Radiolabeling and Immunological Characteristics of In-house Anti-Leukemic Monoclonal Antibodies(Anti-CALLA, Anti-JL-1 Antibodies) (국산 항 백혈병 항체(항 CALLA, 항 JL-1)의 동위원소 표지 및 면역학적 특성에 관한 연구)

  • Recently murine monoclonal antibodies have been studied actively for radioimmuno-scintigraphy and radioimmunotherapy, especially on patients with leukemia and lymphoma. In this research, we studied radiolabeling and immunologic characteristics of two in-house anti-leukemic monoclonal antibodies(anti-CALLA & anti-JL-1 antibodies) to make the basis for their clinical application. Each antibody was radiolabeled successfully with $^{99m}Tc$ by pretargeting transchelation method and with $^{125}I$ by lodogen method. We also studied cell binding assay, Scatchard analysis and modulation phenomenon. $^{125}I$ showed 90% labeling efficiency for each anti-body which was satisfactory, but $^{99m}Tc$ showed labeling efficiency below 70%, for which we need better labeling method. In cell binding assay, the immunoreactivity(IR) was low for $^{99m}Tc$-labeled antibodies. Scatchard analysis showed satisfactory data for both binding affinity. The affinity constant and antibody binding sites per cell are around $10^9M^{-1}$ and $10^4$, respectively. There was no modulation phenomenon in cases of $^{125}I$ or $^{99m}Tc$ labeled antibodies. We expect that two anti-leukemic monoclonal antibodies may be useful in diagnosis and therapy for leukemia and lymphoma patients.

Growth and Flowering of Calla (Zantedeschia elliottiana cv. 'Black Magic') with various mulching materials and GA3 treatments (멀칭재료와 GA3처리에 따른 유색칼라(Zantedeschia elliottiana cv. 'Black Magic')의 생육 및 개화반응)

  • This experiment had been conducted to determine optimal culture conditions for the growth and cut flowers of Calla (Zantedeschia 'Black Magic' 'Golden Affair') in highlands. Tubers of calla 'Black Magic' was treated with 0, 100, and $200mg{\cdot}L^{-1}$ concentration of $GA_3$. ABA content in tubers and roots were twice higher ($20-25pmol{\cdot}mL^{-1}$) in control than in $GA_3$ treated tubers and roots. ABA contents in tubers was not consistent but ABA contents in roots increased with increasing $GA_3$ concentration. However ABA is in inverse proportion to GA. It was mean that GA of high contents was ABA of low contents. Calla 'Golden Affair' was treated with different mulching materials (Control, black PE, Reflective film, Rice hull) and $GA_3$ concentrations (0, 50, 100 and $200mg{\cdot}L^{-1}$). It was largest in the length of flower at mulching of reflective film, Rice hull and it was greatest in the number of cut flowers at the treated with $200mg{\cdot}L^{-1}$ $GA_3$, Therefore the proper condition was at $200mg{\cdot}L^{-1}$ $GA_3$ with reflective film mulching.

  • We investigated the development of a micropropagation protocol for multiplication of calla 'Gagsi' by using shoots as explant. The callus was induced on Murashige and Skoog (MS) basal medium containing cefotaxime antibiotics (25, 50, 100 mg/L). Also, MS basal medium with NAA 0.5 mg/L and BA 1.0 mg/L was used. The callus induction and browning rates were compared by treatment supplemented cefotaxime 25, 50 and 100 mg/L in basal MS medium. The callus induction rate was 10.5 % and browning rate was also, 10.5 % on the MS containing 25 mg/L. In the MNB containing cefotaxime, the callus induction rate was 34.5 % and browning rate was 27.0 %. The cefotaxime experiment has been widely used in previous studies. It is thought that it will help establish the mass multiplication system by positively affecting the growth and browning reduction of calla plants.

Growth response of Calla (Zantedeschia) to root zone environmental conditions in Highland (고랭지에서의 근권환경에 따른 유색칼라(Zantedeschia)의 생육반응)

  • This experiment has been carried out to determine optimal culture conditions for the production of cut flowers and tubers of Calla (Zantedeschia 'Golden Affair' and 'Black Magic') in highlands. Treatments consisted of various levels of root zone environments, Results are as follows: Calla 'Golden Affair' were grown with different mulching materials such as Non-mulching, Black film, reflective film, Rice hull. Mulching materials resulted in no difference in the number of cut flowers but flower length was highest in reflective film. Calla 'Black Magic' were treated with various soil water content, soil type and watering time. Number of cut flowers and flower quality were greatest when the plants were watered at -80 kPa soil water content. No symptoms of soft rot (Erwinia carotovora) was observed at this soil water content. The occurrence of soft rot was observed with similar percentage according to soil type and the soil water content. When Calla 'Black Magic' were watered at the time of soil surface drying, growth was greater compared to others. Air temperature and PPF affected plant growth and photosynthesis. Photosynthetic rate was greatest at $25^{\circ}C$ and PPF $200{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$, while lowest at $28^{\circ}C$ and PPF $800{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$. Soft rot did not occur without regard to dipping treatment (0, 5, 10, 24, 48 hours) when the day and night temperature were maintained at $25^{\circ}C$ and $20^{\circ}C$, respectively.

Growth and flowering as affected by tuber hardness, GA3 concentrations and treatment duration in Calla (Zantedeschia)

  • This experiment was carried out to determine optimal culture conditions for the production of cut flowers and tubers of Calla (Zantedeschia 'Black Magic') in highlands. To achieve the research purpose, growth of 'Black Magic' Calla influenced by tuber hardness (3.3 and $6.0kg/cm^2$), $GA_3$ concentration (0, 100 and $200mgL^{-1}$), duration of $GA_3$ treatment (24 hr, 12 hr and 30min before planting) were investigated. When tubers have high hardness, those were not severely injured by soft rot disease regardless of $GA_3$ concentrations and treatment durations. Tubers with low tuber hardness showed more than 90% of soft rot occurrence when treated with $200mgL^{-1}$ $GA_3$ for 24 hrs before planting. However, soft rot did not occur when tubers were treated with $200mgL^{-1}$ $GA_3$ for 12 hrs before planting. In conclusion, $GA_3$ treatment results showed soft rot occurrence statistically significant degree in accordance with the bulbs hardness. In addition, the yield of the cut flowers is the result received the greatest effect in accordance with the bulbs in size and appeared to not be determined in accordance with the $GA_3$ treatment concentration and hardness bulbs. To obtain flowers without soft rot symptom, tubers (as $6.0kg/cm^2$) should be completely dried after $GA_3$ treatment.

Effect of harvesting time and night temperature on tuber production of calla (Zantedeschia) (수확시기와 야간온도가 유색칼라(Zantedeschia)의 구근생산에 미치는 영향)

  • This experiment was carried out to determine optimal culture conditions for the production of tubers of Calla (Zantedeschia elliottiana 'Golden Affair' and 'Black Magic') in Korea highland. In vitro produced plantlets and tuberlets of Calla 'Golden Affair' and 'Black Magic' were planted plastic film greenhouse and grown for 100, 120, 140 days, with different night temperature treatments ($0{\sim}10^{\circ}C$ : no heating, 10, $15^{\circ}C$). In both cultivars, tuber size(tuber diameter, tuber height) and tuber weight increased with increasing cultivation period when the night temperature was maintained at $10^{\circ}C$. The largest tuber diameter in vitro produced plantlets was 5.8cm in 'Black Magic' and 3.2cm in 'Golden Affair', and daily tuber growth rate was 1.110g in 'Black Magic' and 0.092g in 'Golden Affair' under the culture conditions. Consequently we think that tuber harvest date was Oct. 30 and night temperature was $10^{\circ}C$ and no heating that was proper method of tuber production. However we had selection of $10^{\circ}C$ treatment for tuber production because it appeared freezing damage occasionally in highland late in October.

A New Screening Method for the Selection of Calla Lily Zantedeschia aethiopica Cultivars Resistant to Calla Lily Soft Rot (칼라의 무름병 저항성 품종 선발을 위한 검정 방법 개발)

  • This experiment was carried out to establish the screening methods for the selection of Zantedeschia aethiopica cultivars resistant to calla lily soft rot caused by Pectobacterium carotovorum subsp. carotovorum. A center of leaf and petiole tissues of four Z. aethiopica cultivars were wounded and inoculated with three different inoculum concentrations, $1{\times}10^7cfu/mL$, $1{\times}10^8cfu/mL$, and $1{\times}10^9cfu/mL$, of EccNHRI-21 isolate. And they were evaluated at 4, 18, and 26 hours after inoculation. The lesion of maceration was developed well in the leaf tissues inoculated with $1{\times}10^9cfu/mL$ concentration. And evaluation of resistance in 18 hours after inoculation was correlated with field resistance positively. Using this method, ten Z. aethiopica commercial cultivars and four wild types were screened. 'Crowbrough' and 'White Cutie' were selected as highly resistant genotypes and 'Mont Blanc' and 'Silky White' as resistant genotypes. 'Wedding March' and 'Kiwi Blush' were evaluated as moderate resistant and the others including 'Childsiana' were susceptible. And all of four wild types were evaluated as more than moderate resistant. In spite of control through cultural, biological and chemical methods, the use of resistant cultivars is most efficient to overcome calla lily soft rot. This newly developed screening method was helpful for breeding new varieties resistant to calla lily soft rot.

Growth and Tkber Development of 'Black Magic' Calla Lily as Affected by the Rain Shelter and Tuber Size (유색칼라 생육 및 구근 비대에 미치는 비가림 재배와 구근 크기의 효과)

  • Growth and tuber development of ‘Black Magic’calla lily as affected by the rain shelter with 50% shading and the tuber size were investigated. Tubers of five grades in size were cultivated in the rain shelter or in the open field (control). Days to emergence of shoots in the rain shelter was accelerated by 4.2 days as compared to the open field. Emergence ratio was higher as the tuber was larger, tubers grown in the rain shelter showed the significance in the growth characteristics as compared to the control. There were no significant differences in the number of flowers per tuber between the rain shelter and the open field. Flower quality was 12.2 cm longer than that in the open field. flowering characteristics was improved with the increasing tuber size. Low infection of soft rot disease of 3∼22% was found in the rain shelter as compared to the that of 19∼83% in the open field. Thus tubers produced under the rain shelter showed improved quality, Even if the small tubers with diameters of 0.5∼1.0 cm were cultivated in the rain shelter, the weight and diameters of tuber harvested after approximately 7 months were 50.2 g and 5.7 cm, respectively.

  • 칼라(Zantedeschia spp.)는 남아프리카가 원산인 구근식물로 천남성과에 속하며 7개의 원종과 2개의 아종이 있다. 원예학적으로는 두 그룹으로 분류되며 Z. aethiopica 종이 속한 그룹은 습지생육형으로서 보통 백색칼라로 불리고, Z. albomaculata, Z. rehmannii, Z. jucunda, Z. ellittiana 및 Z. pentlandii 5종이 포함된 건지생육형 그룹은 유색칼라로 불린다. 그 외에 두 그룹의 중간 형태인 Z. ordorata 종이 있다(Funnell, 1993).
