Seasonal variation in fatty acid composition in various parts of broccoli cultivars

  • To evaluate seasonal variation in fatty acid composition in broccoli, 12 commercial cultivars of broccoli were grown in spring and fall season at the field of NIHHS, and their floret, leaf and stem parts were used for the fatty acid composition analyses. Among 14 fatty acids detected in broccoli, linolenic, palmitic and linoleic acids were major fatty acids comprising more than 80% of total fatty acids in both the seasons and all the parts. Likewise, stearic and oleic acids were also present in considerable amount while remaining fatty acids; caproic, lauric, myristic, pentadecanoic, palmitoleic, heptadecanoic, arachidic, behenic and lignoceric acids showed their minor compositional ratio. Among the three parts, stem exhibited highest SFAs (49.681% in spring and 50.717% in fall season) compared to MUFA and PUFA, while highest compositional ratio of PUFAs were observed in leaves (62.588% in spring and 68.931% in fall season), which indicates leaves as a good source of health beneficial fatty acids. In contrast, floret part exhibited highest SFA (48.786%) and PUFA (57.518%) in spring and fall seasons, respectively. Major fatty acids; palmitic, linoleic and linolenic acid showed lowest cultivar dependent variation (below 10%) and leaf showed least variation in both the seasons compared to floret and stem. Our results suggest that all the fatty acids are significantly influenced by genotype of cultivars (C), plant parts (P) and growing seasons (S). Among the 14 fatty acids, myristic and palmitic acid showed highest positive or negative correlationship with oleic (r=$0.912^{**}$) and linolenic acid (r=-$0.933^{**}$), respectively. The most abundant fatty acid, linolenic acid, showed either negative or no correlation ship with other fatty acids while palmitic acid, a second major fatty acid, exhibited either positive or negative correlation ship.

  • By modified method yellow corvenia(called $Y_3$) was prepared with $4\%$ salt, $4\%$ KCl, $6\%$ sorbitol, $0.5\%$ lactic acid and $4\%$ alcohol extract of red pepper to improve the quality of fermented sea food. In this study, changes of volatile compounds and fatty acid composition obtained from modified fermented yellow corvenia($Y_3$) were experimented during fermentation, comparing with conventional fermented yellow corvenia(called $Y_1,\;20\%$ of salt contents). Total lipid of yellow corvenia was composed of $78.1\%$ of neutral lipid, $21.2\%$ of phospholipid and $0.7\%$ of glycolipid. And monoeonoic acid was held $37.4\%$ of fatty acid composition of total lipid and saturated fatty acid ($34.8\%$), polyenoic acid ($27.7\%$) were followed. Saturated fatty acid($C_{14:0},\;C_{16:0},\;C_{18:0}$) in $Y_1,\;Y_3$ increased, polyenoic acid ($C_{22:6}\;C_{22:5}\;C_{20:5}$) decreased while monoenoic acid($C_{16:1}\;C_{18:1}$) in those was little fluctuated during fermentation. Thirty-three kinds of volatile component in whole volatile compounds obtained from $Y_1,\;Y_3$ at 90 days fermentation were identified, and composed of some hydrocarbons (8 kinds), alcohols (7 kinds), acids (6 kinds), aldehydes(4 kinds), sulfides(2 kinds), ketones (2 kinds), one of phenol and 3 kinds of the other components. Among the whole volatile compounds 2-ethoxy ethanol and was held $79.35\%$ in $Y_3$, whereas nonadecane was held $75.85\%$ in $Y_1$. During fermentation 8 kinds of volatile acids, 5 kinds of amines and 9 kinds of carbonyl compounds were also detected. Those volatile acid such as acetic acid, isovaleric acid, n-caproic acid, n-butyric acid were the major portion of total volatile acids in $Y_3$ at 90 days fermentation. Meanwhile, carbonyl compounds such as ethanal, 2-butanone and butanal were the major ones, while TMA held the most part of volatile amines in $Y_3$ during fermentation. From the result of sniff test, the components which are believed to contribute to the characteristic flavor of fermented product $Y_1,\;Y_3$ are deduced to be volatile acid, carbonyl compounds and amines in order. Conclusively, there was little difference in composition of volatile components, but merely a little difference in content of those between $Y_3$ and $Y_1$.

  • The changes of the content of organic acids, carbon dioxide, alcohols and carbonyl compounds of the various Kimchis which were made of cabbage with green onion, garlic, ginger or red pepper and fermented at $12^{\circ}C-16^{\circ}C$ were investigated. Nonvolatile organic acids identified were lactic, succinic, fumaric and malic acid. Volatile organic acids identified were acetic, formic, propionic, butyric, valeric, n-caproic and n-heptanoic acid. Carbonyl compounds identified were acetaldehyde and acetone. The content of lactic acid was increased with fermentation, and higher in Kimchis containing red pepper, garlic and green onion. The content of acetic acid was increased with fermentation, especially in Kimchi containing garlic. The content of carbon dioxide was higher in Kimchi containing garlic. Alcohols identified in all Kimchis was only ethanol. Carbonyl compounds had no direct effect on off-flavor of Kimchi.

Quality Characteristics of Jocheong Containing Various Level of Letinus edodes Extracts (표고버섯 추출액을 이용한 조청의 품질특성)

  • Jocheong was prepared by adding Lentinus edodes extract to improve its quality and to give some functional properties. Contents of crude protein, ash and crude lipid were similar to those of control, while carbohydrate content was decreased. Total mineral content were $1,916.03\~2,674.24mg/kg$ that was no difference between test samples. From HPLC determination of free sugars, Jocheong was found to contain maltose as the highest sugar, followed by glucose and fructose. In amino acid analysis, seventeen amino acid were identified and quantified. Glutamic acid in Jocheong was major amino acid. The major fatty acids in Jocheong $(0\%,\;control)$ were linoleic acid, palmitic acid, oleic acid, myristic acid and caproic acid. There was no significant differences in fatty acid composition, pH and reducing sugar content among the Jocheong samples. The viscosity and solid contents tended to decrease with the addition of Lentinus edodes extract. Increasing the ratio of mushroom extract in Jocheong tended to decrease the lightness, yellowness and redness in Hunter's color value. Although sensory value decreased with increasing Lentinus edodes extracts, use of mushroom extracts (7: 3; saccharification liquids: Lentinus edodes extracts) is recommended for making Jocheong.

  • Free fatty acids of two brands of domestic butter were isolated directly by a modified silicic acid column chromatography, and were analyzed by gas liquid chromatography. $C_{18}$ FFA congeners $(C_{18_0},\;C_{18:1},\;C_{18:2}\;and\;C_{18:3})$ were the predominant components (52.83% in brand A and 47.50% in brand B), followed by $C_{16}$FFA (29.39% in brand A and 30.52% in brand B) and $C_{14}$FFA (11.85% in brand A and 13.76% in brand B). The other FFA were present as minor components (0.29-3.87%). Concentrations of four FFA $(C_4,\;C_6,\;C_{10}\;and\;C_{12})$FFA which would be expected to contribute strongly to hydrolytic rancidity off-flavors were below individual threshold level, except $C_4FFA$ (56 ppm) in butter B.

Flavor Components in Sun-Dried Ray (마른 가오리의 풍미성분(風味成分))

  • To investigate the flavor components of sun-dried ray, Raja porasc, the contents of such as free amino acids, nucleotides and their related compounds, total creatinine, betaine, TMAO, fatty adds, and volatile compounds were analysed. The content of total free amino acids was 1773.3mg% on dry basis and the abundant amino acids were taurine, lysine, leucine, alanine, glycine, glutamic acid, proline, and valine in order and these amino acids accounted for 80% of total free amino acids in sun-dried ray. In the nucleotides and their related compounds, hypoxanthine was the most abundant. Ammonia-N was the most abundant, resulting 26.4% of extractive nitrogen which was 1965.6mg% and next free amino acid-N, total creatinine-N, and nucleotide-N in order. In the fatty acid composition of total lipid and neutral lipid, polyenoic acid was abundant holding 42.0%, 45.3% respectively. In phospholipid, the ratio of polyenoic acid and saturated fatty acid were similar to 39% respectively. The predominant fatty acids in total lipid, neutral lipid, and phospholipid were $C_{16:0},\;C_{22:6},\;C_{18:1},\;C_{18:0},\;and\;C_{20:4}$. In the volatile compounds, isocaproic acid, caproic acid, isobutyric acid, and butyric acid were the major portion of the 8 kinds of volatile fatty acids detected and 2-butanone, pentanal, and 2-methylpropanal were the major portion of the 8 kinds of volatile carbonyl compounds detected and also trimethylamine was the major portion of 2 kinds of volatile amines detected. It was presumed from the results that the reciprocal action of taste compounds such as free amino acids, nucleotides and their related compounds total creatinine, betaine, and TMAO and volatile compounds such as volatile carbonyl compounds, volatile fatty acids, volatile amines, and ammonia played an important role for the characteristics of flavor of sun-dried ray.

복분자 열매의 화학성분 및 휘발성 향기성분

  • 최근 복분자(산딸기)의 효능을 이용한 일반식품 및 기능성 식품 등의 제품 개발이 활발히 추진되고 있다. 따라서 본 연구에서는 식품차원의 연구분야 일환으로서 먼저 복분자의 일반성분, 유리당, 유리아미노산, 무기성분 및 향기성분 등을 보고한 후 앞으로 생리 활성 연구 분야에 초점을 두고 연구를 진행할 하여 학문적 자료 및 산업에 응용할시 기초자료로 활용하고자 한다. 복분자의 수분은 5.39%, 총당은 17.3%, 환원당은 8.6%, 조단백질 10.6%, 조회분은 4.5%, 조지방은1.7%, 조섬유는 3.9%이고, 유리당 함량은 sucrose 1.52%, fructose 3.98%, glucose 1.24%이고, 유기산 함량은 citric acid 10.2%, oxalic acid 6.29%로 가장 많이 함유하고 있었고, 그 다음은 malic acid 1.94%였다. 총 아미노산은 7914.1mg%이고, 필수아미노산 함량은 2622.5mg%로 나타났다. 복분자에 가장 많이 함유되어 있는 아미노산은 aspartic acid 1260.3%, 그 다음은 glutamic acid 1054.3% 순으로 함유하고 있었다. 무기성분 K가 387mg으로 가장 많이 함유되어 있었고, 복분자에서 총 52종의 성분이 화인되었는데 확인된 성분들은 관능기별로 분류하면 amylalcohol를 포함한 alcohol류 11종, valeric acid를 포함한 acid류 13종, hexanal를 포함한 carbonyl류 20종, 2-heptanone를 포함한 hydrocabon류 5종 그리고 methyl palmitate를 포함한 ester류가 3종 이었다. 관능기별로 확인된 성분들의 총 peak area%를 살펴보면 alcohol류 11.56%, acid류 38.87%, hydrocarbon류 2.89%, ester류 0.80%, 총 70.32%가 확인되었다. Alcohol류에서 linalool가 3.78%, acid류에서는 caproic acid류가 14.40%, carbonyl류에서 2-hydroxy-4-methoxyacetophenone이 2.99%, hydrocabon류에서는 aromadendrene가1.59% 그리고 ester류에서는 methyl palmitate가 0.43%으로 가장 많이 함유하고 있었다.

  • Polyesteramide(PEA) has been synthesized with different molar ratio of 6-aminocaproic acid(ACA) and oligo butylene terephthalate(OBT) prepared from Dimethyl terephthalate(DMT) and 1, 4-butanediol(BD) by the melt polymerization. Oligomer and polymer molecular structure were characterized by IR spectra, H'-NMR spectra. Also the thermal properties were examined by thermogravimetric analysis(TGA) and differential scanning calorimetry(DSC). DSC and TGA results exhibited that the$ T_m$ s were detectable in the vicinity of $170-220^{\circ}C$ and the Ta's were detectable in the vicinity of $380-390^{\circ}C$ . X-ray diffraction analysis results show that the synthesized polyesteramide has high degree of crystallinity.

Co-expression of Gamma-Aminobutyrate Aminotransferase and Succinic Semialdehyde Dehydrogenase Genes for the Enzymatic Analysis of Gamma-Aminobutyric Acid in Escherichia Coli

  • Gamma-aminobutyric acid (GABA) aminotransferase (gabT) and succinic semialdehyde dehydrogenase (gabD) genes from Pseudomonas fluorescens KCCM 12537 were cloned into a single pETDuet-1 vector and co-expressed in Escherichia coli BL21(DE3) simultaneously. The mixture of both enzymes, called GABase, is the key enzyme for the enzymatic analysis of GABA. The molecular mass of the GABA aminotransferase and succinic semialdehyde dehydrogenase were determined to be 52.8 and 46.7 kDa following computations performed with the pI/Mw program, respectively. The GABase activity between pH 6.0 and 9.0 for 24 h at $4^{\circ}C$ remained over 75%, but under pH 6.0 decreased rapidly. The GABase activity between 25 and $35^{\circ}C$ by the treatment at pH 8.6 for 30 min remained over 80%, but over $35^{\circ}C$ decreased rapidly. When the activity against GABA was defined as 100%, the purified GABase activity against 5-aminovaleric acid having a similar structure to GABA showed 47.7% and GABase activity against ${\beta}$-alanine, ${\varepsilon}$-amino-n-caproic acid, $_L$-ornithine, $_L$-lysine, and $_L$-aspartic acid showed between 0.3 to 2.3%. The GABA content was analyzed with this co-expressed GABase, compared with the other GABase which was available commercially. As a result, the content of GABA extracted from brown rice, dark brown rice, and black rice were $26.4{\pm}3.5$, $40.5{\pm}4.7$ and $94.7{\pm}9.3{\mu}g/g$, which were similar data of other GABase in the error ranges.

  • A strain of Streptomyces sp. which producing a metal containing proteolytic enzyme was isolated from soil and some properties of this enzyme were investigated. The following results were obtained. 1. Optimal pH and temperature of this enzyme were pH7.0 and 37$^{\circ}C$ 2. This enzyme was easily inactivated with heat treatment: for example, by the treatmentat 37$^{\circ}C$ for 100 minutes the activity of the enzyme was decreased to about 50 per cent of intial actively but this enzyme was stable at neutral pH. 3. The activity of this enzyme was not inhibited by $Mg^{++}$, $_Mn^{++}$, $Ca^{++}$, Pb$^{++}$, or $Ba^{++}$ ect. but strongly inhibited by Hg$^{++}$, Co$^{++}$, Ag$^{+}$, Cu$^{++}$, or Cd$^{++}$. 4. This enzyme was strongly inhibited by EDTA but was not inhibited by oxalic acid, citric acid, 2, 4-dinitrophenol, $\varepsilon$-aminocaproic acid, cysteine, or thiourea.
