Sixty-seven isolates of bacterial spot pathogen (Xanthomonas spp.) collected from six provinces of Korea were tested for the identification of their pathotypes and determination of their distribution throughout Korea in an effort to genetically manage the disease. Near isogenic lines of Early Calwonder (Capsicum annuum) pepper plants carrying $Bs_1$, $Bs_2$ and $Bs_3$, and PI235047 (C. pubescens) were used as differential hosts. Race P1 was found to be predominant, followed by race P7, and races P3 and P8 were also observed. This is the first report of races P7 and P8 in Korea. The races P7 and P8 were differentiated from the former races P1 and P3, respectively, on the basis of their ability to elicit hypersensitive reactions to PI235047.

We evaluated 209 Capsicum accessions from the National Agrobiodiversity of RDA, for their reaction against of anthracnose disease caused by Colletotrichum acutatum. Two hundred nine accessions of Capsicum were almost composed with Capsicum annuum var. annuum originated from 37 countries. The percentage of infection due to C. acutatum ranged from 11.1% to 100% on immature and matured pepper fruits at 21 days after inoculation. On immature fruits of pepper, one hundred seventy three accessions of tested pepper were found infected with C. acutatum. Out of the 173 accessions, eighty five accessions were susceptible to anthracnose with dark brown to black lesions and slowly increased in size at 21 days after inoculation. Only thirty six accessions showed resistance against C. acutatum. On mature fruits of pepper, one hundred ninety eight accessions were infected with C. acutatum at 21 days after inoculation. Twenty one accessions showed a resistance reaction against C. acutatum. The results of this work indicated that all of resistance accessions to C. acutatum were have a genetic potential for the resources of resistance can be further used in pepper breeding programme against anthracnose and also demand more detailed investigation in this.
2002년 경남 하동군 진교면 노지재배 포장과 2003년 진주시 문산읍 촉성재배 비닐하우스 고추에서 이상증상이 발생하였다 병징은 지제부의 줄기가 수침상으로 물러지고 부패하면서 암갈색으로 변하고 시들면서 말라 죽는다. 병반부에 흰색의 곰팡이가 솜털처럼 생기고, 병반 부위와 토양 표면에 갈색의 둥근 균핵이 많이 형성된다 병원균의 균사생장 및 균핵형성은 감자한천배지에 배양시 3$0^{\circ}C$에서 가장 양호하였다. 균핵은 갈색 또는 암갈색으로 크기는 1.0∼2.7 mm이었다. 균사 특유의 clamp connection이 관찰되었고 균사 폭은 3.6∼10.2 $\mu\textrm{m}$이었다. 본 병원균을 Sclerotium rolfsii로 동정하였으며 고추 흰비단병으로 명명할 것을 제안한다.
An Efficient in vitro regeneration system and an optimized Agrobacterium mediated transformation protocol are described, based on the use of young seedling cotyledons of Capsicum annuum L. Optimal regeneration efficiency can be obtained by cultivating cotyledon explants on media containing 4 mg/L benzyladenine and 0.1 mg/L indolacetic acid. The effect of antibiotics used to eliminate Agrobacteria, as well as the toxic level of some generally used selection agents (kanamycin, geneticin, hygromycin, phosphinotricin and methotrexate) in regenerating pepper tissues were determined. To enable the comparison of different selection markers in identical vector background, a set of binary vectors containing the marker genes for NPTII, HPT, DHFR and BAR respectively, as well as the CaMV 35S promoter/enhancer-GUS chimaeric gene was constructed and introduced into four different Agrobacterium host strains.
A species of Heterobasidion was encountered during a diversity study of endophytic fungi from healthy root tissues of chili pepper (Capsicum annuum L.) in Korea. The fungal species (CNU081069) was identified as Heterobasidion araucariae based on phylogenetic analyses of the internal transcribed spacer and translation elongation factor gene sequences. Morphological descriptions of the endophytic isolate matched well with the previous references and supported the molecular identification. The fungus Heterobasidion araucariae CNU081069 is new to Korea.
Efficient plant regeneration has been achieved via organogenesis in the red pepper plant (Capsicum annum L.). Shoots were induced from seed explants of cultivar 'Friendship' on Murashige and Skoog's (MS) basal medium supplemetned with; NAA or IAA, and BAP or zeatin. Seed explants on the medium supplemented with 0.1-0.3 mg/L IAA and 2-5 mg/L zeatin for 2 weeks vigorously formed normal shoots in more than 90% of the explants. When these were transferred to MS medium containing 0.5-1.0 mg/L GA, 90-100% of the shoots have elongated within 1-2 weeks. The elongated shoots rooted in media supplemented with 0.3 mg/L NAA. It was revealed that this method is very rapid and efficient regeneration system for red pepper and regenerated plants can be obtained after only 5-6 weeks of culture.
Extracellular capsidiol, sesquiterpenoid phytoalexin, in the medium of pepper (Capsicum annuum L.) suspension cells was not identified from control cells, but highly accumulated in the elicitor-induced cells within 6 hours after the addition of 0.05$\mu\textrm{g}$/$m\ell$ cellulase. Capsidiol production in elicitor-induced cells was markedly suppressed by cytochrome P450 inhibitors, such as ancymidol and ketoconazole demonstrating that biosynthesis of capsidiol is catalyzed by at least on hydroxylation enzyme in the biochemical pathway. Based on protein electrophoresis, two bands, 23.0kDa and 27.5kDa, were identified as newly synthesized polypeptides in the elicitor-induced suspension cells, suggesting that pepper cells which were subjected to elicitor treatment activate specific gene(s) for capsidiol biosynthesis in cultured cells.
Potato virus X (PVX) resistance in potato is one of the best-characterized resistance models, however little is known in pepper. To evaluate the resistance to PVX in Capsicum annuum, a total of eleven pepper accessions were used for resistance screening against two PVX strains, USA and UK3. None of them were resistant against strain UK3, whereas four resistant genotypes were found against strain USA, three of which were further characterized. Two unlinked dominant genes were identified for both genotypes Bukang and Perennial; resistance in the genotype CV3 seemed to be conferred by two complementary dominant genes. These results demonstrated that the resistance to PVX in C. annuum is different from that in potato. This is the first report on genetic analysis of PVX resistance in C. annuum.
We have constructed a molecular linkage map of pepper (Capsicum spp.) in an interspecific $F_2$ population of 107 plants with 320 RFLP, 136 AFLP, and 46 SSR markers. The resulting linkage map consists of 15 linkage groups covering 1,720 cM with an average map distance of 3.7 cM between framework markers. Most RFLP markers ($80\%$) were pepper-derived clones and these markers were evenly distributed all over the genome. Genes for defense and biosynthesis of carotenoids and capsaicinoids were mapped on this linkage map. By using 30 primer combinations, AFLP markers were generated in the $F_2$ population. For development of SSR markers in Capsicum, microsatellites were isolated from two small-insert genomic libraries and the GenBank database. This combined map provides a starting point for high-resolution QTL analysis, gene isolation, and molecular breeding.
We have constructed a molecular linkage map of chili pepper (Capsicum spp.) in an interspecific (C. annuum cv. TF68 x C. chinense cv. Habanero) F$_2$ population of 107 plants with 150 RFLP and 430 AFLP markers. The resulting linkage map consists of 11 large (206-60.3 cM) and 5 small (32.6- 10.3 cM) linkage groups cover-ing 1,320 cM with an average map distance between framework markers of 7.5 cM. Most (80%) of the RFLP markers were pepper-derived clones and these markers were evenly distributed across the genome. By using 30 primer combinations, 444 AFLP markers were generated in the F$_2$population. The majority of the AFLP markers clustered in each linkage group, although PstI/MseI markers were more evenly distributed than Eco RI/MseI markers within the linkage groups. Genes for biosynthesis of carotenoids and capsaicinoids were mapped on our linkage map. This map will provide the basis of studying secondary metabolites in pepper.
