Proteomic analysis reveals that the protective effects of ginsenoside Rb1 are associated with the actin cytoskeleton in β-amyloid-treated neuronal cells

  • Background: The ginsenoside Rb1 (Rb1) is the most abundant compound in the root of Panax ginseng. Recent studies have shown that Rb1 has a neuroprotective effect. However, the mechanisms underlying this effect are still unknown. Methods: We used stable isotope labeling with amino acids in cell culture, combined with quantitative mass spectrometry, to explore a potential protective mechanism of Rb1 in ${\beta}$-amyloid-treated neuronal cells. Results: A total of 1,231 proteins were commonly identified from three replicate experiments. Among these, 40 proteins were significantly changed in response to Rb1 pretreatment in ${\beta}$-amyloid-treated neuronal cells. Analysis of the functional enrichments and protein interactions of altered proteins revealed that actin cytoskeleton proteins might be linked to the regulatory mechanisms of Rb1. The CAP1, CAPZB, TOMM40, and DSTN proteins showed potential as molecular target proteins for the functional contribution of Rb1 in Alzheimer's disease (AD). Conclusion: Our proteomic data may provide new insights into the protective mechanisms of Rb1 in AD.

A Surgical Treatment of the Esophageal Foreign Body (10 cases report) (수술적 처치가 필요했던 식도이물에 관한 임상적 고찰)

  • Ten cases with esophageal foreign body were treated surgically from July 1980 to October 1995 at the Departme t of Thoracic and Cardiovascular Surgery, Chungnam National University Hospital. The mean age was 45.3 years, with a range from 25 to 71. Out of ten cases, 6 were female and four were male. Common symptoms were dysphagia, fever, foreign body sensation and neck pain. Three cases of foreign bodies were of fish bones, two of bubble paclcage of drugs, one case of a Eeer bottle cap, one of a piece glass, one of a bathtub plug, one of chicken and one of a bean. The diagnosis was established by esophagography using a water soluble contrast material and esophagoscopy. Among of ten cases, two had esophageal stricture due to the ingestion of Aye at a young age. One case had experienced psychological problems. All foreign bodies were removed by surgical procedures. Five cases were treated by cervical esophagostomy, one case by right thoracotomy, one case by retrograde bougienation through gastrostomy and two cases by cervical incision and drainage for cervical abscess. Three cases developed pos operative esophageal leaks which healed spontaneously and transient hoarseness developed in one case. One case developed traumatic pneumothorax and subcutaneous emphysema which was treated by closed thoracostomy. There were no operative deaths. C(ocean J Thorac Cardiovasc Surg 1997;30:1117-20)

Comparison and analysis of men's classic-fit and slim-fit shirt patterns - Utilizing the 3D virtual try-on system - (남성의 클래식핏과 슬림핏 셔츠 패턴 비교 분석에 관한 연구 - 3D 가상착의를 활용하여 -)

  • This study aims to compare the ease of shirt patterns in the men's wear market that have been constantly developing, and to analyze the fitness and appearance through the 3D virtual try-on system. The study selected three industrial patterns and two educational materials of classic-fit and slim-fit shirts for a total of ten items. The experiments involved virtual wearing, a stress contact point perspective map, and appearance evaluations. First, the ease differed significantly in the chest, waist, and sleeve cap height according to the patterns. Second, based on the stress, contact point, and transparency, there was ease in the order of Nam, D-brand, J-brand, Park, and S-brand in the classic-fit shirts while the order of D-brand, Nam, J-brand, Park, and S-brand resulted in the slim-fit shirts. Third, in the appearance evaluation, higher points resulted in the order of J-brand, Park, S-brand, D-brand, and Nam in both classic-fit and slim-fit. The results showed that proper ease differs greatly by shirt-fit and target age. Also, the shirts with more ease tended to be evaluated less favorably in the appearance evaluation. Based on the results, the study suggests that the proper ease in chest circumference is 18cm for the classic-fit shirts and 6 cm for the slim-fit shirts.

Delimitation of Russula Subgenus Amoenula in Korea Using Three Molecular Markers

  • Distinguishing individual Russula species has been difficult due to extensive phenotypic plasticity and obscure morphological and anatomical discontinuities. Due to highly similar macroscopic features, such as the presence of a red-cap, species identification within the Russula subgenus Amoenula is particularly difficult. Three species of the subgenus Amoneula have been reported in Korea. We used a combination of morphology and three molecular markers, the internal transcribed spacer (ITS), 28S nuclear ribosomal large subunit (LSU), and RNA polymerase II gene (RPB2), for identification and study of the genetic diversity of Russula subgenus Amoenula in Korea. We identified only two species in Korea (R. mariae and R. violeipes); these two species were indistinguishable according to morphology and LSU, but were found to be reciprocally monophyletic species using ITS and RPB2. The markers, ITS, LSU, and RPB2, have been tested in the past for use as DNA barcoding markers, and findings of our study suggest that ITS and RPB2 had the best performance for the Russula subgenus Amoneula.

Application of Modified-WASP5 for Daecheong Dam Watershed Management (대청댐 유역관리를 위한 수정-WASP5 모형의 적용)

  • The objective of this study was to develop and apply a water quality simulation model for the evaluation of ungaged watershed. The Modified WASP5 consisted of three sub-models, LOAD-M, DYN-M, and EUT-M. LOAD-M, an empirical model, estimates runoff loadings using point and non-point source data of villages. Daecheong Dam watershed was selected for the research to calibrate, verify and application of Modified-WASP5. LOAD-M model was established using field data collected from all items of water quality and water quantity gaging stations of the watersheds, and was applied to the ungauged watersheds, taking the watershed properties under consideration. The result of water quality simulation using ModifiedWASP5 shows that the observed BOD data of Yongpo and Daechong Dam in 1999 were 0.8 mg/L and 1.0 mg/L, and simulated data were 0.9 mg/L and 0.9 mg/L, respectively. In case of 1999, average BOD concentrations were 0.8 mg/L and 1.0 mg/L. Simulated concentration showed 1.1 mg/L and 1.5 mg/L, respectively. Generally, the simulation results were in good agreement with the observed data. This study was focused on formulating an integrated model for evaluating ungauged watersheds. Even though simulation results varied slightly due to limited availability of data, the model developed in this study would be a useful tool for the assessment and management of ungauged watersheds.

  • Global warming is happening now and inevitable. Everyone knows that immediate action should be taken to slow it down, but uncertain about the effective solution. Despite global efforts to reduce greenhouse gas emissions, sea levels are rising gradually. In 2013, Cornell University announced the Climate Action Plan(CAP) to make the campus greener, to reduce waste, and to ensure efficient use of resources. In particular, they set a goal of reducing energy use by 2050 and making carbon emissions to zero. Accordingly, the purpose of this study is to analyse the case of the master plan of Cornell Tech campus and its major buildings. Mainly, The House, faculty and student housing of Cornell Tech and the world tallest certified passive house, will be the main precedent that shows the architectural planning of passive house. Passive house technology, which was thought to be possible only in single-family houses, can be applied to high-rise buildings. If any passive house technology of The House project is actively introduced into the dormitory projects of domestic universities that are about to be built or renovated, it will be a good opportunity for the university to take the lead in preparing for global warming.

A Statistical Review on States Relating to Operation of Radiotherapy Equipments in Seoul National University Hospital (서울대학교병원의 방사선치료장비 운용 통계에 관한 고찰)

  • To analyze the states of operation of radiotherapy facilities in the period from 1979 to 1992 and to get the base for efficient operation and maintenance of the radiotherapy facilities. Data on the records of annual number of patients operated by each facility; span of suspension of operation, the cost and span of repairing, and parts of oui-of-order in the period from 1979 to 1992 were analyzed. We made a comparative analysis of average annual number of patients, annual span of the suspension of operation, annual cost ratio of repair, span of repairing per break down, and total number of broken parts. We could get following annual number of patients(day), span of the suspension of span ($\%$)(day). annual cost ratio fo repair($\%$), span of repairing per break down(Min-Max, day), and number of broken parts from this analysis. 1. Cobalt unit (Picker C-9) : 10,389(43), 0.4(0.83) 0.07, 1hr-2, 3 2. Linac(Clinac 6/100) : 11,492(50), 4.0(9.57), 0.98, 1hr-30, 12 3. Linac(Clinac 18) : 9.115(44), 12.7(30.5), 3.54, 1hr-108. 41 4. Simulator(Picker Ther-X) : 2,017(9), 0.51(1.3), 0.24, 1hr-2, 7 5. RTP(Capentec Cap-plan) : 528(2), 0.4(0.93), - hrs, - The conclusion obtained from statistical analysis above are as follows. 1. The rate of operation of Cobalt unit($99.6\%$) was higher that of Linear Accelerators ($87.3\%$). The rates operation of Simulator and RTP computer were very close to that of Cobalt unit. 2. In order to raise up the working ratio of accelerator. it is desirable that we keep our engineer to learn a sufficient technical skill and the equipment agent to stock sufficient spare parts. 3. In order to maintain Linear Accelerator efficiently, it is desirable to have annually $2.3\%$ of the purchase price of equiment for repairing.

Comparative Bioavailability and Metabolism of Two Capsule Formulations of Fluoxetine in Human Volunteers (플루옥세틴 캅셀제의 지원자에 대한 생체이용율 및 대사율 비교)

  • Fluoxetine is a nontricyclic antidepressant which blocks serotonin reuptake selectively. Its N-demethyl metabolite, norfluoxetine is also selective inhibitor of serotonin uptake . This study was carried out to compare the bioavailability of Myung-in fluoxetine (20mg/cap.) with that of Prozac$^{\circde{R}}$. The bioavailability was conducted on 24 healthy volunteers who received a single dose (80mg) of each drug in the fasting state, in a randomized balanced 2-way crossover design. After closing, serial blood samples were collected for a period of 48 hours, Plasma was analyzed for fluoxetine and norfluoxetine by a sensitive and validated HPLC assay. The major pharmacokinetic parameters ($AUC_{0-48\;hr}$, Cmax, Tmax , $AUC_{inf.}$, MRT. $T_{1/2}$, Vd and Cl) were, calculated from the plasma fluoxetine concentration-time data of each volunteer. The microcomputer program, 'WinNonlin' was used for compartmental analysis. A two-compartment model with first-order input, first-order output and no lag time was chosen as the most appropriate pharmacokinetic model. The data were best described by using a weighting factor of $1/y^2$. Though the plasma fluoxetine concentrations of Myung-in fluoxetine were higher than those of Prozac$^{\circde{R}}$ at all observed time from 7.9% to 16.9% (P<0.05 at 6.7 and 10 hr), the bioavailability of Myung-in fluoxetine appeared to be bioequivalent with that of Prozac$^{\circde{R}}$. There were no statistical significant differences between the two drugs in all pharmacokinetic parameters including $AUC_{0-48\;hr}$ of norfluoxetine.

Establishment and Characterization of Clonal Cell Lines from Zebrafish, Danio rerio (제브라피쉬(Danio rerio) 배아로부터 동형세포주 확립)

  • Three types of clonal cell lines were isolated according to their size and phenotype from the adherent cell populations in long-term liquid cultures from the embryonic fibroblast cells of Zebrafish, Danio rerio. All kind of cell lines were well proliferated. The size and number of clonal cell lines derived colonies from stable embryonic cells were significantly increased in the presence of NAC and A2P conditioned medium from the cell lines. The stable cell lines and clonal cell lines were cap-able of well proliferation in vitro. These cell lines have been maintained in continuous culture without change in characteristics. A majority of the clonal cells (80%) was shown a normal chromosomal complement (50 chromosomes, 2N) in according with FACs analysis. Majority of cells were positive to vimentin staining and none of them were positive for nestin and Oct -4 by immunocytochemistry. These results indicate that the clonal cell lines obtained from cultured cells are fibroblasts and may be extremely useful in genetic manipulation for further nuclear transfer and fish cloning.

  • The concept of community capacity is regarded as the ability of people and communities to do works associated with the determinant factors and indicators of the circumstances of socio-economic, environmental and physical contexts. Building capacity of communities to effectively analyze our problematic issues and planning of community development is often required to scrutinize current status of community of socio-economic and infrastructural capacity development with GIS. We consider community development as a planned effort to build assets that increase the capacity of communities. Spatial asset mapping is the process enabling to identify and make inventories of tangible and intangible assets. This mapping requires developing a capacity inventory that collects individual organizational and community capacities in view of human, socio-cultural, natural, financial, digital, and physical capacity. The purpose of this research is not only designed to suggest a new concept capacity building, but also proposes a more creative framework of asset-based community cap linking to parcel-based spatial asset mapping and capacity mapping process.
