  • It is well known that p38 mitogen-activated protein kinase (p38MAPK) participates in cellular responses to mitogenic stimuli, environmental and genotoxic stresses, and apoptotic agents. Although there are several reports on p38MAPK in relation to cell growth and apoptosis, the exact mechanism of p38MAPK-mediated cell growth regulation remains obscure.(omitted)

영지버섯의 항산화 효능과 암세포 생장저해도 (Antioxidant activity and Cancer cell growth inhibition of Ganoderma lucidum)

  • 영지버섯 자실체를 열수추출과 주정추출을 하여 항산화 효능 및 간암 및 위암세포의 생장 저해도를 분석하였다. 항산화 효능을 실시한 결과, 대조군인 Trolox, BHA보다 높은 항산화 효능을 보인 것은 ASI 7004, 7014이며, 이 두 균주는 열수추출물과 주정 추출물에서 모두 높은 항산화 효능을 나타냈다. 나머지 균주는 대조군인 ABTs보다 대체적으로 높은 효능을 보였다. 또한 암세포 생장저해도를 알아보기 위해서 열수, 주정 추출물을 간암세포인 HepG2에 농도별로(100, 200, $400{\mu}g/ml$) 처리하여, 세포 생장 저해도(MTT assay)를 측정한 결과, 열수 추출물에서는 ASI 7002, 7011, 7014, 7020이 농도 의존적으로 간암세포의 생장을 저해하는 것을 알 수 있었다. 또한 주정 추출물에서는 ASI 7011, 7019가 농도 의존적으로 간암세포 생장을 저해하는 것으로 나타났다. 위암세포인 AGS에 농도별로(100, 200, $400{\mu}g/ml$)처리하여, 세포 생장 저해도(MTT assay)를 측정한 결과, 열수 추출물에서는 ASI 7001, 7002, 7019, 7020이 농도 의존적으로 위암세포의 생장을 저해하는 것을 알 수 있었다. 또한 주정 추출물에서는 ASI 7001, 7002가 농도 의존적으로 위암세포 생장을 저해하는 것으로 나타났다.

In vitro and in vivo Evaluation of the Antitumor Efficiency of Resveratrol Against Lung Cancer

  • Lung cancer remains a deadly disease with unsatisfactory overall survival. Resveratrol (Res) has the potential to inhibit growth of several types of cancer such as prostate and colorectal examples. In the current study, we evaluated in vitro and in vivo anticancer efficiency of Res in a xenograft model with A549 cells. Cell inhibition effects of Res were measured by MTT assay. Apoptotis of A549 cells was assessed with reference to caspase-3 activity and growth curves of tumor volume and bodyweight of the mice were measured every two days. In vitro cytotoxicity evaluation indicated Res to exert dose-dependent cell inhibition effects against A549 cells with activation of caspase-3. In vivo evaluation showed Res to effectively inhibit the growth of lung cancer in a dose-dependent manner in nude mice. Therefore, we believe that Res might be a promising phytomedicine for cancer therapy and further efforts are needed to explore this potential therapeutic strategy.

Ubiquitination of p53 is Involved in Troglitazone Induced Apoptosis in Cervical Cancer Cells

  • Peroxisome proliferator-activated receptor gamma (PPAR-${\gamma}$), a ligand-dependent nuclear transcription factor, has been found to widely exist in tumor tissues and plays an important role in affecting tumor cell growth. In this study, we investigated the effect of PPAR-${\gamma}$ on aspects of the cervical cancer malignant phenotype, such as cell proliferation and apoptosis. Cell growth assay, Western blotting, Annexin V and flow cytometry analysis consistently showed that treatment with troglitazone (TGZ, a PPAR-${\gamma}$ agonist) led to dose-dependent inhibition of cervical cancer cell growth through apoptosis, whereas T0070907 (another PPAR-${\gamma}$ antagonist) had no effect on Hela cell proliferation and apoptosis. Furthermore, we also detected the protein expression of p53, p21 and Mdm2 to explain the underlying mechanism of PPAR-${\gamma}$ on cellular apoptosis. Our work, finally, demonstrated the existence of the TGZ-PPAR-${\gamma}$-p53 signaling pathway to be a critical regulator of cell apoptosis. These results suggested that PPAR-${\gamma}$ may be a potential therapeutic target for cervical cancer.

속수자 추출물의 HT-29 대장암세포 증식에 대한 억제효과 (Inhibitory effects of Euphorbiae lathyridis Semen extract on cell growth in HT-29 human colon cancer cells)

  • Objectives. In this study, we investigate that methanol extract of Euphorbiae lathyridis Semen contributes to growth inhibitory effect on the HT-29 human colon cancer cells. Methods. Euphorbiae lathyridis Semen (ELS) was extracted with 80% methanol. HT-29 cells were treated with different concentrations of ELS extract for 24-72 hrs. Growth inhibitory effect was determined by MTT assay. Cell apoptosis was determined by surveying caspases cascades activation using Western blot. Cell cycle arrest was analyzed by flow cytometry with PI staining. Results. Exposure to ELS extract showed in inhibitory effects on HT-29 cell growth as a dose-dependent manner. Cell growth inhibition by ELS extract was related with induction of cell apoptosis with DNA fragmentation through the activation of caspases-3, caspase-9 and PARP cleavage. Conclusion. ELS extract significantly inhibited cell growth and induced cell apoptosis in HT-29 human colon cancer cells, therefore, These results suggest that ELS extract can be used as chemoprevention agent of colon cancers.

사람의 암세포주 및 정상세포주에서 역전사 효소의 억제에 의한 세포 성장의 제한 (Arrest of Cell Growth by Inhibition of Endogenous Reverse Transcription Activity in Cancer and Somatic Cell Lines)

  • 이 연구는 여러 종류의 암세포주(A-549, AGS, HCT-116, MDA-MB-231 및 U 87-MG)와 정상세포주(MRC-5 및 MSC)에 RNA를 DNA로 전환시킬 수 있는 역전사 효소의 억제 처리 후 세포의 성장에 미치는 영향을 비교 조사하였다. 각 세포주에 efavirenz (EFA) 역전사 효소 억제제를 1주일 동안 처리하였을 때, 세포 성장의 반억제농도(IC50) 값은 암세포주보다 정상세포주에서 더 높은 값을 나타냈다. 결정된 IC50 값에 따라 15 µM 농도로 EFA를 1주일 동안 처리하였을 때, 역전사 효소 및 말단소립 복원 효소의 활성은 EFA 처리군에서 비처리군에 비하여 유의적으로(p<0.05) 감소하였다. 그러나, 역전사 효소 및 말단소립 복원효소의 활성은 정상세포주에서는 검출되지 않았다. 15 µM EFA를 처리한 후, 암세포주와 정상세포주에서 세포성장율을 비교하였을 때, EFA 처리는 모든 세포의 성장을 억제하였는데, 정상세포주보다 암세포주에서 세포의 성장율이 현저하게(p<0.05) 감소하였다. 또한, 역전사 효소의 억제가 세포의 노화 및 사멸에 미치는 영향을 분석하기 위해 노화 관련 ß-galactosidase 효소의 활성을 분석하였을 때, EFA 처리가 노화관련 효소 활성이 점점 증가하는 것으로 보아, EFA 역전사 효소의 처리는 세포 노화 및 세포 사멸을 유도하는 것을 알 수 있었다. 이상의 결과를 바탕으로, 역전사 효소의 억제는 정상세포보다는 암세포의 성장을 더 억제하는 것을 알 수 있었지만, 항암치료 등에서 응용되기 위해서는 세포에서 역전사 효소의 기능과 역할에 대해서는 좀 더 심도있는 연구가 필요할 것으로 생각된다.

산채 및 해조분말을 첨가한 우리밀 밀가루 열수출물의 항돌연변이성 및 암세포 성장 억제효과 (Antimutagenicity and Cytotoxicity Effects of Woorimil Wheat Flour Extracts Added with Wild Herb and Seaweed Powder)

  • The antimutagenic and cancer cell growth inhibitory effects of woorimil contained herb and seaweed powders were examined. While woorimil itself showed only 40% antimutagenic effect on S. typhymurium TA98 against 4NQO(0.15 g/plate), water extracts of mountain herbs and seweeds including Comfrey, wormwood, Kale, Angelica utilis and pine leaves showed 80~90% antimutagenicity. On the other hand, these extracts along with woorimil showed 68 to 80% antimutagenic activities. Low antimutagenic activities of less than 50% were shown when these extracts were tested on TA98 against Trp P 1(0.5 g/plate), but high antimutagenic activities of 80~93.3% were shown on TA100. Water extracts of Capsella bursa pastoris and Allium grayi exhibited 60~80% of the activites in cytotoxicity tests of woorimil water extracts(0.5mg/ml) on human lung carcinoma cell. A549 showed 10% cell growth inhibitory effect. When mixed with Comfrey and Angelica utilis extracts, it showed 23~25% inhibition and other extracts showed only 12~23% inhibition. Cytotoxicity test of woorimil extracts on human liver cancer cell Hep3B revealed 20% inhibition. The additions of pine needle extracts, Angelica utilis and Comfrey showed 33%, 29% and 25% inhibition, respectively. But other extracts showed only 20% inhibition.

siRNA-mediated Inhibition of hTERC Enhances Radiosensitivity of Cervical Cancer

  • Background: To investigate the influence of telomerase activity, apoptosis, radiosensitivity of cervical cancer after siRNA-mediated knockdown of telomerase RNA and evaluate in vivo growth with gene interference. Methods: We studied siRNA-targeting-telomerase RNA transfection into the Hela cell line. Expression of hTERC mRNA was detected by RT-PCR and telomerase activity was measured by the TRAP assay. Growth inhibition was determined by MTT assay and radiosensitivity of the cervical cancer cells was examined by colony formation assay. In addtion, effects of hTERC inhibition in vivo were studied by injection of siRNA-transfected Hela cells into nude mice. Results: The hTERC siRNA effectively downregulated the expression of hTERC mRNA and also reduced the telomerase activity to 30% of the untreated control vlaue. The viability of hTERC siRNA transfected Hela cells was reduced by 44.7% after transfection. After radiation treatment, the radiosensitivity of Hela cells with hTERC knockdown was increased. In vivo, the tumors developing from the hTERC siRNA-transfected cells were of reduced size, indicating that the hTERT siRNA also depressed the tumorigenic potential of the Hela cells. Conclusions: Our results supported the concept of siRNA-mediated inhibition of telomerase mRNA which could inhibit the expression of hTERC and telomerase activity. Furthermore, radiosensitivity was upregulated after knockdown the hTERC in vivo and in vitro.

Evaluation of Cytotoxicity Effects of Chalcone Epoxide Analogues as a Selective COX-II Inhibitor in the Human Liver Carcinoma Cell Line

  • Objectives: Study of the mechanisms involved in cancer progression suggests that cyclooxygenase enzymes play an important role in the induction of inflammation, tumor formation, and metastasis of cancer cells. Thus, cyclooxygenase enzymes could be considered for cancer chemotherapy. Among these enzymes, cyclooxygenase 2 (COX-2) is associated with liver carcinogenesis. Various COX-2 inhibitors cause growth inhibition of human hepatocellular carcinoma cells, but many of them act in the COX-2 independent mechanism. Thus, the introduction of selective COX-2 inhibitors is necessary to achieve a clear result. The present study was aimed to determine the growth-inhibitory effects of new analogues of chalcone epoxide as selective COX-2 inhibitors on the human hepatocellular carcinoma (HepG2) cell line. Methods: Estimation of both cell growth and the amount of prostaglandin E2 (PGE2) production were used to study the effect of selective COX-2 inhibitors on the hepatocellular carcinoma cell. Cell growth determination has done by MTT assay in 24 h, 48 h and 72 h, and PGE2 production has estimated by using ELYSA kit in 48 h and 72 h. Results: The results showed growth inhibition of the HepG2 cell line in a concentration and time-dependent manner, as well as a reduction in the formation of PGE2 as a product of COX-2 activity. Among the compounds those analogues with methoxy and hydrogen group showed more inhibitory effect than others. Conclusion: The current in-vitro study indicates that the observed significant growth-inhibitory effect of chalcone-epoxide analogues on the HepG2 cell line may involve COX-dependent mechanisms and the PGE2 pathway parallel to the effect of celecoxib. It can be said that these analogues might be efficient compounds in chemotherapy of COX-2 dependent carcinoma specially preventing and treatment of hepatocellular carcinomas.

Overexpression of Cyclin L2 Inhibits Growth and Enhances Chemosensitivity in Human Gastric Cancer Cells

  • Cyclin L2 is a novel member of the cyclin family, recently implicated in the regulation of cell cycle progression and/or transcriptional regulation. The present study was undertaken to investigate the effects of overexpression on tumor cell growth and chemosensitivity in human gastric cells in vitro. Cyclin L2 was transfected into human gastric cancer cell line BCG823 and expressed with a mammalian expression vector pcDNA3.1. The effects and mechanisms of cyclin L2 on cell growth, cell cycling and apoptosis were studied. Compared to control vectors, overexpression of cyclin L2 inhibited the growth of BCG823 cells and enhance their chemosensitivity to fluorouracil, docetaxel and cisplatin. The anti-proliferative effects of cyclin L2 could be due to G0/G1 arrest and apoptosis. Cyclin L2 induced G0/G1 arrest and apoptosis involved upregulation of caspase-3 and down regulation Bcl-2 and survivin. The results indicated that overexpression of cyclin L2 protein may promote efficient growth inhibition and enhance chemosensitivity to chemotherapeutic agents in human gastric cancer cells by inducing G0/G1 cell cycle arrest and apoptosis.
