Effect of Seafood Amino Acid Fertilizer and Korean Effective Microorganisms on the Leaf Quality of Perilla frutescens var. japonica (해양부산물 아미노산액비 및 유용미생물(KEM) 시용이 들깻잎의 품질에 미치는 영향)

  • This study was carried out to investigate the effect of KEM and SAF appication on contents change of fatty acids and organic acid of perilla(Perilla Frutescens Britton). Contenst of squalene in perilla leaves on control and KEM/SAF treated were 3.39 mg and 4.22 mg, respectively. Therefore the squalene quantity of KEM/SAF treated leaves was 24.2% more than that of control. A total 6 fatty acids in perilla leaves were analyzed in this study. Percentage of the saturated and unsaturated fatty acid in perilla leaves were 20 and 80%, respectively. Contents of phytosterols in perilla leaf such as campesterol and sitosterol were 2.0 and 20.0 mg, respectively. Therefore sitosterol content was 10 fold more than that of campesterol. The KEM/SAF application on perilla leaf was effective on the change of squalene or phytosterol contents. However effect of that was negligible on the change of fatty acid content.

Analysis of the General and Functional Components of Various Soybeans (여러 종류 콩의 일반 및 기능성 성분 분석)

  • The objective of this study was to analyze the general and functional components of various soybeans, used for producing soy sauce, tofu, bean sprouts, and for cooking with rice. The moisture, crude protein, crude fat, and ash content of soybeans were in the ranges of 5.50~6.16%, 38.49~41.08%, 14.89~21.89%, and 4.89~5.86%, respectively. Mineral and functional components varied by sample and showed no relationship with usage type. Somyeong for bean sprouts had the highest level of Ca (406.36 mg/100 g), and Sinpaldal for soy sauce and tofu had the highest level of Mg (247.79 mg/100 g). However, Jinpum No. 2 for soy sauce and tofu had the lowest level of Ca (199.51 mg/100 g), and Seonhuk had the lowest level of Mg (90.03 mg/100 g) among the soybeans tested. The isoflavone content in soybeans was in the range of 97.54~402.00 mg/100 g. Somyeong for bean sprouts had the highest level and Seonheuk for cooking with rice had the lowest level. Total oligosaccharides were in the range of $5,838.52{\sim}9,345.90{\mu}g/100g$. Sucrose content was approximately 50% of total oligosaccharides in all samples. Raffinose content was $516.28{\sim}806.95{\mu}g/100g$, and stachyose content was $2,047.13{\sim}3,454.10{\mu}g/100g$. Phytosterols including bracassisterol, campesterol, stigmasterol, and ${\beta}$-sitosterol ranged from 19.25~35.34 mg/100 g. ${\beta}$-sitosterol represented 50% of total phytosterols in almost all samples, followed by campesterol, stigmasterol and bracassisterol. The phytic acid content in soybeans tested was around 2%, except for Sinpaldal No. 2 and Jinpum No. 2 at 0.86% and 1.65%, respectively. The dietary fiber of soybeans was in the ranges of 24.20%~29.20%.

Research on the Quality Properties of Olive Oils Available in Korea (국내 유통되는 올리브오일의 품질특성 실태조사)

  • An investigation of various olive oils available in Korea was carried out to assess their quality properties such as color, oxidative stability, fatty acid composition, tocopherol content, sterol content and benzo(a)pyrene content. In color measurement, by using a Lovibond color scale and Hunter color difference meter, both a and b values of extra virgin olive oil were higher than those of pure olive oil by Tintometer (Lovibond PFX995). However, extra virgin olive oil showed higher a value and lower L value than pure olive oil by the Hunter color difference meter. In the rancimat test, the induction period of extra virgin olive oil $(38.03{\sim}8.47hr)$ was longer than that of pure olive oil $(32.40{\sim}9.94hr)$. In fatty acid composition, C18:1 $(72.01{\sim}78.53wt%)$ was present in the greatest amount, with lesser amounts of C18:2 $(4.88{\sim}10.36wt%)$ and C18:3 $(0.56{\sim}1.09wt%)$. The tocopherol content ranged from ${\alpha}-Toc\;4.09{\sim}13.89mg/100g$, ${\beta}-Toc\;0.57{\sim}1.34mg/100g$, and ${\gamma}-Toc$ $3.41{\sim}8.03mg/100g$, and ${\alpha}-tocopherol$ was found to be the main isomer in all oil samples. Therefore, there was little difference in the fatty acid composition and tocopherol content among the different types of olive oils. In sterol content, ${\beta}-sitosterol$ $(124.52{\sim}19.33mg/100g)$ and campesterol $(1.10{\sim}0.62mg/100g)$ of extra virgin olive oil were higher than that of pure olive oil $({\beta}-sitosterol\;92.68{\sim}17.44mg/100g,\;campesterol\;0.59{\sim}0.35mg/100g)$. Benzo(a)pyrene was found in almost all samples, with $0.287{\sim}0.106{\mu}g/kg$ in extra virgin olive oil and $1.204{\sim}2.130{\mu}g/kg$ in pure olive oil.

  • Compositional and quantitative changes of 4-desmethylsterols in soybean (Glycine max. cv. Gangrim) seed and pod at six stages of development from flowering were investigated. The content of 4-desmethyl-, 4-monomethyl- and 4,4-dimethylsterols in developing soybean seed and pod was found to vary between 0.16-0.35, 0.04-0.12 and 0.04-0.06%, respectively. The predominating 4-desmethylsterols were sitosterol, campesterol and stigmasterol. The proportion of sitosterol in seed decreased to 57.0% with maturation, while in pod increased up to 88.7%. ${\triangle}^{7,22,25}$ Stigmastatrienol estimated 7.5% was detected in the fully mature seed.

  • A sterol mixture, 3-O-glucosides of these sterols, 6'-O-fattyacyl ester of these sterol glucosides, kaempferol, quercetin and isoquercitrin were isolated from the whole plants of Hypericum ascyron L. The sterols were found to be a mixture of ${\beta}-sitosterol$, campesterol and stigmasterol by GC-MS. The kinds of fatty acids linked at 6'-OH of sterol glucoside ester mixture were shown to be palmitic acid, stearic acid, oleic acid and linoleic acid by GC-MS. Three flavonoids were identified by spectroscopic methods and comparisons of mixed mp and co-TLC with authentic specimens.

  • Free and esterified sterols in neutral lipid fractionated from the free lipid of Korean white ginseng (Panax ginseng C.A. Meyer) were analyzed by TLC and GLC to investigate the contents and composition ratios. Content of each sterol was as follows: ${\beta}$-sitosterol was about 51.60 mg%, stigmasterol, 8.93mg% and campesterol, 2.17mg%. Fine root tended to have slightly higher contents of sterols than main root. Linoleic and palmitic acid as the major fatty acids in esterified sterols occupied 70% of the total composition.

  • The chemical constituents of root bark of Acanthopanax sessiliflorum Seemann(Araliaceae) which is distributed in southern region of Korea are studied. The five kinds of chemical constituents are isolated from MeOH extract of root bark by column fractionation and purified by recrystallization. Substance $I{\sim}III$ were identified as stigmasterol, ${\beta}-sitosterol$ and campesterol. Substance IV is suggested as a lignan substance based on chemical and spectral discussions. The molecular formula of substance $V\;show\;C_{20}H_{18}O_6$ and its melting point is $120.5{\sim}121.5^{\circ}$. Due to the spectral data, it is confirmed that substance V is a (+) sesamin of lignan.

  • One new sterol glucoside, $gomphsterol\;{\beta}-D-glucoside$ 1 along with known compounds, ${\beta}-sitosterol$, stigmasterol, campesterol, $stigmasterol-{\beta}-D-glucoside$, friedelin, 3-epi-friedelinol, allantoin, and $chrysoeriol-7-O-{\beta}-D-glucoside$ have been isolated from the aerial parts of Gomphrena globosa (Amaranthaceae). On the basis of spectroscopic (including 2D NMR) and chemical studies, the structure of 1 was elucidated as $(22E,24S)-24-ethylcholesta-7,9(11),22-trien-3{\beta}-ol-3-O-{\beta}-D-glucopyranoside$. Known compounds are reported for the first time from this plant species.

  • "Ha-Neul-Ta-Ri" (Trichosanthes kirilowii Max., Cucurbitaceae) cultivated in Korea and used in popular medicine, was investigated for sterol composition of the seed oil. This seed oil contains unusual sterols having ${\delta}^{7, 22}_-$, ${\delta}^{7, 24}_-$ and ${\delta}^{7, 22, 25}_-$ di and trienols. ${\delta}^{7, 22}_-$ Stigmastadienol and ${\delta}^{7, 22, 25}_-$ Stigmastatrienol were identified as the major sterols in the seed oil of Trichosanthes kirilowii by gas loiquid chromatographic analysis. The results showed that contents of the seed sterols were campesterol 1.0-1.2%, stigmasterol 1.5-1.7%, ${\delta}^{7}_-$ campstenol 0.6-0.8%, sitosterol 10.6-11.4%, ${\delta}^{7, 22}_-$ stigmastadienol 39.1-41.8%, ${\delta}^{7, 22, 25}_-$ stigmastatrienol 28.9-29.3%, ${\delta}^{7, 25}_-$ stigmastadienol 14.9-15.5% and ${\delta}^{7, 24}_-$ stigmastadienol 0.7-1.0%.

  • A experiment was performed to investigate the stigmasterol and campesterol in the Panax ginseng. Crude sterol fraction obtained from the ginseng was analysed by TLC and quantitative determination of ${\beta}$-sitosterol was performed with colorimetric method. The results were summarized as follwer, 1. It was observed with ethyl ether-petroleum ether extract of the ginseng that there was no evidence of showing the presence of both free capesterol and stigmasterol but that only ${\beta}$-sitosterol was presence in an significant amount. 2. Average amount of the ${\beta}$-sitosterol was 0.20-0.35mg per gram of the sample.
