Studies on Tip-burn of Chinese Cabbage by Ammonium Toxicity (암모니움 Toxicity에 의(依)한 배추의 Tip-burn에 관(關)한 연구(硏究))

  • The effect of N forms ($NO_3{^-}-N$, $NH_4{^+}-N$) and concentrations (4, 8, $16meq/{\ell}$) which were changed at head formation stage on the tip-burn incidence of chinese cabbage was studied under the three levels (0, 8, 16 meq/l) of Ca. All of the plants grown on $NH_4{^+}-N$ showed the symptoms of tip-burn and low yields regardless of Ca levels. Roots of plants grown on $NH_4{^+}-N$ were severely damaged. The pH of the leachate from $NH_4{^+}-N$ pot was decreased to below pH 5. Plants which had been grown on $NH_4{^+}-N$ before the head formation stage, but changed to $NO_3{^-}-N$ were recovered from abnormal growth. But, the reverse showed the tip-burn symptoms. $NH_4{^+}-N$ treatments increased the T-N contents, but lowered K and Ca contents of inner leafblades. Icreased applications of Ca did not affect the T-N and K contents of the inner leafblades. $NH_4{^+}-N$ suppressed Ca translocation into the inner leaves from outer leaves after the head formation stage, but $NO_3{^-}-N$ accelarated it. Ca contents were much lower in leaves showing tip-burn symptoms than in healthy leaves. Internal rot which is a tip-burn symptom occuring after head formation were noted in plants applied with high concentration of $NO_3{^-}-N$ both before and after the head formation stage. Ca contents correlated well with the rates of Ca application, but there was no correlation between ca conents and internal rot incidence. Chinese cabbage tip-burn is not caused by Ca deficiency, as is commonly believed, but rather by the water stress (95% water contents in $NO_3{^-}$-fed plants, 91% in $NH_4{^+}$-fed plants) resulting from root damage caused by ammonium toxicity. Internal rot is also caused by excess applications of nitrogen, and is unaffected by calcium levels.

Chemical Composition and Nutritional Value of Algae Meal Produced from Dairy Cow Wastes as a Feedstuff (젖소폐기물에서 생산된 Algae Meal의 화학적조성 및 사료적가치)

  • Algal meal (cell) was produced from the solution of dairy cow wastes by fermentation of ulothrix. sp. and chlorella sp. Raw wastes mainly feces were diluted with ground water to give dry matter concentration of 0.5 w/v of wastes in 20 l amounts of ten plastic containers. Each containers were covered with plastic nets and vinyl films to protect from the insects and rain. Algea cells were harvested every 3 to 5 days and dried by sunlight and artifitial heat. Dried cells were ground by a feed meal, and analyzed and tested for the chemical composition of dry cell, in vitro DM and protein digestibility and the safty of algae. Protein contents in algae meals, ulothrix (29.37%) and chlorella (29.24%) were similar. However, chlorella contained lower Neutral detergent fiber (5.92%) than ulothrix(20,76%), and higher ash (32.86%) and calcium (12.62%) than ulothrix (28.66% and 6.09%) (P<.01). Ulothrix protein had higher for essential amino acids; valine, isoleucine and phenylalanine, than chlorella (P<.05). Algal fats contained high saturated fatty acids, C16:0 and C18:0, for ulothrix and high unsaturated fatty acids, C18:1 and C18:2, for chlorella (P<.01). In vitro digestibility of. ulothrix tended to be higher for DM, but lower for protein than chlorella. The weight gain and survival percentage were higher for pond fishes (loaches, Misgurnus sp. ) fed diet added chlorella meal than diets added ulothrix meal and control diet (P<.05).

Effects of $\textrm{NO}_3$-N:$\textrm{NH}_4$-N Ratio and Elevated $\textrm{CO}_2$ on Growth and Quality of Lactuca sativa L. in Nutrient Film Technique (NFT재배에서 $\textrm{CO}_2$ 시용과 배양액의 $\textrm{NO}_3$:$\textrm{NH}_4$비율이 결구상추의 생육 및 품질에 미치는 영향)

  • Crisphead lettuce(Lactuca sativa L.) was grown in NFT to investigate the effects of NO$_3$-N and NH$_4$-N ratio in nutrient solution and elevated $CO_2$ treatment in the crisphead lettuce growth. This experiment has been conducted under three different ratios of NO$_3$-N:NH$_4$-N(100:0, 75:25, 50:50) with two $CO_2$ concentration (control, 1500ppm ). The results are as follows; 1. In the case of not controlling pH and EC in nutrient solution, pH was gradually increased in NO$_3$-N:NH$_4$-N=100:0 treatment but rapidly decreased in the nutrient solution 2. Daily changes of NO$_3$-N and NH$_4$-N were observed without controlling the nutrient solution. In the treatments of NO$_3$-N:NH$_4$-N ratios were 75:25 and 50:50, NO$_3$-N absorption rates were 27.7% and 26.1%, while NH$_4$-N absorption rates were 87.9% and 71.2%, respectively. 3. There was little differences in total nitrogen of leaves. However phosphorus, potassium, calcium and magnesium contents were highly shown in the treatment of $CO_2$ 1500ppm and 100:0 ratio of NO$_3$-N:NH$_4$-N. 4. Higher $CO_2$ assimilation rate was shown in plants grown under $CO_2$ 1500ppm and 100:0 ratio of NO$_3$-N:NH$_4$-N. It dropped significantly with the increase of NH$_4$- N rates in nutrient solution. 5. Fresh weight, leaf number, root length and root weight of crisphead lettuce were far better in the treatment of $CO_2$ 1500ppm and 100:0 ratio of NO$_3$-N:NH$_4$-N. Growth differences by $CO_2$ elevation were not shown in other NO$_3$-N:NH$_4$-N treatments. 6. The highest nitrate contents of leaves were shown in NO$_3$-N single treatment but shown the lowest vitamin C contents. Nitrate contents of leaves were decreased by $CO_2$ but the effect was slight treatment.

  • Soil chemical properties of agricultural soils in Korea were investigated at four-years interval in order of paddy, plastic film house, upland, and orchard soils since 1999. Paddy soil samples were taken from the surface 15 cm at 4,047, 2,010, 2,110 and 2,110 sites in all provinces of South Korea in 1999, 2003, 2007 and 2010, respectively. Soil chemical properties in Korea except Jeju province were measured. Soil pH and exchangeable calcium and available silicate contents increased with increasing the application rate of silicate fertilizer and with decreasing its application interval. Soil organic matter content also increased from $22.0g\;kg^{-1}$ in 1999 to $26.0g\;kg^{-1}$ in 2011. Average concentration of available phosphate in 2011 was higher than the upper limit of its optimal range for rice cultivation. However, exchangeable magnesium and available silicate contents were below the lower limit of their optimal ranges, which were 80% and 92% of them, respectively.

Chemical characteristics of wet precipitation in urban and mountainous sites of Jeju Island

  • Wet precipitation samples were collected in Jeju City and Mt. Halla-1100 site (a site at an altitude of 1100 m on Mt. Halla) during 2011-2013, and their major ionic species were analyzed to examine the chemical composition and characteristics. A comparison of ion balance, electric conductivity, and acid fraction of precipitation revealed correlation coefficients in the range of r = 0.950~0.991, thereby implying the high quality of analytical data. Volume-weighted mean pH and electric conductivity corresponded to 4.86 and 25.5 µS/cm for Jeju City, and 4.98 and 15.1 µS/cm for Mt. Halla-1100 site, respectively. Ionic strengths of the wet precipitation in Jeju City and Mt. Halla-1100 site corresponded 0.3 ± 0.5 and 0.2 ± 0.2 mM, respectively, thereby indicating that more than 30 % of total precipitation was within a pure precipitation criteria. The precipitation with a pH range of 4.5 - 5.0 corresponded to 40.8 % in Jeju City, while the precipitation with a pH range of 5.0 - 5.5 corresponded to 56.9 % in Mt. Halla-1100 site, thereby indicating slightly more weak acidity than that in Jeju city. The volume-weighted mean concentration (µeq/L) of ionic species was in the order of Na+ > Cl- > nss-SO42- > NO3- > Mg2+ > NH4+ > H+ > nss-Ca2+ > PO43- > K+ > CH3COO- > HCOO- > NO2- > F- > HCO3- > CH3SO3- at Jeju City area, while it corresponded to Na+ > Cl- > nss-SO42- > NO3- > NH4+ > H+ > Mg2+ > nss-Ca2+ > PO43- > CH3COO- > K+ > HCOO- > NO2- > F- > HCO3- > CH3SO3- at Mt. Halla-1100 site. The compositions of sea salts (Na+, Cl-, Mg2+) and secondary pollutants (NH4+, nss-SO42-, NO3-) corresponded to 66.1 % and 21.8 %, respectively, in Jeju City and, 49.9 % and 31.5 %, respectively, in Mt. Halla-1100 site. The acidity contributions in Jeju City and Mt. Halla-1100 site by inorganic acids, i.e., sulfuric acid and nitric acid, corresponded to 93.9 % and 91.4 %, respectively, and the acidity contributions by organic acids corresponded to 6.1 % and 8.6 %, respectively. The neutralization factors in Jeju City and Mt. Halla1100 site by ammonia corresponded to 29.8 % and 30.1 %, respectively, whereas the neutralization factors by calcium carbonate corresponded to 20.5 % and 25.2 %, respectively. From the clustered back trajectory analysis, the concentrations of most ionic components were higher when the airflow pathways were moved from the continent to Jeju area.

The Study on the Amount and Major Compositions of Excreta from Swine (돼지의 분뇨 배설량 및 분뇨 성분 조사)

  • This work was carried out to investigate the quantity of excreta and its composition in crossbred pigs (Yorkshire ${\times}$ Landrace ${\times}$ Duroc) at different stages of growth. Twelve young piglets (average BW weight of $19.0{\pm}0.33kg$) were used in this study. Pigs were divided into four phases during growing time and two phases during finishing time. The average excreta production for growing pig was 3.46 kg/head/day (feces: 1.07 kg, urine: 2.39 kg). The average moisture contents of feces and urine were 70.54% and 97.39%, respectively. Contents of Calcium, Magnesium, Copper, Plumbum, and Arsenic were 1.00%, 0.26%, 10.47 mg/kg, 2.43 mg/kg, and 1.02 mg/kg, respectively. The concentration of the water pollutants like Biochemical Oxygen Demand ($BOD_5$), Chemical Oxygen Demand (COD), Suspended Solid (SS), Total Nitrogen (TN) and Total Phosphorus (TP), excreted from pig were 96335, 61073, 207466, 8104 and 4209 mg/L in feces and 7364, 7149, 2715, 10110 and 613 mg/L in urine at the end of test, respectively. The daily loading amount of water pollutants ($BOD_5$, COD, SS, TN, and TP, respectively) in pig excreta were 102.1, 61.8, 221.6, 8.7, and 3.9 g/head/day in feces, and 19.3, 16.7, 8.0, 22.2, and 1.3 g/head/day in urine, respectively. The Nitrogen, $P_2O_5$, and $K_2O$ contents in the excreta of pigs were 0.96, 0.83 and 0.42% in feces, and 0.80, 0.09 and 0.53% in urine, respectively. Finally, this work was suggested to give basic information to swine farms.

Effect of Fermented Brown Seaweed Waste (FBSW) on Milk Production, Composition and Physiological Responses in Holstein Dairy Cows (발효미역부산물의 첨가가 Holstein 비유우의 비유성적 및 생리반응에 미치는 영향)

  • This study was conducted to determine effects of fermented brown seaweed waste (FBSW) on milk production, composition and physiological responses as functional feed for 60 days in Holstein dairy cows. A total of 24 Holstein dairy cows (average age 49.33 months, average lactation days 175, Reproduction 2.0) were randomly allocated into control(basal diet), 1% FBSW (180g in basal diet) and 2% FBSW (360g in basal diet) groups with 8 replications for 60 days. Daily milk yield and composition (fat, protein, SNF, MUN) were not affected by FBSW supplementation, but Ca level in milk was significantly increased 4.29 mg/dl and 2.91 mg/dl in 1% and 2% groups compared to control group (p<0.05) at the end of the experiment, respectively. The somatic cell count (SCC) in milk was not significant. The plasma $T_4$ level (concentration) were increased in 1% and 2% FBSW compared to control group at the end of the experiment (p<0.05), but between triiodothyronine ($T_3$) and thyroxin ($T_4$) levels were not significant. Concentrations of plasma glucose in control, 1% FBSW and 2% FBSW groups were 64. 37mg/dl, 66.15mg/dl and 73.02 mg/dl and plasma NEFA level was 0.30~0.32 mEq/dl. Concentrations of BUN tended to be higher for FBSW group than control group. Although WBC, RBC, Hb, Hct, T-B, ALP, and GPT levels were not affected by FBSW supplementation, GOT level was significantly decreased in cows fed 1% FBSE diet compared to control group (P<0.05). Therefore we strongly suggest that the 1% FBSW supplementation in basal diet increases the milk yield and Ca level in Holstein dairy cows.

Novel Gap Junction Molecules, Connexin 37, Enhances the Bystander Effect in HSVtk/GCV Gene Therapy (Herpes Simplex Virus thymidine Kinase/Ganciclovir 유전자 치료에서 새로운 간격결합분자 Connexin 37에 의한 방관자 효과의 증가)

  • Purpose : Gap junction intercellular communication(GJIC) is an important mechanism of the bystander effect in herpes simplex thymidine kinase/ganciclovir(HSVtk/GCV) gene therapy Therefore, we attempted to enhance the bystander effect in vitro by exogenous overexpressing connexin 37(Cx37) in cells to increase GJIC. Methods : NIH3T3 cells were transfected with the Cx37 and HSVtk gene or the HSVtk gene alone by the calcium phosphate method, and we detected their expression from these cells by RT-PCR. GCV-mediated cytotoxicity and the bystander effect of each transfectant was then assessed and compared. Results : Cells transfected with HSVtk became sensitive to low concentration of GCV. We found significantly increased cytotoxicity in HSVtk/GCV gene therapy after introduction of the HSVtk and Cx37 genes together compared with the cytotoxicity seen after introduction of the HSVtk gene in vitro. Co-expression of the HSVtk and Cx37 genes potentiates HSVtk/GCV gene therapy through the bystander effect. Conclusion : These results indicated that the increase of GJIC using Cx37 have potentiated the bystander effect of HSVtk/GCV therapy, and may be a new approach to improve response in suicidal cancer gene therapy.

A Study on The Relation between Psychological Stress and Stress Hormone, Nutritional Status of Patients with Non-Insulim Dependent Diabetes Mellitus (인슐린 비의존형 당뇨병 환자의 심리적 스트레스와 스트레스호르몬 및 영양상태와의 상관관계에 관한 연구)

  • This study estimated the re!ation between psychological stress and stress hormones, nu­t tritional status of patients with non-insulin dependent diabetes mellitus(NIDDM). Psycho­l logical stress such as depr'ession and anxiety in 34 diabetics was analyzedin relation to nutrient intake, blood components such as fasting blood sugar(FBS), hemoglobin AIC, stress hormones a and amino acids. The IeveIs of depression and anxiety were measured by The center for ep­i idemiological studies-depc$pm$111.49pg/ml for total catecholamine ( (norepmephrine and epinephrine) and 233.95 $pm$73.99pg/ml for norepinephrine, 94.03$pm$75. 9 97pg/ml for epinephrine, 13.lS$pm$5.55pl/dl for cortisol and 171.50$pm$62.50pg/ml for gul c cagon respectlveIy. The leveIs of stress hormones in diabetics such as total catecholamine, norep­i mephrine cortisol and glucagon were significantly higher than those in normal control. The lev­el of epmephrine was higher in diabetics but the diffierenee was not significant. The calorie m t take in diabetics was 1762$pm$292keal which is S1.4% lower than Korean recommended dietary a allowances(RDA). Calcium intake was slightly low but other nutrients intakeswere higher than R RDA. The value of fasting blood sugar(FBS), usual fasting blood sugar(usual FBS) which refteet a average FBS during 3 months and hemoglobin Al C in diabetics was 1S4.1S$pm$74.22mg/dl, 177.76$pm$42.77mg/dl and S.S4$pm$2.S2% respec디VeIy. The distribution of plasma amino acids in d diabetics was generally in the normal range. The leveI of anxiety in diabetics was positively cor­related with norepinephrine, concentration and usual FBS. The levels of glucagon, usual FBS a and hemoglobin Aj C were pOSI디veIy correlated with the branched chaimamino acid(BCM : leucme, isoleucme and valine)

Physicochemical Changes of Food Waste Slurry Co-fermented with Pig Manure Slurry (음식물쓰레기와 돈분 액상물의 혼합부숙시 이화학적 특성 변화)

  • To find a feasibility of utilization of food waste slurry (FWS) generated during composting, FWS was combined with pig manure slurry (PMS) in various ratios and the change of nutrient contents and offensive odor of the combined slurries before and after fermentation were studied. The initial pH was 7.67 for PMS and 8.45 for FWS. However, during the fermentation, pH increased in the combined slurries with the higher FWS rate among the treatments while decreased in thosewith higher PMS rate. EC of each slurry sample showed that the difference among combined slurry samples has been reduced during fermentation and became stabilized in $21{\sim}23dS\;m^{-1}$ after 180 days. After 180 days fermentation, total nitrogen (T-N) decreased. T-N of mixture with a half and more FWS decreased up to 0.1%, less than the critical level (0.3%). The contents of O.M., T-N, phosphorus, calcium and magnesium decreased with fermentation while those of potash and salinity increased. From initial fermentation until 30 days, a lot of $NH_3$, as an offensive odor, was produced. However, it decreased steadily, except in higher PMS rate. In terms of producing $50{\mu}g\;ml^{-1}$ of $NH_3$, the top layer took 30 days after fertilization with FWS only, 45 days for utilized treatment with F75 (25 % of PMS), 75 days for utilized with F50 (50%) and F25 (75%) and 90 days for PMS only, respectively. $RNH_2$ also had similar trend with $NH_3$ but it was produced continuously as long fermentation proceeded. In terms of $RNH_2$, the decrease in concentration up to $50{\mu}g\;ml^{-1}$ were; 45 days for FWS only(F100), 105 days for F75 utilization, 120 daysfor F50, 165 days for F25, respectively. ethyl mercaptan was produced in PMS until 180 days after fertilization but it was not produced in FWS. Sensory tests as an integrated test of offensive odor were also done. FWS showed lower than 1 after 30 days from initial fermentation, while PMS had still offensive odor even up to 180 days from initial fermentation. It is probably affected by the continuous production of ethyl mercaptan and amines. However, considering in decrease T-N content caused by volatilization while offensive odor intensity according to official standard of fertilizer is lower than 2. Further study on controlling offensive odor needs to be done.
