Isolation and Characterization of Protease Producing B. amyloliquefaciens JH-35 from Food Waste (음식물 쓰레기로부터 Protease를 생산하는 B. amyloliquefaciens JH-35의 분리 및 특성)

  • BACKGROUND: Recent studies have described the importance of microbes and enzymes that can compost food waste. This study was carried out to improve production of protease of isolated microbes from food waste. METHODS AND RESULTS: Seven bacteria isolated from various sources were screened for protease production by adding skim milk into the agar medium. About 7 microbes producing protease were tested, and strain JH-35 showed the highest protease activity among them. The strain was identified as Bacillus amyloliquefaciens JH-35 based on morphological, cultural, physiological characteristics and 16S rRNA. In the fermentation experiment, the assay B. amyloliquefaciens JH-35 showed the highest protease activity in the condition of 1% glucose, 1.5% yeast extract and 0.2%$ K_2HPO_4$. The optimal condition of culture with temperature $35^{\circ}C$, initial pH of 7 and shaking speed of 200 rpm and 24 hr. CONCLUSION: The protease of the B. amyloliquefaciens JH-35 had its activity at pH 7 and the optimal culture time was 24 hr. Also, B. amyloliquefaciens JH-35 was high salt tolerance. Our results suggest that B. amyloliquefaciens JH-35 from food waste may have the potential to degrade protein and carbohydrate in food waste.

A Study of Glycemic Index, Glycemic Load and Food Sources according to Body Mass Index in Female College Students (여대생의 체질량지수에 따른 Glycemic Index, Glycemic Load와 급원식품에 관한 연구)

  • The purpose of this study was to evaluate nutrients intakes, glycemic index (GI), glycemic load (GL) according to body mass index (BMI) in female college students (n = 320). The study subjects were divided into 3 groups based on their body mass index, an underweight group (BMI < 18.5 kg/$m^2$, n = 55), a normal group (18.5 kg/$m^2$ ${\leq}$ < 23 kg/$m^2$, n = 231), and an overweight group (23 kg/$m^2$ ${\leq}$ BMI < 25 kg/$m^2$, n = 34). The food and nutrition intake data obtained by administering a 3-day food record and were analyzed by using Can pro 3.0 software. Anthropometric measurements were collected from each subject. Body weights and BMI of the underweight group were 45.9 kg, 17.6 kg/$m^2$, those of the normal group were 53.8 kg, 20.5 kg/$m^2$, and those of overweight group were 62.6 kg, 23.8 kg/$m^2$, respectively. The mean daily dietary GI of underweight, normal and overweight groups was 66.2, 65.8 and 66.5, respectively. These differences were statistically non-significant. The mean daily dietary GL of underweight, normal and overweight groups were 159.2, 149.4, and 148.9, respectively. The major food source of dietary GI and GL was rice in the three groups. Dietary GI and GL were not significantly correlated with obesity when adjusted for energy, carbohydrate and dietary fiber intake.

Current Achievement and Perspectives of Seed Quality Evaluation in Soybean (콩 품질평가 현황과 전망)

  • Soybean is one of the most important sources of protein and oil in the world. Recently, emphasis has been laid on the chemical composition of soybean seeds for the processing soybean foods. Improvement of soybean components has been expected to improve food-processing quality for the processed soybean products such as soymilk and various edible ingredients as well as fur the traditional soyfoods. In Korea, soybean breeding research programmes have been focused on the quality of the products derived from soybean with yield stability, and some new modified soybean varieties haying good food-processing quality were developed recently. So the efforts of establishing standard and standardization of products in soybean are important. Three main categories should be considered in view of soybean seed quality; the marketing value such as grain size, shape, and appearance; the eating and processing value such as dehulled ratio, water absorption rate, and benny flavor; the nutritional value such as protein, lipid, and carbohydrate contents. And the new frontiers in research are looking at the functional nutrients in soybeans and how to improve them. In case marketing value, mainly the appearance is evaluated, therefore, each country has an application of standard related to quality. Each determination of standard class, heat-damaged kernels, splits, and soybeans of other colors is made on the basis of the grain when free from foreign materials. But processing value and nutritional value for standardization were not studied in detail till now. In addition, soybean has potential roles in the prevention and treatment of chronic diseases, most notably cancer, osteoporosis, and heart disease. The functional nutrients include a protease inhibitor, phytic acid, saponins, and isoflavones, etc.. It is believed that standardization of soybean quality should perform to overcome the difficulties, relatively high price of domestic soybean products has weakened the competitive power, in the market related to WTO. So, we should focus on further research into the evaluation and establishment of quality-standard in soybean.

Nutritional Quality and Physicochemical Characteristics of Defatted Bovine Liver Treated by Supercritical Carbon Dioxide and Organic Solvent

  • Defatted bovine liver (DBL) is a potential source of protein and minerals. Supercritical carbon dioxide ($SC-CO_2$) and a traditional organic solvent method were used to remove lipid from bovine liver, and the quality characteristics of a control bovine liver (CBL), bovine liver defatted by $SC-CO_2$ ($DBLSC-CO_2$) at different pressures, and bovine liver defatted by organic solvent (DBL-OS) were compared. The $DBLSC-CO_2$ samples had significantly higher (p<0.05) protein, amino acid, carbohydrate, and fiber contents than CBL and DBL-OS. There was a higher yield of lipid from CBL when using $SC-CO_2$ than the organic solvent method. SDS-PAGE analysis demonstrated that the CBL and $DBLSC-CO_2$ had protein bands of a similar intensity and area, whereas DBL-OS appeared extremely poor bands or no bands due to the degradation of proteins, particularly in the 50 to 75 kDa and 20 to 25 kDa molecular weight ranges. In addition, $DBLSC-CO_2$ was shown to have superior functional properties in terms of total soluble content, water and oil absorption, and foaming and emulsification properties. Therefore, $SC-CO_2$ treatment offers a nutritionally and environmentally friendly approach for the removal of lipid from high protein food sources. In addition, $SC-CO_2$ may be a better substitute of traditional organic solvent extraction for producing more stable and high quality foods with high-protein, fat-free, and low calorie contents.

Aflatoxin: Factors Affecting Aflatoxin Production (Aflatoxin과 그 생성(生成)에 관련되는 주요인(主要因))

  • Aflatoxins are toxic and carcinogenic secondary metabolites which are produced by trains of A. flavus and A. parasiticus during their growth on foods and feedstuffs. Aflatoxins are a group of closely related heterocyclic compounds of which $B_1$, $B_2$, and $G_2$ are the major members. Aflatoxins are synthesized via a polyketide pathway in which the general steps are acetate, an-thraquinones, xanthone and aflatoxins. Aflatoxin formation is favored by high moisture or high $a_w$(0.95${\sim}$0.99). The limiting $a_w$ for aflatoxin production on agricultural commodities is 0.83. Optimum temperature for aflatoxin production by the molds is $25{\sim}30^{\circ}C$ and the incubation time for the maximum production of the toxin is 7${\sim}$15 days. The limiting temperatures for aflatoxin production are ${\leq}7.5^{\circ}C\;and\;\geq40^{\circ}C$. Cycling temperatures may or may not stimulate aflatoxin production depending on the amplitude of cycling, substrate and strains of molds. Aflatoxin pro-ducing molds are aerobic organisms and thus have a requirement for oxygen. A decreasing $O_2$ concentration and/or increasing concentrations of $CO_2$ or $N_2$ depress the mold growth and aflatoxin formation. A. flavus grows competitively or associatively in the presence of other microorganisms and occasionally loses the competition with other microorganisms. Some lactic acid bacteria have been shown to reduce growth and aflatoxin production by A. parasiticus. Carbon source is the most important nutritional factors affecting aflatoxin formation by the molds. Sucrose, fructose and glucose are the most favorable carbon sources. Food substrates of plant derived products which have high carbohydrate content such as agricultural commodities and their products are most vulnerable to contamination by aflatoxins.

The Nutritional Status of Various Populations Living in Selected Areas for Model Nutritional Work in Korea (시범영양사업지역 주민들의 영양실태조사 보고)

  • It is very important to collect information on the nutritional status of the Korean population for the development of health promotion programs including nutrition. The purpose of this study was to assess the nutritional status of various population living in selected areas for model nutritional work. Seven hundred eighty households(30 households per each area)from 26 areas participated in this study from November 1 to November 20, 1996. Dietary intake data for two consecutive days were collected at household level by a weighting method. The mean energy intake of the subjects(1,934kcal) was higher than that resulted from the ‘95 Korean National Nutrition Survey(1,839kcal). The proportion of energy derived from cereals was 60.1%. The proportion of total protein intake from animal sources was 49.4%. These results were similar to those found in the ‘95 Korean National Nutrition Survey. Most nutrients(except iron, thiamin, riboflavin, vitamin C, and crude fiber) were higher than the result of the ‘95 Korean National Nutrition Survey. However, the average iron intake was about 68% of the result of ‘95 Korean National Nutrition Survey. This may be due to the adjustment of iron content in rice(3.7mg/100glongrightarrow0.5mg/100g) included in nutrient database for calculating nutrient intakes. The mean energy contribution from carbohydrate, protein, and fat were 64.2%, 16.4% and 19.4%, respectively. Significant differences of nutrient intakes were noted among some areas, which may be due to different food intake patterns according to the needs of the particular area. Therefore, the result of this study indicates that there are significant differences in food and nutrient intakes among the areas, suggesting that nutritional improvement programs may need to be developed differently by areas.

Development and Effects' Analysis of Nutrition Education Program for Diabetes Mellitus at Community Health Center - Focused on Individual Daily Energy Requirements and Food Exchange Units - (당뇨병 성인 대상 보건소 영양교육 프로그램 개발 및 효과 분석 - 개인별 맞춤 하루 필요 에너지 및 식품군 단위수 교육을 중심으로 -)

  • The purpose of this study was to investigate effects of the developed nutrition education program focused on individual daily energy requirements and food exchange units using Food Exchange System for diabetes mellitus at a community health center. Developed the nutrition education program, four weeks' nutrition education including provided twice individual meal as diet therapy (2 hour/lesson/week, 4 week), was provided to 20 diabetic elderly (12 male, 8 female, 50-75 yrs): 1st lesson "Introduction: management of diabetes mellitus", 2nd lesson "6 Food groups and sources of 6 food groups", 3rd lesson "Individual daily energy requirements and food exchange units", and 4th lesson "Food choice for diabetes mellitus". For effects' analysis of the developed program, we assessed the changes in anthropometric characteristics; biochemical characteristics and nutrient intakes using 24 hr recall method. Effects of the developed nutrition education program were as follows: weight was significantly decreased, blood urea nitrogen (BUN) and glycosylated hemoglobin (HbA1c) were significantly decreased, and distribution of subjects in BUN and HbA1c was significantly changed. In protein : fat : carbohydrate (PFC) ratio, it was significantly changed from 15.98 : 16.30 : 66.69 to 17.51 : 18.94 : 64.10. In evaluation of nutrient intakes by Dietary Reference Intakes for Koreans (KDRI), protein, fiber, fat, vitamin E, niacin, folic acid, calcium and zinc were shown significantly positive changes in distribution of subjects according to intake level. The index of nutrition quality (INQ), nutrition adequacy ratio (NAR) and mean nutrition adequacy ratio (MAR) were significantly increased. In conclusion, the developed 4 weeks' nutrition education program focused on individual daily energy requirements and food exchange units using Food Exchange System for diabetes mellitus at community health center may improve the symptom of diabetes mellitus.

Degradation Characteristics of Wood Cellulose by Ruminal Cellulolytic Anaerobic Bacterium Ruminococcus albus F-40 (혐기성 세균 Ruminococcus albus F-40에 의한 목재 cellulose의 분해특성)

  • The degradation mode of lignocellulose by anaerobic ruminal cellulolytic bacterium Ruminococcus albus F-40 was investigated. Birchwood holocellulose and filter paper were incubated as the sole carbohydrate sources with using the Hungate techniques. After 2 or 4 days of incubation, samples were employed for chemical and electron microscopic evaluations. The degradation rate of cellulosic substrates and the adhesion rate of bacteria to the substrates increased proportionally with the decrease of relative crystallinity of cellulose, indicating the preferential breakdown of amorphous cellulose, by this bacterium. X-ray diffraction analyses and polarized light microscopy showed, however, that crystalline cellulose was also degraded by R. albus. FT-IR spectra indicated that not only cellulose but hemicellulose was also degraded by this bacterium. Electron microscopic investigations showed the protuberant structures on the surface of R. albus. These structures were much more significant when bacterial cells were grown in the media containing insoluble substrates, such as cellulose, indicating clearly that bacterial protuberant structures were induced by the substrates. Protuberant structures extended from the bacterial cells adhered tightly to the substrates and numerous vesicles covered the surface of cellulosic substrates affected. Cellulosome-like structures were distributed on the cellulose matrix. Electron microscopic works showed that diverse surface organells of R. albus were involved in the degradation of cellulosic materials. SEM examinations showed the breakdown of cellulose by R. albus was proceeded by severeal routes : short fiber formation, defibrillation and destrafication of cellulose microfibril.

Effects of Dietary Protein on Growth and Lipid Metabolism in Growing Rats (식이단백질조성이 흰쥐의 성장과 지방대사에 미치는 영향)

  • The effect of dietary protein on growth and lipid levels of plasma and liver was studied in weanling male rats fed diets differing protein sources and amino acid balance. Rats were devided into 9 experimental diets which were grouped into 3 categories ; 1) Simple protein category includes gluten-, soy protein isolate-, and casein-containing diet groups, 2) Supplemented category includes casein supplemented with methionine, soy protein isolate supplemented with methionine, and gluten supplemented with lysine and methionine, 3) Mixed protein category includes diet groups containing gluten (2/3), casein (1/3), soy protein isolate (2/3) and casein (1/3), and casein (1/3), soy protein isolate (1/3) and gluten (1/3). The experimental diets composed of 15% protein, 65.8% carbohydrate, 10% fat and 1% cholesterol. The body wt. gain and P.E.R. were greater in rats of supplemented and mixed protein groups than simple protein groups. No statistical differences were found in plasma cholesterol among gluten, soy protein isolate and casein groups. Consumption of diets supplemented with limiting amino acid to gluten or soy protein isolate reduced the plasma cholesterol level by 23.2% and 34.2% respectively. However there was no difference between casein and the supplemented casein groups. The mixed protein groups shows relatively high plasma cholesterol concentration and low liver cholesterol levels. On the other hand gluten group showed low plasma cholesterol and high liver cholesterol levels, which means body cholesterol pool may not have been changed by the dietary protein. Feeding soy protein meal and the supplemented soy protein isolate resulted in lower plasma cholesterol, plasma triglycerides, liver cholesterol and liver triglycerides levels. This hypolipidemic effect is considered to see unique to soy protein isolate. Rats in gluten and the supplemented gluten groups showed lower plasma protein levels and a tendency of fatty liver.

Complete genome sequence of Microbacterium aurum strain KACC 15219T, a carbohydrate-degrading bacterium (탄수화물 분해 세균 Microbacterium aurum KACC 15219T의 유전체 염기서열 해독)

  • The complete genomic information of Microbacterium aurum KACC $15219^T$ (= IFO $15204^T$ = DSM $8600^T$) is described. The genome of M. aurum KACC $15219^T$ contains 3,096 protein coding genes and an average G+C content of 69.9% in its chromosome (3.42 Mbp). This strain can use various carbon sources for growth, including quinic acid. Quinic acid is used as a substrate for the synthesis of aromatic amino acids via the shikimate pathway which are useful in the industry. M. aurum KACC $15219^T$ will provide basis to improve our understanding of this organism and allow more efficient application of the strain to industry.
