Quercetin Relaxed the Smooth Muscle of Rabbit Penile Corpus Cavernosum by Activating the NO-cGMP Signaling Pathway

  • The aim of this study was to investigate the effect and action mechanism of quercetin on penile corpus cavernosum smooth muscle (PCCSM). PCCSM precontracted with phenylephrine (Phe) was treated with four different concentrations of quercetin ($10^{-7}$, $10^{-6}$, $10^{-5}$ and $10^{-4}M$). PCCSM were preincubated with N-Nitro-L-arginine methyl ester hydrochloride (L-NAME) and 1H-[1,2,4]oxadiazolo[4,3-a]quinoxalin-1-one (ODQ) to block nitric oxide synthase and guanylate cyclase, respectively. The changes in PCCSM tension were recorded, and cyclic nucleotides in the perfusate were measured by radioimmunoassay. The interactions of quercetin with phosphodiesterase type 5 inhibitors (PDE5-Is) such as sildenafil, udenafil and mirodenafil, were also evaluated. PCCSM relaxation induced by quercetin occurred in a concentrationdependent manner. The application of quercetin to PCCSM pre-treated with L-NAME and ODQ significantly inhibited the relaxation. Quercetin significantly increased cGMP in the perfusate. Furthermore, quercetin enhanced PDE5-Is-induced relaxation of PCCSM. Quercetin relaxed the PCCSM by activating the NO-cGMP signaling pathway, and it may be a therapeutic candidate or an alternative treatment for patients with erectile dysfunction who do not completely respond to PDE5-Is.

식물추출복합발효물(MP119)이 성기능에 미치는 영향 및 카드뮴 독성에 대한 효과 (Effect of Phyto-Extract Fermented Mixture (MP119) on the Sexual Functions and on the Toxicities of Cadmium)

  • 마카, 홍삼, 남가새 추출물을 일정한 무게 비율로 혼합하여 발효시켜 얻어진 식물추출복합발효물(MP119)이 성기능에 미치는 영향을 조사하였다. ACE(angiotensin converting enzyme)과 PDE(phosphodiesterase)에 대한 저해효과를 통해 혈액순환부진 개선 및 cGMP 농도에 의한 발기능 개선에 미치는 효과를 측정하였다. MP119의 ACE 저해효과를 측정한 결과 $IC_{50}$값이 241.3${\pm}$35.5 ppm, PDE 저해효과를 측정한 결과 $IC_{50}$값이 372.2${\pm}$33.8 ppm으로 나타나 혈관확장으로 인한 혈류량 증가 및 cGMP농도 증가에 영향을 줄 수 있을 것으로 예상된다. MP119는 마우스 정소세포인 TM3 cell에 유의성 있는 독성이 나타나지 않았으며, TM3 cell에 농도별로 처리했을 때 세포배양액으로부터 대조군과 비교하여 최대 20.11% 증가한 testosterone 농도를 확인하였다. 또한 HUVEC에 농도별로 처리했을 때 NO생산량이 MP119 처리 농도가 증가할수록 유의성 있게 증가하였다. 웅성마우스에 MP119를 7일간 경구투여 후 생식장기의 무게와 정자수를 측정하고, MP119를 경구투여한 후 염화카드뮴을 투여하여 생식장기의 무게와 정자수를 측정하였다. 또한 실험쥐로부터 혈청을 분리하여 혈청 내 testosterone과 cGMP를 측정하였다. 그 결과 MP119의 투여를 통해 생식장기의 무게 변화 없이 정자수 증가에 도움을 준다는 것을 확인하였으며, 더불어 혈청 내 testosterone 및 cGMP의 농도 역시 증가하였다. MP119를 투여한 후 염화카드뮴을 투여한 실험군에서 역시 정자수 및 혈청 내 testosterone, cGMP 농도 증가를 나타내었으므로 결과적으로 MP119가 염화카드뮴의 독성예방 및 치료에 효과적임을 확인하였다.

A Possible Significance in Vertebrate Phototransduction of Multi-Protein Signaling Complexes on Raft-Like Membranes

  • Raft is a distinctive membrane domain enriched in a certain class of lipids, cholesterol, and proteins observed on the plasma membrane. Growing evidence has revealed that such membrane domains play key roles in signal transduction, fertilization, development, transmitter release, and so on. Recently, we have isolated raft-like detergent-resistant membrane (DRM) fraction from bovine photoreceptor rod outer segments. Transducin and its effecter, cGMP-phosphodiesterase, elicited stimulus-dependent translocation between detergent-soluble membrane and DRM. This suggested potential importance of such distinct membrane domains in vertebrate phototransduction. Here, we will discuss physiological meaning of the translocation of major components of cGMP cascade to raft-like membrane in phototransduction. We would like to propose a hypothesis that raft-like membrane domains on the disk membrane are the place where cGMP cascade system could be quenched.

흰쥐에서 혈관내피 의존적인 혈관이완과 혈압하강에 대한 propofol의 억제 효과 (Inhibitory effect of propofol on endothelium-dependent relaxation and blood pressure lowering in rats)

  • We studied the effect of propofol (PPF) on the endothelium-dependent vascular responses in isolated rat thoracic aorta. In aortic rings with endothelium, PPF inhibited the phenylephrine (PE)-induced contraction in a concentration-dependent manner. In PE-precontracted preparations, PPF attenuated the endothelium-dependent relaxation by acetylcholine but not by A23187. And PPF did not attenuate the endothelium-independent relaxation by sodium nitroprusside (SNP). The relaxation induced by acetylcholine in PE-precontracted aortic rings was significantly augmented by zaprinast, a cGMP-specific phosphodiesterase inhibitor, and this augmentation was inhibited by PPF. Although SNP-induced relaxation was significantly augmented by zaprinast, this augmentation was not inhibited by PPF. In preparations preconstricted with PE, the PPF-induced relaxation was inhibited by atropine. In addition, PPF attenuated the vasorelaxation by phosphodiesterase inhibitors (IBMX, Ro20-1724 or zaprinast except milrinone). In vivo, the infusion of acetylcholine and SNP showed decreased arterial blood pressure in rats. The pre-injection of PPF inhibited the acetylcholine-induced blood pressure lowering, but not the SNP-induced blood pressure lowering. These results suggest that PPF can attenuate in part the acetylcholine-induced vasorelaxation and blood pressure lowering through the inhibition of the acetylcholine receptor-mediated endothelium-derived relaxing factor by acting on endothelium. It is considered that the inhibitory effect of PPF on the vasorelaxation is due to the decreased level of cGMP which can be attributed to the inhibition of the muscarinic receptor and/or receptor-G-protein interaction.

고핵산 함유 Saccharomyces cerevisiae 균주를 이용한 정미성 효모 추출물의 제조 (Preparation of Flavor-enhancing Yeast Extract Using a Saccharomyces cerevisiae Strain with High RNA Content)

  • 핵산 함량이 높은 변이주 Saccharomyces cerevisiae B24를 산업적으로 활용하기 위해 배양 후 자기소화 혹은 효소 분해법으로 효모 추출물을 제조하였다. 배양액의 균체 농도를 10% (w/w)로 하고 pH 5.0, $50^{\circ}C$에서 서서히 교반하면서 48시간 자기소화시켰을 때 세포 내용물이 효모 추출물로 이전되는 수율은 65% 정도였으나 리보핵산은 정미력이 없는 다른 성분으로 분해되어 정미성 핵산성분인 5'-IMP나 5'-GMP가 거의 검출되지 않았다. 반면 $90^{\circ}C$ 이상의 온도로 B24 배양액 1 L를 열처리하여 핵산 분해 효소를 불활성화시키고 세포벽을 변성시킨 다음 ${\beta}-1,3-glucanase$, phosphodiesterase, adenylic deaminase, protease 등을 순차적으로 작용시켜 정미성 5'-IMP와 5'-GMP가 총 3.2% 함유된 효모 추출물(고형분 함량 70%) 84 g을 얻을 수 있었고 이 때 추출물 수득율은 고형분 기준으로 85% 이었다. 반면 모균주인 S. cerevisiae ATCC 7754를 효소적으로 분해할 때 정미성 5'-IMP와 5'-GMP가 총 2.2% 함유된 효모 추출물(고형분 함량 70%)을 77 g밖에 얻지 못하였다.

Antinociceptive Effect of the Intrathecal Phosphodiesterase Inhibitor, Zaprinast, in a Rat Formalin Test

  • Background: Cyclic guanosine monophosphate (cGMP) and opioid receptors are involved in the modulation of nociception. Although the opioid receptors agonists are active in pain, the effect of an phospodiesterase inhibitor (zaprinast) for increasing the level of cGMP has not been thoroughly investigated at the spinal level. This study examined the effects of intrathecal zaprinast and morphine in a nociceptive test and we also examined the nature of the pharmacological interaction after the coadministration of zaprinast with morphine. The role of the nitric oxide (NO)-cGMP-potassium channel pathway on the effect of zaprinast was further clarified. Methods: Catheters were inserted into the intrathecal space of male SD rats. For the induction of pain, $50{\mu}l$ of 5% formalin solution was applied to the hindpaw. Isobolographic analysis was used for the evaluation of the drug interaction between zaprinast and morphine. Furthermore, NO synthase inhibitor ($_L-NMMA$), guanylyl cyclase inhibitor (ODQ) or a potassium channel blocker (glibenclamide) were intrathecally administered to verify the involvement of the NO-cGMP- potassium channel pathway on the antinociception effect of zaprinast. Results: Both zaprinast and morphine produced an antinociceptive effect during phase 1 and phase 2 in the formalin test. Isobolographic analysis revealed a synergistic interaction after the intrathecal administration of the zaprinast-morphine mixture in both phases. Intrathecal $_L-NMMA$, ODQ and glibenclamide did not reverse the antinociception of zaprinast in either phase. Conclusions: These results suggest that zaprinast, morphine and the mixture of the two drugs are effective against acute pain and they facilitated pain state at the spinal level. Thus, the spinal combination of zaprinast with morphine may be useful for the management of pain. However, the NO-sensitive cGMP-potassium channel pathway did not contribute to the antinocieptive mechanism of zaprinast in the spinal cord.

쥐를 이용한 포르말린 및 열 유발 통증에서 척수강 Sildenafil의 효과에 관한 연구 (Evaluation for the Effects of Intrathecal Sildenafil on the Formalin- and Thermal-induced Nocieption of Rats)

  • Background: Cyclic guanosine monophosphate (cGMP) plays an important role in the modulation of nociception. Although local sildenafil produces antinociception, by increasing cGMP through the inhibition of phosphodiesterase 5, the effect of spinal sildenafil has not been determined. The authors evaluated the effects of intrathecal sildenafil on the nociceptive behavior evoked by formalin injection and thermal stimulation. Methods: Lumbar intrathecal catheters were implanted into rats, with formalin and Hot-Box tests used as nociceptive models. The formalin-induced nociceptive behavior (flinching response) and withdrawal latency to radiant heat were measured, and the general behaviors also observed. Results: The intrathecal administration of sildenafil produced dose-dependent suppression of the flinches in both phases in the formalin test, and increased the withdrawal latency in the Hot-Box test. No abnormal behaviors were noted. Conclusions: Sildenafil, an inhibitor of phosphodiesterase 5, is active against the nociceptive state evoked in the spinal cord by formalin and thermal stimulations. Accordingly, spinal sildenafil may be useful in the management of pain.

Involvement of Spontaneously Formed Cyclic Nucleotides in Cat Gastric Muscle Relaxation

  • Muscle strips and muscle cells from cat stomach were used to investigate whether spontaneously formed cyclic nucleotides were involved in the inhibition of gastric smooth muscle contraction. A phosphodiesterase inhibitor, 3-isobutyl-1-methylxanthine (IBMX), increased the levels of both cyclic GMP (cGMP) and cyclic AMP (cAMP) in resting state cells, while decreasing acetylcholine-induced muscle contraction. Under the influence of IBMX, SQ22536, an adenylyl cyclase inhibitor and methylene blue, a guanylyl cyclase inhibitor completely blocked increases in cAMP and cGMP respectively, without any effect on contraction. However, the combination of SQ22536 and methylene blue completely blocked increases in both cAMP and cGMP levels and stimulated contractions markedly even in the presence of IBMX. Muscle contraction inhibitors such as isoprenaline, vasoactive intestinal polypeptide and sodium nitroprusside also appeared to increase cyclic nucleotide levels which decreased contraction. Which nucleotide increased the most was dependent on the agonist used. Therefore, irrespective of the cyclic nucleotide class, the spontaneous formation of cyclic nucleotides should be considered in evaluating the mechanism of gastric smooth muscle relaxation.

Electrolysis of Physiological Salt Solution Generates a Factor that Relaxes Vascular Smooth Muscle

  • Oxygen-derived free radicals have been implicated in many important functions in the biological system. Electrical field stimulation (EFS) causes arterial relaxation in animal models. We found that EFS applied to neither muscle nor nerve but to Krebs solution caused a relaxation of rat aorta that had been contracted with phenylephrine. In the present study, therefore, we investigated the characteristics of this EIRF (electrolysis-induced relaxing factor) using rat isolated aorta. Results indicated that EIRF acts irrespective of the presence of endothelium. EIRF shows positive Griess reaction and is diffusible and quite stable. EIRF-induced relaxation was stronger on PE-contracted aorta than on KCl-contracted one, and inhibited by the pretreatment with methylene blue. Zaprinast, a cGMP-specific phosphodiesterase inhibitor, potentiated the EIRF-induced relaxation. $N^G-nitro-L-arginine$, NO synthase inhibitor, did not inhibit the EIRF-induced relaxation. Deferroxamine, but not ascorbic acid, DMSO potentiated the EIRF-induced relaxation. These results indicate that electrolysis of Krebs solution produces a factor that relaxes vascular smooth muscle via cGMP-mediated mechanism.

Ethanol로 발기부전을 유도한 흰쥐의 성기능 개선에 오공(蜈蚣)추출물이 미치는 영향 (Effects of Scolopendra on Ethanol-Induced Erectile Dysfunction in Rats)

  • Scolopendra was known to cure erectile dysfunction. This study was aimed to investigate the effects of Scolopendra on the nitric oxide synthase activity, nitrite level, cyclic-GMP and erectile responses in rat's corpus cavernosum penis. The crushed Scolopendra was extracted 3 times, each time with 3 volumes of methyl alcohol at $60^{\circ}C$ for 24 h. The extract was filtered and evaporated under a reduced pressure using a rotary evaporator to yield 14.2 g. Scolopendra extract was oral-administered 50 mg per 1 kg of body weight for 30 days. First, samples were treated with Scolopendra, and then ethanol-treated rats and L-N-Nitroarginine methyl ester (L-NAME) treated rats were fed with the samples. The level of urethral nitrite and nitric oxide synthase activity in the ethanol-Scolopendra double administered rats were as high as in the normal group, while the one in the ethanol-treated group was decreased. The level of urethral cyclic-GMP and guanylate cyclase actiyity in the ethanol-Scolopendra double administered rats were as high as in the normal group, while the one in the ethanol-treated group was decreased. The level of urethral phosphodiesterase activity in the ethanol-Scolopendra double administered rats was as low as in the normal group, while the one in the ethanol-treated group was increased. The erectile response to a cavernous nerve stimulation in L-NAME $(10^{-4})-treated$ rats was restored by the Scolopendra to the similar level seen in the normal group. The electile response to cavernous nerve stimulation in the ethanol-Scolopendra double administered rats were increased as high as in the normal group while the one in the ethanol-treated group was decreased. Scolopendra was effective in restoring the ethanol-induced or L-NAME-induced erectile dysfunction in rats.
