A Study on the Content of Heavy Metals of Marine Fish in Korean Coastal water (우리나라 일부연안 해산 어류 중의 중금속 함량에 관한 연구)

  • This study was carried out to find out the content of injurious heavy metals, such as mercury, lead, cadmium and arsenic of marine fish which had been captured in adjacent sea of Korea. From March, 1 to April, 30 in 1993. 60 sample of fishes were collected the adult fish and young fish. These samples were analyzed by the Mercury Analyzer and Inductively Coupled Plasma Emission Spectro Analyzer. The results of the study were summarized as follows: (1) The content of mercury in Therafra chalcograma was 0.112$\pm$0.034 ppm which was higher than other fishes, but Pseudosciaena manchurica and Pampus argenteus had lower (P<0.05). Although lead content of Trichiurus lepturus was 0.359$\pm$0.056 ppm, which was higher than other fishes. The lead content of Theragra chalcograma had lower to the almost same level. The cadmium content of the Theragra chalcograma was 0.069$\pm$0.010 ppm which was higher than other samples, but Pseudosciaena manchurica had 0.039$\pm$0.020 ppm to lower level. Though there was some fluctuation in the arsenic content which was 0.433~3.752 ppm, the arsenic content of Therafra chalcograma was 3.752$\pm$2.873 ppm which was the higher than any other fishes. But there are not statistical significances. (2) Heavy metal content by the maturity of the fishes: Mercury content of the old Pseudosciaena manchurica was 0.055$\pm$0.15 ppm comparing to the young's result of 0.030$\pm$0.009 ppm (P<0.05). Though there were some differences according to the maturity, but thee was no statistical significance. (3) In view of the correlation of the heavy metal content, for Pseudosciaena manchurica, high correlation was founded to r=0.6437 between mercury and cadmium (P<0.05). Though the content of mercury, arsenic cadmium and lead had positive correlation (r=0.2725) and negative correlation (r=-0.3958), but there was no significance at all. The other fishes were not found correlation between the heavy metal content. Positive correlation was found between age with mercury in Pseudociaena manchurica (r=0.7018, P<0.05). The negative correlation of age with lead content in the Pseudociaena Manchurica was signigicant (P<0.05). The correlation coefficient was r=-0.7623. The age with mercury content in Coloabis saira had high correlation (r=0.7201, P<0.05). Through the above analysis, it can get conclusion that injurious heavy metal content of the fishes in Korea such a mercury, lead, cadmium and arsenic were at the level of natural content. At present, guidelines of maximum residue level allowed at foreign countries about the injurious heavy metal have been used according to the kinds of fish but our government guideline for the marine fishes in only total mercury below 0.7 ppm and lead below 2 ppm. So more aggressive guidelines for the allowance level of heavy metals in marine fishes are required for the safety of foods.

Effects of dietary Cadmium and Protein Levels on the Body Protein Metabolism and Cadmium Toxicity in Growing Rats (식이내 Cadmium과 단백질 수준이 흰쥐의 체내 단백질 대사 및 Cadmium 중독에 미치는 영향)

  • This study were performed to investigate effect of dietary cadmium(Cd) and protein levels on growth, body protein metabolism and Cd toxicity in growing rats. Forty eight male rats of Sprague-Dawley weighing 113$\pm$2g were blocked into 6 groups accoridng to body weight. Dietary protein were given at the levels of 7, 15 and 40% of diet and Cd (200ppm)were either added or not. The result obtained were summerized as follow; 1) Food intake, weight gain, FER PER, liver and kidney weight, weight and length of bones, hematocrit, and hemoglobin content in Cd-added groups were low than those in Cd-free groups. 2) Serum total protein showed no significant difference with Cd addition, but it was significantly lower in low protein diet groups. Liver protein in Cd-added groups was lower than Cd-free groups, and was tend to be increased with increasing dietary protein level. 3) Daily urinary and fecal nitrogen excretions in Cd-added groups were lower than Cd-free groups, and were increased with increasing dietary protein level. 4) Cadmium contents in blood, liver, kidney, and femur were tend to be decreased with increasing dietary protein level. Especially, Cd content in kidney of Cd-added groups was significantly decreased with increasing dietary protein level. 5) Daily urinary and fecal Cd excretions were tend to be increased with increasing dietary protein level, and Cd-added-high protein diet group showed the highest Cd excretion among the Cd-added groups, Cd absorption ration and Cd retention ratio were tend to be decreased with increasing dietary protein level.

Cytoprotective Effect of Galgeuntang Extract on Cadmium-Induced Cytotoxicity (갈근탕 추출물의 카드뮴 독성에 대한 세포 보호효과)

  • Cytotoxicity of cadmium on NIH 3T3 fibroblasts was utilized in order to discover antitoxic compound in Galgeuntang extract in this study. Treatment groups were chosen as follows; control (medium only), $MTT_{50}$ group and five experimental groups. MTT {3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazoliumbromide} method was performed to evaluate the cytotoxicity of cell organelles and $IC_{50}$ was also measured. Accordingly we have examined the detoxification effects of Galgeuntang extract on cadmium-treated NIH 3T3 fibroblasts to observe morphological changes by the light microscopy. Galgeuntang extract showed cytoprotective effects on cadmium-induced cytotoxicity. Furthermore, Galgeuntang showed a dose-dependency in detoxication. The phenolic content of Galgeuntang ethanol extract was higher than that of water content. These results suggest that Galgeuntang extract may be used as a cytoprotective agent against cadmium (II)-mediated cytotoxicity.

Effect of the Aloe arborescens Added-Diet on the Cadmium Toxicity in Rat (알로에 첨가식이가 흰쥐의 카드뮴 독성에 미치는 영향)

  • This study was performed to investigate the effect of Aloe arborescens on the cadmium toxicity in rats. Thirty male Sprague-Dawley strains were divided into five groups consisting of a control group, a cadmium treatment group and 3 aloe(0.5%, 0.75%, 1%) treatment groups and observed for 9 weeks. The weight increment of the cadmium and 0.75% aloe group was higher than that of the cadmium treatment group(p<0.01). The food intake did not show the consistency rule among the experimental groups and the decrement tendency of food intake affected by cadmium feeding group. The decrement tendency of water intake affected by cadmium appeared to be suppressed by aloe treatment, especially cadmium and 0.75% aloe treatment group showed the remarkable increment of water intake. The diet efficiency of the control group was the highest among the experimental groups and that of cadmium and 0.75% aloe group was higher than other aloe treatment groups. The weight of each organ did not show consistency among the experimental groups but only the testicle of cadmium and 0.75% aloe treatment group was heavier than that of the control group. The cadmium accumulation was high in order of kidney>liver>spleen>heart>lung>testicle>brain. The cadmium content of the cadmium treatment group was more than that of cadmium and 0.5% aloe group, cadmium and 0.75% aloe group, cadmium and 1% aloe group. The cadmium content of cadmium and 0.75% aloe group was the lowest among other aloe treatment groups. Therefore, cadmium and 0.75% aloe is the most recommendable aloe treatment to eliminate the cadmium accumulated in organ.

RENAL EXCRETION OF $Na^+$ AND $K^+$ IN CADMIUM-INTOXICATED RATS

  • Changes in urinary $Na^+$ and $K^+$ excretions, renal cortical microsomal $Na^+$ -K-ATPase activity, cortical tissue electrolyte content and plasma aldosterone level were studied in rats treated with CdCl2 (2 mg Cd/kg/day, s.c. injection) for 7-14 days. After 7 days of cadmium exposure, urinary excretion of $Na^+$ was markedly reduced. This change was accompanied by an increase in $Na^+$-$K^+$-ATPase activity, a fall in tissue $Na^+$ content, a rise in tissue $K^+$ content and an elevation of plasma aldosterone level.

The Study on the Concentrations of Lead and Cadmium in of the Inhabitants of Jeon-Ju Indurstrial Area (전주공업단지 주변 주민들의 모발내 Pb 및 Cd함량에 관한 연구)

  • This study was carried out to evaluate the concentrations of heavy metal in hair of the inhabitants of Jeon-Ju industrial area. The major findings of this study are as follows 1. The average content of lead in hair of the inhabitants of Jeon-Ju industrial area was 15.06$\pm$ 4.06 ppm and the content of lead of driver's hair showed higher than that of house wives and students, and showed significance statistically ( P < 0.05). 2. The average content of cadmium by occupation in hair of inhabitants of Jeon-Ju industrial area was 0.76$\pm$ 0.21 ~ 0.52$\pm$ 0.16 ppm and the cadmium concentration in hair of driver's showed the highest level among occupations and showed significance statistically (P < 0.05). 3. Lead and cadmium contents in the hair of the inhabitants of Jeon-Ju industrial area showed higher than that of control area residents and showed significance statistically ( P < 0.05 ). 4. The contents between lead and codmium in hair of Jeon-Ju industrial area residents revealed high degree of correlation ( r = 0.7938).

Cadmium Adsorption and Exchangeable Cations Desorption in Soils: Effects of pH and Organic Matter Content (토양에서 카드뮴의 흡착과 치환성양이온의 탈착 : pH와 유기물함량의 영향)

  • In order to investigate the effects of pH and organic matter content on cadmium adsorption and exchangeable cations desorption in soils, the adsorption isotherms of cadmium and the desorption isotherms of calcium and magnesium on four New jersey soils at four pH values were plotted, and the cadmium partition coefficients (Kd) were also calculated. The slopes of cadmium adsorption isotherms dramatically increased with increasing solution pH. Judging from Langmuir adsorption equations, the maximum adsorption quantities(b) of cadmium at high pH values were much greater than those at low pH values for the same soil. The partition coefficients increased greatly with increasing solution pH. The slopes of regression equations between partition coefficients and pH values were steep in the order of the organic matter content of the soils. The correlation coefficients (r2) between partition coefficient and organic matter content for soils. The correlation coefficients (r2) between partition coefficient and organic matter content for $1\times10^{-4}$M increased from 0.3027 at pH 4.0 to 0.9964 at pH 8.5 and from 0.2093 at pH4.0 at 0.9657 at pH 8.5 for$2\times10^{-4}$M ${Cd(NO_3)}_2$. The desorption quantities of calcium and magnesium decreased with increasing solution pH and increased with- increasing cadmium adsorption.

Removal effect of Cadmium by Polyphenol Compound Extracted from Persimmon Leaves(Diospyros kaki folium) (감잎(Diospyros kaki folium)으로부터 분리한 Polyphenol 화합물의 카드뮴 제거효과)

  • In order to investigate the removal effect of cadmium by polyphenol compound extracted from persimmon leaves(Diospyros kaki folium), animal test was done. Two fractions such as F-1 and F-2 from persimmon leaves were compared with their safety and function. The food intake of group F-1 and F-2 considerably decreased within 1% level. Cadmium addition influenced to rat growth(a tested animal), but food efficiency ratio(FER) wasn't shown any considerable difference in F-1 and F-2, as compared to the control. Cadmium content of liver, kidney and femur considerably decreased within 1% level in F-1 and F-2, compared to the control, cadmium content of liver decreased 25% in F-1, 28% in F-2 3150 decreased 22% md 25% in kidney. In femur, also decreased 53% in F-1 and 59% in F-2 respectively. The test of cadmium content in feces indicate that the content considerably increased within 1% level in both group F-1 and F-2, as compared to the control(42% in F-1, 54% in F-2). As shown the above results, have seen the removal effect of cadmium by polyphenol compound.

Effect of Vitamin E on Cadmium Accumulation and Excretion in Chronic Cadmium Poisoned Rats (만성 카드뮴 중독 흰쥐에서 카드뮴 축적과 배설에 미치는 Vitamin E의 영향)

  • The purpose of this study was to investigate the effect of vitamin E on the cadmium accumulation in body, cadmium excretion and detoxification functions in chronic cadmium poisoned rats. Sprague-Dawley male rats weighing 100$\pm$10 g were randomly assigned to one normal group and three cadmium poisoned groups. Cadmium poisoned groups were classified to vitamin E free diet (Cd-0E group), vitamin E 40 mg/kg diet (Cd-400E group) and 400 mg/kg diet (Cd-400E group) according to the levels of vitamin E supplement. Animals were maintained on 0, 40 mg and 400 mg vitamin E/kg diets for 20 weeks and simultaneously administered 50 ppm Cd$^{2+}$ dissolved in the drinking water. Body weight, food intakes and food efficiency ratio were significantly decreased in all cadmium groups, compared with those of normal group. The accumulation of cadmium in rat liver, kidney and blood was reduced by sufficient vitamin E supplementation. The metallothionein (MT) content in liver and kidney were increased in all cadmium groups compared with that of normal group. The ratio of cadmium absorption and retention were significantly decreased in vitamin E supplementation groups. Accordingly, vitamin E supplementation resulted in an excretion of cadmium in urine and feces and a lowered accumulation of cadmium in liver and kidney. It can be suggested that increased MT synthesis lead to the significant decrease in cadmium absorption and retention ratios.s.

Effect of Chitosan and N, O-Carboxymethyl Chitosan of Different Sources and Molecular Weights on Cadmium Toxicity (급원과 분자량이 다른 Chitosan 과 N, O-Carboxymethyl Chitosan이 Cadmium 중독에 미치는 영향)

  • This study was performed to investigate the effect of chitosan and NOCC from different sources and of different molecular weights on cadmium toxicity. Sprague-Dawley rats were blocked into 26 groups according to body weight, and were raised for 4 weeks. Cadmium chloride was given at the level of 0 or 400 ppm in diet. Various forms of chitosan and NOCC were given at the level of 0 or 4%(w/w) of diet. Cd toxicity was alleviated by various chitosan and NOCC supplements. However, the alleviating effects were different with fiber source(crab and shrimp), type(chitosan and NOCC), and molecular weight (low, medium, and high). Molecular weight had no significant effect. Compared with shrimp-source-fiber-fed groups, crab-source-fiber-fed groups showed lower Cd concentrations in blood and kidney, lower kidney metallothionein concentration, and lower liver and kidney MT-Cd contents. Compared with NOCC -fed groups, chitosan-fed groups showed lower intestine and liver Cd concentrations, lower liver and kidney MT concentrations and MT-Cd content, and higher fecal Cd excretion. Among cadmium-exposed group, low molecular weight shrimp chitosan group showed low Cd concentrations of liver and kidney, high fecal Cd excretions, and the lowest intestine and liver MT concentrations and liver MT-Cd content. In summary, a crab source fiber was more effective than shrimp source fiber, chitosan was more effective than NOCC, and the most effective one in alleviating Cd toxicity was low molecular weight shrimp chitosan.
