carbon tetrachloride intoxicated mice, the elevated serum AST, alkaline phosphatase in thioacetamide intoxicated mice, the elevated serum AST and bilirubin in 1-naphthylisothiocyanate intoxicated mice.
