This study were carried out to find bolting response of cultivation in different regions and to isolate FLC (FLOWERING LOCUS C) homologs in Angelica gigas Nakai. The mean temperature of different regions, ordering in altitude, were as follows: 100 m > 350 m > 530 m > 700 m. The largest amount of rainfall was occurred in the region of 350 m while the longest time of sunshine was occurred in the region of 100 m. The content of soil chemical properties in regions showed pH 6.2 ~ 7.4, T-N 0.17 ~ 26, organic mater $1{\sim}32gkg^{-1}$, $P_2O_5$${151{\sim}664_{mgkg}}^{-1}$, exchangeable potassium and calcium and magnesium were 0.78 ~ 1.15, 3.9 ~ 10.0, ${0.7{\sim}3.2_{cmol}}^{+kg-1}$. L5 line of A. gigas was occurred in bolting at all regions, but the bolting ratio was 60.0% in 700 m region with non-mulching treatment. Manchu of A. gigas was not occurred in bolting at all regions. The accumulation bolting ratio of L5 line by non-mulching was higher than that of mulching as 90.4% and 72.8% in 100 m region. The MADS-box transcription factor FLC is one of the well-known examples as a strong floral repressor. We decided to isolate FLC homologs from A. gigas as a starting point of flowering mechanism research of this plant. We have isolated two RT-PCR products which showed very high amino acid sequence homology to Arabidopsis FLC.

Purpose: Radiation damage is produced and viable cell number is reduced. We need to know the type of cell death by the ionizing radiation and the amount and duration of cell cycle arrest. In this study, we want to identified the main cause of the cellular damage in the oral cancer cells and normal keratinocytes with clinically useful radiation dosage. Materials and Methods: Human gingival tissue specimens obtained from healthy volunteers were used for primary culture of the normal human oral keratinocytes (NHOK). Primary NHOK were prepared from separated epithelial tissue and maintained in keratinocyte growth medium containing 0.15 mM calcium and a supplementary growth factor bullet kit. Fadu and Hep-2 cell lines were obtained from KCLB. Cells were irradiated in a /sup 137/Cs γ-irradiator at the dose of 10 Gy. The dose rate was 5.38 Gy/min. The necrotic cell death was examined with Lactate Dehydrogenase (LDH) activity in the culture medium. Every 4 day after irradiation, LDH activities were read and compared control group. Cell cycle phase distribution and preG1-incidence after radiation were analyzed by flow cytometry using Propidium Iodine staining. Cell cycle analysis were carried out with a FAC Star plus flowcytometry (FACS, Becton Dickinson, USA) and DNA histograms were processed with CELLFIT software (Becton Dickinson, USA). Results: LDH activity increased in all of the experimental cells by the times. This pattern could be seen in the non-irradiated cells, and there was no difference between the non-irradiated cells and irradiated cells. We detected an induction of apoptosis after irradiation with a single dose of 10 Gy. The maximal rate of apoptosis ranged from 4.0% to 8.0% 4 days after irradiation. In all experimental cells, we detected G2/M arrest after irradiation with a single dose of 10 Gy. Yet there were differences in the number of G2/M arrested cells. The maximal rate of the G2/M ranges from 60.0% to 80.0% 24h after irradiation. There is no significant changes on the rate of the G0/G1 phase. Conclusion: Radiation sensitivity was not related with necrosis but cell cycle arrest and apoptosis. These data suggested that more arrested cell is correlated with more apoptosis.
Compressive and tensile strengths of commonly being used mortar and mortar specially hardened by adding 1%, 2% and 3% of $CaCl_2$ were compared under the different mixing ratio of mortar and ages to investigate the effect of $CaCl_2$ which is being used as a promotor for coagulation and hardness, on the strength of mortar. The results obtained were as follows: 1. The compressive strength of mortar hardened by addition of $CaCl_2$ was higher than that of commonly being used mortar. The highest compressive strength of mortar was obtained when 2% of $CaCl_2$ were added. 2. The tensile strength of mortar, which was made by adding $CaCl_2$ and aged for seven days, was higher than that of commonly being used mortar. The highest tensile strength of mortar was obtained when 1% of $CaCl_2$ was added and aged for 28 days. And the tensile strength of mortar with 2% of $CaCl_2$ was lower than that of commonly being used mortar. 3. When the amount of $CaCl_2$ added was higher than 3%, the mortar was abruptly hardened and thereby occurred crack was considered lowering strength of mortar. 4. The rich mix was effective for the increasing the compressive and tensile strength before seven days of age and less effective after seven days of age. Therefore, the addition of one to two per cent of $CaCl_2$ would be effective for promoting initial strength of mortar during winter season.
In this study sodium dodecyl sulfate (SDS), which was anionic surfactant, was added for forming micelles to remove iron ion that could be contained with small amount in industrial water. Then aggregates binding between iron ions and micelles were rejected by a ceramic microfiltration membrane. As result of SDS concentration effect on removal rates of iron and SDS in modified iron solution, the removal rate of iron was the highest value of 92.26% and the removal rate of SDS was 61.10% a little higher than the result of calcium ion at 8 mM which was CMC (Critical micelle concentration) of SDS. As final resistance of membrane fouling $R_f$ increased the more at the higher SDS concentration, it showed the highest value at 4 mM and the lowest at 10 mM of SDS. The final permeate flux $J_{180}$ had the highest value and the largest total permeate volume could be finally acquired at SDS 10 mM. In case of CMC 8 mM, low $R_f$ was shown as same as that of 10 mM until 80 minutes of operation, and tended to increase dramatically to 120 minutes and increase slowly again until 180 minutes.

This study was conducted to investigate the physicochemical properties of raw and blanched $Haetsun$ vegetables. The proximate composition, reducing sugar, free amino acid, organic acid, vitamin C and mineral composition were compared between raw and blanched $Haetsun$ vegetables. After blanching treatment, the moisture content of $Haetsun$ vegetables was increased, but the crude ash, crude fat, crude fiber, and crude protein content decreased in all samples. $C.$$sinensis$ contained the highest level of reducing sugar with 1,518.16 mg% among all samples, and the content of the reducing sugar was decreased after blanching in all samples. Raw and blanched $Haetsun$ vegetables contained all essential amino acids except tryptophan. The vitamin C content of blanched samples was reduced 29~88% compared with raw samples. All $Haetsun$ vegetables contained high levels of potassium and calcium regardless of blanching. From these results, even though this study confirmed that $Haetsun$ vegetables were rich in vitamin C and minerals, their high amount of nutrient was reduced by blanching. Therefore, blanching conditions which can keep food value of $Haetsun$ vegetables must be established, because most of the nutrient composition of $Haetsun$ vegetables decreases by blanching.
Encapsulated Aspergillus niger was prepared in order to inspect the effect of oxygen supply on the production of citric acid. A. niger cells which had been immobilized in the calcium alginate capsule grew and mycellia penetrated through the capsule membrane after two days of cultivation and covered over all of the capsule after eight days. The mycellia became loose when the nitrogen source was sufficient of oxygen was deficient. The larger amount of encapsulated cells were put into a given growth medium, the smaller quantity of citric acid was produced. The increase of volumetric oxygen transfer coefficient from 1.8 $hr^-$ to 2.55 $hr^-$ in the flask culture accelerated cell growth rate but did not influence the production of citric acid. The high oxygen supply rate($k_La:\;150\;hr^-$) in the concentric air lift reactor hastened the growth of cells and hindered the production of the citric acid. The reduction of nitrogen source level in the growth medium in the concentric air lift reactor increased citric acid production by 40 percent of that of flask cultivation and the culture period was shortened by 3 days. The variation of the geometry of the concentric air lift reactor did not influence the growth rate of encapsulated cells and production rate of citric acid.
This study was conducted to investigate the amount of fat, vitamins and minerals in milk can be affected by hydrodynamic cavitation since cavitation is recently focused on as one of the sterilization methods. Vitamins (vitamin A, $B_2$), minerals (Ca, P, Mg, Zn) and fat contents were measured according to Ministry of Food and Drug Safety's Processing Standards and Component Specifications of Animal Product. Vitamin A & $B_2$ contents decreased approximately 23% & 19%, respectively after cavitation. Minerals content showed no change after cavitation. Also, P and Mg contents were reduced after cavitation to 2 mg/100g and 0.1 mg/100g, showing no change. Milk fat content was 3.46% before cavitation and 3.41% after cavitation, displaying no difference. Hydrodynamic cavitation process displays a possibility to replace existing pasteurizing method, as it does not change vitamin or mineral contents in milk. Therefore, a more clear and systematic research on hydrodynamic cavitation pasteurization is needed to distribute excellent quality milk and to emphasize physiochemical properties and quality change of existing heat pasteurization process.
The purpose of this study was to evaluate a new biodegradable membrane - atelocollagen as a guided tissue regeneration barrier on the dehiscence defects adjacent to the dental implants. 3 beagle dogs were selected for this study and all the mandibular premolars($P_1,P_2,P_3&P_4$) were extracted. Twelve weeks after the extraction, the edentulous ridges were formed to be placed the titanium plasma-sprayed IMZ implants. Four implant osteotomies were performed on each side of the mandible. The osteotomies were placed facially in the edentulous ridges to approximate an actual dehiscence defect as closely as possible, The standardized dehiscence defects were created 3 mm in width and 4 mm in height by osteotomy. A total 24 implants were placed. e-PTFE, ateloco11agen and $Collatape^{(R)}$ were placed to cover the defects and the one defect served as a control, not covered any membrane. By random selection, three dogs were sacrificed at 2 weeks, 4weeks and 8 weeks after fixation with 3% glutaraldehyde. A week before sacrificing, 8-week dog was infused intravenously with oxy-tetracycline 30mg/kg. The left mandibular blocks were used for full decalcified histologic preparation and the right mandibular blocks were selected for undeca1cified preparation, At 2 weeks, the regenerated bone of e-PTFE and atelocollagen groups appeared to be more dense than other groups and the percentage of bone defect fill was highest for e-PTFE and follwed by ateloco1lagen group. However, the $Collatape^{(R)}$ and control groups showed a little new bone formation. $Collatape^{(R)}$ was almost degraded within 2 weeks. At 4 weeks, the regenerated new bone were much greater and denser than at 2 weeks for e-PTFE and ateloco11agen group. Although a part of atelocollagen bagan to be degraded at the margin and surrounded by foreign body giant cells related to foreign body reaction, it was generally intact and the regenerated new bone was shown much more than at 2 weeks. The amount of new bone in $Collatape^{(R)}$ and control groups at 4 weeks were similar to that of 2 weeks group. At 8 weeks, the regenerated bone was matured and observed along the implant fixture. Direct new bone formation and calcium deposits beneath the e-PTFE were observed. No further bone growth was seen in the $Collatape^{(R)}$ and control groups. In reflected fluoromicrcocopic observation, the osteogenic activity was pronounced between e-PTFE membrane and the old bone. High osteogenic activity was also observed in atelocol1agen group. This study suggested that the ateloco11agen as well as e-PTFE could be used for guided tissue regeneration on dehiscence defects adjacent to the dental implants. But the $Collatape^{(R)}$ was completely resorbed within 2 weeks and was not a suitable membrane for guided bone regeneration.
In analysis of inorganic components of Miscanthus sinensis Andersson var. sinensist, Phosphate increased with the progress of growing stage. in other way potassium, calcium and magnesium was inclined to decrease. The total nitrogen content in the Miscanthus sinensis Andersson var. sinensist was increased until second age cultivated stage, but in its third year, it was decreased apparently. In other way the total carbon content of Miscanthus sinensis was not changed until second year cultivation. But rapid increasing was happen in its third year cultivation stage. 3 year cultivated Miscanthus sinensis Andersson var. sinensist which has a plentiful carbon source can be used as a good culture media source for the formation of mushroom's fruit body. When rice bran added at the amount of 20% to the keep in 65% moisture Miscanthus sinensis Andersson var. sinensist, the fermentation of culture material was well done and the temperature of $50{\sim}55^{\circ}C$ fittest for thermophilic microorganism growth was maintained for 5 days from 5 days after treatment. Rice bran 20% added to the Miscanthus sinensis Andersson var. sinensist was the fittest fermentation culture media for the Pleurotus ostreatus vinyl-bag type cultivation. When Pleurotus ostreatus was cultivated in this culture media, the yield was increased by 60% than whole Miscanthus sinensis Andersson var. sinensist containing culture media. The fermentation Miscanthus sinensis Andersson var. sinensist culture media could be substituted by 20% the sawdust in existing culture media(sawdust 50%+cottonseed bark 30%+cottonseed meal 20%).
