Characterization of human cardiac mesenchymal stromal cells and their extracellular vesicles comparing with human bone marrow derived mesenchymal stem cells

  • Cardiac regeneration with adult stem-cell (ASC) therapy is a promising field to address advanced cardiovascular diseases. In addition, extracellular vesicles (EVs) from ASCs have been implicated in acting as paracrine factors to improve cardiac functions in ASC therapy. In our work, we isolated human cardiac mesenchymal stromal cells (h-CMSCs) by means of three-dimensional organ culture (3D culture) during ex vivo expansion of cardiac tissue, to compare the functional efficacy with human bone-marrow derived mesenchymal stem cells (h-BM-MSCs), one of the actively studied ASCs. We characterized the h-CMSCs as CD90low, c-kitnegative, CD105positive phenotype and these cells express NANOG, SOX2, and GATA4. To identify the more effective type of EVs for angiogenesis among the different sources of ASCs, we isolated EVs which were derived from CMSCs with either normoxic or hypoxic condition and BM-MSCs. Our in vitro tube-formation results demonstrated that the angiogenic effects of EVs from hypoxia-treated CMSCs (CMSC-Hpx EVs) were greater than the well-known effects of EVs from BM-MSCs (BM-MSC EVs), and these were even comparable to human vascular endothelial growth factor (hVEGF), a potent angiogenic factor. Therefore, we present here that CD90lowc-kitnegativeCD105positive CMSCs under hypoxic conditions secrete functionally superior EVs for in vitro angiogenesis. Our findings will allow more insights on understanding myocardial repair.

Methylene Blue-stained Interstitial Cells are Electrically Active in the Myenteric Board Freshly Prepared from the Murine Small Intestine

  • Many gastrointestinal muscles show electrical oscillation, so-called 'slow wave', originated from interstitial cells of Cajal (ICCs). Thus, a technique to freshly isolate the cells is indispensable to explore the electrophysiological properties of the ICCs. To apply an enzyme solution on the serosal surface for cell isolation, the intestine was inverted and 0.02% trypsin solution and 0.04% collagenase solution were applied to serosal cavity. After the enzyme treatment, mucosal layer was removed and longitudinal muscle layer was gently separated from the rest of tissue. The thin layer was stretched in the recording chamber and mounted on an inverted microscope. Using ${\beta}-escine$, perforated whole cell patch clamp technique was used. Under a microscope, the tissue showed smooth muscle cells and interstitial cells around the myenteric plexus. Under voltage clamp condition, three types of membrane potential were recorded. One group of interstitial cells, which were positive to methylene blue and CD34, showed spontaneous outward current. These cells had bipolar shape and were considered as fibroblast-like cells because of their peculiar shape and arrangement. Another group, positive to c-kit and methylene blue, showed spontaneous inward current. These cells had more rounded shape and processes and were considered as ICCs. The third, positive to c-kit and had granules containing methylene blue, showed quiet membrane potentials under the voltage-clamp mode. These cells appeared to be resident macrophages. Therefore, in the freshly isolated thin tissue preparation, methylene blue could easily identify three types of cells rather than morphological properties. Using this method, we were able to study electrical properties of fibroblast and residential macrophage as well as myenteric ICCs.

Enhancing effect of Canavalia gladiata DC semen on the hematopoietic expansion and function of stem cells (도두(刀豆)의 조혈증진 작용에 관한 연구)

  • Objective : Canavalia gladiata DC semen (CGS) have been used to improve hematopoietic activity. In the current study, we investigated whether CGS regulate hemato-potentiating function using hematopoietic stem cells (HSCs) as a testing system. Methods : HSCs isolated from femur in mice with leukopenia and thrombocytopenia induced induced by CTX. Then, Real-time PCR was performed to measure the mRNA expression and hematopoietic related gene (EPO, IL-3, SCF, c-kit, GM-CSF), the phoaphorylation of GATA-1 and STAT-5a/b were observed by ELISA method, and the number of granulocyte erythrocyte monocyte macrophage colony-forming units (CFU-GEMM) and erythroid burst forming units (BFU-E), semisolid clonogenic assay was performed. Result : When HSCs were treated with CGS, the expression of hematopoietic related genes (EPO, IL-3, SCF, c-kit, and GM-CSF) were significantly increased at the levels of mRNA as well as production in HSCs. Additionally, CGS enhanced phosphorylation of STAT-1 and signal transducer and activator of transcription-5a/b (STAT-5a/b) in HSCs. Furthermore, CGS significantly enhanced the growth rate of granulocyte erythrocyte monocyte macrophage colony-forming units (CFU-GEMM) and erythroid burst forming units (BFU-E) in vitro. Conclusion : These result suggest that CGS has hematopoietic enhancement via hematopoietic cytokine-mediated GATA-1/STAT-5a/b pathway.

An AHP/DEA Hybrid Model for Efficiency Evaluation of Container Terminal (컨테이너터미널 효율성 평가를 위한 AHP/DEA 통합모형)

  • In this study, we compared the efficiency of container terminals using DEA. To do this, we designed an AHP/DEA hybrid model using AHP and DEA, and evaluated the efficiency by comparing the container terminal operation company in Gwangyang(KEC, KIT, GICT) and Busan(HBCT, DPCT, KBCT, UPT, Gamman, PNC, PNIT, HJNC, HPNT). The proposed model can control the number of selected promising container terminal by applying DEA-AR model. This model can also improve the credibility of analysis by using objective weights through the AHP application to efficiency evaluation data and normalizing the evaluation data to apply AHP and DEA. The model assumes inputs to be container crane, transfer crane, yard tractor, and reach stacker and output as container traffic. The result shows that DPCT was an efficient DMU.

  • Genonlic DNA from Metagonimn vokogawci and Metagonimw Miyata type was amplified by polymerase chain reaction based on the random amplification of polymorphic DNA (RAPDI technique. Eight random 10-mer oligonucleotide primers (OPA-02, 5-TGCCGAGCTG-3; OPA-09, 5-GGGTAACGCC-3; OPA-17, 5-GTGATCGCAG-3; OPA-11, 5-CAATCGCCGT-3; OPA-13, 5-CAGCACCCAC-3; OPA-17. 5-GACCGCTrGT-3; OPA-19, 5-CAAACGTCGG-3; OPA-20, 5-GTTGCGATCC-3) WITH A G+C CONTENT FO 60-70% (Kit A. Operon Technologies Inc., California, USAI could produce distinguishable banding patterns between the two Metngonimus species. From the results of this study, it was suggested that Metcsonimus Miyata type has a different DNA sequence from M. WOkQgGUIGi. Quality Evaluation of Five Commercial Enzyme Linked Immunosorbent Assay Kits for Detecting Aflatoxin B1 in Feedstuffs

  • The objective of this study was to evaluate the quality of five commercial enzyme linked immunosorbent assay (ELISA) kits (A, B, C, D, and E) from different suppliers for detecting aflatoxin $B_1$ ($AFB_1$). $AFB_1$-free corn samples supplemented with different levels of $AFB_1$ (5, 10, and $20{\mu}g/kg$) were used as positive controls and 6 replicates of each control sample were tested to evaluate the accuracy and precision of these kits. In addition, we also evaluated the performance of these ELISA kits for $AFB_1$ in 30 feed samples, including corn, distillers dried grains with soluble, wheat samples, soybean meal, and poultry feed, which were verified by high performance liquid chromatography. Results showed that the coefficients of variation ranged from 1.18% to 16.22% in intra-plate and 2.85% to 18.04% in inter-plate for the determination of $AFB_1$. The half maximal inhibitory concentration for five kits ranged from 3.72 to $7.22{\mu}g/kg$. The quantitation limits of $AFB_1$ were all under the legal limit in China but somewhat inconsistent with kit instructions. Although the recovery rate of four of the five kits were either less than 90% or more than 110%, all these values were acceptable in practice. Two kits had high false positive rates (C and E). In conclusion, our results revealed that the qualities of five tested ELISA kits were significantly different.

Effects of the Acupuncture or Pharmacopunture Treatment for the Histologic and Morphologic Recovery on the Mouse with the Burn Skin (흰쥐의 화상부위에 시술된 침과 소염약침이 조직회복에 미치는 영향)

  • Objectives: Burn is a severe dermal injury caused by heat. We were to investigated the effects of acupuncture or pharmacopunture treatment for the histologic and morphologic recovery on the mouse with the 3rd grade burn skin. Methods : We divided into 3 groups. One was a control group(n=3) that was not treated any treatments. Another was a acupuncture group(n=3) that was treated only acupuncture. The other was a pharmacopuncture group(n=3) that was treated only pharmacopuncture. We made a 3rd grade burned skin with the stainless steel heating apparatus. We made a treatment for the mice for a week(2 times for a day, totally 14 times treatments). We observed a dermal morphologic recovery on the mice and a histopathological photographs of the burn skin and subcutaneous tissue with H&E stain, Masson's trichrome stain, and VEGF, FGF and c-kit immunohistochemical stain. Results : The pharmacopuncture group were a better morphologic recovery than control group and acupuncture group. And the pharmacopuncture group were a better histopathological recovery than control group and acupuncture group on the burn skin and subcutaneous tissue with H&E stain, Masson's trichrome stain. And the pharmacopuncture group were a better histopathological recovery than control group and acupuncture group on the burn skin and subcutaneous tissue with VEGF, FGF and c-kit immunohistochemical stain. Conclusions : We suggest that the pharmacopuncture treatment is a better histologic and morphologic recovery than the no treatment or the acupuncture group with the burned mouse skin.

  • Gastrointestinal stromal tumor is documented on every part of the gastrointestinal tract. It develops in the stomach and the small intestine most commonly, and also in the esophagus. A 44 year-old male patient was admitted due to dysphagia and weight loss. Chest CT showed about $15\times11\times11cm$ sized, well-defined, and lobulated soft tissue mass with central necrosis was noted in the posterior wall of lower esophagus throughout the lesser curvature of upper stomach. We performed the distal esophagectomy and total gastrectomy using thoracoabdominal incision. The tumor was positive at CD117 (c-kit) and CD 34, and was diagnosed as malignant GIST of the distal esophagus and upper stomach. The patient is on routine follow up at the out patient department for nineteen months up to now.

Study on Heat-shield Property of Surface-treated Inorganic Oxide Particles (표면처리된 무기산화물 입자의 열차단 특성에 관한 연구)

  • In this study, we produced heat-shield coating materials using surface-treated Ga-doped ZnO (GZO) and investigated the dispersity of particle, visible light transmittance, ultraviolet light cut off, infrared light cut-off, heat-shielding property by surface-treating compounds and treatment conditions. In the case of using IPA or acryl binder for heat-shield coating, the dispersity of inorganic oxide particles was poor but in the case of using surface-treated inorganic oxide particles by hybrid compound having urethane (urea) group, acryl group and silica, dispersity of particle, visible light transmittance and haze were improved. We used the measurement kit and sunlamp for measuring heat-shielding property and confirmed that the internal temperature of the measurement kit using heat-shield film was lower more than $4.8^{\circ}C$ in comparison with using PET film for itself.
